The Study Of Drug Interactions During Separation Using Gas Chromatography – Mass Spectrometry, Leanne Mocniak 2016 Seton Hall University

The Study Of Drug Interactions During Separation Using Gas Chromatography – Mass Spectrometry, Leanne Mocniak

Capstone Projects/Undergraduate Honors Theses

There are numerous existing techniques for forensic toxicological testing, but each one has limitations. Although there have been significant improvements in these techniques over the past 10 years, it still remains very difficult to successfully identify a wide range of drugs from one sample using a single method [12]. This difficulty arises from the major differences in molecular structure, volatility and stability in solution between these drugs. The overall goal of this study is to develop a method using gas chromatography (GC) coupled with triple quadrupole mass spectrometry (GC/MS/MS) to separate and accurately identify 10 different drugs of abuse from …

Dendrimer Modified Silica Nanoparticles As Fluorescent Chemosensors For The Detection Of Copper And Cyanide Ions, Alicia Luhrs 2016 University of San Francisco

Dendrimer Modified Silica Nanoparticles As Fluorescent Chemosensors For The Detection Of Copper And Cyanide Ions, Alicia Luhrs

Master's Theses

A series of silica nanoparticle-based fluorescent chemosensors were synthesized for the sensitive and selective detection of Cu2+ ions in aqueous solution and subsequent CN- ion detection. Silica nanoparticles (SNP) were modified with branching polyamidoamine (PAMAM) dendrimers, which function as selective turn-off Cu2+ ion chelators and platforms for the attachment of rhodamine isothiocyanate (RITC) or fluorescein isothiocyanate (FITC) fluorescent dyes. In the presence of Cu2+ ions, the emission of the fluorescent dye is quenched by Forster resonance energy transfer (FRET) and is dependent upon dendrimer generation, pH, fluorescent dye, and surface charge. The quenching effectiveness was analyzed …

Application Of An Ionic Liquid Column To The Analysis Of Flavor And Fragrance Ingredients, Nicole L. Curto 2016 Seton Hall University

Application Of An Ionic Liquid Column To The Analysis Of Flavor And Fragrance Ingredients, Nicole L. Curto

Seton Hall University Dissertations and Theses (ETDs)

Traditional, polar (polyethylene glycol/wax) stationary phase gas chromatography columns pose challenges for flavor and fragrance analysis particularly in regards to thermal instability at high temperatures, degradation when exposed to water, unchanging selectivity, and relatively short shelf lives. Recently, capillary columns using ionic liquids as stationary phases have become available. Ionic liquid columns offer a potential combination of high polarity and high temperature stability with unique selectivity. An in depth discussion about the history and development of column phase characterization, with specific emphasis on selectivity and polarity, will allow for a critical look at the polarity scale currently employed to characterize …

A Pilot Study Analyzing Solvents In Electronic Cigarette “E-Liquid” Using High Performance Liquid Chromatography, Austin Matranga, Olivia Sellers, Matthew Gartner 2016 Chapman University

A Pilot Study Analyzing Solvents In Electronic Cigarette “E-Liquid” Using High Performance Liquid Chromatography, Austin Matranga, Olivia Sellers, Matthew Gartner

Student Scholar Symposium Abstracts and Posters

Introduction: Electronic cigarettes, also known as an e-cigarette, are battery-powered devices that use a heating element to convert a liquid (“e-liquid”) into an inhalable aerosol. Their advertised use is as a nicotine delivery system minus the harmful chemicals. Since “e-liquids” and the electronic cigarettes are not regulated by the Food and Drug Administration, it is not well known under what conditions, if any, carcinogenic compounds are produced. There has been little research on the toxicity of electronic cigarettes. The aim of this study was to determine if formaldehyde or acetaldehyde are formed from the e-liquid by the high heat of …

Lead-Free Gunshot Residues As Forensic Evidence, Lashaundra A. Fambro 2016 Kennesaw State University

