Radix Oryzae Glutinosae (ROG) has been used as a traditional anhidrotic agent in China. Ten samples were systematically assessed based on four aspects of their chemical profiles and antioxidant activity. The former was achieved using ultraviolet-visible spectra (UV), infrared absorption spectra (IR), proton nuclear magnetic resonance spectroscopy (1H NMR), and inductively coupled plasma-mass spectrometry (ICP-MS) fingerprints, and antioxidant activity was assessed using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay, the 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) assay, and the ferric reducing antioxidant power (FRAP) assay. In this study, the QuEChERS and UHPLC-MS/MS method using a biphenyl stationary phase was developed and applied for the rapid separation of 12 anthocyanosides in grapes. Efficient separation was achieved for a wide range of mono- and un-glycosylated anthocyanosides. The linear ranges of the target anthocyanosides were 0.5 to 500 ng mL−1. The intra- and inter-day precisions were both below 11.8%. Anthocyanosides in grapes from different sources were successfully quantified. Thus, the developed QuEChERS and UHPLC-MS/MS method provide an attractive alternative for the quantification of anthocyanosides in grapes.

A simple fluorescence method to quantitatively monitor malachite green (MG) was introduced based on the conformation switch effect of MG-aptamer (Apt) and the fluorochrome of SYBR Green I (SGI). SGI can embed in the stem–loop structure of Apt to produce fluorescence. However, the fluorescence will be quenched when MG is introduced because of the specific binding of Apt to MG. Under the optimal conditions, the fluorescence method displayed excellent linear relationship between the fluorescence quenching efficiencies and the MG concentrations ranging from 0.02 μmol L−1 to 1.0 μmol L−1. Bromelain, a cysteine protease found in pineapple, is known to exert protective effects against non-alcoholic fatty liver disease (NAFLD); however, the underlying mechanism is unclear. In this study, we aimed to investigate the molecular mechanisms underlying the beneficial effects of bromelain using in vivo and in vitro models. C57BL/6 mice were fed a high-fat diet (HFD) with or without bromelain (20 mg/kg/day) for 12 weeks. Herbal medicine (HM) is a type of medicine that uses active ingredients made from plants to treat diseases and maintain health and wellbeing. Due to its increasing worldwide usage, the possibility of HMs and conventional drugs being concurrently used is high, potentially leading to adverse events resulting from herb-drug interactions. Despite the safety concerns regarding such interactions, few studies have been conducted for assessing clinical consequences of using HMs with conventional drugs in real-world settings. The Buffalo National River (BUFF) was established as the first national river in 1972 and later designated as an Extraordinary Resource Water and Natural and Scenic Waterway. This thesis seeks to address the potential threat of elevated nutrients and its association with freshwater mussel declines in the BUFF through laboratory and in situ exposures. Water and sediment chemistry data were collected, and relevant thresholds were used to extrapolate lab-to-field toxicity during low-flow events. Acute toxicity was not observed; however, low-concentration chronic toxicity may be influencing mussel assemblages with rising temperatures. Smelting activities by the American Smelting and Refining Company (ASARCO) lead to the substantial release of metal pollution into the El Paso, Texas, and border cities region. To-date few studies have examined metal accumulation within vertebrate taxa in these areas. As reptiles are currently experiencing population declines globally for a variety of reasons including pollution, lizards were chosen to be evaluated for trace element (As, Cu, Cr, Cd, Pb, and Zn) concentrations. Acute myeloid leukemia (AML) is a commonly diagnosed cancer in smokers. When current or former smokers have AML, they have worse survival compared to never smoking patients. This has been observed clinically for decades, but then it is unknown how smoking leads to worsened AML survival. Smoking causes oxidative stress and altered DNA methylation that persists for decades in peripheral blood mononuclear cells, but these changes from smoking have not been evaluated in the context of AML. In this paper, the concentration of As, Cd, Cr, Cu, Pb, Zn was investigated in soil and Ambrosia artemisiifolia L. sampling from polluted cite near the enterprises for the production and processing of batteries in the city of Dnipro in Ukraine. The obtained results of the study provided to assess plants regarding bio-monitoring and phytoremediation. Although Ambrosia artemisiifolia L. is a weed that causes serious allergic reactions in humans, this species can also have a high bioaccumulative capacity regarding metals. This study assessed toxicity of the crude ethanolic pod extracts of Swartzia madagascariensis Desv in Wistar rats. Forty (40) Wistar rats aged 8–10 weeks were orally administered with crude extracts from pods of S. madagascariensis. Chemical analysis of serum and histopathology of liver and kidney from test animals were performed to determine the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and creatinine in serum. In addition, serum albumin levels were also determined. The levels of ALT, AST and creatinine were found to be high in groups treated with crude extracts compared to the control group, suggesting some damage in …

