Investigating Visinin-Like Protein 1 And Ubiquitinated Visinin-Like Protein 1 As Mild Traumatic Brain Injury Biomarkers, Yueming Wu

Theses and Dissertations--Chemistry

Traumatic brain injury (TBI) continues to be a significant cause of morbidity and mortality worldwide. Despite significant progress in understanding the complex pathophysiology of TBI, the underlying mechanisms remain poorly understood. The primary brain damage is acute and irreversible. However, secondary brain injuries often develop gradually over months to years, creating an opportunity for critical therapeutic interventions. In the past decade, research on TBI biomarkers has seen significant progress. This progress has been driven by the diverse nature of TBI pathologies and the challenges they present for evaluation, management, and prognosis. TBI biomarker proteins resulting from axonal, neuronal, or glial …

Plant Sentience Revisited: Sifting Through The Thicket Of Perspectives, Paco Calvo, Miguel Segundo-Ortin 2023 University of Murcia

Plant Sentience Revisited: Sifting Through The Thicket Of Perspectives, Paco Calvo, Miguel Segundo-Ortin

Animal Sentience

In our target article (Segundo-Ortin & Calvo 2023), we proposed the intriguing possibility of plant sentience, drawing parallels with non-human animal studies. This response aims to sift through the rich thicket of perspectives offered by our commentators. To do so, we assess the risks of employing double standards, as well as the tendencies of anthropomorphizing and zoomorphizing in plant studies. We also emphasize the need for clarity in linguistic and conceptual terms, examine the neurophysiological evidence for plant sentience, and discuss the ethical implications of such recognition.

Scale-Free Behavioral Dynamics Directly Linked With Scale-Free Cortical Dynamics, Sabrina A. Jones, Jacob H. Barfield, V. Kindler Norman, Woodrow L. Shew 2023 University of Arkansas, Fayetteville

Scale-Free Behavioral Dynamics Directly Linked With Scale-Free Cortical Dynamics, Sabrina A. Jones, Jacob H. Barfield, V. Kindler Norman, Woodrow L. Shew

Physics Faculty Publications and Presentations

Naturally occurring body movements and collective neural activity both exhibit complex dynamics, often with scale-free, fractal spatiotemporal structure. Scale-free dynamics of both brain and behavior are important because each is associated with functional benefits to the organism. Despite their similarities, scale-free brain activity and scale-free behavior have been studied separately, without a unified explanation. Here, we show that scale-free dynamics of mouse behavior and neurons in the visual cortex are strongly related. Surprisingly, the scale-free neural activity is limited to specific subsets of neurons, and these scale-free subsets exhibit stochastic winner-take-all competition with other neural subsets. This observation is inconsistent …

Selective Gsk3b Overexpression In Medial Prefrontal Cortex Camkiia Cells Produces Sex-Specific Changes In Ethanol Self-Administration, Anxiety-Like Behavior, And Working Memory, Sam Gottlieb, Zachary Tatom, Michael F. Miles 2023 Virginia Commonwealth University

Selective Gsk3b Overexpression In Medial Prefrontal Cortex Camkiia Cells Produces Sex-Specific Changes In Ethanol Self-Administration, Anxiety-Like Behavior, And Working Memory, Sam Gottlieb, Zachary Tatom, Michael F. Miles

Graduate Research Posters

Background: Glycogen synthase kinase 3 beta (Gsk3b) is central in a gene network regulated by ethanol in mouse medial prefrontal cortex (mPFC). GSK3B abundance/activity modulations regulate ethanol consumption, suggesting GSK3B could be a target in treating alcohol use disorder. However, the critical cell type in this ethanol response is not yet known, nor have sex differences in GSK3B modulations been fully investigated. Here we report results of GSK3B overexpression selectively in CamKIIa+ mPFC cells on multiple behaviors.

Methods: Mice underwent stereotaxic injections to overexpress GSK3B in mPFC CamKIIa+ cells or control virus (n=4-6/sex/virus). We assayed working …

Alzheimer’S Disease Genetics And Short-Chain Fatty Acid Treatment In Studies Of The Murine Gut Microbiome, Diana Zajac

Theses and Dissertations--Physiology

Elucidating the relationship of the gut microbiome in Alzheimer's Disease (AD) risk and pathogenesis is an area of intense interest. Since 60 to 80% of AD risk is related to genetics and APOE alleles represent the most impactful genetic risk factors for AD, their mechanism(s) of action are under intense scrutiny.

First, I conducted a study on APOE targeted replacement mice to investigate the impact of APOE alleles on the murine gut microbiome. The relative abundance of bacteria from the family Ruminococacceae and related genera increased with APOE2 status. The relative abundance of the class Erysipelotrichia increased with APOE4 status, …

Artificial Light at Night Disrupts Pain Behavior and Cerebrovascular Structure in Mice

Jacob R. Bumgarner

Circadian rhythms are intrinsic biological processes that fluctuate in function with a period of approximately 24 hours. These rhythms are precisely synchronized to the 24- hour day of the Earth by external rhythmic signaling cues. Solar light-dark cycles are the most potent environmental signaling cue for terrestrial organisms to align internal rhythms with the external day. Proper alignment and synchrony of internal circadian rhythms with external environmental rhythms are essential for health and optimal biological function.

