Genome-wide association studies (GWAS) have become a very effective research tool to identify genetic variants of underlying various complex diseases. In spite of the success of GWAS in identifying thousands of reproducible associations between genetic variants and complex disease, in general, the association between genetic variants and a single phenotype is usually weak. It is increasingly recognized that joint analysis of multiple phenotypes can be potentially more powerful than the univariate analysis, and can shed new light on underlying biological mechanisms of complex diseases. In chapter 1, I compared the performance of eight multivariate phenotype association tests. The motivation to conduct this power comparison paper is as follows. For nearly 15 years, genome-wide association studies (GWAS) have been widely used to identify genetic variants associated with human diseases and traits. Any counting system is prone to recording errors including underreporting and overreporting. Ignoring the misreporting pattern in count data can give rise to bias in the estimation of model parameters. Accordingly, Poisson, negative binomial and generalized Poisson regression have been expanded in some instances to capture reporting biases. However, to our knowledge, no program has been developed to allow users to apply all of these models when needed. Understanding what factors influence wildlife movement allows landscape planners to make informed decisions that benefit both animals and humans. New quantitative methods, such as step-selection functions, provide valuable objective analyses of wildlife connectivity. This paper provides a framework for creating a step-selection function and demonstrates its use in a case study. The first section provides a general introduction about wildlife connectivity research. The second section explains the math behind the step-selection function using a simple example. To date, genome-wide association studies (GWAS) have identified a large number of common variants that are associated with complex diseases successfully. However, the common variants identified by GWAS only account for a small proportion of trait heritability. Many studies showed that rare variants could explain parts of the missing heritability. Since the well-developed common variant detecting methods are underpowered for rare variant association tests unless sample sizes or effect sizes are very large, investigation the roles of rare variants in complex diseases presents substantial challenges. Despite antenatal care presenting opportunities to identify and monitor women at risk, use of recommended antenatal care services remains. Barriers preventing use of antenatal services vary between countries, and limited knowledge exists about the link between geographical settings and antenatal service use. The objective of this cross-sectional quantitative study was to explore geographical variations and investigate how social demographic characteristics affect use of antenatal care for women in Sierra Leone using the Andersen behavioral model. Linear regression is a widely used method for analysis that is well understood across a wide variety of disciplines. In order to use linear regression, a number of assumptions must be met. These assumptions, specifically normality and homoscedasticity of the error distribution can at best be met only approximately with real data. Quantile regression requires fewer assumptions, which offers a potential advantage over linear regression. In this dissertation, we aim to address three important questions in practice, which can be solved through complex survival models. The first project focuses on studying the longitudinal fitness effect on cardiovascular disease (CVD) mortality. In the second project, we study the disease-death relation between CVD and all-cause mortality and evaluate important covariate effects on the disease or death transitions. In the third project, we compare antiretroviral treatment (ART) for HIV patients and consider both treatment effect and side effect of the drugs. Structural Equation Modeling (SEM) is a framework of statistical methods that allows us to represent complex relationships between variables. SEM is widely used in economics, genetics and the behavioral sciences (e.g. psychology, psychobiology, sociology and medicine). Model complexity is defined as a model’s ability to fit different data patterns and it plays an important role in model selection when applying SEM. Cryo-electron microscopy (cryo-EM) is a structure determination method for large molecular complexes. As more and more atomic structures are determined using this technique, it is becoming possible to perform statistical characterization of side-chain conformations. Two data sets were involved to characterize block lengths for each of the 18 types of amino acids. One set contains 9131 structures resolved using X-ray crystallography from density maps with better than or equal to 1.5 Å resolutions, and the other contains 237 protein structures derived from cryo-EM density maps with 2-4 Å resolutions. This capstone is a data-driven investigation into New York City's rat problem. By using publicly available government data to map rat activity in NYC, I identified several socio-economic variables that correlate with rat populations at the community district, borough, and city-scale. I used these findings (mainly that rat problems are linked to lower incomes) as the basis of an investigation, which includes interviews with residents, experts, and city officials. Prof. Bobby Corrigan, urban rodentologist and formerly with the NYC Department of Health criticizes the city's efforts for the first time on the record.

Movement and habitat selection by Greater Sage-grouse (Centrocercus uropasianus) is of great interest to wildlife managers tasked with applying conservation measures for this iconic western species. Current technology has created small and lightweight GPS (Global Positioning Systems) transmitters that can be attached to sage-grouse. Using GIS software and statistical programs such as Program R, land managers can analyze GPS location data to assess how sage-grouse are geospatially interacting with their habitats. Increased availability of data and accessibility of computational tools in recent years have created an unprecedented upsurge of scientific studies driven by statistical analysis. Limitations inherent to statistics impose constraints on the reliability of conclusions drawn from data, so misuse of statistical methods is a growing concern. Hypothesis and significance testing, and the accompanying P-values are being scrutinized as representing the most widely applied and abused practices. Background: Previous research has developed an observational tennis serve analysis (OTSA) tool to assess serve mechanics. The OTSA has displayed substantial agreement between the two health care professionals that developed the tool; however, it is currently unknown if the OTSA is reliable when administered by novice users.

Purpose: The purpose of this investigation was to determine if reliability for the OTSA could be established in novice users via an interactive classroom training session.

Methods: Eight observers underwent a classroom instructional training protocol highlighting the OTSA. Following training, observers participated in two different rating sessions approximately a week apart. Each observer …

MicroRNA (miRNA) expression varies in association with different tissue types and in diseases. Having been found in body fluids including blood and cerebrospinal fluid (CSF), miRNAs constitute potential biomarkers. CSF miRNAs have been proposed as biomarkers for neurodegenerative diseases; however, there is a lack of consensus about the best candidate miRNA biomarkers and there has been variability in results from different research centers, perhaps due to technical factors. Red Drum, Sciaenops ocellatus, are highly sought after by sport fishermen in Mississippi coastal waters. In 2016, Mississippi anglers made over 180,000 fishing trips targeting Red Drum, making it the second most targeted marine species. The current Fishery Management Plan of the Gulf of Mexico Fishery Management Council, prohibits harvest of Red Drum in federal waters. Monitoring of the stock in Mississippi state waters occurs at sites that are almost exclusively estuarine, using gear types selective for juvenile fish. Introduction: Differential scanning calorimetry (DSC) is used to determine thermally-induced conformational changes of biomolecules within a blood plasma sample. Recent research has indicated that DSC curves (or thermograms) may have different characteristics based on disease status and, thus, may be useful as a monitoring and diagnostic tool for some diseases. Since thermograms are curves measured over a range of temperature values, they are often considered as functional data.
