The Pet Mobility Project, sponsored by Terri Smith and Jasper Jackson, is an initiative aimed at providing a custom mobility aid to a pet in the San Luis Obispo community. Amber Rappozo’s young German Shepherd, LeDoux, was selected as the project’s recipient. LeDoux suffers from hindleg weakness and, as a result, has limited mobility. This document provides a comprehensive overview of the Pet Mobility Project, detailing the motivation, background, and design approach behind the development of a LeDoux’s custom mobility aid.

The goal of this project is to develop a low-cost and accurate force measurement system and method of measurement to be used in the testing of grown cardiac tissue samples. The primary stakeholder in this project is Dr. Luke Perreault, a biomedical engineering professor at California Polytechnic State University San Luis Obispo who is studying grown cardiac tissue rings, and any of his student lab personnel. Other stakeholders include other researchers in the tissue engineering field who require a similar form of testing to the methods developed in this project.

Peripheral artery disease (PAD) affects an estimated 200 million people worldwide and is characterized by arterial narrowing that reduces perfusion to the limbs. Collateral arterioles can naturally restore blood flow through a remodeling process called arteriogenesis, making it a promising therapeutic target. However, the femoral artery ligation (FAL) model used to study arteriogenesis requires surgical access deep to the inguinal fat pad, and approaches for handling the fat pad vary widely across surgeons and protocols. Uncontrolled tipping during incisor retraction with clear aligners continues to limit orthodontic treatment. Different countermeasures have been proposed to improve the predictability of aligners, such as power ridges and attachments, while their efficacy requires further investigation. This work investigated the biomechanics of the overcorrection in reducing the tipping angle and moment during incisor retraction with a clear aligner, through in vitro experiments and finite element analysis (FEA). Specifically, the influence of overcorrection angle (1 deg), retraction distance (0.15 mm), and the combination of both were assessed through in vitro experiments and the finite element simulation. Peripheral Artery Disease (PAD), caused by plaque buildup and narrowing of the arteries, reduces blood flow to the extremities. Promoting collateral arteriogenesis can help redirect blood flow. However, simple growth of collaterals may not be enough to reverse PAD. Instead, the ability for the collaterals to vasodilate and control blood flow is important, meaning assessing collateral function is important for the arteriogenesis assesment. Although microscopy techniques can measure vasodilation, contrast limitations in larger vessels such as collaterals make blood flow measurements difficult. Peripheral nerve stimulation is clinically used to manage pain, rehabilitate motor function, and promote nerve regeneration. The sciatic nerve is often studied for stimulation due to its accessibility and clinical relevance. A conventional nerve cuff electrode is the least invasive method for nerve stimulation, but the mismatch of mechanical properties of the electrode with the nerve tissue can cause damage to the nerve and the electrode itself. Congenital hemiparesis is a unilateral motor impairment stemming from brain injury in utero or early postnatally. Hemiparesis can be difficult to detect in early infancy with current clinical tools. Yet, early identification of motor asymmetry could play a key role in the design of effective early intervention and rehabilitation strategies. Embedded system security in implantable medical devices (IMDs) remains an underexplored challenge due to the unique constraints of these systems. Specifically, pacemakers are optimized for ultra-low power consumption to maximize device lifespan, are highly resource constrained, and demand reliable and correct operation to prevent severe health consequences. These systems rely on sensing electrodes, called leads, to detect cardiac activity and stimulate the patient’s heart when appropriate. Therefore, pacemaker lead integrity is a critical issue, as malfunction or compromise can result in device failure jeopardizing patient safety.

Microalgae are a promising renewable source of biofuel due to their high lipid content and rapid growth rates; however, algae cells must be electroporated (lysed) to release these lipids. The system developed in this project applies short (40 μs–10 ms), high-intensity (up to 33 kV/cm) pulsed electric fields (PEF) across cell membranes to enable electroporation.
Our lab-scale PEF chamber generates uniform, high-intensity electric fields using parallel-plate electrodes with sub-millimeter spacing. The system is designed to efficiently lyse microalgae cells for lipid extraction and biofuel production.

