The Role Of Glycogen Synthase Kinase 3 Β (Gsk-3Β) At The Cardiac Myofilament, Marisa Jean Stachowski
Dissertations
Altered kinase activity and localization is gaining appreciation as a mechanism contributing to cardiovascular disease. Previous work suggests that the kinase glycogen synthase kinase 3 β (GSK-3β) regulates contractile function of the myofilament, but this work was performed in vitro. The goal of this work was to investigate GSK-3β’s in vivo role in regulating myofilament function, the mechanisms involved, and the translational relevance in disease. GSK-3β’s myofilament localization was confirmed in wild type mouse and human tissue via western blot of myofilament enriched tissue and immunofluorescence. These experiments also revealed that GSK-3β phosphorylated at Y216 binds to the z-disc. We …

Using The Mulligan Mobilization With Movement And Fibular Repositioning To Treat High School Patients With A Grade One Lateral Ankle Sprain, Alexander Zettlemoyer, Rich Patterson, Russell Baker, Alan Nasypany
Clinical Practice in Athletic Training
Ankle injuries are common among high school athletics with the highest incidence rates in boys and girls basketball and boys football. Novel treatment paradigms like the Mulligan Mobilization with Movement (MWM) in conjunction with fibular repositioning tape have shown promising results in reducing pain, disability, and time lost due to lateral ankle sprains (LAS). However, research focusing on Mulligan treatment strategies and ankle swelling following a LAS is limited. Therefore, the purpose of this case series was to evaluate the combined effects of the MWM and fibular repositioning taping on swelling, function, and pain in high school patients with an …

Casting And Splinting Options For Pseudo-Jones Avulsion Fractures: An Evidence-To-Practice Review, Amber M. Kingsley, Aysha B. Reyes, Zachary K. Winkelmann
Clinical Practice in Athletic Training
Avulsion fractures of the fifth metatarsal bone occur when a portion of the bone is broken or pulled off the rest of the bone. These fractures are often seen in athletes, especially those of a young age. Avulsion fractures of the fifth metatarsal bone are commonly treated non-surgically with immobilization of the foot. There are several different options for immobilization of the foot with no clear result as to which immobilization method is the most effective. The purpose of this evidence-to-practice review was to summarize a systematic review on comparative outcomes of immobilization interventions on avulsion fractures of the fifth …

Suspension Training As An Effective Intervention To Improve Core Stability: A Critically Appraised Topic, S Stiltner, M Boham, D Melrose
Clinical Practice in Athletic Training
Clinical Scenario: The core is a functional unit connecting the upper and lower extremities consisting of abdominal, back, and hip musculature. Core stability is a determining factor in patient motor performance and provides optimal force production and precision of movement. A highly functional core is essential in injury prevention and rehabilitation. Traditional means of core stability training are performed on a stable surface. Emerging research suggests patients may benefit from core stability training on unstable mediums such as is used with suspension training. Clinical Question: Does suspension training improve core activity in an active population? Search Strategy: Key words used …

Analysis Of The Antimicrobial Activity Of The Novel Chemotherapeutic Drug, Tpp1, Against Pseudomonas Aeruginosa And The Hydrogel Delivery Of Water-Soluble Antimicrobial Compounds, Alex Gasper
Senior Independent Study Theses
Cancer is one of the leading causes of death in the world, and it is commonly linked with bacterial infections that often complicate treatments. Recently, chemotherapeutics have been developed that are able to act as anti-cancer agents using delocalized lipophilic cations (DLCs) that are able to specifically target mitochondrial membranes of cancer cells. TPP1 is a newly developed chemotherapeutic drug that has activity against bladder cancer and melanoma cell lines in vitro. In order to determine if TPP1 has antimicrobial activity, TPP1 was tested against a common bacteria, Pseudomonas aeruginosa, to determine if antimicrobial activity was present. This testing was …

