Microglia/macrophage activation has been associated with the pathogenesis of various neurological diseases including human immunodeficiency virus encephalitis, transmissible spongiform encephalitis, and Alzheimer's disease (AD). In vitro studies have indicated a role for TNFα in activating these cells which leads to their migration, proliferation, and secretion of proinflammatory cytokines and chemokines that may potentially damage brain tissue. In the current study, we analyzed the phenotype of microglia and macrophages enriched from wild type and TNFα deficient mice infected with a neurovirulent murine retrovirus. Viral infections of the central nervous system (CNS) in infants are rare; however, they are associated with high morbidity and mortality rates. These virus infections often induce strong innate immune responses in the brain including: the production of cytokines and chemokines, the activation of astrocytes and microglia and the recruitment of macrophages. Innate immune responses are often initiated by toll-like receptors (TLR). Several studies have demonstrated that toll-like receptor 7 (TLR7) can be stimulated by single-stranded RNA from multiple viruses. Herpes simplex virus type-1 (HSV-1) morphogenesis occurs in multiple stages within infected cells. Initially, the virion capsid assembles within the nucleus and buds through the nuclear membrane into the cytoplasm. Within the cytoplasm, additional tegument proteins attach to the capsid and the fully tegumented capsids bud into trans-Golgi network (TGN) derived vesicles. Enveloped virions are ultimately secreted to extracellular spaces. The process by which the cytoplasmic capsids bud into TGN-derived vesicles is not well understood. The prevalent model calls for specific interactions among viral tegument proteins and membrane proteins and glycoproteins embedded within TGN membranes. To further investigate the roles …

ABSTRACT In 1999, there were reports of a new type of lesion in the hides of captive reared alligators from Florida. Similar lesions were first reported from alligator hides in Louisiana in 2001; however, it wasn’t until 2002 that small epizootics became apparent. In 2002, the Louisiana Department of Wildlife and Fisheries began a collaborative effort with the Louisiana State University School of Veterinary Medicine (LSU SVM) to help elucidate the etiology of “PIX” disease, later renamed Lymphohistiocytic Proliferative Syndrome of Alligators (LPSA). Brucella melitensis is a facultative intracellular bacterial pathogen that causes abortions in goats and sheep and Malta fever in humans. The zoonotic disease brucellosis causes severe economic losses in the Mediterranean region and parts of Africa, Asia, and Latin America. With the completion of the genomic sequences of B. abortus 2308 and B. melitensis 16M, no classical virulence factors were found; and the chromosomes were virtually identical. However, in B. melitensis, a putative hemagglutinin gene was identified which is absent in B. abortus. West Nile virus (WNV), a member of the genus Flavivirus transmitted by mosquitoes, first appeared in the New York in 1999. Within five years WNV was detected throughout the contiguous 48 states causing disease in reservoirs and accidental hosts alike. In Louisiana, WNV was first detected in 2001 with one human case, ten equine cases, and six dead birds reported. The introduction of WNV into Louisiana presented an unique opportunity to observe an emerging disease unfold, so a study was launched to gain insight into the epidemiology of WNV in Louisiana. Brugia infections occur via the bite of an infected mosquito. Third stage infective larvae (L3) deposited on the skin during feeding migrate into the bite wound, through skin and into the lymphatic system. It is hypothesized that L3 excretory/secretory products (ES) are important in this initial phase of the infection. A model for these early migrations has been established by inoculating L3s into the dermis (ID) of the permissive gerbil host. Extracts of adult Haemonchus contortus were purified and used as a vaccine against the blood feeding parasite in goats as previously studied in sheep. The proteins used were H11 and H-gal-GP, hidden gut antigens from the microvillar membrane of the gut of the worm and combined with Quil A as adjuvant, then administered to a group of goats kept on concrete then artificially infected with H. contortus. The control group received Quil A injections and also infected. This study was performed to analyze the effects of the H11/H-gal-GP vaccine when given to goats as compared to sheep. Salmonella is an important intestinal pathogen in horses capable of infecting populations without demonstrating clinical illness. This study was performed to determine the prevalence of Salmonella fecal shedding among racehorses in Louisiana. Three serial fecal samples were collected from 429 Thoroughbred horses housed at four racetracks. Feces were tested for Salmonella by microbiologic culture with selective primary enrichment and delayed secondary enrichment (DSE). Samples were also evaluated for the presence of Salmonella by polymerase chain reaction (PCR) using genus-specific oligonucleotide primers. The drive to exercise voluntarily likely results from complex interactions between genes in many organ systems and various psychological parameters, such as motivation and the perception of fatigue. Reproducible variations in exercise intensity and duration are well established in laboratory rodents, but the genes responsible remain largely unknown. Also, to date, studies addressing the adaptive changes to exercise that might prevent dietary-induced obesity have focused primarily on energy intake and nutrient oxidation/partitioning, as opposed to genetics. Gopher tortoise (Gopherus polyphemus) populations have experienced precipitous declines from habitat loss, and human and disease related mortality. The goal of this study was to characterize the health status of free-ranging Louisiana gopher tortoises. Gopher tortoises were collected during two distinct trapping seasons: fall (August-October 2002) and spring (April-June 2003). Captured tortoises were given a physical exam and the carapace and plastron length and width, weight, and body temperature were recorded. Hysteroscopic insemination in mares with delayed uterine clearance (DUC mares) is controversial. While some authors proposed that insemination with a reduced volume and number of spermatozoa may reduce post-mating endometritis, others proposed that the hysteroscopic procedure is inflammatory and should not be used in DUC mares. Development of therapies designed at stopping the progression of osteoarthritis, require a method of evaluating efficacy. The murine pathogen Helicobacter hepaticus has important confounding effects on research. Neonatal fostering has been studied in our laboratory for elimination of infection in mice. The purpose of our study was to examine fostering of pups from experimentally infected dams in male-absent parturition, and to determine the significance of gender and time on quantity of bacterial colonization in the cecum and feces of C57BL/6 mice. Approximately 20 C57BL/6 mice were fostered per day from one to four days of age. None of the C57BL/6 pups tested positive by PCR in fecal or cecal samples through four days of age. This …