We aim to develop an efficient and non-invasive strategy for monitoring of drugs and their metabolites via human skin. Probe sampling was combined with thermal desorption-electrospray ionization tandem mass spectrometry (TD-ESI/MS/MS) to characterize trace terbinafine, which was secreted on patient’s skin after ingesting terbinafine tablets. The terbinafine ion signals were directly detected in the samples collected from different skin regions and the signals were monitored for 8 weeks. The detection and location of terbinafine via the skin suggest that the methods are useful in rapidly and noninvasively collecting the molecular information of the ingested drug on skin for pharmacokinetic studies.

Tetracycline was reported to trigger intense lucigenin chemiluminescence (CL). Based on its significant quenching to lucigenin-tetracycline CL, procyanidin was detected. The mechanism of the lucigenin-tetracycline CL and its quenching by procyanidin were proposed. Under the optimum conditions, the calibration curve is linear in the range of 10-1000 nM for procyanidin with a detection limit of 4.4 nM. The RSD for nine replicate determinations of procyanidin was less than 1.7%. Moreover, this method was satisfactorily applied to the determination of procyanidin in human urine samples and health food capsule of grape seed.

Mycophenolate mofetil (MMF) and enteric-coated mycophenolate sodium (EC-MPS), specific inhibitors of lymphocyte proliferation, are commonly used as adjuvant therapy with calcineurin inhibitor agents after kidney transplantation. After administration, MMF and EC-MPS are hydrolyzed to mycophenolic acid (MPA), the active form of the drug, which must be monitored due to its narrow therapeutic window, drug–drug interactions, and large intra-and inter-individual pharmacokinetic variability despite a fixed dose. Monitoring plasma MPA level is recommended to maintain the drug within the therapeutic window, optimize its efficacy, and minimize side effects. Camphorataimide B (CamB) has anticancer activities against several tumors. Here, we aimed to investigate the mechanism(s) by which CamB inhibits metastasis of colorectal cancer (CRC) cells. Low-dose CamB did not affect the cell viability and cell cycle progression of CRC cells, but significantly inhibited the metastatic potentials of CRC cells. Mechanically, CamB reduced the protein and mRNA expression of BMP4, and inhibited Smad and FAK/Src/Akt signaling. CamB also decreased Snail levels by promoting its degradation via proteasome and thereafter reduced Snail-mediated BMP4 transcription. Moreover, CamB considerably inhibited the in vivo metastasis of DLD-1 cells in xenograft mice.

On analyzing the results of cell-based assays, we have previously shown that perilla (Perilla frutescens) leaf extract (PLE), a food supplement and orally deliverable traditional Chinese medicine approved by the Taiwan Food and Drug Administration, effectively inhibits SARS-CoV-2 by directly targeting virions. PLE was also found to modulate virus-induced cytokine expression levels. Methamphetamine (MAMP) is a highly addictive illicit drug typically abused for its nervous system stimulating effects. Conversely, methamphetamine has therapeutic use treating attention deficit hyperactivity disorder, controlling appetite and assisting with weight loss, and is available as the pure l-isomer in over-the-counter (OTC) nasal inhalers due to its decongestant activity. Because l-methamphetamine (l-MAMP) is available in OTC form, forensic guidelines require a sample to contain greater than 20% d-methamphetamine (d-MAMP) when classifying results as consistent with illicit MAMP use. Farm animals in concentrated animal feeding operations (CAFOs) are routinely implanted with synthetic and natural hormones to enhance growth and production, contributing significantly to environmental steroid hormone contamination.
