Concentration Of Leukotrienes B4 And C4 In Human Periradicular Lesions, Elisabetta Cotti

Loma Linda University Electronic Theses, Dissertations & Projects

The 5-lipoxygenase metabolites of arachidonic acid such as leukotrienes (LT) B4 and C4, are potent mediators of inflammation. The purpose of this investigation was to determine the concentrations of LTB4 and LTC4 in human periradicular lesions. Specimens from asymptomatic and symptomatic patients were obtained and immediately frozen in liquid nitrogen. Periradicular tissues from unerupted third molars as well as chronically inflamed gingival tissues were also obtained, frozen and were used as negative and positive controls respectively. The concentrations of LTB4 and LTC4 were determined by the reverse phase high pressure liquid chromatography. Representative …

The Viral Envelope Gene Is Involved In Macrophage Tropism Of A Human Immunodeficiency Virus Type 1 Strain Isolated From Brain Tissue, Zhen-Qian Liu, Charles Wood, Jay Levy, Cecilia Cheng-Mayer Dec 1990

The Viral Envelope Gene Is Involved In Macrophage Tropism Of A Human Immunodeficiency Virus Type 1 Strain Isolated From Brain Tissue, Zhen-Qian Liu, Charles Wood, Jay Levy, Cecilia Cheng-Mayer

Nebraska Center for Virology: Faculty Publications

Human immunodeficiency virus type 1 (HIV-1) strains isolated from the central nervous system (CNS) may represent a subgroup that displays a host cell tropism different from those isolated from peripheral blood and lymph nodes. One CNS-derived isolate, HIV-lSF128A , which can be propagated efficiently in primary macrophage culture but not in any T-cell lines, was molecularly cloned and characterized. Recombinant viruses between HIV-1SF128A and the peripheral blood isolate HIV-ISF2 were generated in order to map the viral gene(s) responsible for the macrophage tropism. The env gene sequences of the two isolates are about 91.1% homologous, with variations …

Trace Element Studies On Karachi Populations Part V: Blood Lead Levels In Normal Healthy Adults And Grammar School Children, W W. Manser, R Lalani, S Haider, M A. Khan Jul 1990

Trace Element Studies On Karachi Populations Part V: Blood Lead Levels In Normal Healthy Adults And Grammar School Children, W W. Manser, R Lalani, S Haider, M A. Khan

Department of Biological & Biomedical Sciences

Blood lead levels of healthy Karachi population were estimated. Mean levels for males, females, soldiers and school children were 34.4, 31.8, 29.9 and 38.2 micrograms/dl respectively. About 93% cases of either sex had elevated lead levels, of whom 30% males and 10% females had levels above the safety limits (40 micrograms/dl). Soldiers living in relatively pollution free area though had levels lower than the rest of the population but 91% had levels over 25 micrograms/dl and only two had acceptable levels. Ninety-two percent children showed levels above 25 micrograms/dl with a large number having levels over 40 micrograms/dl. A very …

The Growth Of Simian Virus 40 (Sv40) Host Range/Adenovirus Helper Function Mutants In An African Green Monkey Cell Line That Constitutively Expresses The Sv40 Agnoprotein., Terryl P. Stacy, Michele Chamberlain, Susan Carswell, Charles N. Cole Jul 1990

The Growth Of Simian Virus 40 (Sv40) Host Range/Adenovirus Helper Function Mutants In An African Green Monkey Cell Line That Constitutively Expresses The Sv40 Agnoprotein., Terryl P. Stacy, Michele Chamberlain, Susan Carswell, Charles N. Cole

Dartmouth Scholarship

The simian virus 40 T-antigen carboxy-terminal mutants, dlA2459 and dlA2475, are cell line and temperature dependent for growth and plaque formation in monkey kidney cells. Although these mutants did form plaques on BSC-1 cells at 37 degrees C, they were about fivefold less efficient for plaque formation than wild-type simian virus 40. These mutants did not grow in CV-1 cells and did not synthesize agnoprotein in those cells. CV-1 cells which constitutively express the agnoprotein were permissive for mutant plaque formation. However, late mRNAs, virion proteins, and progeny virion yields did not accumulate to wild-type levels during mutant infection of …

