Enhanced tumor delivery of plasmid DNA with electric pulses in vivo has been confirmed in many preclinical models. Intratumor electrotransfer of plasmids encoding therapeutic molecules has reached Phase II clinical trials. In multiple preclinical studies, a reduction in tumor growth, increased survival or complete tumor regression have been observed in control groups in which vector or backbone plasmid DNA electrotransfer was performed. This study explores factors that could produce this antitumor effect. The specific electrotransfer pulse protocol employed significantly potentiated the regression. Approximately 1.7 million Americans experience a traumatic brain injury (TBI) each year. Concussive injuries are a subset of TBI in which blows to the head cause the brain to collide against the interior of the skull. Damage to the neurons, supporting cells, and surrounding extra cellular matrix resulting from these collisions can lead to permanent physical, cognitive, and psychological impairment. We believe the prevalence and clinical significance of concussive injures warrants research investment. Demand exists for brain-machine interfaces that offer a wide range of sensory feedback along with volitional motor control to individuals with limited control of natural sensory or motor function. As these sensorimotor devices are developed, it is necessary to improve the interaction between the prostheses and higher-level cortical structures. Optimizing these somatosensory stimulation parameters will require the use of a high-throughput experimental design. Optogenetics is a developing field in neuroscience, where nerves are stimulated through optical instead of electrical signals, allowing for a more selective action of nerve populations [Aravanis et al, 2007]. The objective of this dissertation was to more fully understand the role of the human brain in the production of lower extremity rhythmic movements. Throughout the last century, evidence from animal models has demonstrated that spinal reflexes and networks alone are sufficient to propagate ambulation. However, observations after neural trauma, such as a spinal cord injury, demonstrate that humans require supraspinal drive to facilitate locomotion. To investigate the unique nature of lower extremity rhythmic movements, electroencephalography was used to record neural signals from the sensorimotor cortex during three cyclic ankle movement experiments. Maintaining dynamic balance is an important component of walking function that is likely impaired in chronic stroke survivors, evidenced by an increased prevalence of falls. Dynamic balance control requires maintaining the center of mass (COM) within the base of support during movement. During walking, dynamic balance control is achieved largely by modifying foot placement to adjust the base of support. However, chronic stroke survivors have difficulty with both precision control of foot placement, as well as reduced control of COM movement. Certain areas of the body, such as the hands, eyes and organs of speech production, provide high-bandwidth information channels from the conscious mind to the outside world. The objective of this research was to develop an innovative wearable sensor device that records signals from these areas more conveniently than has previously been possible, so that they can be harnessed for communication. A novel bioelectrical and biomechanical sensing device, the wearable endogenous biosignal sensor (WEBS), was developed and tested in various communication and clinical measurement applications.

An estimated 0.5% of world’s population has been diagnosed with epilepsy. Of these patients 20-30% will be unable to achieve seizure control with anti-epileptic drugs. Vagus nerve stimulation (VNS) may be an appropriate treatment option for some patients with pharmaceutically refractory, partial-onset seizures.

VNS therapy uses a helical electrode to interface between the implantable pulse generator and the vagus nerve. Permeabilization of cell membranous structures by nanosecond pulsed electric field (nsPEF) triggers transient rise of cytosolic Ca2+ concentration ([Ca2+]i), which determines multifarious downstream effects. By using fast ratiometric Ca2+ imaging with Fura-2, we quantified the external Ca2+ uptake, compared it with Ca2+ release from the endoplasmic reticulum (ER), and analyzed the interplay of these processes. We utilized CHO cells which lack voltage-gated Ca2+ channels, so that the nsPEF-induced [Ca2+]i changes could be attributed primarily to electroporation. Stretch is known to result in an electrically less stable ventricular substrate, yet the reported effects of stretch on measured electrophysiological parameters have been inconsistent and even contradictory. The goal of this study was to evaluate the effects of acute mechanical stretch on cardiac electrical features thought to be key in generation of arrhythmia, namely restitution of action potential duration (APD), electrical memory, and onset of alternans.

The steady-state visual evoked potential (SSVEP) is an electroencephalographic response to flickering stimuli generated in significant part by activity in primary visual cortex (V1). SSVEP signal-to-noise ratio is generally low for stimuli that are located in the visual periphery, at frequencies higher than 20 Hz, or at low contrast. Because of the typical "cruciform" geometry of V1, large stimuli tend to excite neighboring cortical regions of opposite orientation, likely resulting in electric field cancellation.
