A Comparison Between The Dhia Test For Somatic Cell Counts Compared To Porta Side Test On The Individual Quarters Of The High Scc Cows At The Cal Poly Dairy, Melissa Zimmer

Dairy Science

The objective of my senior project is to determine if California Mastitis Test (CMT) and the Porta Side Somatic Cell Count (SCC) Test is equivalent to the DHIA laboratory somatic cell test. The data was taken from the Cal Poly Dairy on October 19th, 2010. Samples were taken from eight Holstein cows during milking time and the tests were run in the barn. These eight cows were the highest somatic cell count cows at the Cal Poly Dairy on the previous test day October 9, 2010. During milking the cows were pre and post dipped before the samples were taken. …

A Novel Method For Evaluating The Release Of Fermentable Sugars From Cellulosic Biomass, Rafael Jiménez-Flores, Gina Fake, Jennifer Carroll, Elizabeth Hood, John Howard

Dairy Science

Efficient hydrolysis of cellulosic biomass into free sugars for the production of bioethanol would allow utilization of an abundant and renewable natural resource. However, complex microcrystalline cellulose in biomass resists digestion. A preferred method to deconstruct cellulose is using enzyme cocktails, but the presence of multiple enzymes with synergistic outcomes for this process complicates direct biochemical analysis as a reliable indicator of deconstruction. Methods to detect release of free sugars rather than direct measurement of enzymatic activity are available but are time-consuming, and do not reflect the production of metabolic inhibitors of microbial growth which may interfere with the subsequent …

The Impact Of Antioxidant Addition On Flavor Of Cheddar And Mozzarella Whey And Cheddar Whey Protein Concentrate, I. W. Liaw, H. Eshpari, P. S. Tong, M. A. Drake

Dairy Science

Lipid oxidation products are primary contributors to whey ingredient off-flavors. The objectives of this study were to evaluate the impact of antioxidant addition in prevention of flavor deterioration of fluid whey and spray-dried whey protein. Cheddar and Mozzarella cheeses were manufactured in triplicate. Fresh whey was collected, pasteurized, and defatted by centrifugal separation. Subsequently, 0.05% (w/w) ascorbic acid or 0.5% (w/w) whey protein hydrolysate (WPH) were added to the pasteurized whey. A control with no antioxidant addition was also evaluated. Wheys were stored at 3 °C and evaluated after 0, 2, 4, 6, and 8 d. In a subsequent experiment, …

Growth Hormone Alters Lipid Composition And Increases The Abundance Of Casein And Lactalbumin Mrna In The Mac-T Cell Line, Tasha L. Johnson, Brent A. S. Fujimoto, Rafael Jiménez-Flores, Daniel G. Peterson May 2010

Growth Hormone Alters Lipid Composition And Increases The Abundance Of Casein And Lactalbumin Mrna In The Mac-T Cell Line, Tasha L. Johnson, Brent A. S. Fujimoto, Rafael Jiménez-Flores, Daniel G. Peterson

Dairy Science

The MAC-T cell line has been used extensively to investigate bovine mammary epithelial cell function. A lactogenic phenotype is generally induced in this cell line by a combination of dexamethasone, insulin and prolactin and has typically been assessed by milk protein production. Few studies have focused on identifying other factors that may affect milk protein synthesis in the MAC-T cell line, and none have considered the lipid class distribution of MAC-T cells as a component of the lactogenic phenotype. Growth hormone (GH) has been shown to increase milk protein synthesis both in vivo and in mammary cell models, and has …

Lipid Rafts In The Bovine Milk Fat Globule Membrane Revealed By The Lateral Segregation Of Phospholipids And Heterogeneous Distribution Of Glycoproteins, Christelle Lopez, Marie-Noëlle Madec, Rafael Jiménez-Flores

Dairy Science

This study reveals the lateral organisation of the milk fat globule membrane (MFGM). Using confocal laser scanning microscopy (CLSM) and a lipid soluble molecule, an exogenous phospholipid and two lectins as fluorescent probes we located triacylglycerols in the core of fat globules and investigated the organisation of the polar lipids and glycoproteins of the MFGM, in situ in milk. Lipid rafts corresponding to the lateral segregation of sphingolipids in liquid-ordered phases surrounded by liquid-disordered domains composed by the glycerophospholipids were observed in the MFGM. These lipid rafts which correspond to rigid sphingolipid-rich domains have a circular shape at room temperature. …

Investigating The Protective Properties Of Milk Phospholipids Against Ultraviolet Light Exposure In A Skin Equivalent Model, Ashley Russell, Andrea Laubscher, Rafael Jiménez-Flores, Lily H. Laiho

