Temperature-sensitive (Ts) plasmids are useful tools for genetic engineering, but there are currently none compatible with the gram positive, thermophilic, obligate anaerobe, Clostridium thermocellum. Traditional mutagenesis techniques yield Ts mutants at a low frequency, and therefore requires the development of high-throughput screening protocols, which are also not available for this organism. Recently there has been progress in the development of computer algorithms which can predict Ts mutations. Most plasmids currently used for genetic modification of C. thermocellum are based on the replicon of plasmid pNW33N, which replicates using the RepB replication protein. This dissertation presents work towards the development of a multiple finger, worn, dynamic display device, which utilizes a method of texture encoded information to haptically render graphical images for individuals who are blind or visually impaired. The device interacts directly with the computer screen, using the colors and patterns displayed by the image as a means to encode complex patterns of vibrotactile output, generating the texture feedback to render the image. In turn, the texture feedback was methodically designed to enable parallel processing of certain coarse information, speeding up the exploration of the diagram and improving user performance. Injury to the medial malleolus, the distal end of the tibia and one of the bones comprising the ankle joint, can occur in various loading scenarios. Open reduction/internal fixation (ORIF) to reattach the malleolar fragment to the proximal tibia can be achieved via various devices, however small fragments are particularly challenging to treat. In this study, computational finite element analysis (FEA) was utilized to investigate the fixation of transverse medial malleolar fractures by two cancellous screws or by a new fixation device, the Medial Malleolar Sled™. This study aims at designing and implementing a single channel stand-alone Brain-Controlled Switch (BCS) device, which records the electroencephalography (EEG) signals from the scalp using electrodes, amplifies it, eliminates interferences (associated with the EEG signals) and processes the EEG signals to extract and decode temporal signal features to determine user’s intention of regulating an external switch. The design of our “brain-controlled switch” device is implemented using a bio-potential amplifier and a microcontroller. Halorhodopsin (HR), a light-activated chloride ion pump, demonstrates potential for use as the scaffolding in an artificial retina. Retinal implants are needed to restore vision to people afflicted with ophthalmic diseases, such as age-related macular degeneration (AMD) and retinitis pigmentosa (RP). A protein-based chloride ion-patch would be utilized by the eye to create an influx of chloride ions, similar to ion concentrations in healthy retinas during the conversion of light stimuli to electrochemical signals. The device simulates the signals produced by each neuronal population during the control of a horizontal saccade and allows for observing and recording. It will serve as a valuable teaching tool in the field of neural control. Furthermore, the device will have applications in the realm of diagnosing and properly treating brain injury. The large quantity of alarms characteristic of clinical settings have resulted in nursing staff suffering from a phenomenon termed ‘alarm fatigue’. Alarm fatigue is desensitization via habituation – as these commonly heard noises cause caregivers to have delayed or no response to alarms. Alarm management was ranked number one on the annual top ten list of technology hazards afflicting healthcare institutions published by the ECRI Institute for 2012. Accurate tracking of hand grip force is an important consideration needed for a robust understanding in the study of human biomechanics. One aspect where it proves useful is in hand-arm vibration, such as from gripping a power tool. Biomechanical risk factors are physical stressors that act on the neuromuscular structures of the human body and are present in all occupational settings. Repetition and intensity of tasks performed can magnify the effects of the biomechanical risks that a worker is exposed to.
After reviewing the current literature on ergonomics in laparoscopic surgery it was clear that a comprehensive approach to quantitatively reporting on the biomechanical risk factors associated with surgical hand tools had never been performed. Oral feeding competency is a primary requirement for preterm infant hospital release. Currently there is no widely accepted method to objectively measure oral feeding. Feeding consists primarily of the integration of three individual feeding events: sucking, breathing, and swallowing, and the objective of feeding coordination is to minimize aspiration. The purpose of this work was to quantify the infant feeding process from signals obtained during bottle feeding and ultimately develop a measure of feeding coordination. Electrospun materials have potential use in many biomedical applications such as soft tissue replacements or as scaffolds to target drug delivery to local sites. Electrospinning is a polymer processing technique that can be used to create materials composed of fibers with diameters ranging from the micron to the nanoscale. We investigated the effects of microfibrous and nanofibrous electrospun polycaprolactone (EPCL) on innate, cell-mediated, and humoral components of the immune system. Results demonstrated that in both young (12 week) and old (6 month) mice, EPCL had no effect on various immune parameters. With its lack of immunotoxicity, EPCL presents an excellent …

Miniaturized multi-electrode arrays are MEMS devices that have found use as neural prosthetics (Neuro-MEMS). As implants they can interface with neurons as sensors or actuators and help compensate for loss of sensory input, motor control, or cognitive function. The micro-electrodes, studied here, were developed in-house. They have a vertically aligned gold nanowire array and are mounted on a sturdy fine gauge titanium needle. Hence, the bill of materials and design characteristics encourage its use as neural probe.
