Transendothelial hyperpermeability caused by numerous agonists is dependent on heat shock protein 90 (Hsp90) and leads to endothelial barrier dysfunction (EBD). Inhibition of Hsp90 protects and restores transendothelial permeability. Hyperacetylation of Hsp90, as by inhibitors of histone deacetylase (HDAC), suppresses its chaperone function and mimics the effects of Hsp90 inhibitors. In this study we assessed the role of HDAC in mediating lipopolysaccharide (LPS)-induced transendothelial hyperpermeability and acute lung injury (ALI). We demonstrate that HDAC inhibition protects against LPS-mediated EBD. Inhibition of multiple HDAC by the general inhibitors panobinostat or trichostatin provided protection against LPS-induced transendothelial hyperpermeability, acetylated and suppressed Hsp90 …

Galactose is a simple sugar that at supraphysiological concentrations accelerates aging and age-related complications, which lead to impaired mitochondrial functions. MitoNEET is a small mitochondrial membrane protein with a molecular mass of 12.2 kDa that functions in diabetes, iron metabolism, regulation of oxidative phosphorylation (OXPHOS) and reactive oxygen species (ROS) homeostasis. MitoNEET knockdown increases cellular respiration rates and ROS levels similar to galactose treatment. Pioglitazone, an antidiabetic drug, binds to mitoNEET and ameliorates galactose toxicity. Cellular mitoNEET levels, exposure of cells to galactose medium, and pioglitazone treatment directly influence cellular respiration. To elucidate the role of mitoNEET in galactose induced …

Hydrogen sulfide (H2S) is receiving increasing interest, as much as nitric oxide (NO) and carbon monoxide have received previously, to understand its physiological functions as it meets all the criteria to define as a third gasotransmitter. Endogenous synthesis from L-cysteine via cystathionine-γ-lyase (CSE) and cystathionine-β-synthase (CBS) and the function of H2S as an inhibitor of smooth muscle contraction in gastrointestinal tract are known. This thesis examines situations in which computer hacking might be considered ethical. It addresses fundamental questions regarding the motivation and consequences of ethical hacking. The paper is organized into three sections. The first section discusses the history of hackers, classifies them according to their motivational background. The second part of the paper comprehensively describes the features of the ethical or “white hat” hacker group, and explores the positive and negative behaviors of ethical hackers in relation to their ethical principles. Detecting meaningful structure in neural activity and connectivity data is challenging in the presence of hidden nonlinearities, where traditional eigenvalue-based methods may be misleading. We introduce a novel approach to matrix analysis, called clique topology, that extracts features of the data invariant under nonlinear monotone transformations. These features can be used to detect both random and geometric structure, and depend only on the relative ordering of matrix entries. We then analyzed the activity of pyramidal neurons in rat hippocampus, recorded while the animal was exploring a 2D environment, and confirmed that our method is able to detect geometric organization using …

