Surface-Enhanced Raman Scattering For Enantiomeric Discrimination, Thanyada Sukmanee
Surface-Enhanced Raman Scattering For Enantiomeric Discrimination, Thanyada Sukmanee
Chulalongkorn University Theses and Dissertations (Chula ETD)
Surface-enhanced Raman scattering (SERS) substrates with intended surface modification were developed for enantiomeric analysis by taking the consideration of asymmetric electric field and molecular interaction between a probe molecule and enantiomeric analyte. In the first study, discrimination between enantiomers is achieved by performing tip-enhanced Raman scattering using a silver tip which is chemically modified by an achiral para-mercaptopyridine (pMPY) probe molecule. The different relative intensities of the pMPY spectra were obtained when three pairs of enantiomers containing hydroxy and/or amino groups were used as analytes. Enantiomeric discrimination can be attained by the asymmetric electric field of the tip apex co-existed …

Pyrrolidinyl Peptide Nucleic Acid Probe Capable Of Crosslinking With Dna, Penthip Muangkaew
Chulalongkorn University Theses and Dissertations (Chula ETD)
DNA interstrand cross-linking (ICL) is a phenomenon in which two strands of the DNA (most often as part of the same DNA duplex) bind together via a covalent bond. This process prevents the DNA strand dissociation, which is essential for its biological functions, and thus is extremely damaging to cells. To overcome the problem of non-specificity associated with general DNA cross-linking agents, a cross-linkable probe in which the reactive group is placed on the probe that can specifically recognize part of the DNA target. The strong binding affinity and biological stability of the pyrrolidinyl peptide nucleic acid (acpcPNA) inspired us …

Tetrabromomethane-Mediated Desulfurization For Synthesis Of Isothiocyanates From Amines, Saharat Techapanalai
Chulalongkorn University Theses and Dissertations (Chula ETD)
Isothiocyanate considered as an important building block for pharmaceutical industry. Traditional methods for preparation of isothiocyanate involved the desulfurization of dithiocarbamate salt from amine using oxidizing agent. Although those methods are efficient, however, all of them require the use of toxic reagent, strong oxidizing agent, multiple step synthesis, harsh condition and large amount of metal catalyst. Therefore, in this research, we develop two mild methods to synthesize isothiocyanates from amines using low toxic reagent. For the first process, we were able to demonstrate the use of photocatalyst, safranin O to convert dithiocarbamate salt of 4-bromoaniline into 4-brophenyl isothiocyanate in 48% …

Diabetes mellitus occurs from deficiencies in insulin secretion or insulin resistance which leads over time to high risk for long-term macro-and microvascular complications. Diabetes mellitus type 2 contributes to more than 90% of all cases worldwide. One approach of treatment for this disorder is utilizing α-glucosidase inhibitors. Certain sulfonamides and chalcones are considered as viable candidates that are effective in reducing hyperglycemia. Forty-seven sulfonamide chalcones were synthesized by the Claisen-Schmidt reaction with various substituents on the A, B, and C-rings furnishing the desired products with 50-95 %yield. All compounds were-tested for α-glucosidase inhibitory activity. Twenty-nine compounds exhibited a very strong …

Electrochemical Paper-Based Immunosensor For Biomarkers Detection, Suchanat Boonkaew
Chulalongkorn University Theses and Dissertations (Chula ETD)
This dissertation focused on the development of electrochemical paper-based immunosensor for determining essential biomarkers by using a label-free immunoassay. This dissertation can be divided into four main parts. In the first part, an origami paper-based analytical device (oPAD) was developed for c-reactive protein (CRP) detection. oPAD was designed to combine the preparation of immunosensor and detection step into a single device. A gold nanoparticle modified graphene screen-printed carbon electrode (AuNPs/G/SPCE) was utilized to enhance the sensitivity of the electrode. The developed sensor was successfully applied to determine CRP in a certified serum sample. For the second part, an electrochemical paper-based …

Surface-Enhanced Raman Scattering Using Fluorescence-Quenched Carbon Quantum Dots For Mercury Ion Detection, Andaru Dena Prasiwi
Chulalongkorn University Theses and Dissertations (Chula ETD)
Mercury ion (Hg2+) is one of the most challenging problems due to its high toxicity at low concentration and bioaccumulative effects affecting human health. Herein, we propose a strategy to exploit the fluorescence quenching of carbon quantum dots (CQDs) combined with surface-enhanced Raman scattering (SERS) technique to detect Hg2+. CQDs exhibit strong fluorescence emission interfering SERS measurement. However, with the presence of Hg2+, fluorescence from CQDs can be quenched, and the SERS spectrum can be collected. In this work, SERS spectra of rhodamine 6G (R6G) incorporated with CQDs and Hg2+ (0.100‒100 ng/L) were studied. The SERS spectra of R6G was …
Paper-Based Device For Urinary Oxalate Detection, Kanphitcha Mueangdech
Paper-Based Device For Urinary Oxalate Detection, Kanphitcha Mueangdech
Chulalongkorn University Theses and Dissertations (Chula ETD)
In this work, a simple method for urinary oxalate determination was developed using testing paper modified with reddish-brown Fe(III)-curcumin complex. Oxalate in sample interacted with Fe(III) ions on the surface of the testing paper to form a Fe(III)-oxalate complex in solution, resulting in a decrease of the amount of Fe(III)-curcumin complex on the paper surface. The amount of the remaining Fe(III)-curcumin complex on the testing paper was inversely proportional to the oxalate level in the sample solution. A change of the testing paper color from reddish-brown to yellow could be observed and the color intensity was determined by ImageJ program. …

Synthesis Of Nitrogen-Doped Carbon Dots From Aromatic Amino Acids, Nattapon Siengdung
Chulalongkorn University Theses and Dissertations (Chula ETD)
Nitrogen-doped carbon dots (NCDs) were synthesized from amino acids via the hydrothermal method under an acidic condition. L-histidine, L-phenylalanine, L-tyrosine, L-tryptophan, and glycine were used as a precursor which NCDs synthesized from those precursors were labeled as His-NCDs, Phe-NCDs, Tyr-NCDs, Trp-NCDs, and Gly-NCDs, respectively. Spectroscopic and microscopic characterizations were operated to investigate optical properties, chemical structures, and morphologies of NCDs. The results show that all NCDs show a spherical shape with an average diameter of 4.11±1.20 nm. Their energy gap between the highest occupied molecular orbitals (HOMO) and the lowest unoccupied molecular orbitals (LUMO) obtained from fluorescence spectroscopy can be …

Development Of Paper-Based Analytical Devices For Simultaneous Naked-Eye Detection Of Heavy Metal Ions, Pornphimon Kamnoet
Chulalongkorn University Theses and Dissertations (Chula ETD)
Paper-based analytical device was fabricated by a polymer screen printing and wax printing methods. The various types of device were designed for simultaneous heavy metal ions detection in synthetic unknown and real water samples. The colorimetric method was applied to detect the heavy metal ions using 1-(2-pyridylazo)-2-naphthol, bathophenanthroline, 1, 5-diphenylcarbazide, bathocuproine, dimethylglyoxime, dithizone, and 4-(2-pyridylazo) resorcinol as complexing agents. However, complexing agents require very highly selectivity. To improve the selectivity for heavy metal ion detection, a device was designed with pretreatment zones and the masking agents were applied to remove interfering ions. The proposed method was successfully applied for simultaneous …
