Introduction: Despite the commonality of fasting, there still remains a lack of scientific research, specifically in regard to the impact of fasting on cardiovascular physiology. Thus, the goal of this research project was to further explore the relationship between fasting and cardiovascular physiology. As numerous bodies of research have characterized the adipocytokine adiponectin (ACRP30) as an anti-diabetic compound, this work serves as an analysis to partly elucidate the potential usage of supplementary adiponectin as a therapeutic compound in the treatment of type II diabetes mellitus. In this work, a meta-analysis was conducted to centralize large amounts of data on adiponectin’s insulin sensitizing characteristics in conjunction with like-minded studies utilizing metformin treatment as the diabetic standard of care. INTRODUCTION: For safe and efficient fireground performance, firefighters (FF) must consume air supplied by the self-contained breathing apparatus (SCBA) efficiently while sustaining high work rates. Firefighter research and fire department physical ability assessments typically quantify performance solely on task completion time (i.e., work rate), failing to consider the critical element of air consumption efficiency, which subsequently impacts the SCBA’s functional work duration. Circadian rhythms are approximately 24-hour cycles of behavior, physiology, and gene expression. In mammals, these circadian rhythms are generated by clocks located in nearly every tissue in the body. The function of circadian clocks is to synchronize physiology and behavior with environmental cycles such as the light-dark cycle. After menopause, when circulating levels of estrogens are very low, women are more susceptible to obesity comorbidities such as metabolic syndrome and cardiovascular disease, suggesting that estrogens regulate these processes. Estrogens could protect females from metabolic dysfunction by regulating circadian rhythms. Consumption of diets that are high in fat contribute to obesity. …

In the U.S., anxiety is recognized as an increasing range of mentally and physically debilitating psychiatric health disorders with significant economic repercussions. Over the last 20 years, several novel anti-anxiety therapies have entered the drug development pipeline, but none have made it to market.
The work in this dissertation focused on structurally modifying valerenic acid (VA), a structurally unique carboxylated sesquiterpene acid found in Valeriana officinalis. VA is putatively reported to have allosteric modulatory activity of the human GABAA receptor, a ligand-gated ion channel responsible for attenuating neurotransmissions. Recent evidence suggests that the gut microbiome could play a role in skeletal muscle plasticity, providing novel treatments for muscle wasting diseases and/or performance enhancements. I first sought to determine if the gut microbiome is necessary for skeletal muscle adaptation to exercise. Forty-two, four-month old, female C57Bl/6J underwent nine weeks of weighted wheel running or remained in cage with a locked wheel, without or without the administration of antibiotics (treated). In response to wheel running, I found that antibiotic depletion of the microbiome led to a blunted hypertrophic response in the soleus muscle as measured by normalized muscle wet weight …

Plants have evolved a sophisticated immune system to defend themselves against pathogens. This immune response can be triggered in response to pathogen-associated molecular patterns (PAMP) or specialized effectors that are recognized by the plant resistance (R) proteins. The latter, commonly referred to as effector-triggered immunity (ETI), is well known to induce broad-spectrum resistance throughout the plants. This phenomenon known as systemic acquired resistance (SAR) is regulated by several chemical signals including salicylic acid (SA), and free radical nitric oxide (NO) and reactive oxygen species (ROS). Although systemic lupus erythematosus (SLE) is known as the autoimmune disease that triggers inflammation and bone impairment, the mechanisms leading to the decrease in bone regeneration remain inadequately characterized. Patients with SLE have been observed high prevalence in bone loss due to host immune responses that may induce host antimicrobial activities. Here, this study used systemic and local lipopolysaccharide (LPS) administration for mimicking infectious condition in FcγRIIB−/− mice as a lupus murine model. There has been a higher prevalence of developing anxiety due to frequent episodes of stress among adults in recent years. Chronic anxiety can contribute to the prevalence of elevated blood pressure and hypertension. High anxiety and stress also contribute to overactivation of the sympathetic nervous system which can be quantified by increased muscle sympathetic nerve activity (MSNA). Sympathetic overactivation can lead to vasoconstriction and loss of arterial elasticity. Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease. To date, there are no significant disease-modifying treatments, and one limiting factor in treatment is the amount of time it takes for a patient to receive a diagnosis of ALS. This study examined multiple mouse models before symptom onset to help identify early changes in a reflex circuit of ALS mice. Dorsal root stimulation of the sacral spinal cord in multiple models ALS mouse models showed changes in the resulting ventral root compound action potential amplitude, latency, and ability to maintain synaptic depression. Using forelimb asymmetry analysis along with Montoya Staircase methodology we tested how a fluoxetine simvastatin ascorbic acid drug cocktail would affect recovery from a demyelinating event in a rat model, with the hypothesis that if administered then it would help female recovery but hinder male recovery. It was found that a fluoxetine simvastatin and ascorbic acid drug cocktail did not significantly enhance recovery from a demyelination model injury in female rats, and that the same drug cocktail significantly slowed male rat recovery from the same type of injury. Multiple sclerosis (MS) affects an estimated one million people in the US alone (Wallin et al., 2019). We modeled MS in rats using lysolecithin injection into the corpus callosum to diminish motor function unilaterally through demyelination, meanwhile treating the injury with Fluoxetine, Ibuprofen, and ascorbic acid (FIAA) to increase neurogenesis and oligodendrogenesis. Motor function was assessed using the Montoya Staircase test pre- and post-surgery. Motor capabilities recovered in the contralateral limb, but not in the ipsilateral, and recovery was not significantly affected by treatment. Evidence for sentience in animals distantly related to humans is often sought in observations of behavioral and neural responses to noxious stimuli that would be painful in humans. Most proposed criteria for painful sentience in “lower” animals such as decapod crustaceans have no necessary links to the affective (“suffering”) component of pain. The best evidence for painful affect in animals is learned aversion to stimuli associated with noxious experience, and conditioned preference for contexts associated with relief from aversive consequences of noxious experience, as expressed in voluntary behavior. Such evidence is currently lacking for any invertebrate except octopus.

Cognitive impairment is a serious global health issue due to its widely complicated pathologies that are linked to many diseases, including type 2 diabetes. Yet, detailed mechanisms of cognitive impairment associated with type 2 diabetes are unknown. Evidence suggests type 2 diabetes is associated with disrupted glucose metabolism, elevated methylglyoxal, and axon initial segment (AIS) shortening. Subtle changes of AIS could significantly contribute to neuronal dysfunction in neurological diseases (e.g., cognitive impairment). Utilizing in vitro dissociated mouse cortical cultures, we initially discovered elevated methylglyoxal shortens the AIS length of the immunofluorescent marker AnkyrinG (Griggs et al., 2021).