Lead-Free Gunshot Residues As Forensic Evidence, Lashaundra A. Fambro

Master of Science in Chemical Sciences Theses

The analysis of gunshot residue (GSR) has been forensically relevant for many years and several methods are well characterized in scientific literature. However, the manufacture and distribution of lead-free alternatives to small caliber firearms ammunition is increasing rapidly as these types of rounds are more environmentally friendly and safer for use in indoor shooting ranges. Removing lead-containing compounds (e.g., lead styphnate) from the primer mixture eliminates a significant chemical marker vital to the legal precedent governing GSR analysis. The American Society for Testing and Materials (ASTM) standard ASTM E1588-95 is no longer applicable to such residues. With this emerging market, …

Quantifying The Use Of Chloroform Vapor Exposure To Improve The Adhesion Of Au Thin Films Onto Pmma, Kathleen T. Krist 2016 James Madison University

Quantifying The Use Of Chloroform Vapor Exposure To Improve The Adhesion Of Au Thin Films Onto Pmma, Kathleen T. Krist

Senior Honors Projects, 2010-2019

The metallization of Au onto plastics is an important processing step in applications such as the aerospace and automotive industries, the field of microelectronics, and the fabrication of microfluidic devices. While its corrosion resistance and excellent electrical and thermal conductivity make Au a useful choice, its inertness results in poor adhesion to polymer surfaces. Previous studies have indicated that exposing commercially available poly(methyl methacrylate) (PMMA) sheets to chloroform vapor following Au deposition significantly improves adhesion. In this study, we utilized electron-beam evaporation and magnetron sputtering to deposit Au thin films onto 1.50 mm thick PMMA and exposed the samples to …

Extracting (1,3/1,6)-Β-Glucans From Saccharomyces Cerevisiae: A Fungal Immunotherapeutic, James C. Elliott 2016 East Tennessee State University

Extracting (1,3/1,6)-Β-Glucans From Saccharomyces Cerevisiae: A Fungal Immunotherapeutic, James C. Elliott

Undergraduate Honors Theses

The goal of this research was the development of a method to extract pure (1,3/1,6)-β-glucans from Saccharomyces cerevisiae. These β-glucans are of pharmaceutical importance because an animal’s immune system can recognize glucan molecules, and these molecules can act as immunomodulators, essentially turning on the immune system. The problem in the past has been that previously published methods produce β-glucans with low side chain lengths and few branching occurrences. This issue was tackled by a multivariable approach that reduced extraction steps, initial sample size, and concentrations of reagents used. This method has been shown to produce greater yields of β-glucans while …

Microdialysis Studies Using Porcine Pancreatic Elastase To Guide Mathematical Modeling Of Microdialysis Sampling For In Vivo Measurements., Mason W. Young

Biomedical Engineering Undergraduate Honors Theses

Microdialysis sampling uses a semi-permeable membrane to allow solute collection by diffusion. When used in conjunction with other instruments, analytes in question can be quantified. Matrix Metalloproteinases (MMPs) are enzymes involved in numerous biological processes where they serve the role of degrading extracellular matrix. Microdialysis sampling, in coordination with further analysis methods, can be used in order to measure the activity of these enzymes in a region of the body instantaneously. The intention of this project is to determine ways of measuring in vitro activity of elastase from porcine pancreas using determined activity values and the collection of elastase products. …

Staphylococcal Nuclease And Ubiquitin Local Folding Energies And Rates Using Peps-Hdx-Esi-Ms, Julie Rhee

Chemistry & Biochemistry Undergraduate Honors Theses

In this study, Protein Equilibrium Population Snapshot Hydrogen-Deuterium Exchange Electrospray Ionization Mass Spectrometry (PEPS-HDX-ESI-MS) was applied to study the local regions of model proteins, staphylococcal nuclease and ubiquitin. The hydrogen deuterium exchange (HDX) has become a key technique for studying the structural and dynamic aspects of proteins in solution. This technique creates a rapid exchange between all of the exchangeable hydrogen ions with deuterium when the protein is exposed to a solvent. The PEPS method is an equilibrium-based method used to determine the populations of the closed native and open denatured states of a protein. By combining the applications of …