Maternal Immunomodulation Of Neonatal Alloantigen Response, Leh Chang Jun 1990

Maternal Immunomodulation Of Neonatal Alloantigen Response, Leh Chang

Loma Linda University Electronic Theses, Dissertations & Projects

Remarkable success has been achieved in the transplantation of allogeneic cardiac grafts into newborn infants at Loma Linda University Medical Center. The superior graft survival rate documented in these patients has not correlated with the degree of immunosuppression rendered, or the selection of genetically matched donors. However, the clinical success has correlated with the age of the recipient at the time of receiving a transplant. Patients receiving an allograft within the first few weeks of life are unique in that they seem to accept the alloantigens of their cardiac graft while responding aggressively to antigens in their environment. These observations …

Characterization Of Hsd::Mudx128 Operon Fusion Mutant Of Escherichia Coli K-12, Marjorie Ann T. Reyno Jun 1990

Characterization Of Hsd::Mudx128 Operon Fusion Mutant Of Escherichia Coli K-12, Marjorie Ann T. Reyno

Loma Linda University Electronic Theses, Dissertations & Projects

The study of the regulation of gene expression in complex genes can be facilitated by the use of operon fusions which place a well characterized lacZ gene under the control of a promoter of interest. An hsd::MudX128 operon fusion mutant of E. coli K-12 isolated by Prakash (1986) was observed to have a high β-galactosidase activity (1000 units), ten to twenty times higher than other hsd::MudX mutants. This β-galactosidase activity was suppressed to a very low level (25 units) by the introduction of an F’ plasmid, F'101, which carries the 98 min to 2 min region of the …

Citrate As A Siderophore In Bradyrhizobium Japonicum., Mary Lou Guerinot, Erik J. Meidl, Ora Plessner Jun 1990

Citrate As A Siderophore In Bradyrhizobium Japonicum., Mary Lou Guerinot, Erik J. Meidl, Ora Plessner

Dartmouth Scholarship

Under iron-limiting conditions, many bacteria secrete ferric iron-specific ligands, generically termed siderophores, to aid in the sequestering and transport of iron. One strain of the nitrogen-fixing soybean symbiont Bradyrhizobium japonicum, 61A152, was shown to produce a siderophore when 20 B. japonicum strains were screened with all six chemical assays commonly used to detect such production. Production by strain 61A152 was detected via the chrome azurol S assay, a general test for siderophores which is independent of siderophore structure. The iron-chelating compound was neither a catechol nor a hydroxamate and was ninhydrin negative. It was determined to be citric acid via …

Mechanism Of Escape Of Endogenous Murine Leukemia Virus Emv-14 From Recognition By Anti-Akr/Gross Virus Cytolytic T Lymphocytes., Hillary D. White, Michael D. Robbins, William R. Green Jun 1990

Mechanism Of Escape Of Endogenous Murine Leukemia Virus Emv-14 From Recognition By Anti-Akr/Gross Virus Cytolytic T Lymphocytes., Hillary D. White, Michael D. Robbins, William R. Green

Dartmouth Scholarship

It was previously shown that spleen cells from endogenous ecotropic murine leukemia virus emv-14+ AKXL-5 mice fail to stimulate an anti-AKR/Gross virus cytolytic T-lymphocyte (CTL) response in a mixed lymphocyte culture with primed C57BL/6 responder spleen cells, whereas spleen cells from AKXL strains carrying the very similar emv-11 provirus do stimulate a response (Green and Graziano, Immunogenetics 23:106-110, 1986). We wished to determine whether the lack of response with AKXL-5 spleen cells was at the level of recognition between effector cell and target cell and whether the relevant mutation was within the emv-14 provirus. It is shown here that EMV-negative …

