Electronic Theses and Dissertations

T-cell therapies have been gaining popularity in recent years due to their cancer fighting potential. With remission rates improving in this field of immunotherapy, the demand for T-cell therapies has also increased; however, the cell processing techniques for these therapeutic products have yet to rise to the level of demand. The manufacturing process takes too long and a significant amount of processed cell batches can fail to meet safety requirements. These limitations of cell processing can be detrimental to patients seeking T-cell therapies. While current products have improved the time it takes to manufacture these therapeutic products, there is still …

Acoustofluidic Delivery Of Gene Editing Compounds For Improved Immunotherapy Processing., Riyakumari K. Patel 2022 University of Louisville

Acoustofluidic Delivery Of Gene Editing Compounds For Improved Immunotherapy Processing., Riyakumari K. Patel

Electronic Theses and Dissertations

Cell-based immunotherapies are a new generation of “living-drug” treatments for cancer and other diseases. Chimeric antigen receptor (CAR) T-cell cancer therapy has shown promising results in lymphoma and B-cell malignancies. Currently, there are six FDA-approved CAR-T drugs on the market, and all of them use viral transfection for reprogramming. While viral transfection is effective, there are safety concerns due to inconsistent transfection that limit the use of CAR-T therapy. Current non-viral transfection techniques generally have lower transfection efficiency than viral transfection. Additionally, these techniques can be toxic, time-consuming, non-transportable, and expensive. To address these limitations, a novel 3D printed acoustofluidic …

Study Of Porosity Of Gelatin-Alginate Hydrogels To Model Brain Matter For Studying Traumatic Brain Injuries, Apolline Vincent

Honors Scholar Theses

Traumatic brain injuries (TBIs) affect brain tissue and neuronal signaling, leading to many side effects including death. Research on TBIs is limited by the lack of accurate brain matter models to study the physiological and cellular reaction. Alginate-gelatin hydrogels have been designed and modified to mimic the mechanical properties of the brain to act as an accurate in vitro model. This Honors thesis aims to verify the porous microstructure of gelatin-alginate hydrogels through Scanning Electron Microscopy (SEM) to understand how gelatin and alginate affect the microstructure. I measured the pore sizes of each hydrogel using ImageJ, compared the average pore …

Effects Of Orthopedic Implant And Host Bone Properties On Stress-Shielding Induced Osteopenia, Scott Jason Jan Baskerville 2022 Florida Institute of Technology

Effects Of Orthopedic Implant And Host Bone Properties On Stress-Shielding Induced Osteopenia, Scott Jason Jan Baskerville

Theses and Dissertations

Stress shielding is a term commonly used in literature to describe a bone density reducing phenomenon which occurs due to long-term exposure of bone tissue to sub-homeostatic levels of mechanical stimuli. A number of mechanobiological models ranging in complexity and purpose have been developed with the general overall intent of predicting bone remodeling behavior under a set of prescribed conditions. A previously developed heuristic model is expanded in the present work through model calibration with physiological data and the development of a reduced bone-implant-construct (BIC) mechanical model configurable for patient factors such as age, gender, and associated bone properties. The …

Cellular Migration As A Function Of Magnetic Field Intervention, Amy Marie Vecheck 2022 Florida Institute of Technology

Cellular Migration As A Function Of Magnetic Field Intervention, Amy Marie Vecheck

Theses and Dissertations

This work aims to assess cellular migration in three-dimensional structures as a function of Zeeman resonance. By using Zeeman resonance to initialize intersystem crossing of electrons, also known as the Radical Pair Mechanism, bioenergetics processes would be modulated. Processes such as oxygen consumption rate and extracellular acidification rate are indicative of cellular respiration and glycolysis and can be altered based on the orientation of the magnetic field applied: parallel and perpendicular, respectively. This study focused on the parallel orientation and looked to used Zeeman resonance to increase flavin adenosine dinucleotide, which results in preventing the breakdown of F-actin. Cellular viability …

Neural Correlates Of Multisensory Integration For Feedback Stabilization Of The Wrist, Aaron J. Suminski, Raymond C. Doudlah, Robert A. Scheidt

Biomedical Engineering Faculty Research and Publications

Robust control of action relies on the ability to perceive, integrate, and act on information from multiple sensory modalities including vision and proprioception. How does the brain combine sensory information to regulate ongoing mechanical interactions between the body and its physical environment? Some behavioral studies suggest that the rules governing multisensory integration for action may differ from the maximum likelihood estimation rules that appear to govern multisensory integration for many perceptual tasks. We used functional magnetic resonance (MR) imaging techniques, a MR-compatible robot, and a multisensory feedback control task to test that hypothesis by investigating how neural mechanisms involved in …

Quantifying Shear Stresses In Tissue Engineered Aortic Heart Valves, Raj Nitin Dave 2022 Clemson University

Quantifying Shear Stresses In Tissue Engineered Aortic Heart Valves, Raj Nitin Dave

All Theses

Present heart valve prosthesis have limitations such as capability to grow, repair and remodel post implantation. Tissue engineering offers to be a promising alternative to overcome these limitations. Maturation of seeded human cells on the valve subjected to favorable growth conditions in the bioreactor is critical to the success of tissue engineered heart valves. Mechanical stress and strain which results from the pressure and flow conditions in the bioreactor plays a critical role on the developing valve tissue and are currently unknown. The goal of this research is to relate the magnitude of wall shear stress (WSS) within the heart …

Fabrication Of Hydrogel Microparticles For Acute Treatment Of Penetrating Brain Injuries, Isabell Foulger

