Behavioral health organizations often face the challenge of high employee turnover rates, which can critically affect their ability to treat mental illnesses successfully. High counselor turnover at one such organization, BHOX, threatens the provision of timely access to quality mental care. This qualitative case study examined counselor turnover and its impact on the quality of behavioral health services. The Baldrige Excellence Framework was used to guide this descriptive case study of a nonprofit behavioral health organization on the East Coast of the United States, which serves men and women struggling with mental health disorders and substance abuse addictions. Commercial running power (RP) meters may provide endurance runners and coaches with improved precision for monitoring running intensity. Cyclists rely upon the relationship between cycling power and metabolic demand to regulate physiological intensity during training and racing, independent of changes in the physical environment (e.g., terrain, wind). Research demonstrates strong reliability and validity for a specific RP meter to indicate metabolic demand within controlled running conditions. Chronic systemic inflammation, known as inflammaging is considered a hallmark of aging and associated diseases. While adipose tissue has long been considered only as a caloric reservoir regulating systemic energy homeostasis, research in the past couple of decades substantiate its endocrine function to secrete an array of inflammatory mediators and hormones, which have physiological effects on multiple organ systems, and contribute to inflammaging. Redistribution of adipose tissue from subcutaneous to visceral depots, changes in the immune profile and the chronic inflammatory state are among the major sources of adipose tissue dysfunction with aging.

Among the earliest changes to occur in Alzheimer’s disease, metabolic dysfunction and chronic neuroinflammation are now known to be major driving forces in disease progression. The paradigm of ‘immunometabolism’ seeks to bridge these two facets, positing that metabolic transformations are indispensable in determining the response of immune cells, such as microglia – the brain’s resident immune population. Proinflammatory stimulation of microglia leads to a shift away from mitochondrial respiration towards a dramatic upregulation of the glycolysis pathway for energy production. Influenza-A virus (IAV) infects yearly an estimated one billion people worldwide, resulting in 300,000–650,000 deaths. Preventive vaccination programs and antiviral medications represent the mainstay of therapy, but with unacceptably high morbidity and mortality rates, new targeted therapeutic approaches are urgently needed. Since inflammatory processes are commonly associated with measurable changes in the cell membrane potential (Em), we investigated whether Em hyperpolarization via TREK-1 (K2P 2.1) K þ channel activation can protect against influenza-A virus (IAV)-induced pneumonia. Previous work has examined the temporal and behavioral aspects of proximate fitness on life history outcomes particularly regarding highly perturbed environments (i.e., climate and land use change, resource extraction, agricultural erosion, etc.). My work focuses on the less examined spatial aspect of these perturbed environments. Metabolic-associated fatty liver disease (MAFLD) affects 1 in 3 people in the US and is a healthcare burden on the order of billions of dollars. MAFLD is characterized by pathological lipid accumulation in the liver, excess production of triglycerides and cholesterol, and export of LDL particles that raise serum lipid levels. These metabolic perturbations can lead to the development of liver fibrosis, chronic inflammation, and insulin resistance. Diagnosing MAFLD prior to fibrosis is crucial for successful intervention and prevention of advanced liver disease and other metabolic complications. It has recently become evident that the bacterial stringent response is regulated by a triphosphate alarmone (pGpp) as well as the canonical tetra- and pentaphosphate alarmones ppGpp and pppGpp [together, (p)ppGpp]. Often dismissed in the past as an artifact or degradation product, pGpp has been confirmed as a deliberate endpoint of multiple synthetic pathways utilizing GMP, (p)ppGpp, or GDP/GTP as precursors. Tracing how free-ranging organisms interact with their environment to maintain water balance is a difficult topic to study for logistical and methodological reasons. Social context alters cognitive performance, but the mechanisms for this remain unclear. One possibility is that stressors associated with social contexts mediate changes in cognitive performance by influencing attention. We conducted two experiments to investigate how social context affects performance on an attention task in capuchin monkeys (Cebus [Sapajus] apella). We additionally validated and measured physiological stress through salivary cortisol. In experiment one, capuchins were exposed to three conditions where they were either isolated or paired with an affiliative or agonistic social partner separated by a barrier. Birds can cope with energy-shifting challenges by redirecting energy and behavior to maintain energetic homeostasis. Nestlings allocate energy to support their rapid growth and developmental period; thus, their challenges and the subsequent energetic demands can be much different than an adult. At a baseline level, energy is constantly shifting in response to regularly occurring challenges; however, more substantial challenges can initiate the production of glucocorticoid hormones. The rise in e-cigarette use is likely due to the overwhelming evidence regarding the addictive properties of nicotine and the damage caused by traditional tobacco cigarettes. Most e-cigarettes, however, still contain nicotine and are associated with a rise in nicotine-related intoxications (Belkoniene et al., 2019). I investigated the effects of diet and sex on zebrafish bone metabolism. Zebrafish were subjected to either a high-calorie or low-calorie diet over a five-week period. After this diet, zebrafish scales were used to measure alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRAP) enzyme activity with a fluorescent substrate. The enzyme activities were markers of the osteoblasts (ALP) and the osteoclasts (TRAP). These data were compared among sex and diet of the zebrafish using a repeated measures ANOVA statistical test. I also measured the number of resorption lacunae per scale (another indicator of osteoclast activity). The results showed higher TRAP …

Neurodegenerative diseases such as multiple sclerosis affect millions of people around the globe, and promising treatments to counter its progress have been studied extensively. In our study, we used Dawley rats and tested them using the Montoya Staircase test to evaluate their ability to use their forelimbs. After training, surgery was performed using lysolecithin to induce focal demyelination on the rats. Abnormal glycosylation can impact cardiac proteins, affecting signaling, contraction, and metabolism, which leads to compromised cardiac function. O-GlcNAcylation, the only form of intracellular glycosylation, involves adding a single GlcNAc molecule to proteins via enzymes: O-GlcNAc Transferase (OGT) for addition and O-GlcNAcase (OGA) for removal. The Ednie/Bennett lab has previously demonstrated that in both chronic (OGT KO) and acute (OGT inhibition) models of reduced cardiomyocyte O-GlcNAcylation, a disruption of Ca2+ homeostasis occurs. The intraflagellar transport(IFT) system, composed of two large multimeric IFT complexes, is universally present in all cilia and facilitates bidirectional cargo transport. While the role of IFT in primary cilia and eukaryotic motile cilia has been demonstrated, the association between IFT and mammalian motile ciliogenesis remains largely unknown. This experiment will aim to explore the heterodimerization of Dopamine (D2) and Adenosine (A2A) receptors in cells when bound to different ligands. Borroto-Escuela et al. (2018) investigated the dimerization of the A2AR-D2R in rats when administered with cocaine, and they found that disrupting the A2AR-D2R complex restores the cocaine reward system in rats. This was a strong influence as to why we chose to investigate A2AR-D2R dimerization in Chinese hamster ovary (CHO) cells.