Somatostatin (SST) is a neuropeptide hormone that regulates the release of secondary hormones. Evidence suggests SST plays a neuromodulatory role due to its distribution throughout the central nervous system. Interestingly, SST has been suggested to affect the carotid body, the small peripheral chemoreceptors that regulate breathing. It has been shown that the peripheral chemoreflex sensitivity to CO2 and hypoxia is reduced by SST in humans (Pedersen et al., 1999; Pandit et al., 2014). Hypoxic events encountered during the perinatal period leading to pathologic decreases in fetal cerebral oxygen availability are detrimental resulting in neonatal hypoxic-ischemic encephalopathy (HIE). This work focused on the characterization of a rat model of neonatal HIE for use in the preclinical evaluation of infrared light therapy (IRL). Consistent with results from previous IRL efficacy studies using a rat model of adult global brain ischemia, IRL treatment attenuated cerebral injury resulting from hypoxia-ischemia in the neonatal rat. The advances made in science over the past several decades increased the number of methods available for the classification of muscle fibers, resulting in many fiber type classes and their corresponding characteristics. Prostate cancer development is uniquely dependent on the androgen receptorâ??s (ARâ??s) transcriptional regulation in response to hormone binding. Current therapies directly target AR acting as an antagonist at androgen binding sites. However, once androgen-dependence is lost, meaning the patient has progressed into a late-stage hormone resistant phenotype, all current treatments are essentially ineffective. Direct-developing frogs lack, wholly or in part, a wide range of larval features found in metamorphosing species and form adult-specific features precociously, during embryogenesis. Most information on thyroid regulation of direct development relies on hormone manipulations; the ontogeny of many thyroid axis components has not been fully described. This analysis examines differentiation of the median eminence of the hypothalamus and production of thyroid-stimulating hormone (TSH) by the pituitary of the direct-developing frog Eleutherodactylus coqui. The mammalian/mechanistic target of rapamycin complex 1 (mTORC1) signaling pathway appears to be the primary regulator of protein synthesis and growth. While resistance exercise paradigms are often divided into hypertrophy (HYP) and strength (STR) protocols, it is unknown whether these protocols differentially stimulate mTORC1 signaling. The purpose of this study was to examine mTORC1 signaling in conjunction with circulating hormone concentrations following a typical lower-body HYP and STR resistance exercise protocol. The successful regeneration of muscle tissue is dependent upon the infiltration of phagocytic CD14++CD16- monocytes that support the proliferation and differentiation of myogenic precursor cells. Spinal manipulative therapy (SMT) is a therapeutic procedure employed by various healthcare practitioners for alleviating acute and chronic musculoskeletal complaints. This form of treatment is also delivered to enhance the performance and augment the rehabilitation of athletes. However, despite research findings alleging the strength-modulating effects of SMT alongside numerous professional athletes’ positive anecdotal claims concerning its results, the physiological processes to explain its effects remain largely unexplained. To investigate further into the mechanism in which these defects are obtained, local brain insulin-like growth factor 1 (Igf1) was investigated in relation to the hypothyroid brain. Many mammalian species live in complex social groups that profoundly affect the behavior and ecology of their members. In ring-tailed lemurs (Lemur catta) these groups are characterized by female dominance, where females have priority over males for food, grooming, and space. Among females there is also a linear dominance hierarchy. L. catta have a strong matrilineal social core due to females staying within their natal group and males dispersing at sexual maturity. Vagal tone, a measure of parasympathetic activity via the vagus nerve, is known to be associated with positive emotion because it promotes social engagement and self-soothing behavior. Heart rate variability (HRV), especially high frequency oscillation, is a direct measure of vagal tone, and has been used in previous studies to test the correlation between vagal tone and positive emotion. The purpose of this study was to determine physiological differences in drought response among Garry oak (Quercus garryana Douglas ex Hook. [Fagaceae]) seedlings that grow in distinct populations in Washington state, in order to aid in restoration efforts by informing growers of the potential differences between the acorn collection sites. Acorns from six Washington populations east of the Cascades, as well as one population from Whidbey Island, were collected and grown in containers under controlled conditions. The impact of restraint stress on changes in cytokine expression and oxidative stress markers in hippocampal (Hipp) and hypothalamic (Hyp) regions of the brain, were assessed in a rat model. In addition, immune and inflammatory variables were assessed in peripheral blood. Male Wistar rats were acutely (30 min) or repeatedly (30 min/d for 14 consecutive days) exposed to a restraint stress, or maintained as non-stressed controls. In order to determine if mutations made to phospholemman (PLM) could increase PLM binding to the Na/K-ATPase (NKA) and cause positive inotropy we performed scanning mutagenesis of the transmembrane domain of PLM and measured FRET between each mutant and NKA. We observed increased binding to NKA for several PLM mutants compared to WT, including L27A, L30A, and I32A. In isolated cardiomyocytes, overexpression of WT PLM increased the amplitude of the Ca2+ transient compared to GFP control. Ca2+ transient amplitude was further increased by L30A PLM overexpression. L30A mutation also delayed Ca2+ extrusion and increased the duration of the cardiomyocyte contraction. …

As the central pacemaker, the suprachiasmatic nucleus (SCN) has long been considered the primary regulator of blood pressure circadian rhythm; however, this dogma has been challenged by the discovery that each of the clock genes present in the SCN is also expressed and functions in peripheral tissues. The involvement and contribution of these peripheral clock genes in the circadian rhythm of blood pressure remains uncertain.
