Background

Intramuscular fat (IMF) and intramuscular connective tissue (IMC) are often seen in human myopathies and are central to beef quality. The mechanisms regulating their accumulation remain poorly understood. Here, we explored the possibility of using beef cattle as a novel model for mechanistic studies of intramuscular adipogenesis and fibrogenesis.

Methods

Skeletal muscle single-cell RNAseq was performed on three cattle breeds, including Wagyu (high IMF), Brahman (abundant IMC but scarce IMF), and Wagyu/Brahman cross. Sophisticated bioinformatics analyses, including clustering analysis, gene set enrichment analyses, gene regulatory network construction, RNA velocity, pseudotime analysis, and cell-cell communication analysis, were performed to elucidate …

Physiological signals are used in engineering and the sciences to determine the state of functionality of certain physiological systems such as the human endometrium. Many women use the basal body temperature method to track ovulation, or their fertile window. However, temperature tracking alone is subject to environmental factors. Non-invasive wearable devices can be tools in recording cycle-related physiological features relevant to women’s health research. This research uses the Empatica E4 wristband to track changes in the physiological features during sleep across the menstrual cycle of ovulating and non-ovulating females. Breast density screenings are an accepted means to determine a patient's predisposed risk of breast cancer development. Although the direct correlation is not fully understood, breast cancer risk increases with higher levels of mammographic breast density. Radiologists visually assess a patient's breast density using mammogram images and assign a density score based on four breast density categories outlined by the Breast Imaging and Reporting Data Systems (BI-RADS). There have been efforts to develop automated tools that assist radiologists with increasing workloads and to help reduce the intra- and inter-rater variability between radiologists. We examined a key aspect of sensorimotor skill: the capability to correct performance errors that arise mid-movement. Participants grasped the handle of a robot that imposed a nominal viscous resistance to hand movement. They watched a target move pseudo-randomly just above the horizontal plane of hand motion and initiated quick interception movements when cued. On some trials, the robot's viscosity or the target's speed changed without warning coincident with the GO cue. We fit a sum-of-Gaussians model to mechanical power measured at the handle to determine the number, magnitude, and relative timing of submovements occurring in each interception attempt. When …

Background: Coarctation of the aorta (CoA; constriction of the proximal descending thoracic aorta) is among the most common congenital cardiovascular defects. Coarctation-induced mechanical perturbations trigger a cycle of mechano-transduction events leading to irreversible precursors of hypertension including arterial thickening, stiffening, and vasoactive dysfunction in proximal conduit arteries. This study sought to identify kinetics of the stress-mediated compensatory response leading to these alterations using a preclinical rabbit model of CoA.

Limb loss poses a significant challenge to mobility and the performance of daily activities. Currently, nearly half of the amputees using powered prosthetics abandon their device due to inadequate limb coordination and movement control abilities. In the healthy system, passive and neural dynamics facilitate the synchronized activation of muscles necessary for coordinated movement. However, in the amputee, the system is disrupted. The goal of this research is to use neural and mechanical dynamics to inform the design of prosthetic devices in order to restore intuitive control and coordinated limb movement.

Cadaveric specimens are a necessary, albeit limited, resource for training medical students on basic surgical skills. The availability of surrogate 3D-printed organs would readily allow access to resources that could reduce or potentially eliminate the need for cadaveric specimens or, at a minimum, provide students the opportunity to practice with 3D-printed surrogates before transitioning to those specimens. Neutrophils are the body’s front-line defenders against foreign insult and are key players in a variety of inflammatory conditions. This body of work examines the role of neutrophils in promoting pathology in three distinct inflammatory contexts. In the pro-inflammatory state provoked by breast cancer, neutrophils decondense their nuclei and release cytotoxic web-like structures known as neutrophil extracellular traps (NETs). NETs form most commonly via histone modifications facilitated by the enzyme PAD4. Ehlers-Danlos syndrome, classical type, (cEDS) is a hereditary connective tissue disorder causing excessive elasticity and fragility of the connective tissue and problems with wound healing. Most cases of cEDS are caused by haploinsufficiency for collagen V. Collagen V regulates collagen fibril diameter. In cEDS fibroblast migration is impaired and integrin expression is altered.

The effects of collagen V on collagen gel ultrastructure and how it alters its mechanical properties were measured using scanning electron microscopy (SEM) and rheology respectively. The hearing quality provided by cochlear implants are poorly predicted by computer simulations. A realistic cochlear anatomy is crucial for the accuracy of predictions. In this study, the standard multipolar stimulation paradigms are revisited and Rattay’s Activating Function is evaluated in a finite element model of a realistic cochlear geometry that is based on µ-CT images and a commercial lead. The stimulation thresholds across the cochlear fibers were investigated for monopolar, bipolar, tripolar, and a novel (distant) bipolar electrode configuration using an active compartmental nerve model based on Schwartz-Eikhof-Frijns membrane dynamics. Ischemic tissue disease is caused by a lack of circulation / blood supply to tissue. This can be treated by introducing a number of angiogenic (pro-blood vessel forming) factors into the tissue. This work presents strategies for ischemic tissue treatment utilizing a novel proangiogenic self-assembling peptide hydrogel platform. To demonstrate the utility of this platform, its use alone as an angiogenic therapeutic (both alone as a self-assembling hydrogel and with two-component systems), and its ability to vascularize implants is explored.