This project developed and evaluated an optical sensing system for detecting changes associated with glucose concentration. The system combined a laser-diode, cuvette sample holder, photodiode, resistive-feedback transimpedance amplifier, high-resolution analog-to-digital converter, and microcontroller. Parametric testing evaluated the effects of input current and feedback resistance on transimpedance gain, output range, and linearity. Firmware was developed to configure the ADC, average repeated conversions, monitor measurement variation, convert raw digital counts into voltage using a source-meter calibration equation, and compare sample measurements with a water reference. A cuvette enclosure maintained alignment between the laser-diode, sample, and photodiode while reducing external optical interference. Testing …

Deep-sea fishing presents significant challenges for individuals with upper-limb differences, particularly when using prosthetic devices that are not designed for recreational fishing activities. This project developed a custom prosthetic fishing rod attachment for a user who had lost her forearm and desired a safe, comfortable, and independent method of fishing. Through interviews with the sponsor and an iterative design process, a lightweight aluminum attachment was created to interface directly with the user's existing prosthetic quick-connect system.

Cardiovascular catheter systems rely on laser-cut hypotubes to balance flexibility and structural integrity required for minimally invasive surgical procedures. Optimizing these mechanical characteristics remains a significant challenge. This study aims to investigate the influence of key laser-cut geometric parameters on the mechanical performance of hypotubes through finite element analysis.
Commercially representative laser-cut hypotubes were modeled and a finite element model was developed. Tissue engineering is widely used in research for investigating cellular proliferation, behavior, and responses to various stimuli. However, the predictive value of preclinical studies using cell culture plates is limited by the inability to recapitulate the complexity of the physiological microenvironment. Binocular dysfunctions are more prevalent in the persistent post-concussive symptoms (PPCS) population than in the general population. The most prevalent binocular disorder is convergence insufficiency (CI), affecting 3-17% of the general population and up to 10 times as many people with PPCS. CI makes it difficult to fuse or maintain fusion on targets at near, and its symptoms include double or blurry vision and headaches when performing close-range tasks such as reading, which can exacerbate PPCS symptoms. Lipid-based nanocarriers have emerged as a cornerstone technology for RNA therapeutics, enabling effective intracellular delivery for applications ranging from vaccination to gene regulation. However, current manufacturing approaches, particularly microfluidic-based platforms, face inherent limitations in scalability, throughput, and structural tunability due to their reliance on confined channel geometries and restricted mixing architectures. Entrainment of cerebellar nuclear (CN) cells via cerebellar transcranial alternating current stimulation (ctACS) has been reported in animals under ketamine/xylazine anesthesia. Our main objective was to demonstrate modulation of CN activity in unanesthetized, freely moving animals using ctACS. Multi-channel carbon-fiber electrodes were implanted into the interpositus nucleus for recording multi-unit (MU) activity, and thin-film electrodes were implanted subcutaneously over the posterior cerebellum for stimulation. A frequency-domain-based metric was developed to quantify modulation from MU signals. The results demonstrated modulation in a wide range of frequencies (4 Hz-300 Hz) as in anesthetized animals. Binocular vision relies on the coordination of near response triad composed in part by the vergence and accommodative systems. Binocular dysfunctions linked to these systems impact the quality of life of those affected by complicating day-to-day tasks and leading to further health concerns. This robustness of histopathology classification models under adversarial and real-world perturbations resembling clinical artifacts is being investigated.
Using whole-slide images from the CAMELYON17 cohort, four representative architectures—ResNet-18, ResNet-50, HIPT-2MLP, and ViT-B/16 —are benchmarked across controlled pixel-level distortions and artifact-like transformations. Adversarial methods include iterative Fast Gradient Sign, Projected Gradient Descent, Salt-and-Pepper noise, and the Adversarial Watermark—Stain Shift (AWSS). Three defense strategies—Randomized Smoothing, Adversarial Training, and an Artifact Detector—are evaluated for their ability to preserve diagnostic accuracy and model reliability. Structured perturbations consistently degrade performance, with transformer-based models showing the greatest sensitivity. The benchmark developed here offers a reproducible framework for …

