Research and design of drugs for treatment against microbial infections require the study of pathogenic proteins involved during infection and replication. Drugs can be designed to interfere with the interaction network of these pathogenic proteins and inhibit the infection process. Determination of the structure and ligands of the target proteins in microbes is essential for designing mechanistic-based drugs. Two Chlamydia proteins involved in host cell invasion were investigated in this study. Purpose: to find, modify, and/or create grade-level appropriate labs and demonstration activities for 5th-8th graders, with a specific focus on activities hat involve easily available components. In enzymatic and synthetic catalytic oxidations, high-valent iron-oxo intermediates play a vital role as the active oxidant. In this regard, many synthetic metal catalysts are designed as biomimetic models to resemble the active site of Cytochrome P450 enzymes (P450) which are the predominant oxidation catalysts in nature. High-valent transition metal-oxo intermediates play essential roles as active oxidizing species in enzymatic and biomimetic catalytic systems. Extensive research has been conducted on a variety of transition metal catalysts being studied as models for the ubiquitous cytochrome P450 enzymes. This dissertation exploits properties inherent to azide-alkyne cycloaddition and applies practical solutions to difficult problems. Chapter II addresses structure-property relationships in glassy azido-alkyne matrices by varying the identity of the central linkage within tetrapropargyl bis-aniline-type crosslinkers, and by the addition or omission of Cu(I) catalyst. This systematic study showed that an ether or methylene linkage yielded lower melting tetrapropargyl crosslinkers that were soluble in, and produced homogeneous, networks when cured with, a standard azido resin, di(3-azido-2-hydroxypropyl) ether of bisphenol-A; in contrast, a sulfone linkage yielded a relatively insoluble crosslinker and poorly dispersed, heterogeneous networks when reacted with the same …

Amino acids are also considered highly labile during organic matter recycling, making them useful proxies for organic carbon cycling. One type of fluorinated compounds is fluoroalkene-based peptidomimetics which are structurally similar to amide-containing compounds. They have been prepared to improve the pharmaceutical properties such as bioactivity, target specificity, and metabolic stability of the bioactive amides.

Bomb calorimetry was explored as a new method for determining the cation exchange capacity (CEC) of clays. Smectite clays were modified with several alkyl ammonium salts varying in number of carbons and the spatial orientation of the carbons about the central nitrogen atom. The clays used, standards purchased from the Source Clay Repository, have CECs of 44, 80, 88, and 120 meq/100 g. Theoretically, the combustion energy of the organo-clays should be approximately the same for each salt.
