An important pillar of precision medicine for oncology is the ability to identify patients who respond to treatment early into their therapy. Positron emission tomography (PET) allows physicians and researchers to measure changes in tumor behavior prior to noticeable differences in morphology.
Objective: Determine the utility of multiple tracers for PET in assessing early changes in tumor activity that result from treatment.

Background: The configuration of necrotic areas within the retinal pigmented epithelium is an important element in the progression of age-related macular degeneration (AMD). In the exudative (wet) and non-exudative (dry) forms of the disease, retinal pigment epithelial (RPE) cells respond to adjacent atrophied regions by secreting vascular endothelial growth factor (VEGF) that in turn recruits new blood vessels which lead to a further reduction in retinal function and vision. Anaerobic digestion (AD) is a microbiologically coordinated process with dynamic relationships between bacterial players. Current understanding of dynamic changes in the bacterial composition during the AD process is incomplete. The objective of this research was to assess changes in bacterial community composition that coordinates with anaerobic codigestion of microalgal biomass cultivated on municipal wastewater. An upflow anaerobic sludge blanket reactor was used to achieve high rates of microalgae decomposition and biogas production. Selectively permeable biological membranes composed of lipophilic barriers inspire the design of biomimetic carrier-mediated membranes for aqueous solute separation. This work imparts selective permeability to lipid-filled pores of silica thin film composite membranes using carrier molecules that reside in the lipophilic self-assemblies. The lipids confined inside the pores of silica are proven to be a more effective barrier than bilayers formed on the porous surface through vesicle fusion, which is critical for quantifying the function of an immobilized carrier. Patients with unilateral dysvascular transtibial amputation (TTA) adopt movement compensations that are required to maintain balance and achieve ambulation in the absence of ankle plantar flexion, and result in increased and asymmetric joint loading patterns. As a result, this population is at an increased risk of overuse injuries, such as low back pain (LBP). Clinical gait analysis is used to guide diagnostics in movement retraining following amputation, and is performed using instrumented (research based) or observational analyses (clinically based). Red blood cells (RBCs) are the most abundant cellular element suspended in blood. Together with the usual biconcave-shaped RBCs, i.e., discocytes, unusual-shaped RBCs are also observed under physiological and experimental conditions, e.g., stomatocytes and echinocytes. Stomatocytes and echinocytes are formed from discocytes and in addition can revert back to being discocytes; this shape change is known as the stomatocyte–discocyte–echinocyte (SDE) transformation. Conventional lens-based (far-field) fluorescence microscopy is a widely used imaging technique with spatial resolution up to 150–350 