The modern human environment on Earth is no longer …

The Effects Of Age On Working Memory Capacity And Neural Function, Jasmin P. Joshi 2023 Claremont McKenna College

The Effects Of Age On Working Memory Capacity And Neural Function, Jasmin P. Joshi

CMC Senior Theses

Do age-related differences in working memory capacities predict specific neural responses? Theories of working memory (WM) suggest that there are different types of WM processes that are associated with specific neural networks and WM declines with age. Event-related potentials (ERPs) evoked by certain tasks are also associated with specific cognitive functions. This study investigates whether specific types of working memory capacities are related to corresponding neural indices of attention and context updating (P3; visual oddball task) or semantic (N400; word-pair paradigm task) processes. In addition, it examines whether age affects this relationship. Younger adults (n=30; ages 18-23) and older adults …

To Laugh Or Not To Laugh: That Is The Question Of Humor Techniques And Sex Differences, Yu-Chen Chan 2023 Tsing Hua University, Hsinchu

To Laugh Or Not To Laugh: That Is The Question Of Humor Techniques And Sex Differences, Yu-Chen Chan

Journal of Educational Technology Development and Exchange (JETDE)

A humor structure comprises two essential stages: the setup and the punch line. The punch line stage is to provide the incongruity resolution that creates amusement in humor. The current article aimed to look into how humor is amusing and how it differs between the sexes. Functional and effective connectivity analyses in cognitive and affective neuroscience have facilitated the implications of humor comprehension, appreciation, and laughter responses. The processing of incongruity-resolution humor revealed effective connectivity from the amygdala to the precuneus (amygdala → precuneus). Conversely, the processing of nonsense humor demonstrated effective connectivity from the amygdala to the inferior frontal …

Mediators Between Job Stress And Health Quality Of Life Among Adults With Irritable Bowel Syndrome, Aida M. Benitez-Rexach

Walden Dissertations and Doctoral Studies

Irritable bowel syndrome (IBS) is a functional gastrointestinal affliction that affects millions around the globe, impacting health-related quality of life (HRQoL). The biopsychosocial model, Folkman and Lazarus’ transactional model of stress, Leventhal’s common sense model of self-regulation health and illness, and Bronfenbrenner’s human ecology theory are relevant to conceptualize interactions among environmental factors, biopsychosocial responses, and health-related quality of life (HRQoL). This quantitative, cross-sectional, complex correlational study sought to provide more understanding of how and whether IBS symptoms of pain, cognitive functioning, and interpersonal functioning serve as mediators between work stress and HRQoL. A total of 133 English-speaking volunteers from …

Sense And Sensitivity: Spatial Structure Of Conspecific Signals During Social Interaction, Keshav Ramachandra 2023 West Virginia University

Sense And Sensitivity: Spatial Structure Of Conspecific Signals During Social Interaction, Keshav Ramachandra

Graduate Theses, Dissertations, and Problem Reports (ETD)

Organisms rely on sensory systems to gather information about their environment. Localizing the source of a signal is key in guiding the behavior of the animal successfully. Localization mechanisms must cope with the challenges of representing the spatial information of weak, noisy signals. In this dissertation, I investigate the spatial dynamics of natural stimuli and explore how the electrosensory system of weakly electric fish encodes these realistic spatial signals. To do so In Chapter 2, I develop a model that examines the strength of the signal as it reaches the sensory array and simulates the responses of the receptors. The …

Spatial Processing Of Conspecific Signals In Weakly Electric Fish: From Sensory Image To Neural Population Coding, Oak Everette Milam

Graduate Theses, Dissertations, and Problem Reports (ETD)

In this dissertation, I examine how an animal’s nervous system encodes spatially realistic conspecific signals in their environment and how the encoding mechanisms support behavioral sensitivity. I begin by modeling changes in the electrosensory signals exchanged by weakly electric fish in a social context. During this behavior, I estimate how the spatial structure of conspecific stimuli influences sensory responses at the electroreceptive periphery. I then quantify how space is represented in the hindbrain, specifically in the primary sensory area called the electrosensory lateral line lobe. I show that behavioral sensitivity is influenced by the heterogeneous properties of the pyramidal cell …

Sex Differences In Endothelial Function, Metabolism, And Sleep: Responses To Disrupted Circadian Rhythms, Olga Hecmarie Meléndez-Fernández 2023 West Virginia University

Sex Differences In Endothelial Function, Metabolism, And Sleep: Responses To Disrupted Circadian Rhythms, Olga Hecmarie Meléndez-Fernández

Graduate Theses, Dissertations, and Problem Reports (ETD)

Circadian rhythms are endogenous cycles that recur approximately every 24 h, and manifest in virtually every physiological and behavioral process among the vast majority of organisms. These rhythms, sustained by the central circadian clock, the suprachiasmatic nucleus (SCN) of the hypothalamus, allow for optimal timing of biological processes that ensure organisms’ homeostasis, and are entrained to precisely 24 h by daily exposure to light. Disruption of the molecular mechanisms that drive these rhythms have negative consequences on physiology and behavior that may impair survival. The molecular mechanisms that underlie these effects are widely studied and linked to multiple diseases and …

