The objective of this project was to convert a Sarastro 2000 confocal laser scanning microscope (CLSM) into a system capable of far-field two-photon excitation (TPE) imaging for the use of the PSU Biology department. TPE microscopy operates on the ability of fluorophores to accept two photons each with half the energy of a desired transition in a single quantum event via a virtual energy state and then emit a higher energy photon upon relaxation. Purpose: The mechanical behavior of heart tissue plays a crucial role in elucidating cardiac physiology and pathophysiology. Accurate measurement of the mechanical properties of cardiac and arterial tissues is indispensable for advancing computer simulations and developing medical devices. This study aimed to assess the passive mechanical properties of cardiac and arterial tissue in a calf model using a paired comparison method.

In engineering education, students often face feelings of inadequacy, leading to academic struggles and potential dropout. This dissertation investigates the impact of interactive course materials on students' confidence and self-efficacy in problem-solving, focusing on an Engineering Materials class at Louisiana Tech University. Over four quarters, involving seven sections and 218 students, a 13-question Likert scale survey was administered repeatedly, alongside demographic data and textbook usage surveys. The study aims to compare students’ attitudes and beliefs when not using a textbook versus when using an interactive web-native book. Patients diagnosed with glioblastoma multiforme (GBM) continue to face a dire prognosis. Developing accurate and efficient contouring methods is crucial, as they can significantly advance both clinical practice and research. This study evaluates the AI models developed by MRIMath© for GBM T1c and fluid attenuation inversion recovery (FLAIR) images by comparing their contours to those of three neuro-radiologists using a smart manual contouring platform. The mean overall Sørensen–Dice Similarity Coefficient metric score (DSC) for the post-contrast T1 (T1c) AI was 95%, with a 95% confidence interval (CI) of 93% to 96%, closely aligning with the radiologists’ scores. Objective: Bioprosthetic valves using either porcine or bovine pericardium have been widely used for transcatheter heart valve replacements. However, producing bioprosthetic valves is not a sustainable solution. Acquiring animal tissue is often not readily available and costly and requires time-consuming modifications. Moreover, its primary tissue failure requires reoperation after roughly 15 years. The suggested replacement of porcine and bovine leaflets with biocompatible polymers appears to be an attractive alternative to bioprosthetic valves. In this study, engineered fiber-reinforced polymers were developed, which are less degenerative and offer greater hemocompatibility in relation to mechanical valves.

Movement can affect the way we make sense of complex visual information. To investigate this issue, we designed an experiment to assess changes of plaid motion perception threshold after a period of sensorimotor contingency experience. We found that movement training facilitates combination of elementary motion cues into a global motion percept. No changes in perceptual thresholds are observed in a passive visual condition. A Bayesian model suggested a reduction, after training, of the cross-talk between two gratings with unbalanced contrasts in corresponding sensory channels. In this thesis, we have applied Orthogonal Projective Non-Negative Matrix Factorization (opNMF) to identify spatial patterns of white matter hyperintensities (WMH) within UK Biobank's imaging data. Our selection criteria excluded subjects with a history of neurological, mental, and specific cerebrovascular conditions, allowing us to focus on WMH patterns in a healthy aging population. We have interrogated the association of location-specific WMHs with a variety of demographic, clinical, and genetic factors. Our multivariable regression analysis evaluates the strength and nature of the associations between these factors and WMH distribution. Tumor-associated macrophages (TAMs) play a substantial role in sculpting the tumor microenvironment (TME) into an immunosuppressive bastion that undermines immune surveillance and enables tumor progression. They therefore represent a promising therapeutic target to make the TME more conducive for anti-tumor immunity and greatly improve treatment outcomes in cancers that are highly resistant to conventional forms of therapy. Machine learning is commonly used in biomedical image analysis, as it allows automated image segmentation and identification that minimizes the need for tedious human involvement. Drosophila melanogaster is often used as a cardiac disease model, where optical coherence microscopy (OCM) is used to image and analyze its beating dynamics. As OCM often generates a large volume of images, automated image segmentation is necessary to quantify the heart beating efficiently.