Summer pasture-associated obstructive pulmonary disease (SPAOPD), a naturally occurring airway disease of horses, is characterized by clinical exacerbation associated with exposure to pasture environment during the summer. Aeroallergens are believed to trigger exacerbation of SPAOPD, cytokines are likely associated with the anamnestic response to aeroallergens, and endothelin (ET)-1 is a potential mediator of airway obstruction. The SARS-Coronavirus (SARS-CoV) is the etiological agent of the severe acute respiratory syndrome (SARS). The SARS-CoV spike (S) glycoprotein mediates membrane fusion events during virus entry and virus-induced cell-to-cell fusion. Investigations, described herein, have focused on the genetic manipulation of the SARS-CoV S glycoprotein in order to delineate functional domains within the protein. This was accomplished by incorporating single point mutations, cluster-to-lysine and cluster-to-alanine mutations, as well as carboxyl terminal truncations into the protein and investigating these mutants in transient expression experiments. An Edwardsiella ictaluri strain with disruption of ureG was identified through the use of signature tagged insertion mutagenesis as being attenuated for virulence in the channel catfish host. Sequencing of the flanking regions surrounding the insert showed that the gene was part of a urease gene complex that included ureE, ureF, ureG, ureD, ureI, and an ammonium transporter homologue. The ureG gene encodes a GTP-binding accessory protein which is thought to function in energy-dependent urease assembly. Urokinase (uPA) and urokinase receptor (uPAR) are present in the urinary tract of people, mice, rats and cows. The presence of urokinase and urokinase receptor in the urinary tract of dogs has not been documented. Documenting the presence of uPA and uPAR in the urinary tract of healthy dogs will help determine the role these proteins play in pathology of the urinary tract in further research. Urine was collected from fifty-four healthy dogs. A fluorescence assay was developed using the fluorogenic substrate Z-Gly-Gly-Arg-AMC, HCl. Intraoperative reductions in serum fentanyl levels in dogs with induced moderate hypothermia and transdermal fentanyl patches (TDF) in place has been documented. The impact of such reductions has not been evaluated nor has the anesthetic sparing effect of the TDF. Reductions in serum levels may be the result of either a decreased uptake of fentanyl from the dermal depot, or a biophase shift. The common eland is a useful model species for studies on rare tragelaphine antelopes. In Chapter 3 of the present study, effects of components/attributes of protein-free embryo culture media on the in vitro development of in vitro-derived bovine embryos were evaluated. During the course of their disease, leprosy patients may experience two types of inflammatory reactions- erythema nodosum leprosum (ENL) or reversal reaction (RR). Thalidomide is effective treatment for ENL, but not for RR. Using concentrations of thalidomide similar to that achieved in the treatment of ENL, we investigated thalidomide’s effect on reactions, viability of M. leprae, and integrity of plasma membranes. These studies were conducted to elucidate the regional analgesic effect that is observed clinically after treatment of orthopedic disorders with application of extracorporeal shock waves in horses. Regional analgesia after treatment with extracorporeal shock waves presents a concern because it may eliminate protective limiting mechanisms and may place equine athletes with predisposing lesions at risk of sustaining career- or life-ending injuries. Direct percutaneous application of non-focused extracorporeal shock waves to palmar digital nerves in the pastern area of horses resulted in decreased sensory nerve conduction velocities compared with untreated control nerves at 3, 7, and 35 days after treatment. Cranial cruciate ligament (CCL) rupture is a common injury in the dog and major cause of degenerative joint disease. The pathophysiology of CCL rupture in the dog is well described. Osteoarthritis secondary to CCL rupture causes severe pain and lameness. There are many surgical techniques accepted for dogs with CCL rupture. A commonly performed technique is an extracapsular repair with a lateral fabella-tibial suture (LFS) using large diameter nylon leader line (NLL). The purpose of this study was to investigate methods to induce earlier onset of cyclicity in transitional mares. Two experiments were conducted evaluating the effect of follicular aspiration to advance the onset of cyclicity, more succinctly define criteria for selection of mares for follicular aspiration and to compare aspiration to deslorelin treatment for initiating cyclicity in transitional mares. In Experiment 1, anestrous mares were assigned to control (n=6) or follicular aspiration (n=11). The control mares were monitored twice weekly, until ovulation was detected. Brucellosis and pseudorabies lead to abortion in pregnant sows and are perpetuated by feral swine reservoirs. A multivalent oral vaccine for these diseases would improve vaccination and eradication programs worldwide. Previous studies have shown that the rough attenuated Brucella strains RB51 and VTRS1, when administered subcutaneously to swine, stimulate host immune responses, transiently colonize tissues, and provide partial protection against virulent B. suis infection in pregnant sows. The leading viral killer of commercially produced channel catfish is Channel Catfish Virus (CCV). Studies conducted to evaluate DNA vaccination against CCV compared encoded gene, dose, multiple DNA vaccines, and immune response to vaccination. Genes were selected (ORFs 1 and 3 [immediate early genes], ORFs 6, 19, and 46 [membrane genes], and ORF59 [putative major envelope glycoprotein gene], cloned into a plasmid, and expressed in mammalian and fish cell culture to detect predicted molecular weight proteins.
