Planet earth has now entered into its sixth mass extinction as a result of expanding human populations, posing an unprecedented threat to biologic diversity. Climate change, habitat fragmentation, wildlife trade, pollution, and biological invasions are just a few consequences of human activities that have directly contributed to unprecedented species extinctions. While widespread amphibian declines have gained more recent attention, it has been proposed that reptiles may be in even greater danger of extinction worldwide. Unfortunately, many reptiles are destined to go extinct before many of their basic biologic traits, including reproductive methods, have even been determined.

The objectives of this study were to investigate concentration dependent effects of canine prostatic fluid (PF) on in vitro seminal parameters of cooled canine semen. Sperm motility parameters, plasma membrane integrity and stability, acrosome integrity and DNA fragmentation were measured after the addition of 0%, 10%, 25%, or 50% PF to extended semen of fertile dogs. Assessments were made at 0 h pre-cooling, at 24, and 48 h of cooled storage (4 °C), and after freezing and thawing followed by incubation (37 °C) at 0, 4 and 24 h. The objectives of this study were to determine what effects iodixanol would have on total and progressive motility, plasma membrane integrity (viability), acrosome integrity and DNA structure when present during cryopreservation of equine spermatozoa,. We hypothesized that the addition of iodixanol would improve post-thaw values for measured parameters. Ejaculates from six stallions were collected, centrifuged at 900 x g for ten minutes to remove supernatant, and suspended to 200 x 106 cells/ml with 0%, 2.5% and 5% iodixanol in an egg-yolk based extender and cryopreserved.
