Adipose-derived Stem Cells (ASC’s) provide an optimal source for potentially graft replacing, tissue-engineered vessel constructs due to their ease of extraction and ability to be patient-specific. In order to make ASC ring constructs a more viable replacement for grafts, they need to have mechanical properties similar to native arteries, which is largely influenced by the extracellular matrix protein collagen. TGFβ-1 is known to stimulate collagen production in ASC’s, so this study explores the effect that TGFβ-1 has on the resulting thickness and tensile strength of ASC ring constructs.

Polymer hydrogelshave shown to exhibit improved properties upon the addition of nanoparticles; however, the mechanical underpinnings behind these enhancements have not been fully elucidated. Moreover, fewer studies have focused on developing an understanding of how polymer parameters affect the nanoparticle-mediated enhancements. In this study, we investigated the elastic properties of silica nanoparticle-reinforced poly(acrylamide) hydrogels synthesized using crosslinkers of various lengths. Crosslinker length positively affected the mechanical properties of hydrogels that were synthesized with or without nanoparticles. This retrospective study shows that the majority of patients’ correlations between PSA and Testosterone during the on-treatment period is at least 0.90. Model-based duration calculations to control PSA levels during off-treatment are provided. There are two pairs of models. In one pair, the Generalized Linear Model and Mixed Model are both used to analyze the variability of PSA at the individual patient level by using the variable “Patient ID” as a repeated measure. In the second pair, Patient ID is not used as a repeated measure but additional baseline variables are included to analyze the variability of PSA.

Abdominal aortic aneurysms (AAA) are localized expansions of the aorta wall that continue to grow until they reach a critical size and fatally rupture. This growth is driven by the chronic disruption, degradation, and subsequent loss of aortal wall elastic fibers by matrix metalloproteinases (MMPs) secreted by inflammatory cells recruited to the aorta wall following an injury stimulus, and the inherent inability of vascular smooth muscle cells (SMCs) to naturally repair or regenerate elastic fibers. Stem cell fate has been linked to the mechanical properties of their underlying substrate, affecting mechanoreceptors and ultimately leading to downstream biological response. Studies have used polymers to mimic the stiffness of extracellular matrix as well as of individual tissues and shown mesenchymal stem cells (MSCs) could be directed along specific lineages. In this study, we examined the role of stiffness in MSC differentiation to two closely related cell phenotypes: osteoblast and chondrocyte. Computational models are useful tools to study the biomechanics of human joints. Their predictive performance is heavily dependent on bony anatomy and soft tissue properties. Imaging data provides anatomical requirements while approximate tissue properties are implemented from literature data, when available. We sought to improve the predictive capability of a computational foot/ankle model by optimizing its ligament stiffness inputs using feedforward and radial basis function neural networks. While the former demonstrated better performance than the latter per mean square error, both networks provided reasonable stiffness predictions for implementation into the computational model.

The mortality rate for patients requiring mechanical ventilation is about 35% and this rate increases to about 53% for the elderly. o facilitate high-throughput proteomic analyses we have developed a modified FASP protocol which improves the rate at which protein samples can be processed prior to mass spectrometry. Adapting the original FASP protocol to a 96-well format necessitates extended spin times for buffer exchange due to the low centrifugation speeds tolerated by these devices. However, by using 96-well plates with a more robust polyethersulfone molecular weight cutoff membrane, instead of the cellulose membranes typically used in these devices, we could use isopropanol as a wetting agent, decreasing spin times required for buffer exchange from an hour to 30 minutes. We have developed a miniature human liver (liver-sinusoid-on-a-chip) model using a dual microchannel separated by a porous membrane. Primary human hepatocytes and immortalized bovine aortic endothelial cells were co-cultured on opposite sides of a microporous membrane in a dual microchannel with continuous perfusion. Primary human hepatocytes in this system retained their polygonal morphology for up to 26 days, while hepatocytes cultured in the absence of bovine aortic endothelial cells lost their morphology within a week. In many in vitro experiments, primary cells are harvested from an animal species to undergo experimental manipulation and subsequent analysis. Cell sorters are a luxury to have after cell harvesting to ensure pure populations of cells. Recently, it was discovered that different cell types adhere to cell culture dishes at different strengths. Accurate antigen detection is imperative for clinicians to diagnose disease, assess treatment success, and predict patient prognosis. The most common technique used for the detection of disease-associated biomarkers is the enzyme linked immunosorbent assay (ELISA). In an ELISA, primary antibodies are incubated with biological samples containing the biomarker of interest. Then, detectible secondary antibodies conjugated with horseradish peroxidase (HRP) bind the primary antibodies. Upon addition of a color-changing substrate, the samples provide a colorimetric signal that directly correlates to the targeted biomarker concentration. We are using numerical solutions for the Navier-Stokes equations and the concentration - diffusion equation to model fluid flow and reactant distribution in serpentine type channels for micromixers/microreactors development. These mixers exploit centripetal forces on the fluid to induce cross-sectional fluid mixing, aka Dean flows. Various modifications are used to increase the mixing character of these crosssectional flows. We found that the performance of these mixers exceeds that of unmodified channels and we currently assess their performance relative to other state of the art methodologies used to induce mixing on the microscale.

Establishing the role that mechanics play in nerve cell (e.g. neurons) function requires experimental testing. Microfluidic based experiments are commonly used to study neuron growth and function, and studies have found mechanics to play an important role in neuron health. External loads can be applied to a microfluidic device using a motor, which presumably influences the mechanical environment of the cells. While a motor can easily apply known displacements, a “load cell” is necessary to measure corresponding forces.
