Underlying Mechanism Of Ovarian Reserve Preservation From Gonadotoxicity, Yi Luan 2022 University of Nebraska Medical Center

Underlying Mechanism Of Ovarian Reserve Preservation From Gonadotoxicity, Yi Luan

Theses & Dissertations

Cancer therapies often lead to premature ovarian insufficiency by ovarian damage and follicle depletion. However, the oocyte death pathway induced by chemotherapies has been debated. Here we clarified the precise mechanism of ovarian depletion induced by cyclophosphamide and doxorubicin. Cyclophosphamide induced direct apoptosis in oocytes rather than overactivation of primordial follicles. Dormant oocytes instead of activated oocytes with high PI3K activity were more sensitive to cyclophosphamide. Although the ABL inhibitor, GNF2, has been proposed to ameliorate the apoptosis of primordial follicles against cyclophosphamide, GNF2 did not prevent the loss of primordial follicles induced by cyclophosphamide in the systems tested. Oocyte-specific …

With wearable technology growing in popularity and sophistication, there remains a need to determine the validity of these devices by independent observers. Validation studies of wearable technology can involve large amounts of data, with data preparation techniques that are not always clearly established. This can make attempts to reproduce the results difficult and does not allow researchers to gain guidance in how to perform their own analyses if they wanted to perform a similar study. Therefore, this paper details the process that was utilized to prepare and …

Atrial Fibrillation In Aging: Mechanisms And Potential Therapeutics, Mengmeng Chang 2022 University of South Florida

Atrial Fibrillation In Aging: Mechanisms And Potential Therapeutics, Mengmeng Chang

USF Tampa Graduate Theses and Dissertations

Atrial fibrillation (AF) is one of the most common cardiac arrhythmias seen in the clinics, and currently available antiarrhythmic pharmacotherapies in AF are not very effective. Although AF has been recognized as an aging-mediated disease, our understanding of the electrophysiological pathways that link aging and AF remain incomplete, which limits breakthroughs in the development of novel antiarrhythmic treatments for this disease. Studies have shown that aging increases the generation of reactive oxygen species (ROS) in the heart, and high levels of ROS have been associated with development of AF in animals and patients. Additionally, in some forms of AF, the …

Effects Of Titanium Dioxide Nanoparticle Injections On Tissue Function And Morphology In Goldfish (Carassius Auratus), Bryan Joel Patterson

Graduate Theses

Fish gills are sensitive to a variety of environmental factors because they regulate the exchange of oxygen and ions between the fish and the external environment. This study examined the effects of polyacrylic acid (PAA) coated titanium dioxide nanoparticle injections (TiO2 NPs) on goldfish (Carassius auratus) over 14 days. Following the experimental period, we examined gill histological samples and looked for signs of an inflammatory response through blood glucose levels and relevant gene expression. TiO2 injected fish developed a more robust ILCM (interlamellar cell mass) compared to the non-injected controls, which remained elevated over 14 days post injection. …

Integrative Physiology-Coupled Pilot-Centered Flight Simulation, Shawn C. Harrison

Electrical & Computer Engineering Theses & Dissertations

Maintaining the pilot’s physiological performance envelope within the limits of human capabilities may be crucial for avoiding hazardous physiological episodes in fighter aircraft that compromise safety. The main physiological episode of interest is impaired pilot respiration, better known as hypoxia caused by a high fraction of inspired oxygen (FiO2) at high altitudes and variation in accelerative gravitational forces (g-forces). Integrated into fighter aircraft is an Onboard Oxygen Generating System (OBOGS) developed to mitigate the necessity of gaseous and liquid oxygen cannisters [1]. OBOGS act as a life support in hypoxic environments by providing oxygen-rich air, thereby oxygenating the …

Establishing The Efficacy Of Non-Cellular Components Of Adipose-Derived Stromal Vascular Fraction In Promoting Angiogenesis., Daniel Benson 2022 University of Louisville

Establishing The Efficacy Of Non-Cellular Components Of Adipose-Derived Stromal Vascular Fraction In Promoting Angiogenesis., Daniel Benson

Electronic Theses and Dissertations

Microvascular disease is hallmarked by pathophysiological conditions such as endothelial senescence, intimal thickening which impairs vasodilation, and regression of the capillary beds causing tissue ischemia in the myocardium or in peripheral vascular networks. Adipose-derived stromal vascular fraction (SVF) has previously demonstrated the ability to revascularize tissue. Increasing evidence shows that regenerative cells elicit their therapeutic benefit by paracrine mechanisms, leaving open extracellular vesicles (EVs) as a potential crux of the cell therapy paradigm. To test this idea, three types of gelatin methacrylate hydrogels were employed: SVF gels, EV gels derived from SVF, and blank control gels, which were used in-vitro …