Design And Validation Of Artificial Feeders To Study Feeding Preferences And Growth Of Hematophagous Juvenile Sea Lamprey (Petromyzon Marinus), Gracie Li-Ting-Wai
Theses and Dissertations (Comprehensive)
The parasitic and invasive land-locked sea lamprey Petromyzon marinus populations contributed to the collapse of the Great Lakes fisheries in the mid-1900s, while native anadromous populations are currently under threat and require conservation efforts for protection. Despite the increasing concern for both populations of this species, the juvenile stage of the sea lamprey’s life cycle has not been well studied. This is due, in part, to ethical concerns raised around holding live host fish with juveniles and the lack of an alternative feeding method to maintain these juveniles in the laboratory. The feeder should be designed to minimize fouling of …

The Exposome, Metabolic Dysfunction And Silicosis: A Pulmonary And Pathophysiology Study, Janet A. Thompson
Graduate Theses, Dissertations, and Problem Reports (ETD)
The exposome encompasses the concept that an individual’s health is a combination of genetic, environmental, and occupational exposures. A worker’s exposomal profile could potentially influence their susceptibility to hazardous occupational exposures. Over 35% of adults in the United States have metabolic dysfunction (MetDys), yet interactions between MetDys and hazardous occupational exposures are generally unknown. MetDys is associated with HFWD-consumption and obesity and induces a chronic low grade systemic inflammation and alters serum adipokine profiles. Silicosis, a progressive inflammatory lung disease, results from occupational inhalation exposure to respirable crystalline silica particles. This research tests the hypothesis “Inflammation resulting from HFWD-induced …

Microvascular Consequences Of Engineered Nanomaterial Inhalation Exposure And Subsequent Reproductive Outcomes, Krista Lee Garner
Graduate Theses, Dissertations, and Problem Reports (ETD)
Nano-titanium dioxide (nano-TiO2) is a commonly utilized engineered nanomaterial found ubiquitously in consumer products. While benefits of ENM utilization in are undeniable, nanoparticle inhalation has been linked to cardiovascular consequences and altered microvascular responses. Given these outcomes, it is important to explore vulnerable timepoints, such as gestation, as robust vascular adaptations are essential for maternofetal health.
The first aim of this dissertation was to assess myogenic responsiveness following maternal nano-TiO2 inhalation. Fetal aortic responses were evaluated on gestational day (GD) 20 to assess how exposure may impact the F1 generation. Myogenic responsiveness within the uterine radial arteries …

Amelioration Of Mitochondrial Bioenergetic Dysfunction In Diabetes Mellitus: Delving Into Specialized And Non-Specific Therapeutics For The Ailing Heart, Andrya Jean Durr
Graduate Theses, Dissertations, and Problem Reports (ETD)
Morbidity and mortality of the diabetic population is influenced by many confounding factors, but cardiovascular disease (CVD), remains the leading cause of death. Mitochondrial dysfunction is central in the development of cardiac contractile dysfunction, with decreased mitochondrial bioenergetic function, increased dependence on free fatty acid utilization, and a decrease in glucose utilization having been shown to contribute to contractile dysfunction. Strategies targeting the amelioration of mitochondrial bioenergetic function are attractive for limiting diabetes-induced heart failure, and preserving health-span. The goals of this dissertation were to assess two mitochondrial-centric approaches for the amelioration of mitochondrial and cardiac contractile dysfunction in diabetes …

Sleep And Cardiovascular Reactivity To An Acute Virtual Stressor, Alaina Grace Tiani
Graduate Theses, Dissertations, and Problem Reports (ETD)
Inappropriate response and magnitude of cardiovascular reactivity (CVR) to stress is a proposed mechanism through which environmental stressors are linked with poor cardiovascular health outcomes (Chida & Steptoe, 2010; Krantz & Manuck, 1984). Studies of reactivity to various laboratory tasks commonly control for factors known to influence CVR such as smoking, medication use, caffeine intake, and BMI; however, few have considered the influence of sleep on CVR. In order to determine whether sleep characteristics need to be assessed and considered in studies of CVR, this study aimed to examine the association between indices of sleep quality and the magnitude and …