The quantitation of opiates was conducted from the livers of sixty-four possible overdose cases that were brought into the Erie County Medical Examiner’s Office between 2013 and 2015. Results showed that the opiate drugs could successfully be quantitated using the liver analysis. Generally …

Curing Of Polymer Thermosets Via Click Reactions, Mark Richard Brei 2016 University of Southern Mississippi

Curing Of Polymer Thermosets Via Click Reactions, Mark Richard Brei

Dissertations

In the first project, an azide functional resin and tetra propargyl aromatic diamines were fabricated for use as a composite matrix. These systems take already established epoxy/amine matrices and functionalize them with click moieties. This allows lower temperatures to be used in the production of a thermoset part. These new systems yield many better mechanical properties than their epoxy/amine derivatives, but their Tgs are low in comparison.

The second project investigates the characterization of a linear system based off of the above azide functional resin and a difunctional alkyne. Through selectively choosing catalyst the linear system can show …

Immobilization Of Heteropolyacids In Silica Gel, Opeyemi Adetola 2016 East Tennessee State University

Immobilization Of Heteropolyacids In Silica Gel, Opeyemi Adetola

Electronic Theses and Dissertations

Silica gels containing incorporated heteropolyacids (HPAs) were synthesized in acidic media by co-condensation of tetraethoxysilane (TEOS) with phosphotungstic or phosphomolybdic acids using sol-gel technique. Effect of the synthesis conditions on their structure and morphology was studied. Yields of modified materials were some lower as compared to non-modified silica gels. All materials were mesoporous but contained micropores in their structures. Presence of bands of Keggin’s structures in FT-IR spectra along with absence of XRD patterns of crystalline HPAs confirmed their fine incorporation into silica network. Particle sizes of modified materials were 800-1100 nm excepting for W-containing sample obtained with trimethylstearylammonium chloride. …

Chemoselectivity is the preferential reaction of a reagent with a select functional group among other plausible reactions. This dissertation describes chemoselective reagents and methods explored to develop more accurate, efficient and high-throughput means to quantify metabolites containing either thiol, carboxylic acid or 1,2-diol moieties directly from biological mixtures. Chapter 1 discusses how dysregulation of thiol- or carboxylic acid-containing metabolites is either the cause or effect of cellular dysfunction. Quantifying such metabolites is important in discovery of pathways to understand etiology of diseases. Chapter 1 also discusses the need for sustainable syntheses using renewable resources and describes the traditional methods for …

Facilitating Conceptual Learning In Quantitative Chemistry, Sarah R. Johnson 2016 East Tennessee State University

Facilitating Conceptual Learning In Quantitative Chemistry, Sarah R. Johnson

Electronic Theses and Dissertations

Traditional chemistry laboratory courses have a manual consisting of “step-by-step” experiments; instructions are given to complete experiments, requiring minimal information/concepts processing to be successful. This experience leaves students unprepared for the real-world, where critical thinking skills are needed to conduct research. This study focused on building analytical techniques, conceptual knowledge, and critical thinking skills used to solve research problems. A new quantitative chemistry laboratory manual was developed to transition students from traditional to inquiry-based experiments, requiring analytical method development. Data showed students having less difficulty using the new manual (0.8281 average difficulty) on method development exam questions and experiments, compared …

Development Of Electrochemical Sensors Suitable For In Vivo Detection For Neurotransmitters, Mengjia Hu 2016 University of Arkansas, Fayetteville

Development Of Electrochemical Sensors Suitable For In Vivo Detection For Neurotransmitters, Mengjia Hu