Toxicity Of Protease-Resistant Domains From The Delta-Endotoxin Of Bacillus Thuringiensis Subsp. Israelensis In Culex Quinquefasciatus And Aedes Aegypti Bioassays, Mary Ann Pfannenstiel, William C. Cray Jr., Graham Couche, Kenneth W. Nickerson Jan 1990

Toxicity Of Protease-Resistant Domains From The Delta-Endotoxin Of Bacillus Thuringiensis Subsp. Israelensis In Culex Quinquefasciatus And Aedes Aegypti Bioassays, Mary Ann Pfannenstiel, William C. Cray Jr., Graham Couche, Kenneth W. Nickerson

Papers in Microbiology

The mosquitocidal glycoprotein endotoxin of Bacillus thuringiensis subsp. israelensis was digested with chymotrypsin to yield protease-resistant domains which were then separated from smaller protease digestion products by high-performance liquid chromatography. Once purified, the domains no longer bound wheat germ agglutinin, a lectin which binds N-acetylglucosamine (GlcNAc) and GlcNAc oligomers. Purified protease-resistant domains were as toxic for Culex quinquefasciatus larvae as intact solubilized toxin. In separate experiments, the toxicity of chymotrypsin-digested endotoxin for Aedes aegypti larvae was reduced fivefold or more. A model is presented in which GlcNAc-containing oligosaccharides are required for toxicity for A. aegypti larvae but not C. …

The Isolation And Partial Characterization Of The Lipopolysaccharides From Rhizobium Leguminosarum Biovar Trifolii Strains Tb4, Tb104, And Tb112, Jill K. Miller Jan 1990

The Isolation And Partial Characterization Of The Lipopolysaccharides From Rhizobium Leguminosarum Biovar Trifolii Strains Tb4, Tb104, And Tb112, Jill K. Miller

Masters Theses

The lipopolysaccharides (LPSs) from Rhizobium leguminosarum biovar trifolii TB4 and two transposon (Tn5) symbiotic mutants, R. leguminosarum bv. trifolii TB104 and TB112, were isolated and partially characterized. Bv. trifolii TB4 is a derivative of R. leguminosarum bv. trifolii ANU843. The two mutant strains elicited incompletely developed clover nodules that exhibited low bacterial populations and very low nitrogenase activity. The LPSs were extracted using the hot phenol-water method and purified with gel filtration column chromatography using Sepharose 4B with an elution buffer consisting of EDTA and triethylamine. The hexose compositions were determined by gas chromatography of the alditol acetate derivatives. LPS …

Simian Virus 40 Host Range/Helper Function Mutations Cause Multiple Defects In Viral Late Gene Expression., Terryl Stacy, Michele Chamberlain, Charles N. Cole

Dartmouth Scholarship

Simian virus 40 (SV40) deletion mutants dlA2459 and dlA2475 express T antigens that lack the normal carboxy terminus. These mutants are called host range/helper function (hr/hf) mutants because they form plaques at 37 degrees C on BSC-1 and Vero monkey kidney cell lines but not on CV-1p monkey kidney cells. Wild-type SV40 can provide a helper function to permit growth of human adenoviruses in monkey kidney cells; the hr/hf mutants cannot. Progeny yields of hr/hf mutants are also cold sensitive in all cell lines tested. Patterns of viral macromolecular synthesis in three cell lines (Vero, BSC-1, and CV-1) at three …

Significance Of Premature Stop Codons In Env Of Simian Immunodeficiency Virus, Toshiaki Kodama, Dawn P. Wooley, Yathirajulu M. Naidu, Harry W. Kestler Iii, Muthiah D. Daniel, Yen Li, Ronald C. Desrosiers

