In the United States, due to the increasing prevalence of cardiovascular disease, over 900,000 cardiac surgeries are performed annually, with projections exceeding 1.3 million by 2029. Many post-surgical patients require continuous in-hospital cardiac monitoring, which is typically conducted using electrocardiograms (EKGs). However, traditional EKG systems such as bedside cardiac monitors and cardiac telemetry monitors rely on wired leads that can easily tangle, degrade signal quality, and require frequent readjustments or replacements due to wire breakage, and for the patient, this can cause discomfort from prolonged use. Traumatic Brain Injury (TBI) is a serious public health concern that leads to the death or disability of thousands of individuals in the US annually. Progression of TBI involves primary damage from the initial, mechanical injury and secondary damage, from nonmechanical, pathological dysfunction. The secondary damage mechanisms can be better understood, diagnosed, and treated through the quantification of TBI biomarkers. The two biomarkers of interest in this study are pTau and GFAP. The male and female mouse models showed similar significant increase in GFAP following injury, as well as a general lack of increase in pTau expression following the injury. …

Our research team developed a PVNS for Epilepsy device which is intended to act as a preventative measure against seizures for patients with epilepsy. The device is a less invasive alternative to the VNS device on the market using electroacupuncture at the Shenmen and Xin points along the posterior pinna to stimulate the vagus nerve.

Pediatric epilepsies, particularly those that are drug-resistant or genetically driven, represent some of the most complex neurological disorders encountered in childhood. Central to their pathophysiology is a disruption in the delicate balance between cortical excitation and inhibition (E/I), often resulting from impaired GABAergic interneuron function. This imbalance manifests as aberrant network dynamics and altered neural oscillations, giving rise to seizures and long-term cognitive impairments. The capacity of temporal interference (TI) stimulation to target deep brain regions without affecting nearby surface electrodes remains uncertain. Using artifact-free optical recording, we compare excitation patterns and thresholds in hippocampal neurons stimulated by “pure” and amplitude-modulated sine waves, representing TI waveforms near electrodes and at the target, respectively. We show that pure 2- and 20-kHz sine waves induce repetitive firing at rates that increase up to 60–90 Hz with stronger electric fields. Beyond this limit, action potentials merge into sustained depolarization, resulting in an excitation block. Modulating the sine waves at 20 Hz aligns firing with amplitude “beats” and …

Understanding plasma-liquid interactions and the impact of the electrical conductivity (σ) of liquid on plasma is important for plasma applications in water treatment and medicine. Ultrapure water, phosphate-buffered saline (PBS) solution, biological media, and sodium chloride (NaCl) solutions at various concentrations were used in this study to provide a broad range of conductivity values ranging from 0.055 μS cm−1 to 222 mS cm−1. Hand function is often impaired following neurological trauma or in conditions such as stroke, limiting independence and the ability to perform activities of daily living (ADLs). Current clinical assessments rely heavily on subjective rating scales that lack the resolution to detect subtle changes in motor performance, which can limit access to rehabilitation. This study presents an innovative, cost-effective strategy to develop a flexible, enzyme-free biosensor for the sensitive detection of uric acid (UA). Utilizing electrochemically modified cobalt nanostructured on laser-induced graphene electrodes (CoNCs/LIG), this approach surpasses traditional noble metal-based electrocatalysts in sensitivity and affordability. The one-step electrochemical modification method is efficient and straightforward, enabling the uniform deposition of hierarchical flower-like cobalt nanostructures. These structures synergistically enhance the performance of the LIG, resulting in a broad detection range of 5 to 700 µM with a sensitivity of 6.75 µA µM-¹ cm-² and a low detection limit of 3.66 µM for UA. …

Traumatic brain injury (TBI) is a major cause of neurological impairment, often leading to variable recovery and uncertain prognosis in the neurocritical care setting. There is a pressing clinical need for robust, physiologically grounded biomarkers to inform prognosis and therapeutic decision-making in acute TBI. This thesis investigates neurovascular coupling (NVC), the physiological coordination between neuronal activity and cerebral blood flow, as a candidate biomarker for brain function and recovery after injury.

Durable dielectric encapsulation is crucial for the long-term function of implantable neural electrodes; however, the reliability of curved, thermoformed coatings remains underexplored. This study addresses that gap by comparing planar (non-thermoformed) and thermoformed interdigitated neural electrodes (IDEs) encapsulated in Polyimide-2611. Both device types underwent accelerated in vitro aging in phosphate‑buffered saline (PBS) at 67 °C for approximately 91 days, simulating two years of physiological exposure. Post-accelerated aging evaluation included electrochemical impedance spectroscopy (EIS) from 0.1 Hz to 100 kHz to monitor moisture‑induced leakage, and high‑voltage breakdown testing under saline immersion to assess dielectric strength. Background: Parkinson’s disease (PD) is a progressive neurodegenerative disorder affecting ~1 in 1000 individuals over age 45. Freezing of Gait (FOG), a debilitating symptom in late-stage PD, affects up to 63% of patients and contributes to falls and reduced quality of life. While deep brain stimulation (DBS) of the subthalamic nucleus (STN) helps some symptoms, FOG remains difficult to treat. Given the complexity of basal ganglia circuitry, the role of related basal ganglia nuclei, such as the Globus Pallidus internus (GPi), in FOG deserves further investigation.