Graduate Theses and Dissertations

The electrochemical method of redox cycling was exploited to achieve new discoveries in neurotransmitter detection and to advance its suitability toward in vivo use. Redox cycling has advantages in signal amplification, selectivity of species based on their electrochemical reaction mechanisms, and limited or no background subtraction. Distinction of dopamine from norepinephrine in a mixture with an electrochemical method at unmodified electrodes was demonstrated for the first time in vitro. This ability resulted from a series of fundamental studies of redox cycling behavior of the catecholamines (dopamine, norepinephrine and epinephrine) using different electrode configurations. Taking advantage of the ECC’ mechanism associated …

Lipid Content And Biomass Analysis In Autotrophic And Heterotrophic Algal Species, Addie M. Lauder, Daniel P. Jones, Thomas E. Walker, Todd Allen

Montview Journal of Research & Scholarship

Biofuels are a form of renewable energy derived from living matter, typically plants. The push for biofuels began in order to decrease the amount of carbon dioxide (CO2) released into the atmosphere, as biofuels are essentially carbon neutral. The idea is the same amount of CO2 the plants took in to perform photosynthesis will then be released in the burning of the biofuels. Algae is an excellent source of biofuels because it grows quickly and is versatile in terms of the type of fuel it can produce. The two most common mechanisms for algae growth are heterotrophic or photoautotrophic. Heterotrophically …

Binding Of Oxygen To Human Hemoglobin Within The Erythrocyte Using Icam Spectrophotometry, Kyle K. Hill 2016 University of Nebraska-Lincoln

Binding Of Oxygen To Human Hemoglobin Within The Erythrocyte Using Icam Spectrophotometry, Kyle K. Hill

Department of Chemistry: Dissertations, Theses, and Student Research

Many of the spectrophotometric techniques used to determine the properties of intracellular human hemoglobin cannot be utilized due to the turbidity of erythrocyte suspensions. An Integrating Cavity Absorption Meter, or ICAM, allows for absorption measurements of strongly scattering samples in the visible-light region of the spectrum. The spectrum of oxygenated hemoglobin within erythrocytes is significantly different from the absorption spectrum of oxygenated hemoglobin in solution. Studies of the oxygen binding to hemoglobin in erythrocytes allowed the four sequential binding constants (Adair constants) to be determined and compared with those of hemoglobin in solution. The Adair constants for hemoglobin in solution …

Photocatalytic Properties Of G-C3n4–Tio2 Heterojunctions Under Uv And Visible Light Conditions, Rachel Fagan, Declan Mccormack, Steven J. Hinder, Suresh Pillai

Articles

Graphitic carbon nitride (g-C3N4) and titanium dioxide (TiO2) were chosen as a model system to investigate photocatalytic abilities of heterojunction system under UV and visible light conditions. The use of g-C3N4 has been shown to be effective in the reduction in recombination through the interaction between the two interfaces of TiO2 andg-C3N4. A simple method of preparing g-C3N4 through the pyrolysis of melamine was employed, which was then added to undoped TiO2 material to form the g-C3N4–TiO2 system. These materials were then fully characterized by X-ray diffraction (XRD), Brunauer Emmett Teller (BET), and various spectroscopic techniques including Raman, X-ray photoelectron …

Photocatalytic Reduction Of Fumarate To Succinate On Zns Mineral Surfaces, Ruixin Zhou, Marcelo I. Guzman 2016 University of Kentucky

Photocatalytic Reduction Of Fumarate To Succinate On Zns Mineral Surfaces, Ruixin Zhou, Marcelo I. Guzman

Chemistry Faculty Publications

The reductive tricarboxylic acid (rTCA) cycle is an important central biosynthetic pathway that fixes CO2 into carboxylic acids. Among the five reductive steps in the rTCA cycle, the two-electron reduction of fumarate to succinate proceeds nonenzymatically on the surface of photoexcited sphalerite (ZnS) colloids suspended in water. This model reaction is chosen to systematically study the surface photoprocess occurring on ZnS in the presence of [Na2S] (1–10 mM) hole scavenger at 15 °C. Experiments at variable pH (5–10) indicate that monodissociated fumaric acid is the primary electron acceptor forming the monoprotic form of succinic acid. The following …
