The very potent antibacterial activity of cecropin B makes it a likely candidate to prevent and/or treat Mannheimia haemolytica 1:A infection in the upper respiratory tract (URT) of cattle. The purpose of this study was to ascertain if the URT could be transfected with a gene coding for the antimicrobial peptide cecropin B. Estimation Of The Effect Of Misclassifications On Diagnostic Test Performance In Two Persistent Bovine Viral Infections, Kimberly Orr

Validation of diagnostic assay performance is hampered where no gold standard exists. Bayesian estimates of the sensitivity and specificity were applied to studies of two persistent bovine infections: bovine immunodeficiency virus (BIV), and bovine viral diarrhea virus (BVDV). BVDV pathogenesis is well described. Losses of $20 million in 1992 alone led the Danish government to mandate eradication. Two enzyme-linked immunosorbent assay (ELISA) tests were developed; one antibody-based to detect exposure, one antigen-based to detect persistent infection. Bayesian estimates of the sensitivity and specificity were 96.6% and 99.2%, and 98.2% and 99.8% for the antibody and antigen ELISA, respectively. Maximum Likelihood …

The host-parasite interaction during early filarial nematode migration is poorly understood. The objective of this study was to develop a model of early cutaneous filarid migration using Brugia pahangi in the jird (gerbil) host and measure the histologic and cytokine responses during this period. Male gerbils were intradermally inoculated in the left hindlimb with 100 B. pahangi L3 then necropsied at 3 hours, 24 hours, 3 days, 7 days, and 28 days post-infection. Larvae were recovered and tissues collected for histology and cytokine measurement. At 3 hours, most larvae (96.3%) were recovered from tissues associated with the infection site. Migration …