Auditory Nerve Interface (ANI) devices, currently in developmental stages, represent an emerging neuroprosthetic approach to restore hearing in severe sensorineural loss patients by directly stimulating the auditory nerve via the Utah Slanted Electrode Array (USEA). Conventional implant leads are comprised of multiple wires made from materials such as platinum-iridium and MP35N stainless steel that are overmolded in an insulative material. Background: Large bifrontal defects pose unique reconstruction challenges due to their complex curvature and mechanical requirements. This case demonstrated how computer-aided design/manufacturing (CAD/CAM) enabled precise single-piece polymethyl methacrylate (PMMA) implant fabrication, thereby overcoming traditional limitations. Case presentation: A 25-year-old male who had undergone bifrontal decompressive craniectomy suffered a severe traumatic brain injury. The autologous bone flap had been temporarily stored in a subcutaneous fat area of the abdomen for 3 months to preserve its viability. Pain is a hallmark of Sickle Cell Disease (SCD), a genetic hemoglobinopathy characterized by chronic hemolytic anemia, vaso-occlusion, and organ damage. While acute vaso-occlusive crises (VOC) have been extensively studied, the mechanisms underlying chronic pain and its interplay with brain function remain unclear. This dissertation investigates the relationships between cerebral blood flow (CBF), fluid cognition, and pain sensitivity in adults with SCD, utilizing advanced imaging techniques and validated assessment tools.

Predicting compound-protein interactions (CPIs) plays a crucial role in drug discovery. Traditional methods, based on the key-lock theory and rigid docking, often fail with novel compounds and proteins due to their inability to account for molecular flexibility and the high sparsity of CPI data. Here, we introduce ColdstartCPI, a framework inspired by induced-fit theory, which leverages unsupervised pre-training features and a Transformer module to learn both compound and protein characteristics. ColdstartCPI treats proteins and compounds as flexible molecules during inference, aligning with biological insights. Evaluating the effectiveness of transcranial direct current stimulation (tDCS) is essential for guiding its integration into therapeutic and performance-enhancement applications. In our laboratory, we investigate the efficacy of tDCS across multiple experimental models, including both animal and human studies. I have contributed significantly to the execution and analysis of these experiments, which include studies in rats and healthy human participants aimed at evaluating whether electrical stimulation of the motor cortex can enhance motor learning. Non-invasive electrotherapies offer promising treatments for a wide range of conditions such as pain, headaches, depression, addiction, cognitive decline, wound healing, and drug delivery. However, patient compliance remains low due to the high initial cost, cumbersomeness and inconvenience of existing technology compared to pharmaceuticals. Current electrotherapy devices are often bulky and require users to connect disposable electrodes, initiate programs, and manage charging cycles, which hinders widespread adoption.

Intrinsic neural timescales (INTs) characterize the persistence of neural activity over time and offer insight into the brain’s temporal hierarchy. This thesis evaluates and compares six autocorrelation-based INT metrics—AR(1) Coefficient (Autoregressive Coefficient at 1), First Zero Lag, Lag at ACF = 1/e, Sum of Positive ACFs, ACF at Half Lag, Exponential Decay Constant (τ)— using resting-state fMRI data from 400 cortical regions across a large, lifespan sample (ages 6– 85) from the NKI-Rockland dataset. Neuroimaging studies have revealed consistent abnormalities in functional connectivity within specific neural networks that may serve as biomarkers for major depressive disorder (MDD). It is important to find inexpensive, non-invasive techniques that target these biomarkers to make diagnosis easier and more objective. EEG microstates are quasi-stable potential topographies that are thought to reflect the quasi-stable network activity of the underlying neural generators. Fast Scan Cyclic Voltammetry (FSCV) is a widely used electrochemical technique for real-time measurement of the brain’s chemical messengers, including the molecule dopamine, with high temporal resolution. Currently the financial burden of performing FSCV is quite high, ranging from $8,000 to $20,000+ making the barrier to entry nearly insurmountable for laboratories and classrooms at small institutions. The purpose of this project was to develop a Do-It-Yourself (DIY), portable, and cost-effective FSCV system for use in laboratory and classroom settings. Spinal cord injury (SCI) is a devastating, life changing event that greatly impacts quality of life. SCI can have body-wide, multi-system consequences including dysfunction within the urogenital system. Challenges with the urinary system include urinary incontinence, urine retention, and detrusor-sphincter dyssynergia all of which require maintenance through the regular use of catheters. Sexual dysfunction in men, who comprise nearly 80% of the SCI population, includes erectile dysfunction, anejaculation, retrograde ejaculation, poor expulsive force during ejaculation, and poor sperm quality. Implantable drug delivery devices have many benefits over traditional drug administration techniques and have attracted a lot of attention in recent years. By delivering the medication directly to the tissue, they enable the use of larger localized concentrations, enhancing the efficacy of the treatment. Passive-release drug delivery systems, one of the various ways to provide medication, are great inventions. However, they cannot dispense the medication on demand since they are nonprogrammable. In this thesis, we have applied Orthogonal Projective Non-Negative Matrix Factorization (opNMF) to identify spatial patterns of white matter hyperintensities (WMH) within UK Biobank's imaging data. Our selection criteria excluded subjects with a history of neurological, mental, and specific cerebrovascular conditions, allowing us to focus on WMH patterns in a healthy aging population. We have interrogated the association of location-specific WMHs with a variety of demographic, clinical, and genetic factors. Our multivariable regression analysis evaluates the strength and nature of the associations between these factors and WMH distribution. The Labor Assist Device (LAD) was designed to reduce the muscle fatigue experienced by women during childbirth to decrease the number of unnecessary cesarean deliveries and injuries to nurses. The purpose of this thesis is to develop a protocol for a future study to assess the effectiveness of the LAD at reducing muscle fatigue. While multiple parameters were investigated in this pilot study, the major parameter needed to develop the future protocol is the muscle fatigue rate in women contracting their muscles to simulate pushing during labor. However, many standardized assessment tools rely on human raters for performance assessment, which is resource-intensive and subjective. However, inadequate methods for open suturing skills training necessitate efficient and objective methods to develop skills. To address this need, medical training simulators for objective surgical skills training are gaining popularity for their formative assessment.
