Enabling Breeding For Targeted Heat Requirement In Peach, Omer Atagul 2022 Clemson University

Enabling Breeding For Targeted Heat Requirement In Peach, Omer Atagul

All Theses

Peach [Prunus persica (L.) Batsch] germplasm varies in heat requirement and bloom time. This variability can be utilized in breeding for targeted environment or overall climate resilience. Development of molecular tools to aid in targeted selection of dormancy related traits is needed to support breeding for these traits. This project furthers our understanding of heat requirement accumulation diversity and genetic control in peach.

Peach bloom date (BD) is determined by the dynamic relationship of temperature requirements, from the sequential fulfillment of chill requirement (CR) and heat requirement (HR). Knowledge of HR and its influence on BD would provide deeper …

Managing Stress Tolerance On Warm-Season Putting Greens In The Transition Zone, Thomas Edward Walton 2022 University of Arkansas, Fayetteville

Managing Stress Tolerance On Warm-Season Putting Greens In The Transition Zone, Thomas Edward Walton

Graduate Theses and Dissertations

The use of warm-season putting greens in the transition zone has increased in recent years. Ultradwarf bermudagrass (Cynodon dactylon (L.) Pers x C. transvaalensis Burtt-Davy) is the most prevalent warm-season putting green selection in the transition zone, however, newly developed greens-type zoysiagrass (Zoysia spp.) cultivars represent another potential selection for golf courses. The two major limitations of warm-season grasses in the transition zone are a general lack of cold- and shade-tolerance. Protective covers are essential to protect ultradwarf bermudagrass putting greens in the winter months. Unfortunately, golf courses can still experience winterkill underneath protective covers. Two field trials were conducted …

Genetic Control Of Prickles And Plant Height In Blackberry, Carmen Johns 2022 University of Arkansas, Fayetteville

Genetic Control Of Prickles And Plant Height In Blackberry, Carmen Johns

Graduate Theses and Dissertations

The fresh-market blackberry (Rubus L. subgenus Rubus Watson) industry has been expanding for the past two decades. Blackberry market growth can be attributed to many factors including expanded production regions, improved production systems, and release of superior cultivars. Blackberry breeding is a time- and labor-intensive process which would benefit from implementation of molecular markers for highly heritable, important traits. Next generation sequencing, a high-quality reference genome, and software capable of analyzing this complex genome were applied in a genome-wide association study (GWAS) to reveal maker-trait associations related to prickles and plant height variation in fresh-market blackberry. In 2020 and 2021, …

Traits have been selectively bred for hundreds of years in order to produce a beautiful flower as well as essential agricultural products. Recently interest in the sunflower as a food and fuel crop has led to selective breeding of seeds that produce high yields of sunflower oil. To further push the limits of this plant's genome, researchers have used mutagenesis to force mutations in hopes of developing novel modifications that could increase yield. With the entire genome mapped, the goal for many research …

Marker-Trait Association Mapping Of Sorghum Mosaic Virus Resistance In Sugarcane, Chenie Zamora 2022 Louisiana State University and Agricultural and Mechanical College

Marker-Trait Association Mapping Of Sorghum Mosaic Virus Resistance In Sugarcane, Chenie Zamora

LSU Master's Theses

Mosaic is a potentially devastating disease of sugarcane that had severe economic impact on Louisiana’s sugarcane industry in the 1920’s and has caused periodic problems ever since. The disease is vectored by migrating aphids that transmit Sugarcane mosaic virus (SCMV) and Sorghum mosaic virus (SrMV) in a non-persistent manner. Mosaic in Louisiana is currently caused by strains of SrMV. Mosaic has been controlled primarily by developing resistant cultivars. Recently, mosaic was detected in some near-commercial clones in the cultivar selection program. This prompted extensive screening of breeding program parent populations that detected infiltration of susceptibility and revealed the need for …

Center for Plant Science Innovation: Faculty Publications

Plasmodiophora brassicae is a soil-borne pathogen that attacks the roots of cruciferous plants and causes clubroot disease. CircRNAs are noncoding RNAs, widely existing in plant and animal species. Although knowledge of circRNAs has been updated continuously and rapidly, information about circRNAs in the regulation of clubroot disease resistance is extremely limited in Brassica rapa. Here, Chinese cabbage (BJN 222) containing clubroot resistance genes (CRa) against P. brassicae Pb4 was susceptible to PbE. To investigate the mechanism of cicRNAs responsible for clubroot disease resistance in B. rapa, circRNA-seq was performed with roots of ‘BJN 222’ at 0, 8, and 23 days …

Genetic Characterization Of Resistance To Bacterial Panicle Blight And Sheath Blight In Rice Using Qtl Linkage Analysis And Qtl-Seq, John Christian E. Ontoy

