Soybean aphid (Aphis glycines Matsumura) has been a major pest of soybean [Glycine max (L.) Merr.] in North America particularly in the northern United States and three Canadian provinces. At least four biotypes of soybean aphid have been confirmed in the United States. Identification of soybean aphid resistance sources in early-maturing soybeans and genetic characterization of new sources of resistance will facilitate to expand the gene pool of soybean aphid resistance and thus will help to develop soybean aphid resistant cultivars. Gene stacking is a method used in biotechnology by which multiple genes can be placed at a single genomic site, thereby simplifying plant breeding. In this approach, DNA nucleases are used for excising selectable marker genes (SMG), which are the unneeded components of transgenic plants. The goal of this project is to evaluate the effectiveness of the nuclease I-SceI in excising DNA in plants. Specifically, this study tests heat-inducible I-SceI through the use of a heat-shock promoter (HS) in order to control SMG excision by heat application. Since 1964, the University of Arkansas blackberry breeding program has worked to improve fruit quality and shipping capabilities. A major limitation in blackberry fruit is postharvest handling potential for the shipping market. Maintaining fruit firmness in storage is crucial. Red drupelet reversion (or simply reversion) is also an important postharvest disorder in which drupelets change from black to red during storage. It is hypothesized that reversion is increased when fruit is picked at hot temperatures and exposed to a rapid change of temperature. Plants have an intriguing tripartite genetic system: Nuclear genome 9 Mitochondria 9 Plastids and their interactions may impact germplasm breeding. In grapevine, the study of cytoplasmic genomes has been limited, and their role with respect to grapevine germplasm diversity has yet to be elucidated. In the present study, the results of an analysis of the cytoplasmic diversity among 6073 individuals (comprising cultivars, interspecific hybrids and segregating progenies) are presented. Genotyping by sequencing (GBS) was used to elucidate plastid and mitochondrial DNA sequences, and results were analyzed using multivariate techniques. CYP82e4, CYP82e5, and CYP82e10 mutant alleles that minimize the conversion of nicotine to nornicotine have been introgressed into numerous existing low converting (LC) burley varieties and parental lines developed by the Kentucky-Tennessee tobacco breeding program. A backcross breeding protocol was utilized, with the objective being the creation of "e3" varieties that differed from their LC counterparts only for nornicotine and nitroso-nornicotine content. Field studies were conducted in Kentucky and Tennessee during the 2013 growing season, with 17 prospective parental lines and 20 prospective commercial varieties grown and compared to their original counterparts. Competition between plants for pollinators can have serious impacts on plant reproduction; these impacts depend on many factors, such as plant abundance, plant diversity, floral abundance, pollinator abundance, and pollinator preference. The way pollinators move among and between coflowering species can tell us more about how these factors affect competition. In this study, we examine the movement patterns of flower visitors to Mimulus ringens and coflowering species in Northeastern Ohio through several types of observations. The first objective of this study was to compare two methods of generating DH lines in tobacco (Nicotiana tabacum L.). C. zeae-maydis orients hyphal growth towards stomata (stomatal tropism) and forms infectious structures (appressoria) that are necessary for successful infection. Quantitative trait loci (QTL) utilizing a pedigree-based analysis (PBA) approach was performed in the peach breeding program for the first time. The pedigree consisted of seven-F1 populations, their parents, ancestors, and cultivars. Flesh firmness and titratable acidity (TA) were examined in 2011, 2012, and 2013. For TA, a consistent and strong QTL was identified on the proximal end of linkage group (LG) 5 of the peach genome. For flesh firmness, two QTLs were located on LG 4.
