Metabolomics provides direct insights into cellular physiology, yet it faces greater analytical challenges compared to genomics and transcriptomics due to the chemical diversity, instability, and environmental sensitivity of metabolites. Conventional bulk metabolomics averages signals across cell populations, thereby masking cellular heterogeneity critical for understanding disease mechanisms. Recent breakthroughs in single-cell metabolomics (SCM), driven by advances in mass spectrometry, microfluidics, isotope tracing, and spatial omics, have enabled the detection of metabolic diversity at unprecedented resolution. SCM has uncovered cell-type-specific biomarkers, revealed metabolic reprogramming in cancer and immunity and revealed disease progression. Per- and polyfluoroalkyl substances (PFAS) are a class of synthetic chemicals known for their high stability and bioaccumulation potential, leading to their widespread presence in the environment and human tissues. Exposure to PFAS has been associated with significant health concerns, including neurotoxicity, hepatotoxicity, and metabolic disturbances. Microplastics have become widely dispersed in our environment, and even in the air we breathe. The present research aims at identifying and determining the amounts of microplastics present in atmospheric fallout over Lahore city, which is characterized by a large population and several manufacturing industries. In order to find out the existence of microplastics, samples of atmospheric fallout were collected from various sites across hustling municipalities of Lahore that included urbanized settlements. Human health may be at risk if drinking water contains excessive amounts of heavy metals (HMs). To determine risks related to heavy metals, fifteen water samples were taken every month between July and September, 2024 from the communities of Abobo (Site A), Osara (Site B) and Osaragada (Site C) close to Confluence University of Science and Technology, Osara. These samples were tested for ten HMs (Cu, Fe, Ni, Zn, Co, Cd, As, Cr, Hg, Pb) with the aid of Atomic Absorption Spectroscopy. Background and Purpose: Mercury is a pervasive environmental contaminant, and human epidemiologic studies have linked chronic low-level exposure to increased autoimmune disease-risk. Dysregulated lymphocyte activation and impaired signaling tolerance are central mechanisms in human autoimmunity. Protein phosphorylation governs lymphocyte differentiation and function, alterations in phosphoproteomic networks may represent a signature of immune disruption. This study evaluates how low dose mercury exposure modifies B and T cell abundance and intracellular signaling patterns in BALB/c and Diversity Outbred (DO) mice.

Francisella tularensis is a highly infectious intracellular bacterium and the causative agent of tularemia, with pulmonary infection resulting in severe disease and high mortality if untreated. While protective immunity to F. tularensis has been largely attributed to CD4⁺ T cell–mediated IFN-γ responses, the contribution of γδ T cells to mucosal immunity and vaccine-induced protection remains poorly defined. The objective of this study was to characterize the role of γδ T cells during primary pulmonary infection with F. tularensis live vaccine strain (LVS) and following intranasal immunization with inactivated F. tularensis (iFt) prior to LVS challenge.

Autoimmune disorders disproportionately affect females, a bias largely attributed to estrogen's modulation of B lymphocyte development and tolerance. Bisphenol A (BPA), a common plastic monomer, shows estrogenic activity and acts as an endocrine disruptor and epigenetic modifier. This thesis examines how BPA may increase autoimmune risk by mimicking estrogen such that it can excite estrogen receptors which rescue autoreactive B lymphocytes from apoptosis. While direct research linking BPA to B lymphocyte-mediated autoimmunity is limited, understanding BPA’s course of action in B lymphocytes is critical for addressing the rising prevalence of autoimmune diseases and its possible link to the increased and …

This thesis examines the disproportionate burden of lead exposure carried by low income, POC communities. The systemic nature of this problem is a symptom of a longstanding legacy of environmental injustice in the United States. Decades of federal neglect are reflected in the higher statistics of lead exposure and poisoning in predominantly black communities. While it is understood that lead exposure poses a serious threat to physical health and early cognitive development, there is a discouraging lack of urgency to remove the toxin from non-wealthy communities. The Environmental Influences on Child Health Outcomes Fetal Growth Studies (ECHO-FGS) birth cohort data was used to identify chemical mixtures associated with adverse outcomes. This biomonitoring study associated specific chemical mixtures in maternal serum with low birth weight or childhood obesity (Pearce et al., 2021). Mixture Profiles 1, 3, and 4, comprised of different observed concentrations of DDE, PCB153, BDE47, and PFOS were prioritized. Mixtures were tested to determine whether they perturbed mitochondrial metabolism and energy sensing pathways. This paper examines the hidden environmental, geopolitical, economic, and social costs of the cobalt supply chain. Fueled by the demand for battery technology and electric vehicles, cobalt has become an essential element to the green energy transition. With the largest share of global cobalt deposits, the Democratic Republic of the Congo bears a disproportionate share of the industry’s harms. Cobalt mining in the DRC is marked by environmentally degrading practices and inhumane working conditions, compromising Congolese health, safety, and well-being. This research was conducted to explore the possibility of constructing floating treatment wetlands (FTWs) without using plastic materials. FTWs are floating platforms that hold plants on the water’s surface, letting roots dangle within the water column, to help remove contaminants from contaminated water, especially in stormwater ponds. Many conventional FTWs are made with plastic components, such as mats and plastic cup containers, which can release microplastics into the environment over time. The US population faces simultaneous public health threats from both aging and environmental pollutants. Chief among these is hexavalent chromium [Cr(VI)], whose toxic effects in peripheral tissues are well-established but its effects on the brain are still understudied. Following previous results indicating hippocampal metals dyshomeostasis after Cr(VI) exposure, we aim to assess Cr(VI)’s impact on the blood-brain barrier (BBB), a key structure for brain protection. Pharmaceutical contaminants are increasingly prevalent in aquatic environments, yet their sublethal effects on ecologically relevant behaviors remain poorly understood, particularly in estuarine systems. Selective serotonin reuptake inhibitors (SSRIs), such as fluoxetine, are of particular concern due to the conserved role of serotonin in regulating behavior across vertebrates. Here, we investigated how acute exposure to environmentally relevant concentrations of fluoxetine influences anti-predator behavior in Gulf killifish (Fundulus grandis), a key estuarine fish commonly found in saltmarsh habitats. Coral reefs are biologically diverse marine ecosystems that provide numerous goods and ecosystem services. These sessile organisms are vulnerable to anthropogenic impacts, habitat degradation, climate change, and pollution, particularly Petroleum hydrocarbon contamination. Previous research has characterized coral species' sensitivity to hydrocarbons, using a continuous passive dosing system, but the specific histological impacts of petroleum hydrocarbons remain underexplored. Valproic acid (VPA) is a common medication used to treat epilepsy, known for its broad-spectrum efficacy but also associated with hepatotoxicity and metabolic disturbances. β-glucan, a natural polysaccharide with hepatoprotective properties, may mitigate druginduced liver damage. The study sought to assess hepatorenal toxicity of VPA and investigate the potential protective and therapeutic effects of β-glucan. Forty male rats were randomly distributed into four groups. The treated group received VPA (500 mg/kg), β-glucan (50 mg/kg), or combinations for 14–21 days via intraperitoneal injection. Serum biochemical parameters (ALT, AST, ALP, urea, creatinine, lipids) were measured. Histopathological evaluation of hepatic tissue was performed. …