Dairy Science

Current research on bioactive molecules in milk has documented health advantages of bovine milk and its components. Milk Phospholipids, selected for this study, represent molecules with great potential benefit in human health and nutrition. In this study we used confocal reflectance and multiphoton microscopy to monitor changes in skin morphology upon skin exposure to ultraviolet light and evaluate the potential of milk phospholipids in preventing photodamage to skin equivalent models. The results suggest that milk phospholipids act upon skin cells in a protective manner against the effect of ultraviolet (UV) radiation. Similar results were obtained from MTT tissue viability assay …

Algae As An Economical Protein Source For Dairy Cattle Nutrition, Brian Azevedo Dec 2009

Algae As An Economical Protein Source For Dairy Cattle Nutrition, Brian Azevedo

Dairy Science

Dairying continues to change with economic and environmental demands of today’s fast moving society. Feed costs continue to rise and now account for a large majority of expenses on dairies; especially protein, which has become expensive to purchase. I believe that Algae can serve as a possible solution to aid in alleviating this problem. Algae grow rapidly and consume undesirable carbon dioxide to release oxygen to the environment, making it an environmentally friendly source of feed. There are particular strands of algae that are high in protein. During the course of this project, I intend to discover how effective protein …

Usage Of The Penn State Forage Separator For Evaluating Particle Size Of Tmrs, Ashley Rene Garcia Dec 2009

Usage Of The Penn State Forage Separator For Evaluating Particle Size Of Tmrs, Ashley Rene Garcia

Dairy Science

Particle size has many effects on the cow. Adequate forage particle length is necessary for proper rumen function. Dairymen are also having a hard time measuring particle size. Proper particle size distribution of feeds is an important part of ration formulation. Management of forage particle size starts with harvesting forages at the proper stage of maturity. Chopping the crop at the right length helps to achieve the desired particle length in a TMR. Measuring particle length of the forages is only a portion of the solution. Analyzing the TMR particle size is the main goal in measuring the distribution of …

Concentrate Levels And Saccharomyces Cerevisiae Affect Rumen Fluid-Associated Bacteria Numbers In Dairy Heifers, G. J. Lascano, G. I. Zanton, A. J. Heinrichs Dec 2009

Concentrate Levels And Saccharomyces Cerevisiae Affect Rumen Fluid-Associated Bacteria Numbers In Dairy Heifers, G. J. Lascano, G. I. Zanton, A. J. Heinrichs

Dairy Science

Total viable rumenbacteria counts through the use of colony-unit forming assays lack accuracy because they only include culturable bacteria capable of initiating cell division. Thus, bacterial counts can be underestimated. The use of fluorescent characteristics of cell membranes allows flow cytometry to enumerate and distinguish dead from live bacteria cells. The objective of this experiment was to investigate the viable and total ruminal bacteria counts when 3 levels of forage:concentrate in diets were fed at restricted levels with the addition of Saccharomycescerevisiae (YC). Three cannulated post-pubertal Holstein heifers (age 18 ± 1.0 months) were fed corn silage (CS)-based diets in …

Formation Of Cull Cow Program For The Artificial Insemination Class, Brian O. Medeiros

Dairy Science

Cal Poly’s “Learn by Doing” philosophy is one of the great traits that the University holds. However it is this philosophy that is also one of the most expensive and hardest to maintain. “Production agriculture majors use more dollars per unit than any other major in the entire college,” as Dr. Golden stated during his talks about herd cut backs. Never the less the College of Agriculture prides itself on supplying knowledgeable and experienced students to the industry that supports it so much. One of the amazing classes that are offered at the university is the Dairy Science Artificial Insemination …

Rumen Fermentation Pattern Of Dairy Heifers Fed Restricted Amounts Of Low, Medium, And High Concentrate Diets Without And With Yeast Culture, G. J. Lascano, A. J. Heinrichs

Dairy Science

Restricted feeding and highconcentratediets are potential strategies for growing dairyheifers. Ruminal manipulation with additives such as Saccharomyces cerevisiaeyeastculture (YC) has been shown to alter digestibility when added to this type of diet. An experiment was conducted to investigate the ruminal fermentation and in situ digestibility of diets with 3 different levels of forage to concentrate (F:C) fed at restricted intake without and with YC addition. Three cannulated post-pubertal Holstein heifers (age 18.0 ± 1.2 months; body weight 449.6 ± 19.7 kg) were feddiets consisting of corn silage as the sole forage source in a 3 period (35-day) Latin square …