Effect Of Umbilical Cord Essential And Toxic Elements, Thyroid Levels, And Vitamin D On Childhood Development, Jesse Cottrell, Chelsea Nelson, Catherine Waldron, Mackenzie Bergeron, Abigail Samson, Monica Valentovic

Biomedical Sciences

Introduction: The in-utero environment has dramatic effects on childhood development. We hypothesized prenatal levels of inorganic agents, thyroid levels, and Vitamin D effect childhood development.

Methods: Umbilical cord blood was collected from April 3, 2013 to January 30, 2014 and analyzed for 20 different elements, thyroid and Vitamin D. A retrospective review (n = 60) was performed of well-child examinations from birth to 5 years old (y.o.).

Results: There were associations with calcium and 4 month BMI (p = <0.01), 12 month language (p = 0.03); Magnesium and 6 month language (p = 0.04) and gross motor skills at 5 years old (y.o.) (p = 0.03); Copper and 12 month fine motor (p = 0.02); Zinc with fine motor (p = <0.01) and language (p = 0.03) at 2 y.o.; Manganese was associated with language development at 2 y.o. (p = 0.02); Molybdenum and fine motor at 12 months of age (p = 0.02); Selenium with gross motor (p = 0.04) and BMI (p = 0.02) at 5 y.o.; Lead with cognitive function at 4 months (p = 0.04) and 2 y.o. (p = 0.01); Mercury with gross motor at 4 months (p = 0.04) and language at 2 y.o. (p = 0.02). Platinum at 12 months of age (p = <.01) as well as multiple associations at 5 y.o. (p = <.01). Thyroid function tests for free T3 were associated with multiple cognitive and physical milestones. T3 Uptake was associated with 5 y.o. gross motor skills (p = 0.02). Total and Free T4 was associated with cognitive development (p = <.01) and fine motor development, respectively. Vitamin D was associated with a delay of fine motor development (p<0.01).

Conclusion: There were multiple associations between umbilical cord essential and toxic elements, thyroid levels, and Vitamin D on childhood development.

Voltage-Gated Potassium Channel Dependent Mechanisms Of Cardiovascular Adaptation To Chronic Exercise., Sean Raph 2022 University of Louisville, School of Medicine

Voltage-Gated Potassium Channel Dependent Mechanisms Of Cardiovascular Adaptation To Chronic Exercise., Sean Raph

Electronic Theses and Dissertations

Exercise increases cardiac workload, escalating the demand for oxygen. The myocardial vasculature responds to the greater demand for oxygen by increasing blood flow to match the needs of the heart during augmented work. Increasing blood flow requires the coronary arteries dilate (a.k.a. vasodilation); this is mediated predominantly by vascular smooth muscle cell relaxation. Vasodilation is driven through inhibition of calcium influx into vascular smooth muscle cells. The prevention of calcium influx is largely mediated by efflux of potassium via potassium channels causing membrane hyperpolarization, which in turn closes voltage-dependent (or gated) calcium channels (VDCC). Changes in vascular smooth muscle cell …

Importance Of The Microhabitat And Microclimate Conditions In The Northern Gray-Cheeked Salamander (Plethodon Montanus) Across An Elevation Gradient, Trevor Chapman 2022 East Tennessee State University

Importance Of The Microhabitat And Microclimate Conditions In The Northern Gray-Cheeked Salamander (Plethodon Montanus) Across An Elevation Gradient, Trevor Chapman

Electronic Theses and Dissertations

The southern Appalachian Mountains have among the highest salamander diversity in the world, largely due to local speciation in the family Plethodontidae. Plethodontid salamanders (i.e., lungless salamanders) are particularly sensitive to habitat climate conditions due to their reliance on cutaneous respiration, and their immediate environmental conditions (microhabitat) likely influence their dispersion and activity more than the large-scale atmospheric conditions. The Northern Gray-cheeked salamander (Plethodon montanus) is restricted to high elevations in the Appalachian Mountains. Our goal was to investigate the relationship between P. montanus and its microhabitat by examining behavioral preference for climatic conditions, characterizing the microclimate with …