nm. However, this technology cannot discern very small structural features, because the spatial resolution is limited by diffraction to about half of the wavelength of light (λ/2,λ is the wavelength of light). Adaptive optics (AO) imaging tools enable direct visualization of the cone photoreceptor mosaic, which facilitates quantitative measurements such as cone density. However, in many individuals, low image quality or excessive eye movements precludes making such measures. As foveal cone specialization is associated with both increased density and outer segment (OS) elongation, we sought to examine whether OS length could be used as a surrogate measure of foveal cone density. In this study, a curcumin-loaded bacterial cellulose films were developed. Bacterial cellulose films were prepared by culturing Gluconacetobacter xylinus in coconut water-based medium. Curcumin, an active substance found in turmeric (Curcuma longa Linn), was then absorbed into never-dried bacterial cellulose pellicles by immersion in 0.5 and 1.0 mg/ml curcumin solutions, with absolute ethanol as solvent, for 24 hours. The curcumin-loaded bacterial cellulose pellicles were then air-dried. Controlled release of curcumin was achieved in buffer solutions containing Tween 80 and methanol additives, at pH 5.5 and 7.4. งานวิจัยนี้มีวัตถุประสงค์ในการพัฒนาไฮโดรเจลไฟโบรอินไหมไทย (SF) ที่ถูกกระตุ้นด้วยการเติมสารลดแรงตึงผิวประเภทประจุลบชนิดต่างๆ เพื่อประยุกต์ใช้ในการควบคุมการปลดปล่อยเคอร์คูมินและใช้เป็นเจลทากระตุ้นการหายของแผล โดยได้ทำการศึกษาอิทธิพลของสารลดแรงตึงผิวประเภทประจุลบชนิดต่างๆ ได้แก่ sodium octyl sulfate (SOS), sodium dodecyl sulfate (SDS) และ sodium tetradecyl sulfate (STS) ซึ่งมีสูตรโครงสร้างเคมีคล้ายกัน แต่มีความยาวของสายอัลคิลแตกต่างกัน ต่อพฤติกรรมการเกิดเจลไฟโบรอินไหมไทย พบว่าสารลดแรงตึงผิวประเภทประจุลบทั้ง 3 ชนิด สามารถเร่งการเกิดเจล ณ อุณหภูมิ 37oC, pH 7.4 ได้ในช่วงระยะเวลา 14 นาที ถึง 144 ชั่วโมง ขึ้นกับชนิดและความเข้มข้นของสารลดแรงตึงผิว กลไกการเกิดเจลดังกล่าวเป็นการเกิดร่วมกันระหว่างอันตรกิริยาของส่วนที่ไม่ชอบน้ำ อัตรากิริยาของไฟฟ้าสถิตระหว่างโมเลกุลของไฟโบรอินไหมไทยและโมเลกุลของสารลดแรงตึงผิวและการเกิดเจลตามธรรมชาติของไฟโบรอินไหมไทยโดยกลไกดังกล่าวจะกระตุ้นการเกิดโครงสร้างทุติยภูมิชนิด β-sheet ซึ่งมีเสถียรภาพและผันกลับไม่ได้ งานวิจัยนี้ พบว่า ไฮโดรเจลไฟโบรอินไหมไทยที่กระตุ้นด้วย STS 0.09% โดยน้ำหนัก มีระยะเวลาการเกิดเจลรวดเร็ว (20 นาที) เจลมีความคงตัวสูง อัตราการย่อยสลายช้า และสามารถปลดปล่อยเคอร์คูมินได้เนิ่นนาน นอกจากนี้ยังไม่เป็นพิษต่อเซลล์ผิวหนังของหนู เมื่อทดสอบในระดับห้องปฏิบัติการตามมาตรฐาน ISO 10993 part 5 และเมื่อทดสอบประสิทธิภาพของไฮโดรเจลในการยับยั้งการอักเสบและกระตุ้นการหายของแผลชนิดสูญเสียทั้งชั้นในหนูทดลองพบว่า ไฮโดรเจลไฟโบรอินไหมไทยที่ไม่บรรจุและบรรจุเคอร์คูมินสามารถยับยั้งการอักเสบของแผลได้ในช่วง 3 วันแรกของการรักษา และเร่งการหายของแผลโดยการกระตุ้นการสร้างเซลล์เยื่อบุผิวได้ดีเทียบเท่ากับแผลกลุ่มที่รักษาด้วยไฟบรินเจล อันเนื่องมาจากไฟโบรอินไหมมีความเข้ากันได้กับเนื้อเยื่อ และมีอัตราการย่อยสลายที่เหมาะสม ส่วนเคอร์คูมินมีฤทธิ์ในการยับยั้งการอักเสบ จึงสรุปได้ว่า ไฮโดรเจลไฟโบรอินไหมไทยนี้มีประสิทธิภาพในการควบคุมการปลดปล่อยเคอร์คูมินและการกระตุ้นการหายของแผล