Neuroscience, Cell Biology & Physiology Faculty Publications

The location of the translational termination codon for the transmembrane protein (TMP) varies in three infectious molecular clones of simian immunodeficiency virus from macaques (SIVmac). The SIVmac251 and SIVmac142 infectious clones have premature stop signals that differ in location by one codon; transfection of these DNAs into human HUT-78 cells yielded virus with a truncated TMP (28 to 30 kilodaltons [kDa]). The SIVmac239 infectious clone does not have a premature stop codon in its TMP-coding region. Transfection of HUT-78 cells with this clone initially yielded virus with a full-length TMP (41 kDa). …

Linker Insertion Mutants Of Simian Virus 40 Large T Antigen That Show Trans-Dominant Interference With Wild-Type Large T Antigen Map To Multiple Sites Within The T-Antigen Gene., Jiyue Y. Zhu, Charles N. Cole Nov 1989

Linker Insertion Mutants Of Simian Virus 40 Large T Antigen That Show Trans-Dominant Interference With Wild-Type Large T Antigen Map To Multiple Sites Within The T-Antigen Gene., Jiyue Y. Zhu, Charles N. Cole

Dartmouth Scholarship

Linker insertion mutants affecting the simian virus 40 (SV40) large tumor (T) antigen were constructed by inserting a 12-base-pair oligonucleotide linker into restriction endonuclease cleavage sites located within the early region of SV40. One mutant, with the insertion at amino acid 5, was viable in CV-1p and BSC-1 cells, indicating that sequences very close to the amino terminus of large T could be altered without affecting the lytic infection cycle of SV40. All other mutants affecting large T were not viable. In complementation assays between the linker insertion mutants and either a late-gene mutant, dlBC865, or a host range/helper function …

Electrostatic Effects On Airborne Asbestos Monitoring, Roxanne Francis Oct 1989

Electrostatic Effects On Airborne Asbestos Monitoring, Roxanne Francis

Biological Sciences Theses & Dissertations

The affects of electrostatic forces on the Membrane Filter Method (MFM) used for air-borne asbestos monitoring have been investigated for many years. Several studies have indicated that these forces interfere with the collection of asbestos fibers on the membrane filter, which results in an underestimated analysis of the airborne concentration. By varying the electroconductivity of the extension cowls on the filter cassettes, it was speculated that the significance of the electrostatic forces could be statistically analyzed. Four sets of filter cassettes with grounded conductive (GC) extension cowls, ungrounded conductive (UC) extension cowls, grounded nonconductive (GN) extension cowls, and ungrounded nonconductive …

Use Of Trpe/Gag Fusion Proteins To Characterize Immunoreactive Domains On The Human Immunodeficiency Virus Type 1 Core Protein, Michael Windheuser, Gary Tegtmeier, Charles Wood

Nebraska Center for Virology: Faculty Publications

The human immunodeficiency virus (HIV) p24 core protein is one of the most immunogenic of HIV structural proteins. Infected individuals develop high titers of antibodies against p24 early in infection, which makes anti-p24 antibodies important serological markers. However, despite the clinical importance of the anti-p24 response, no systematic study to characterize the antigenic domains on the p24 protein has been reported. We report here on the use of 12 overlapping fragments of the HIV type 1 p24 protein, synthesized in bacteria as TrpE/Gag fusion proteins, to identify at least two and possibly three antigenic domains on the p24 protein. In …

Immunosuppressive Activity Derived From The Uc729-6 Lymphoblastoid And M21-Hpb Melanoma Cell Lines, Charlene J. Repique

Loma Linda University Electronic Theses, Dissertations & Projects

Previous work conducted in our laboratories, using a murine model, suggested that soluble factors secreted by tumor cells suppress lymphocyte responses. To apply this premise to human tumors, we studied the effects of UC729-6 (lymphoblastoid B-cell) and M21-HPB (malignant melanoma) conditioned media (CM) on lymphocyte proliferation assays, as well as on the growth of other cells. The CM was collected at 2-5 day intervals from cultures of UC729-6 and M21- HPB cells in serum-free media. In assays using phytohemagglutinin (PHA) and concanavalin A (ConA), mononuclear peripheral blood cells from healthy human donors showed decreased [3H]-thymidine ([3H]-Tdr) …
