Mutations in mitochondrial aminoacyl-tRNA synthetases are an increasingly recognized cause of human diseases, often arising in individuals with compound heterozygous mutations and presenting with system-specific phenotypes, frequently neurologic. FARS2 encodes mitochondrial phenylalanyl transfer ribonucleic acid (RNA) synthetase (mtPheRS), perturbations of which have been reported in 6 cases of an infantile, lethal disease with refractory epilepsy and progressive myoclonus. Here the authors report the case of juvenile onset refractory epilepsy and progressive myoclonus with compound heterozygous FARS2 mutations. The authors describe the clinical course over 6 years of care at their institution and diagnostic studies including electroencephalogram (EEG), brain magnetic resonance …

While many studies have detailed the complex intricacies of the endothermic immune system, relatively little is known about the immune system of ectotherms--specifically, reptiles. In an attempt to gain more knowledge about the factors affecting reptilian immune function, ornate box turtles (Terrapene ornata) were subjected to chronic stress in the form of high ambient temperatures in a semi-natural environment. It was hypothesized that chronic stress would lead to elevated levels of corticosterone in the blood, which would, in turn, suppress immune activity. It was found that body temperature and body mass in particular were significantly affected by chronic heat stress. …

Cardiovascular Effects Evoked By Airway Nociceptive Reflexes In Healthy And Cardiovascular Diseased Rats, Justin Shane Hooper
USF Tampa Graduate Theses and Dissertations
Acute inhalation of airborne pollutants alters cardiovascular function and has been shown to have its greatest affects on individuals with pre-existing cardiovascular disease. Evidence suggests that pollutant-induced activation of airway sensory nerves via the gating of ion channels is critical to these systemic responses. Here, we have investigated the cardiovascular responses evoked by inhalation of AITC (TRPA1 agonist) and capsaicin (TRPV1 agonist) in healthy Sprague Dawley (SD) and Wistar Kyoto (WKY) rats, and cardiovascular diseased Spontaneously Hypertensive (SH) rats. Inhalation of the agonists by healthy SD and WKY rats caused significant bradycardia, atrio-ventricular (AV) block and prolonged PR-Intervals. Inhalation of …

The Helicobacter pylori energy sensor TlpD determines tactic behaviour under low energy conditions and is important in vivo. We explored protein-protein interactions of TlpD and their impact on TlpD localisation and function. Pull-down of tagged TlpD identified protein interaction partners of TlpD, which included the chemotaxis histidine kinase CheAY2, the central metabolic enzyme aconitase (AcnB) and the detoxifying enzyme catalase (KatA). We confirmed that KatA and AcnB physically interact with TlpD. While the TlpD-dependent behavioural response appeared not influenced in the interactor mutants katA and acnB in steady-state behavioural assays, acetone carboxylase subunit (acxC) mutant behaviour was altered. TlpD was …

Poor Quality Of Instruction Leads To Poor Motor Performance Regardless Of Internal Or External Focus Of Attention, M. Jay Polsgrove, Tom E. Parry, Nathan T. Brown
International Journal of Exercise Science
International Journal of Exercise Science 9(2): 214-222, 2016. The view that external focus of attention provides beneficial performance outcomes when compared to an internal focus of attention has been consistently supported in the movement performance literature. While type of focus has been well investigated, the current study examined the influence of quality of instruction as a variation of the type of focus. Specifically, the purpose of the study investigated how performance-enhancing instructions would differ from performance-neutral instructions on an agility performance. An agility L-run was used to measure performance in the four counterbalanced conditions: Internal-Performance Neutral (INT-PN), Internal-Performance Enhancing (INT-PE), …

Geometric Entropy For Lead Vs Top-Rope Rock Climbing, Phillip B. Watts, Scott N. Drum, Matthew A. Kilgas, Kevin C. Phillips
International Journal of Exercise Science
International Journal of Exercise Science 9(2): 168-174, 2016. The complexity of movement of a rock climber’s center of mass during an ascent has been described as geometric entropy (GE). It has been proposed that lower geometric entropy could represent more fluid and economical movement during climbing. The purpose of the present study was to measure GE during rock climbing ascents under a lead condition (LD), where the climber connects a safety rope to several intermediate anchors during the ascent and under a top-rope condition (TR), where the safety rope is always anchored above the climber. Six experienced rock climbers volunteered …

Acinar cells of the exocrine pancreas are dedicated to synthesize, package and secrete immense quantities of pro-digestive enzymes to maintain proper metabolic homeostasis for the organism. Dysregulation of enzyme secretion in acinar cells can give rise to exocrine diseases including acute pancreatitis (AP), a disease that targets acinar cells, leading to acinar-ductal metaplasia (ADM), inflammation and fibrosis—events that can transition into the earliest stages of pancreatic ductal adenocarcinoma (PDAC). The focus of this thesis is to interrogate transcriptional regulatory networks that are susceptible to AP and the role that these networks play in acinar cell and exocrine pancreas responses. The …

Metformin is the main drug of choice for treating type 2 diabetes, yet the therapeutic regimens and side effects of the compound are all undesirable and can lead to reduced compliance. The aim of this study was to elucidate the mechanism of action of two novel compounds which improved glucose handling and weight gain in mice on a high-fat diet. Wildtype C57Bl/6 male mice were fed on a high-fat diet and treated with novel, anti-diabetic compounds. Both compounds restored the glucose handling ability of these mice. At a cellular level, these compounds achieve this by inhibiting complex I activity in …

An Evaluation Of The Influences Of Extra-Hippocampal Processes On Pattern Separation, Malia L. Anderson
Theses and Dissertations
Long-term declarative memory depends on pattern separation, which reduces the degree of overlap between similar representations, to maintain memory specificity, and on pattern completion, which occurs when a degraded cue is used to retrieve a previously stored memory. Previous studies aimed at evaluating the underlying neuronal substrates of these computational processes have used a mnemonic discrimination paradigm and fMRI to focus on the hippocampus, to the exclusion of cortical processing. We aim to investigate the influences extra-hippocampal processes have on pattern separation in the following two studies. Study 1. Computational models of pattern completion suggest it occurs cortically and results …