งานวิจัยนี้มีวัตถุประสงค์ในการพัฒนาอนุภาคทรงกลมขนาดไมครอนจากเจลาตินเพื่อนำส่งสารสกัดชาเขียว Epigallocatechin 3-gallate (EGCG) ร่วมกับกรดไฮยาลูรอนิคสำหรับรักษาโรคข้อเสื่อม โดยได้ทำการศึกษาเปรียบเทียบสมบัติทางเคมี และกายภาพของเจลาติน 4 ชนิดได้แก่ เจลาตินชนิดเอ (GA) เจลาตินชนิดบี (GB) เจลาตินชนิดเอที่ดัดแปรด้วยเอธิลีนไดเอมีน (GE) และเจลาตินชนิดบีที่ดัดแปรด้วยซัคซินิคแอนไฮไดร์ด (GS) พบว่า GE มีปริมาณหมู่อะมิโนอิสระเพิ่มขึ้นเมื่อเทียบกับ GA ซึ่งเป็นเจลาตินตั้งต้นเนื่องมาจากการคอนจูเกตเอธิลีนไดเอมีนเข้าไปในโครงสร้าง และส่งผลมีค่าศักย์เซต้าเป็นบวกมากขึ้นและมีความชอบน้ำสูง ในขณะที่ GS มีปริมาณหมู่อะมิโนอิสระลดลงเมื่อเทียบกับ GB ซึ่งเป็นเจลาตินตั้งต้นเนื่องจากหมู่อะมิโนอิสระถูกใช้ไปในการคอนจูเกตซัคซินิคแอนไฮไดร์ด อย่างไรก็ตาม ผลสเปกตรัมจากการวิเคราะห์ Fourier-transform infrared spectroscopy ยืนยันการปรากฏของหมู่ฟังก์ชั่นที่แสดงคุณลักษณะเฉพาะของโปรตีนในเจลาตินทั้ง 4 ชนิด เจลาตินทั้ง 4 ชนิดถูกนำมาขึ้นรูปเป็นอนุภาคทรงกลมขนาดไมครอนด้วยเทคนิคอิมัลชั่นชนิดน้ำในน้ำมัน และเชื่อมขวางด้วยความร้อน หรือด้วยสารละลายกลูตารัลดีไฮด์ อนุภาคทรงกลมขนาดไมครอนเจลาตินที่ได้มีขนาดอยู่ในช่วง 43-49 ไมครอน ซึ่งมีความเหมาะสมสำหรับนำไปใช้ฉีดผ่านเข็มฉีดยาได้โดยไม่อุดตัน เมื่อเปรียบเทียบวิธีการเชื่อมขวาง พบว่าอนุภาคที่เชื่อมขวางด้วยสารละลายกลูตารัลดีไฮด์มีความคงตัวมากกว่าอนุภาคที่เชื่อมขวางด้วยความร้อนโดยประเมินจากอัตราส่วนการบวมน้ำและการละลายน้ำของอนุภาค อนุภาคทรงกลมขนาดไมครอน GE ที่เชื่อมขวางด้วยสารละลายกลูตารัลดีไฮด์มีร้อยละการเชื่อมขวางสูงที่สุด เนื่องจากมีหมู่อะมิโนอิสระจำนวนมากสำหรับการเกิดปฏิกิริยาเชื่อมขวางกับหมู่อัลดีไฮด์ของกลูตารัลดีไฮด์ และเมื่อทดสอบอัตราการย่อยสลายทางชีวภาพในระดับห้องปฏิบัติการในสภาวะจำลองร่างกายที่ไม่มีและมีเอนไซม์คอลลาจีเนส พบว่าอนุภาคทรงกลมขนาดไมครอน GE มีอัตราการย่อยสลายช้าที่สุด อนุภาคทรงกลมขนาดไมครอนเจลาตินทั้ง 4 ชนิดไม่มีความเป็นพิษต่อเซลล์ผิวหนังของหนูเมื่อทดสอบในระดับห้องปฏิบัติการตามมาตรฐาน ISO 10993 part 5 และเมื่อนำอนุภาคทรงกลมขนาดไมครอนเจลาตินทั้ง 4 ชนิดไปดูดซับสารสกัดชาเขียว EGCG และทดสอบอัตราเร็วในการปลดปล่อยสาร EGCG ในสภาวะจำลองร่างกายที่ไม่มีและมีเอนไซม์คอลลาจีเนส พบว่าอนุภาค GE สามารถปลดปล่อยสาร EGCG ได้อย่างคงที่และเนิ่นนานที่สุดในทั้ง 2 สภาวะ โดยการปลดปล่อยควบคุมด้วยกลไกการแพร่ของสาร EGCG จากอนุภาคเป็นหลัก ในขณะที่อนุภาค GA GB และ GS ปลดปล่อยสาร EGCG ด้วยกลไกการแพร่และการบวมน้ำหรือการย่อยสลายของอนุภาค จากผลการศึกษา สรุปได้ว่าอนุภาคทรงกลมขนาดไมครอน GE มีประสิทธิภาพเหมาะสมสำหรับการควบคุมการปลดปล่อยสาร EGCG ได้เนิ่นนาน อนุภาคทรงกลมขนาดไมครอน GE ที่กักเก็บสาร EGCG นี้ได้ถูกนำไปผสมกับสารละลายกรดไฮยาลูรอนิคเพื่อเตรียมเป็นยาฉีดสำหรับรักษาโรคข้อเสื่อมในสุนัขต่อไป โดยผู้วิจัยคาดหวังว่ายาฉีดที่พัฒนาขึ้นนี้จะมีประสิทธิภาพในการรักษาโรคข้อเสื่อมได้ดีขึ้น เนื่องจากการออกฤทธิ์ร่วมกันของสารสกัดชาเขียว …

Two specific drug-delivery applications were sought in this work using polyamidoamine (PAMAM) dendrimers. One drug-delivery system used a novel dendrimer hydrogel (DH) for sustained delivery of anti-glaucoma drugs. In this work, PAMAM G3.0 dendrimers were covalently bonded with poly(ethylene glycol) (PEG12000) molecules which were subsequently acrylated, resulting in photocurable DH conjugates. For pharmacological studies, DH were loaded with a solution of intraocular pressure lowering drugs, brimonidine and timolol maleate, and were characterized for in vitro release and ex vivo transport and uptake.
