Automated, Robust Radiomics Biomarker Modeling For Glioblastoma, Eric Nathan Carver
Wayne State University Dissertations
Individualized medicine is possible due to the discovery that a patient’s gene expression can serve as a bio-marker and provide prognostic clinical knowledge. The genetic revolution increased interest in the field of “-omics” which investigates differing classifications of patient characteristics to increase clinician knowledge, which directly improves patient care. Glioblastoma (GBM) is the most aggressive brain cancer and very difficult to treat, resulting in a 15 month median survival. Researchers have discovered several patient specific markers, specifically IDH1 and MGMT methylation, which energized further research for GBM patient specific markers. A relatively new subfield of ‘-omics’ is Radiomics; which aims …

Probing The Effect Of Hyperglycemia On Endothelial Force Generation And Transmission, Jovani J. Gutierrez
Honors Undergraduate Theses
This thesis intends to utilize biomechanics to study the endothelial biomechanical response in a static hyperglycemic microenvironment. Hyperglycemia is a diabetic condition with abnormally high levels of glucose in the bloodstream. The effects of hyperglycemia over time lead to vascular complications resulting in patients being more prone to cardiovascular diseases. Current studies have focused on the molecular mechanisms affected by hyperglycemia; however, the mechanical mechanisms by which hyperglycemia causes vascular structural and functional changes are understudied. Therefore, to study the effects of hyperglycemia in the endothelium, Human Umbilical Vein Endothelial Cells (HUVEC) were cultured under three glucose conditions: normal glucose …

Electronic Theses and Dissertations, 2020-2023
Mobile brain-body imaging (MoBI) seeks to understand human brain and body dynamics during movement and locomotor tasks such as walking with perturbations that challenge balance and lead to adaptation of walking behavior. In this dissertation, I evaluated the long-term electromyography (EMG) recording performance of dry epidermal electrodes for measuring electrical muscle activity. I also evaluated the relationships between the signals recorded from the two sides of dual-sided electroencephalography (EEG) electrodes, a recent advancement in EEG electrode design for measuring electrical brain activity. Last, I investigated adaptation of brain and body responses to small and frequent perturbations during treadmill walking while …

The most critical component of the TDDS is the adhesive, which is responsible for the safety, efficacy and quality of the patch. For drug delivery to successfully occur, the patch must adhere to the surface of the contact area. If a patch has inadequate adhesion, it is likely to fall off before the entire delivery period has been satisfied, leading to risks for the patient and others who may encounter the patch. Despite the critical concerns associated with the adhesive properties of the patches, the adhesion quality and failure mechanisms have not been fully studied. If certain molecules encounter the …

The main motivation for studying damage in bone tissue is to better understand how damage develops in the bone tissue and how it progresses. Such knowledge may help in the surgical aspects of joint replacement, fracture fixation or establishing the fracture tolerance of bones to prevent injury. Currently, there are no standards that create a realistic bone model with anisotropic material properties, although several protocols have been suggested. This study seeks to retrospectively evaluate the damage of bone tissue with respect to patient demography including age, gender, race, body mass index (BMI), height, and weight, and their role in causing …

Diffuse midline glioma, K27M-mutant (DMG) are intractable brain tumors, primarily occurring in the pediatric and adolescent population. Patients have a median survival of less than one year after diagnosis. A lack of therapeutic targets has been a barrier to improvement in patient survival. Irradiation therapy improves symptoms while chemotherapy and surgical intervention, for the most part, have not yet demonstrated utility for treatment. DMG are characterized by a histone H3 mutation that results in the genome-wide loss of epigenetic-repressive marks on K27 and is associated with the misexpression of genes, like Cancer/Testis antigens. A member of this group, transketolase-like 1 …

Peripheral artery disease (PAD) is a disease that affects over 200 million patients worldwide and results in a narrowing of the blood vessels in the lower limbs that is caused by plaque buildup and chronic inflammation. The standard of care for treating this disease is the use of a drug-coated balloon (DCB) to open up the artery through the dilation of the vessel with the balloon and to deliver paclitaxel which prevents the vessel from narrowing after the procedure. In the United States, these DCBs are only approved by the FDA to be used in the artery segments above the …

In recent years, the emerging technology of cold atmospheric pressure plasma (CAP) has grown rapidly along with the many medical applications of cold plasma (e.g., cancer, skin disease, tissue repair, etc.). Plasma-activated liquids (e.g., culture media, water, or normal saline, previously exposed to plasma) are being studied as cancer treatments, and due to their advantages, many researchers prefer plasma-activated liquids as an alternative to CAP in the treatment of cancer. In this study, we showed that plasma-activated-saline (PAS) treatment significantly inhibited tumor growth, as compared with saline, in melanoma, and a low-pH environment had little effect on tumor growth in …

During the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic, alcohol consumption increased markedly. Nearly one in four adults reported drinking more alcohol to cope with stress. Chronic alcohol abuse is now recognized as a factor complicating the course of acute respiratory distress syndrome. and increasing mortality. To investigate the mechanisms behind this interaction, we developed a combined acute respiratory distress syndrome and chronic alcohol abuse mouse model by intratracheally instilling the S1 subunit of SARS-CoV-2 spike protein (S1SP) in K18-human angiotensin-converting enzyme 2 (ACE2) transgenic mice that express the human ACE2 receptor for SARS-CoV-2 and are kept on an …