Mechanistic Insight Into Chronic Stress-Induced Cerebrovascular Dysfunction, Emily Nicole Burrage
Graduate Theses, Dissertations, and Problem Reports (ETD)
Despite decades of research psychological stress remains a prevalent problem in society, often diminishing quality of life and increasing the risk of developing dementia. The brain plays a central role in the response to stress and as it is unable to store energy substrates, the brain relies heavily on dynamically regulated blood flow. Therefore, it is important that there is a tight coupling between neural activity and cerebral blood flow to meet metabolic demands and maintain critical brain function. It has been established that chronic psychological stress has detrimental effects on cerebrovascular function, potentially through increased superoxide anion production through …

Common Garden Study Reveals Frost-Tolerant, Generalist Northern Seed Sources Are Best Suited To Expand Range Of Quercus Rubra, Emily Lindback
Dissertations, Master's Theses and Master's Reports
Climate change is shifting the potential suitable range of northern red oak (Quercus rubra L.) faster than it can migrate, creating an adaptation lag. Quercus rubra is prominent in eastern North American forests and important for its carbon sequestration, ecological roles, and economic significance. Our study addresses which populations of Q. rubra are best suited for climatic conditions at the northern and central range limits. Our findings can inform forestry management policies such as assisted migration, where species are transferred within their native range to mitigate the effects of climate change. We planted over 800 Q. rubra seedlings …

How Exercise Affects The Musculoskeletal System In The Biological Sexes: A Literature Review, Lauren Connolly
Honors Program Theses
Exercise has countless benefits for the human body. Physical activity increases overall health and reduces the risk of developing many chronic diseases. While the entire body is impacted by exercise, the musculoskeletal system is perhaps targeted most directly. Muscle contraction is required to complete any form of exercise, which helps build muscle mass and consequently strengthens bones. The physiology of musculoskeletal growth after exercise is the same for all individuals. However, the magnitude of this growth is not the same for men and women. Men have a profound ability to gain muscle mass that most women are not capable of …

Microglia/astrocyte and B cell neuroimmune responses are major contributors to the neurological deficits after traumatic spinal cord injury (SCI). Bruton tyrosine kinase (BTK) activation mechanistically links these neuroimmune mechanisms. Our objective is to use Ibrutinib, an FDA-approved BTK inhibitor, to inhibit the neuroimmune cascade thereby improving locomotor recovery after SCI. Rat models of contusive SCI, Western blot, immunofluorescence staining imaging, flow cytometry analysis, histological staining, and behavioral assessment were used to evaluate BTK activity, neuroimmune cascades, and functional outcomes. Both BTK expression and phosphorylation were increased at the lesion site at 2, 7, 14, and 28 days after SCI. Ibrutinib …

There are functional benefits to exercise in muscle, even when performed late in life, but the contributions of epigenetic factors to late-life exercise adaptation are poorly defined. Using reduced representation bisulfite sequencing (RRBS), ribosomal DNA (rDNA) and mitochondrial-specific examination of methylation, targeted high-resolution methylation analysis, and DNAge™ epigenetic aging clock analysis with a translatable model of voluntary murine endurance/resistance exercise training (progressive weighted wheel running, PoWeR), we provide evidence that exercise may mitigate epigenetic aging in skeletal muscle. Late-life PoWeR from 22–24 months of age modestly but significantly attenuates an age-associated shift toward promoter hypermethylation. The epigenetic age of muscle …

An Exploratory Comparison Of Subjective Mental Fatigue Following A Task Designed To Replicate The Observation Of Game Film, Anthony Magdaleno, Lenny Wiersma, Barbara B. Meyer
International Journal of Exercise Science
International Journal of Exercise Science 15(6): 25-35, 2022. Laboratory-induced subjective mental fatigue (MF) has been shown to decrease sport-related performance (23, 38), yet there is a lack of research identifying tasks in real-world sport environments that induce MF (37). Since the identification of real-world tasks that induce MF may inform activities undertaken in the daily training and competition environments, the purpose of the current study was to compare changes in MF following a task designed to replicate the observation of game film to changes in MF following completion of a laboratory-based task (e.g., Stroop test). On separate counterbalanced visits, participants …
