Neurodegenerative diseases are thought to be brought on by genetic and environmental factors including exposure to compounds and extracts found in plants. Many substances extracted from plants have been shown to induce cell death in neurons. This experiment tests different plant compounds to determine if they induce death in neurons in cell culture. In particular, we focused on extracts and compound from the Asimina Triloba plant- a plant that had anecdotally been connected with Parkinson's Disease-like effects. Cultured neurons were treated with increasing concentrations of the compounds and extracts. Twenty-four hours after treatment, viability was assessed. Results show that while …

Manipulation of cellular components in synthetic environment has attracted interest for applications ranging from sensors to nanelectronics and from catalysis to biomedical devices. In such applications, biological-based processes such as specific recognition and self-assembly direct the formation of hierarchical structures, all at minimum energetic costs, with high efficiency and reliability.

Non-catalytic protein-carbohydrate interactions are an essential element of various biological events. This dissertation presents the work on understanding carbohydrate recognition mechanisms and their physical significance in two groups of non-catalytic proteins, also called lectins, which play key roles in major applications such as cellulosic biofuel production and drug delivery pathways. A computational approach using molecular modeling, molecular dynamic simulations and free energy calculations was used to study molecular-level protein-carbohydrate and protein-protein interactions. Biofuels produced from lignocellulosic biomass via the fermentation platform are sustainable energy alternatives to fossil fuels. Process Systems Engineering (PSE) uses computer-based tools and methods to design, simulate and optimize processes. Application of PSE tools to the design of economic biorefinery processes requires the development of simulation approaches that can be integrated with existing, mature PSE tools used to optimize traditional refineries, such as Aspen Plus. Including inherited genetic diseases, like lipoprotein lipase deficiency, and acquired diseases, such as cancer and HIV, gene therapy has the potential to treat or cure afflicted people by driving an affected cell to produce a therapeutic protein. Using primarily viral vectors, gene therapies are involved in a number of ongoing clinical trials and have already been approved by multiple international regulatory drug administrations for several diseases. Statin drugs are commonly prescribed to lower cholesterol levels in patients, but recently statins have been under investigation as a potential anti-cancer drug. Statins are HMG-CoA reductase inhibitors and subsequently inhibit the mevalonate pathway and cholesterol synthesis. Since cholesterol trafficking is vital in the growth and metastasis of cancer, statin drugs are being studied to explore their effects on cancer cells. CoQ10, one of the products from the mevalonate pathway, also has shown some potential anti-cancer properties, but its use to combat cancer is controversial. This study investigated natural product inhibitors of blood coagulation Factor XII (FXII). FXII initiates the Kallikrein Kinin-System (KKS) within the intrinsic coagulation pathway. Increased expression of this system leads to proinflammatory and procoagulant activities. Inflammation occurs through downstream activation in the KKS. Following the activation of FXII-dependent pathway, bradykinin (BK) binds to the BK receptor 2 (B2) and produces inflammatory signaling. Abnormally increased BK can result in life threatening Hereditary Angioedema (HAE), characterized by severe reoccurring swelling. FXII is critical for pathogenic thrombosis, but is dispensable for hemostasis. This unique property of FXII makes it an attractive drug target. We …

In microfluidics, efficiency and mixing time are the greatest disadvantages. These parameters hinder the application of microfluidic devices for biochemical and immunological assays. However, once these disadvantages have been overcome by optimizing the parameters of the microfluidic device, it becomes the important analytical tool. In this experiment, various designs of microfluidic devices have been both simulated using COMSOL software, and experimentally verified to obtain the optimized parameter such as depth and velocity for better mixing efficiency. The removal of organic material in free surface gravel bed filters was investigated and modelled. Two pilot wetland units of 7.5m x 1.5m x 0.7m were constructed adjacent to the University of Dar es Salaam waste stabilization ponds to receive maximum organic and hydraulic loading rates of less than 185 kg/ha/d and 110 l/m2/d, respectively. These units received wastewater of domestic characteristics from primary stabilization pond. β-Glucosidases (βgls) primarily catalyze the hydrolysis of the terminal glycosidic bond at the non-reducing end of β-glucosides, although glycosidic bond synthesis (called transglycosylation) can also occur in the presence of another acceptor. In the final reaction step, the glucose product or another substrate competes with water for transfer to the glycosyl-enzyme intermediate. Commercial scale production of biofuels from lignocellulosic feed stocks has been hampered by the resistance of plant cell walls to enzymatic conversion, primarily owing to lignin. This study investigated whether DypB, the lignin-degrading peroxidase from Rodococcus jostii, depolymerizes lignin and reduces recalcitrance in transgenic tobacco (Nicotiana benthamiana). The protein was targeted to the cytosol or the ER using ER-targeting and retention signal peptides. For each construct, five independent transgenic lines were characterized phenotypically and genotypically. There is a compelling need for the development of new sensory and neural prosthetic devices which are capable of more precise point stimulation. Current prosthetic devices suffer from the limitation of low spatial resolution due to the non-specific stimulation characteristics of electrical stimulation, i.e., the spread of electric fields generated. We present a visible light stimulation method for modulating the firing patterns of electrically excitable cells using surface plasmon resonance phenomena. In an attempt to combat the possibility of bacterial infection and insufficient bone growth around metallic, surgical implants, bioactive glasses may be employed as coatings. In this work, silica-based and borate-based glass series were synthesized for this purpose and subsequently characterized in terms of antibacterial behavior, solubility and cytotoxicity. Borate-based glasses were found to exhibit significantly superior antibacterial properties and increased solubility compared to their silica-based counterparts, with BRT0 and BRT3 (borate-based glasses with 0 and 15 mol% of titanium dioxide incorporated, respectively) outperforming the remainder of the glasses, both borate and silicate based, in these respects. Chitosan-based hemostats are promising candidates for immediate hemorrhage control. However, they have some disadvantages and require further improvement to achieve the desired hemostatic efficiency. Here, a series of 1% Ga2O3-containing mesoporous bioactive glass-chitosan composite scaffolds (Ga-MBG/CHT) were constructed by the lyophilization process and the effect of various concentrations of Ga-MBG (10, 30, and 50 wt %) on the hemostatic function of the CHT scaffold was assessed as compared to that of Celox Rapid gauze (CXR), a current commercially available chitosan-coated hemostatic gauze. Microbial biosensors can be used to provide information about the cells’ environment in large-scale fermentations. In this project an oxygen sensitive biosensor is being developed in Escherichia coli to determine what kind of conditions cells are growing in: aerobic or anaerobic. This project specifically studies expression from the fumarate and nitrate reductase (FNR) promoter (PFNR) under aerobic and anaerobic conditions. U.S. Food and Drug Administration (FDA) recommends using several model bacteriophages as surrogates of pathogenic mammalian virus for process validation. As these markers increase in complexity, the role of support substrates grows increasingly important. Limitations in existing support substrates include the potential for increased sensitivity, binding specificity, and bio-stability.