All Theses

Traumatic brain injuries (TBIs) are a type of acquired head trauma that results from an external force to the head. The development and severity of the secondary injury increases the risk of neurodegeneration and reduced cognitive recovery. Glucocorticoids are potent antiinflammatory therapeutics that have been investigated for potential treatment of TBIs, strokes, and other neuroinflammatory pathologies. However, their clinical application has been limited due to their ability to induce a variety of side effects, including adrenal suppression, edema, and reduced plasticity of the hippocampus. The use of a localized drug delivery system would overcome these systemic concerns. In previous studies, …

Design Of A Four Channel Pulsatile Perfusion Bioreactor For Ex-Vivo Study Of Vascular Grafts, Thomas M. Fair

All Theses

Cardiovascular diseases are the leading cause of death in the United States. Atherosclerosis in peripheral arteries is a major contributing factor. Autologous saphenous vein grafts are the most common bypass grafts for treating peripheral artery diseases. A quarter of these grafts fail within a year, and around half have failed within 10 years of the initial surgery. Graft failure is attributed to the development of intimal hyperplasia indicated by the migration and proliferation of vascular smooth muscle cells, fibroblasts, and the deposition of extracellular proteins.

Increases in flow, pressure, the pulse frequency, and the differential of pressure that model an …

Portable U-Bent Optical Fiber Biosensor For Detecting Microcystin-Lr, Quan Dio Anh Le

All Theses

Cyanobacteria are the only known prokaryotes that can photosynthesize. Due to their unique ability, they are important entities in the global carbon and nitrogen cycles. However, two factors can turn cyanobacteria into a nuisance. In general, eutrophication is the process that supplies external nutrients that are needed for cyanobacteria blooms into bodies of water from the surrounding environment. Climate warming intensifies this process by producing carbon dioxide, methane, and nitrous oxide. With cyanobacteria blooms occurring worldwide, they are becoming a bigger threat to human health, and there has been increased interest in detecting cyanobacteria and their toxic byproducts.

For instance, …

Insole Fall Prevention Device, Nick M. Hughes, Andrew M. Slaboda 2022 California Polytechnic State University, San Luis Obispo

Insole Fall Prevention Device, Nick M. Hughes, Andrew M. Slaboda

Biomedical Engineering

Falls among the aging population occur every single day, with 1 in every 5 resulting in some injury and 300,000 hospitalized every year with a hip fracture [1]. The most popular and effective way to mitigate these falls is through physical therapist intervention. However, with the increased popularity in telerehab, many patients at risk for falls cannot accurately convey their gait tendencies to their physical therapists from the comfort of their home or while not in direct contact with the PT. A device like an insole, implanted with force sensors, which measures different parts of a patient’s foot, could convey …

Exploring The Role Of Metabolites In Cancer And The Associated Nerve Crosstalk, Inah Gu, Emory A. Gregory, Casey Atwood, Sun-Ok Lee, Young Hye Song

Biomedical Engineering Faculty Publications and Presentations

Since Otto Warburg's first report on the increased uptake of glucose and lactate release by cancer cells, dysregulated metabolism has been acknowledged as a hallmark of cancer that promotes proliferation and metastasis. Over the last century, studies have shown that cancer metabolism is complex, and by-products of glucose and glutamine catabolism induce a cascade of both pro- and antitumorigenic processes. Some vitamins, which have traditionally been praised for preventing and inhibiting the proliferation of cancer cells, have also been proven to cause cancer progression in a dose-dependent manner. Importantly, recent findings have shown that the nervous system is a key …

Exploring The Effects Of Varied Land Use On Elemental Concentrations Within Streams, Logan Jennings

Biological and Agricultural Engineering Undergraduate Honors Theses

It is well documented that human activity influences the chemistry of surrounding waters. As such, it is possible that there is a link between land use within a watershed and the chemical composition of the stream. The objectives of this study are to determine if varied land use does affect the concentrations of macronutrients and trace elements present in the streams of Northwest Arkansas, and if so, to determine what extent urban and agricultural development are responsible for these changes. Water samples were collected across 19 streams in the Northwest Arkansas region between January and March of 2022. Water samples …

Development Of An Equation To Correlate Brite Tank Temperature And Pressure To Beverage Carbonation Concentration, Devyn Meyer 2022 University of Arkansas, Fayetteville

Development Of An Equation To Correlate Brite Tank Temperature And Pressure To Beverage Carbonation Concentration, Devyn Meyer

Biological and Agricultural Engineering Undergraduate Honors Theses

The Carbo Rock-It™ is an invention created to carbonate beverages at craft breweries. One of the Carbo Rock-It’s main features is that it does not add a significant volume of undissolved gas bubbles to the brite tank in which the beverage is carbonated. This may allow temperature and pressure readings from the brite tank to be used to determine the concentration of dissolved carbon dioxide in the beverage continuously in real time. This eliminates the need for manual samplings, which could save breweries time and allow for more controlled, accurate carbonation. The goal of this project was to develop an …

Influence Of Mouthguard Design On Sports Performance And Injury Prevention, Mohammad Ahmed 2022 Florida Institute of Technology

Influence Of Mouthguard Design On Sports Performance And Injury Prevention, Mohammad Ahmed

Theses and Dissertations

Contact sports are associated with some invisible injuries after the incident when symptoms do not appear until weeks after. This can be dangerous as players may continue to engage in the sport and risk further injury. Athletic mouthguards are used to reduce the risk of injury, but there is insufficient data to quantify the effectiveness of mouthguard design on sports performance and injury prevention. In this work, the thickness and material selection of the mouthguard design were characterized in terms of airway resistance, electromyography muscle activation, and energy absorption. First, the effect of thickness on the condyle and temporal bone …