LSU Master's Theses

Bacterial panicle blight (BPB) and sheath blight (SB) are major diseases of rice causing significant yield reduction in conducive conditions. Few cultivars have shown reliable resistance to these diseases. In this study, a recombinant inbred line (RIL) population derived from the U.S. cultivars Jupiter (moderately resistant) and Trenasse (susceptible) was investigated to identify genomic regions associated with BPB and SB resistance. Phenotypes in BPB and SB, as well as days to heading (DH), of the RIL population were evaluated in the field. DH was found to be correlated to BPB and SB diseases. BPB was also positively correlated to SB. …

Molecular Evidence Of Cryptic Hybridization In The Japanese Nezasa Bamboos (Pleioblastus Section Nezasa), Ryan Long 2022 Jacksonville State University

Molecular Evidence Of Cryptic Hybridization In The Japanese Nezasa Bamboos (Pleioblastus Section Nezasa), Ryan Long

Theses

The genus Pleioblastus is a complex group of Southeast Asian temperate bamboos with 7-21 species, depending on taxonomic authority. This study tests the hypothesis that taxonomic complexity in this group is due in part to hybridization and subsequent backcrossing in natural populations, resulting in a reticulate evolutionary history and a contemporary species assemblage that includes cryptic hybrids. This hypothesis is supported by recent research on the temperate bamboos that revealed intergeneric hybrids involving Pleioblastus and other genera of temperate bamboos (Triplett and Clark, 2021). The objective of the current study was to use data from Amplified Fragment Length Polymorphism (AFLP) …

Estimating Effects Of Radiation Frost On Wheat Using A Field-Based Frost Control Treatment To Stop Freezing Damage, Brenton A. Leske, Ben Biddulph

Grain and Other Field Crops Research Articles

Crop phenotyping experiments have long struggled to have a reliable control treatment that excludes frost and associated freezing damage to plants. Previous attempts used a barrier, such as a removable shelter or cloth to exclude frost. However, these methods were labour intensive and varied in their effectiveness. An automated diesel heater was used to protect field plots of wheat (Triticum aestivum L.) from frost damage. In 2018 and 2019 there were 22 and 33 radiation frost events from July to October at the field site. The heater maintained canopy air temperature above freezing (>0 °C) for the duration of …

Reproductive stage frost poses a major constraint for wheat production in countries such as Australia. However, little progress has been made in identifying key genes to overcome the constraint. In the present study, a severe frost event hit two large-scale field trials consisting of six doubled haploid (DH) wheat populations at reproductive stage (young microspore stage) in Western Australia, leading to the identification of 30 robust frost QTL on 17 chromosomes. The major 18 QTL with the phenotype variation over 9.5% were located on 13 chromosomes including 2A, 2B, 2D, 3A, 4A, 4B, 4D, 5A, 5D, 6D, 7A, 7B and …

Increased map density and transferability of markers are essential for the genetic analysis of fruit quality and stress tolerance in interspecific grapevine populations. We used 1449 GBS and 2000 rhAmpSeq markers to develop a dense map for an interspecific F2 population (VRS-F2) that was derived by selfing a single F1 from a Vitis riparia x ‘Seyval blanc’ cross. The resultant map contained 2519 markers spanning 1131.3 cM and was highly collinear with the Vitis vinifera ‘PN40024’ genome. Quantitative trait loci (QTL) for berry skin color and flower type were used to validate the map. Four rhAmpSeq …

DNA methylation is an important epigenetic modification required for the specific regulation of gene expression and the maintenance of genome stability in plants and animals. However, the mechanism of DNA demethylation remains largely unknown. Here, we show that two SGS3-like proteins, FACTOR OF DNA DEMETHYLATION 1 (FDDM1) and FDDM2, negatively affect the DNA methylation levels at ROS1-dependend DNA loci in Arabidopsis. FDDM1 binds dsRNAs with 50 overhangs through its XS (rice gene X and SGS3) domain and forms a heterodimer with FDDM2 through its XH (rice gene X Homology) domain. A lack of FDDM1 or FDDM2 increased DNA methylation levels …

Motivation: Over the last decade, RNA-Seq whole-genome sequencing has become a widely used method for measuring and understanding transcriptome-level changes in gene expression. Since RNA-Seq is relatively inexpensive, it can be used on multiple genomes to evaluate gene expression across many different conditions, tissues and cell types. Although many tools exist to map and compare RNA-Seq at the genomics level, few web-based tools are dedicated to making data generated for individual genomic analysis accessible and reusable at a gene-level scale for comparative analysis between genes, across different genomes and meta-analyses. Results: To address this challenge, we revamped the comparative gene …