• Microtubules are composed of of a- and b- tubulin heterodimers, vital for cell structure, shape, and intracellular transport • Microtubules are also useful as they separate chromosomes during cell division • Microplastics (< 5 mm) are considered a health threat and have been found throughout the whole body (1) o In several studies, microplastics have been correlated to higher rates cardiorespiratory issues, endocrine disruption, and cancer (1) • Tubulin-binding medications are widely used as chemotherapeutic agents as they are known for disrupting cell division and targeting tubulin (2,3) o In this study, these medications were used to determine whether microtubules play a role in microplastic uptake § Colchicine typically binds to tubulins and arrests polymerization and growth § Nocodazole disrupts polymerization and prevents spindle formation § Vinblastine inhibits microtubule assembly and disruptions miotic spindle formation.

Ashwagandha has been shown to reduce inflammation in humans by lowering pro-inflammatory cytokines and oxidative stress markers, helping regulate immune responses. It is commonly used to support conditions linked to chronic inflammation, such as stress-related disorders, arthritis, and metabolic imbalance. THP-1 macrophages are a part of the body's immune response to inflammation. We use Caenorhabditis elegans (C. elegans) as a model organism to investigate the effects of microplastic and chemical exposure on muscle structure and function.
• Microplastics (MP) are pieces of plastic less than 5 μm, and Nanoplastics (NP) are less than 1 μm. Microplastics are man-made pollutants and have been found in the environment and in the food and water sources, including drinks from plastic bottles (Hu, 2020). • Our work relates environmental contamination to potential biological consequences at the muscular level, providing insight into how pollutants may contribute to degenerative damage.

Amphibians are experiencing population declines due to habitat loss, climate change, and diseases, particularly chytridiomycosis caused by the fungus Batrachochytrium dendrobatidis. Protection from this fungal infection stems from the cutaneous microbiome. Pesticides have been shown to affect amphibians in a variety of ways, including reducing survival, locomotor performance, and accumulation in body tissues. Pesticides may also lead to changes in the cutaneous microbiome affecting amphibians who rely on these organisms as an anti-Bd defense mechanism. Gut microbiota produces a wide range of metabolites and plays a critical role in maintaining host health. Dysregulation of these metabolites can influence host metabolism through systemic circulation, contributing to the development of various diseases, including immunological, neurological and cancer-related disorders. Chromatography coupled with mass spectrometry (MS) has emerged as a powerful analytical approach, offering high sensitivity and resolution for studying gut microbiota-related metabolites. Heavy metals are natural components of the environment and can be relatively non-impactful in low concentrations. However, high concentrations of heavy metal toxicity can result in air, water, and soil contamination that adversely impacts environmental, human, and wildlife health. This study focuses on detecting the presence of heavy metals in soil, Loblolly Pine tree bark, and wood core samples from sites near a landfill in the Tallassee, Alabama, community and a post-agricultural site in Tuskegee, Alabama. Mercury (Hg), an environmental pollutant, can harm human health. Seafood consumption is a major way that people are exposed to Hg and the analysis of human hair is a standard method to measure Hg exposure. This community-based study identified demographic and dietary factors associated with elevated Hg concentrations and health outcomes of individuals with elevated Hg concentrations throughout two coastal South Carolina (SC) counties. We collected hair samples (n = 85) at local barbershops and salons; participants completed an online survey with questions on demographics, diet, and health histories; and we analyzed hair samples using a DMA-80. Mercury is a persistent global contaminant that biomagnifies through aquatic food webs, with dietary and environmental factors serving as the primary drivers of accumulation in fishes. Trophic structure and methylmercury dynamics in Walleye (ogaa; Sander vitreus) and Yellow Perch (asaawens; Perca flavescens) were investigated in the Upper and Lower Red Lake basins in Red Lake, Minnesota, during 2024-2025. Diets of Walleye and Yellow Perch were assessed through stomach dissections, and tissue samples from both species were analyzed for total mercury concentrations.