The Herpes Simplex Virus life cycle contains a number of membrane fusion events that must function properly to ensure a productive infection, including: virus attachment and entry into susceptible cells, de-envelopment at the outer nuclear lamellae, and virus-induced cell-to-cell fusion. A virus-free cell fusion assay was recently developed in order to attempt to understand the underlying mechanisms that are responsible for viral fusion events and was utilized in order to investigate the effect of mutations targeted to the carboxyl terminus of gB. The first event in the infection process of Ostertagia ostertagi in cattle is the process of exsheathment. Before trichostrongylid nematodes can transition from a free-living infective stage larva (L3) on pasture to a parasitic existence within the ruminant host, it must first undergo exsheathment. Exsheathment is the process whereby the L2 cuticle retained from the previous molt is cast from the L3. Exsheathment enables the developmental transition from a free-living stage on pasture to a parasitic existence in the bovine host. Acute laminitis is a severely debilitating disease of the laminae of the equine digit; however, the mechanism(s) of pathogenesis have yet to be fully elucidated. In physiologic states, the endothelium synthesizes substances, such as nitric oxide (NO; vasodilator) and endothelin-1 (ET-1; profound vasoconstrictor), which play a crucial role in vasomotor regulation. The overall hypothesis is that the initiating factor in the onset of acute laminitis is a disruption in the balance between NO and ET-1, which leads to digital vasoconstriction and subsequent laminar ischemic necrosis. To study the protective responses of cyathostome-infected ponies, two challenges were performed employing animals with different histories of exposure to these parasites. The hypothesis developed and to be tested in these experiments was that ponies that had longer exposure to cyathostome contaminated pastures would express acquired resistance to infection. The assumption behind this hypothesis was that helminth-naïve ponies infected with cyathostomes would eliminate the infection using only innate immune responses. Whereas previously exposed ponies would eliminate the infection with acquired immune responses, and these would be more effective in ponies with longer exposure to cyathostomes. Inflammation is a component of the innate immune response. However, severe or prolonged inflammation can be detrimental. Dietary n-3 fatty acid supplementation has proven clinical benefits in chronic inflammatory diseases, most likely due to reduced synthesis of inflammatory mediators and inhibition of lymphocyte proliferation and function. The purposes of this study were to characterize alterations in inflammatory mediator production, and lymphocyte proliferation, in dogs fed a diet consisting of 0.65% n-3 fatty acids (DMB) with an n-6:n-3 fatty acid ratio of 3.4:1. Currently, the control of H. contortus and others is almost entirely based on the use of anthelmintics. Consequently, anthelmintic resistance has developed worldwide and this has become a serious problem in small ruminant nematode parasite control programs. Effects of silicone polymer on equine articular cartilage and synovial fluid have not been evaluated. Gastrointestinal tract disease is the leading natural cause of death in horses and horses with ischemic intestinal disease have the greatest mortality. We hypothesized there is basal synthesis of endothelin-1 (ET-1) in the intestinal tract of healthy horses that is likely involved in regulating vasomotor tone, secretion and motility and that ET-1 synthesis increases with gastrointestinal tract disease, which may be involved in the pathophysiology of these disorders. Plasma ET-like immunoreactivity was increased in horses with naturally-acquired gastrointestinal disease, compared with normal horses; values were greatest in horses with large intestinal strangulation obstruction, enterocolitis and peritonitis. The current pathogenesis of Sudden Infant Death Syndrome (SIDS) is unknown. Elevated endogenous ethanol production by gastrointestinal yeast has been proposed as a possible mechanism for SIDS. To investigate the role of ethanol in SIDS, three studies were undertaken. In the first study, transcutaneous blood gas technology was evaluated for repeated monitoring of arterial blood gases. The second study identified the cardiorespiratory responses to inhaled CO2 challenge in juvenile rats, to determine the age corresponding to the age range of peak incidence of SIDS in human infants. Complete sequencing and annotation of the M. leprae genome has provided new information related to proteins constituting its hypothetical proteome. Since M. leprae can not be grown in vitro, novel approaches are needed to determine which proteins are expressed during infection and whether these proteins are related to pathogenesis. It is important to be able to predict the potential spread of water borne diseases when building dams or redirecting rivers. This study was designed to test whether the use of a growing degree day (GDD) climate model and remotely sensed data (RS) within a geographic information system (GIS), could be used to predict both the distribution and severity of Schistosoma haematobium. This study was designed to characterize changes in the content of adenosine triphosphate (ATP) and cellular permeability of gastric and jejunal mucosa during gastric dilatation-volvulus (GDV). Our hypothesis stated that experimental GDV would result in a decrease in ATP content and cellular integrity of the gastric and intestinal mucosa that may improve after decompression and derotation of the stomach. Fifteen medium-to-large mixed-breed male dogs were randomly divided into three groups: GDV dogs, Ischemia dogs and Control dogs. All dogs were maintained on gas anesthesia for 210 minutes. Substantial funding has established the Mississippi Sandhill Crane National Wildlife Refuge in Southern Mississippi and an artificial insemination / breeding facility at the Audubon Center for Research of Endangered Species in New Orleans, Louisiana to promote species recovery. Photobacterium damselae subspecies piscicida, previously known as Pasteurella piscicida, is an important pathogen of hybrid striped bass and many fish species cultured in brackish water in the United States, Japan, Europe, and the Mediterranean. The purpose of this study is to investigate virulence mechanisms that contribute to the pathogenesis of this organism. The ability of P. damselae to survive/replicate within hybrid striped bass macrophages was evaluated with an in vitro killing assay. Ehrlichia canis is the etiologic agent of “tropical canine pancytopenia”, or canine ehrlichiosis. The impetus for this research was to overcome the lack of any reliable means of elucidating the genetic profiles of these illusive and historically difficult to manipulate organisms. The use of a broad-host range plasmid greatly facilitated the determination of an electro-transformation protocol. The transforming plasmid possesses a chloramphenicol antibiotic resistance gene marker (chloramphenicol acetyltransferase [CAT] gene), and a visual reporter gene marker, the Green Fluorescent Protein (GFP) gene. Achondroplasia and hypochondroplasia are two forms of skeletal dysplasias caused predominantly by single base mutations in the fibroblast growth factor receptor 3 gene (FGFR-3). The mutation for achondroplasia is a G1138A/C substitution and the mutation for hypochondroplasia (occurring about 50% of the time) is a C1620A/G substitution. Recent genetic studies have shown that spontaneous mutations for achondroplasia and hypochondroplasia occur exclusively on the paternally derived chromosome, suggesting that these mutations occur preferentially during spermatogenesis. For unknown reasons, the mutation rates at these FGFR-3 nucleotides appear to occur at a much higher frequency than nucleotide specific mutation rates observed in other …