Investigation Of Regulator Of G Protein Signaling-12 (Rgs12) As A Modulator Of Monoamine Transporters And Behavioral Responses To Cocaine, Allison Noel White

Graduate Theses, Dissertations, and Problem Reports (ETD)

Stimulant use disorder is a psychiatric illness that is characterized by repeated use of stimulant drugs despite harm to the user. There are currently no Federal Drug Administration (FDA)- approved medications for stimulant use disorder, and the number of stimulant use-associated deaths have increased over the past decade. Given the prevalence of and lack of treatments for stimulant use disorder, there is an urgent need to identify new drug targets and treatments for this illness. Our group is investigating Regulator of G protein Signaling-12 (RGS12) as one such potential drug target for stimulant use disorder. Mice lacking RGS12 are insensitive …

Cue Reactivity In Electronic Cigarette Users With Sign-Tracking Or Goal-Tracking Behaviors, Polina Krom 2023 West Virginia University

Cue Reactivity In Electronic Cigarette Users With Sign-Tracking Or Goal-Tracking Behaviors, Polina Krom

Graduate Theses, Dissertations, and Problem Reports (ETD)

Polina Krom

Cue reactivity is an important predictor of addiction course and relapse. However, cue reactivity is only observed after an addiction develops. As such, it is unclear to what degree cue reactivity represents a state stemming from the addiction process versus a trait-like propensity towards developing cue-reward associations. Work in animal models has pointed to important individual differences in trait-like inclination to attribute incentive salience to reward-predictive cues that is associated with addiction-relevant behavioral and neurobiological features. These individual differences manifest as sign-tracking (ST) and goal-tracking (GT) behaviors during Pavlovian conditioning. Little research has attempted to translate ST and …

Effects Of Development On Hpa Function Following Pubertal Stress, Brittany D. Elliott 2023 West Virginia University

Effects Of Development On Hpa Function Following Pubertal Stress, Brittany D. Elliott

Graduate Theses, Dissertations, and Problem Reports (ETD)

For women, two of the greatest risk factors for affective disorders are adversity experienced during puberty and later becoming pregnant. We have created a translationally relevant mouse model where we address these complex risk factors. Previously, we discovered that pregnant mice (dams) that experienced chronic variable stress (CVS) during puberty display a blunted hypothalamic-pituitary-adrenal (HPA) response when exposed to an acute stressor. Interestingly, this alteration only first becomes apparent during pregnancy, which is a sensitive period for these effects due to normative neuroendocrine changes. Further investigation of the mechanisms underlying this dysfunction revealed altered gene expression in the paraventricular nucleus …

Multimodal Neuron Classification Based On Morphology And Electrophysiology, Aqib Ahmad 2023 West Virginia University

Multimodal Neuron Classification Based On Morphology And Electrophysiology, Aqib Ahmad

Graduate Theses, Dissertations, and Problem Reports (ETD)

Categorizing neurons into different types to understand neural circuits and ultimately brain function is a major challenge in neuroscience. While electrical properties are critical in defining a neuron, its morphology is equally important. Advancements in single-cell analysis methods have allowed neuroscientists to simultaneously capture multiple data modalities from a neuron. We propose a method to classify neurons using both morphological structure and electrophysiology. Current approaches are based on a limited analysis of morphological features. We propose to use a new graph neural network to learn representations that more comprehensively account for the complexity of the shape of neuronal structures. In …

Editorial: Microglia In Neuroinflammation, Pinar Ayata, Ido Amit, Carla M. Cuda

Advanced Science Research Center

Microglia are a resident innate immune cell population of the central nervous system (CNS) derived from yolk sac erythro-myeloid progenitors that migrate to the developing brain prior to the formation of the blood-brain barrier. This critical cell population has gained considerable traction in the literature as it is considered a protective barrier from CNS damage and, yet, can also serve as a primary mediator of neuroinflammation. Under normal physiological conditions, microglia perform homeostatic functions, such as parenchymal surveillance, neurotrophic support, pathogen or debris removal, and maintenance of synaptic homeostasis and neuronal plasticity.

Abnormal glycosylation can impact cardiac proteins, affecting signaling, contraction, and metabolism, which leads to compromised cardiac function. O-GlcNAcylation, the only form of intracellular glycosylation, involves adding a single GlcNAc molecule to proteins via enzymes: O-GlcNAc Transferase (OGT) for addition and O-GlcNAcase (OGA) for removal. The Ednie/Bennett lab has previously demonstrated that in both chronic (OGT KO) and acute (OGT inhibition) models of reduced cardiomyocyte O-GlcNAcylation, a disruption of Ca2+ homeostasis occurs. This study delved into the cardiac voltage-gated sodium channel (NaV 1.5) in these models. NaV 1.5 expression was analyzed through western blotting and immunocytochemistry, while its functional activity was …