Ethanol stress assays are commonly used to evaluate microbial tolerance, metabolic adaptation, and fermentation performance. However, manual liquid handling introduces variability across replicate wells and small-volume pipetting steps, limiting reproducibility and throughput. This study developed an automated OT-2 robotic workflow to generate replicated ethanol concentration gradients for high-throughput inhibition assays in engineered thermophilic biofuel strains. Kinetic plate-reader measurements were used to quantify ethanol-dependent growth responses under anaerobic fermentation conditions. Cucurbita pepo leaf extract (CPE) was incorporated into electrospun poly(methyl methacrylate)/ halloysite/chitosan/ CaCO₃ (PMMA/Hal/CS/CaCO₃) composite nanofibers to develop a novel biomaterial for accelerating wound healing rates. The different nanofibrous scaffolds were successfully fabricated and characterized through scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD). SEM analysis revealed uniform, smooth nanofibers, while FTIR and XRD confirmed the integration of CPE into nanofiber matrix, indicating an amorphous structure and effective dispersion of the extract. In vitro agar well-diffusion and antibiofilm assays revealed that the optimized formulation exhibited potent antimicrobial activity against wound-associated pathogens. Background: Breast cancer progression and metastasis are closely linked to alterations in the mechanical properties of tumor cells and their microenvironment. Softer, more deformable cells are often associated with higher metastatic potential. While atomic force microscopy (AFM) is the current gold standard for mechanical characterization, it is limited to surface measurements and can damage 3D cultures. It remains unclear how the mechanical properties evolve over time in normal versus malignant spheroids. Type 2 diabetes mellitus (T2DM) is a global metabolic disorder characterized by insulin resistance and impaired glucose uptake. Despite the availability of pharmacological therapies, limitations such as adverse effects and high costs highlight the need for alternative therapeutic candidates. This thesis presents a comprehensive framework for the automated tracking and visualization of articulatory movements based on magnetic resonance imaging (MRI) data. A well-known data analysis tool for markerless pose estimation, known as DeepLabCut, is investigated for this purpose. The performance of this tool is enhanced through the design and implementation of a pre-processor. DeepLabCut is a markerless pose estimation toolbox based on deep learning, which overcomes the issue of making manual annotations frame-by-frame. Limitations from manually marking the MRI images are addressed by implementing transfer learning with convolutional neural networks to achieve accurate, user-defined articulator tracking without markers. The developing brain is highly susceptible to alcohol-induced toxicity, often resulting in long-term deficits in executive function and behavioral regulation. Inhibitory control impairments are among the most prominent deficits observed in Fetal Alcohol Spectrum Disorder (FASD). This study investigated the neural mechanisms of inhibitory dysfunction using a multimodal MEG–DTI approach in 67 children aged 6–8 years (34 with FASD, 33 controls) who performed a Go/No-Go task. Source-level MEG analyses revealed reduced stimulus-locked cortical activation in the anterior cingulate cortex and significant group-by-hemisphere interactions in the superior parietal cortex and cuneus. Background: The indications for the use of cone beam computed tomography (CBCT) in orthodontics has grown since it was first introduced and dentists are discovering new ways that 3D images enhance diagnosis and treatment planning. Evaluating bone quality and quantity can be measured as bone density and thickness in CBCT imaging and is helpful for temporary anchorage device (TAD) placement in orthodontic treatment. Medium-resolution cryo-electron microscopy (cryo-EM) density maps preserve substantial information about protein secondary-structure organization; however, accurately recovering the topology and connectivity of α-helices and β-strands remains challenging due to noise, structural heterogeneity, and the intrinsic resolution limitations that obscure residue-level detail. Topology determination is a key intermediate step toward building atomic protein models from medium-resolution cryo-EM density maps. It requires identifying the correct correspondence and orientation between secondary-structure elements (SSEs), i.e., α-helices and β-strands, predicted from the amino-acid sequence and those detected in the three dimensional (3D) density map.
