In this dissertation I have studied a novel single-molecule nanosensor that integrates a Solid-State Nanopore (SSNP), milled in silicon nitride with a Double Nanohole (DNH) nanoaperture, milled in gold. This device leverages Self-Induced Back-Action (SIBA) for optical trapping with ssNP electrical sensing and is called the SIBA-Actuated Nanopore Electrophoresis (SANE) sensor. My lab has previously shown that simultaneous acquisition of bimodal optical and direct current (DC) electrical signatures enables more detailed characterization of nanoscopic analytes compared to using electrical data alone, which are typically collected by ssNP sensors. Abstract Title: A Quality Improvement Project to Reduce CAUTIs on Inpatient Units

Objectives:

  1. Participants will be able to understand how interprofessional collaboration contributed to CAUTI reduction on inpatient acute care units.
  2. Participants will be able to understand education nurses completed to reduce CAUTI rates on inpatient acute care units.

Purpose/Aims:

To reduce CAUTI rates for adult inpatients admitted to the acute care units: Progressive Care Unit (PCU), Surgical/Orthopedics, Medical/Oncology, and the Intensive Care Unit (ICU).

In this work, we developed and investigated the cancer cell killing efficacy of different instances using real-time high-sensitivity measurements facilitated by Guided-Mode Resonance (GMR) sensors. The GMR biosensor technology detects shifts in the spectral peak caused by changes in the refractive index of the surrounding medium, offering a label-free method for monitoring cellular events.

A549 cell line wild type and HER2 knockout (lung cancer cells) were exposed to different concentrations of Triton X-100 and ADC (Herceptin with αHFc-CL-DMDM). Heart rate variability (HRV) is a naturally occurring cardiovascular phenomenon referring to the changing timing between consecutive heartbeats. The connection between HRV and overall cardiovascular health and autonomic nervous system function has been well established through prior research and well documented in existing literature. The existing studies, however, included shorter HRV subject recording session, using traditional HRV monitoring methods that do not typically combine electrocardiogram (ECG), seismocardiogram (SCG) and galvanic skin response (GSR) respiration monitoring. The inclusion of longer HRV subject recording may allow for further insight on the possible effects of given observable biological phenomenon on HRV.

Amyotrophic lateral sclerosis (ALS) is a terminal neurodegenerative disease that leads to a lack of independent mobility. One solution uses a unilateral surface EMG (sEMG) interface on the temporalis muscle to provide autonomous control of a powered wheelchair. Limbitless Journey, an EMG-controlled serious game, intends to provide users with a virtual environment to train in before use in a real-world scenario. Posterior Capsule Opacification (PCO) is a common complication post cataract surgery. It is believed that Intraocular Lens (IOL) physical and chemical properties would affect their PCO potential. The tight binding between IOL and posterior lens capsule (PLC) is believed to have a negative impact on PCO formation, as suggested in the “No space, no cell, no PCO” hypothesis. This thesis presents the development, calibration, and initial implementation of transcranial focused ultrasound (tFUS) for neuromodulation in a human study. The primary objectives were to design a system capable of delivering precise and safe ultrasound stimulation to targeted brain regions and to conduct preliminary trials to assess its effects on brain activity. A custom calibration device was developed to ensure accurate measurement of acoustic pressure, addressing challenges such as voltage alterations caused by changes in system impedance. Breast Cancer Related Lymphedema (BCRL) is a common co-morbidity in cancer survivors following neoadjuvant therapies such as chemotherapy, radiation, and/or surgery. It is brought about by the disruption in the lymphatic system (think lymph node biopsy) that leads to a buildup of lymphatic fluid in the arm. Current diagnostic strategies for this condition are merely retroactive, and fairly limited in the parameters that are examined to ensure patient well-being long term. Gait patterns are a class of biometric information pertaining to the way a person moves and poses. Gait information is unique to each person and can be used to identify and reidentify people. The increase in human activity in space has revealed several kinds of physiological challenges brought on by the absence of Earth's gravitational pull. NASA observed that during spaceflight, astronauts experience a 1-1.5% monthly loss in the mineral density of weight-bearing bones, with rehabilitation efforts on Earth often proving ineffective. This issue extends to muscles, the neuro vestibular system, the cardiovascular system, and vision. In breast x-ray imaging scattered radiation adds 50% of harmful radiation dose from anisotropic Compton scattering mechanism. We have been working with double layered inorganic salt materials that can induce Compton scattering to the incident mammographic x ray beams (in 20-30 kVp range) with adequate isotropy (angular control). Typically metal nitrates and alkali halide salt layers are shown here to cause low energy radiation interference zones with high and low photon intensities and local flux heterogeneity in terms of flux covariance. Neural electrode technology has been around for centuries since the times of Galvani. In early electrophysiology experiments metal wires were used to induce contractions in dissected animals. The metal wire electrode has since been a standard tool to both stimulate and record neural activity. In the past two decades, a new strategy for neural stimulation has been formulated based on the emergent field of optogenetics. Biomanufactured 3D-organoids show promise for modeling and treating various conditions, particularly insulin-dependent diabetes mellitus (type-1 diabetes). Pancreatic islet organoids serve as sustainable sources of insulin-producing β-cells, potentially restoring insulin regulation in patients and providing a long-term solution for diabetes. Hydrogel droplet printing is an emerging technique in regenerative medicine due to its advantageous tunability and biocompatibility; however, challenges of consistency, quality control, and scalability persist. Bacterial infections are the leading cause of mortality worldwide due to the lack of accurate diagnosis of the pathogens causing infections. Currently, microbiological and molecular techniques are used to identify pathogenic bacteria. However, these techniques have limitations as they are time-consuming, cost-inefficient, and require trained personnel. Therefore, timely and precise identification of pathogenic bacteria is crucial for public safety. Our lab previously synthesized a fluorescent sensor array (3-hydroxyflavone derivatives) encapsulated in hyaluronic acid-based nanoparticles to identify bacterial species and recognize their Gram status. Though promising, this technique was relatively time-consuming and required high sample and volumes.

A Calibrated Cuff Plethysmography device was built, tested for verification, and used to experiment on human subjects to measure the cardiovascular response of consuming a beet supplement, specifically looking at arterial compliance and pressure-area curves. Each subject was tested four times. A baseline was measured under normal conditions and after five-minute hyperemia conditions. 10 subjects were given 6 ounces of water mixed with either purple Kool-Aid (control), a SuperBeets supplement, or a SuperBeets Sport supplement and after 45 minutes, measurements were taken undergoing normal and hyperemia conditions once more.
