Nicotiana glauca is a medicinal plant used by traditional healers as antibacterial, antifungal, antiviral and anti-inflammatory medicines. The leaves of N. glauca are very rich in indolic alkaloids which give it a larvicidal power which allows us to use it as a bio-insecticide.In the present study we were perfomed in the direct (mortality) and indirect (food attractiveness and pupation) toxic effects of the aqueous extract of N. glauca on the mortality and feeding behavior of the fruit fly Drosophila melanogaster. The treatment was administered to 2nd instar larvae (L2) and the mortality rate was monitored for 15 days. Similarly we …

The Role Of Dha In The Impact Of Trem2 On Microglia Activation And Alzheimer’S Disease, Michael A. Palmieri Iii
Seton Hall University Dissertations and Theses (ETDs)
Alzheimer’s disease (AD) is a neurological disease that is associated with microglia activation. An important receptor involved in microglia activation is Triggering receptor expressed on myeloid cells 2 (TREM2). Recent studies suggest that docosahexaenoic acid (DHA) could reduce the neuroinflammation that is associated with microglia activation. We hypothesized that when DHA concentration was increased, TREM2 expression would decrease, microglia activity would be inhibited, and a resulting decrease in neuroinflammation would be observed. We examined peer-reviewed journal articles from 2017-2022 that investigated the relationship between TREM2 activation and severity of AD symptoms, the protective properties of DHA against AD, and the …

The Role Of Parkin In Mitochondrial Dna, Eliezer Lichter
Theses & Dissertations
Mitochondria are at the center of biological phenomena such as aging and diseases, especially neurodegenerative diseases. While the discovery of mitochondria only came approximately 200 years after the cell was discovered, a lot of progress has been made since. The mitochondrial genome encodes proteins vital for mitochondrial function. These proteins are only a subset of the proteins present in mitochondria; the rest are nuclear encoded. The nucleus also encodes cytosolic proteins vital for mitochondrial maintenance. One of these is Parkin, an E3 ubiquitin ligase that ubiquitinates mitochondrial proteins as mitochondria become depolarized. Its activity has been shown to be involved …

Metoprolol Disrupts Sterol Biosynthesis Through Inhibition Of 7-Dehydrocholesterol Reductase (Dhcr7), Luke B. Allen
Theses & Dissertations
Cholesterol is essential for life. It is particularly important in the brain as it relies on de novo synthesis of cholesterol following the formation of the blood brain barrier (BBB). As such, disrupting sterol biosynthesis during neurodevelopment can have devastating outcomes. The most common post-lanosterol sterol biosynthesis disorder, Smith-Lemli-Opitz Syndrome, arises from a faulty DHCR7 enzyme. DHCR7 has also been shown to be inhibited by several psychotropic medications. Here we assess six beta-blockers and their effects on sterol biosynthesis in vitro. Two beta-blockers, metoprolol and nebivolol strongly inhibit DHCR7 in four separate in vitro models of both mouse and …

Making Emotions Trustworthy, Xiaoyu Ke
Arts & Sciences Graduate Student Theses and Dissertations
I investigate ways by which agents can make their emotions more trustworthy from a perspective that’s informed by recent psychological research on the nature of emotion regulation. This dissertation is a collection of three articles, each addressing one type of cases of untrustworthy emotions in a particular context. Chapter one (Developing Appropriate Emotions) concerns emotions that are untrustworthy because they are irrelevant to the moral or epistemic judgements at hand. By situating the problem in the context of a situationist challenge to neo-Aristotelian virtue ethics, I argue that we can address this problem by developing ameliorative abilities such as emotion …

The early stages of the COVID-19 pandemic and associated stay-at-home orders resulted in a stark reduction in daily social interactions for children and adolescents. Given that peer relationships are especially important during this developmental stage, it is crucial to understand the impact of the COVID-19 pandemic on social behavior and risk for psychopathology in children and adolescents. In a longitudinal sample (N=224) of children (7-10y) and adolescents (13-15y) assessed at three strategic time points (before the pandemic, during the initial stay-at-home order period, and six months later after the initial stay-at-home order period was lifted), we examine whether certain social …

Loss of function lesions in Ubiquitin-specific protease (USP) 7 are associated with intellectual disability, speech delay, autism spectrum disorder, and seizures. While the deubiquitinating activity of USP7 has been characterized in cancer, its role in neurological conditions is not understood. Here, we show that USP7 can regulate the development of the brain by localizing to synaptosomes in the hippocampus and stabilizing neural-specific substrates.

Misophonia sensitivity as measured by the A-MISO-S predicts emotional responses to misophonia trigger videos, but musical sophistication (Gold MSI scores) did not. A measure of real-time responses to videos can capture a meaningful aspect of misophonic experience in the general population. Future research should investigate whether more direct measures of musicality such as perceptual tasks will show a relationship between musicality and misophonic reactions.

Unraveling The Anti-Inflammatory Mechanisms And Efficacy Of Cannabidiol On The Progression Of A Murine Model Of Multiple Sclerosis From The Innate To The Adaptive Immune System To Clinical Symptoms, Christa Marie Frodella
Theses and Dissertations
Cannabidiol (CBD), a non-psychotropic phytocannabinoid with structural similarity to Δ 9 -tetrahydrocannabinol (THC), is currently being investigated as a therapeutic for its immunosuppressive effects. One disease for which CBD is extensively researched is multiple sclerosis (MS), a demyelinating, autoimmune disorder, and its murine model counterpart, experimental autoimmune encephalomyelitis (EAE). The focus of this dissertation aimed to analyze the transcriptomic brain pathways in EAE and its comparison to MS in addition to CBD’s immunosuppressive mechanisms in the innate and adaptive immune systems. Evidence presented here showed that transcriptomic signaling pathways in the EAE brain of mice with clinical symptoms were similar …

Investigating The Impact Of Dividing Attention On Auditory And Visual Object Memory, Sharica Lee, Alexa Salomon, Laura L.S. Werner, Kevin D. Mohawk, Maggie Mcmullin
Undergraduate Research Symposium Posters
Recently, we found that dividing attention reduced recollection and familiarity for visual objects, but a different pattern emerged for auditory object memory: auditory object recollection was not affected by dividing attention. This could be attributable to differing levels of baseline performance with visual memory far exceeding auditory memory. Thus, we attempted to equate baseline performance in both modalities in order to adequately investigate the previous findings.

Fall 2022 Virtual OTD Capstone Symposium
Children with chronic neurological conditions and their families experience various challenges that impact their quality of life and occupational participation as they reintegrate back into their communities (Dumas & Grajo, 2021; Taib et al., 2021). However, rehabilitation for children with chronic neurological conditions focuses on addressing functional impairments and self-care activities rather than transitioning home (Diener et al., 2021). Therefore, children and their families are in need of resources and interventions to address their occupational impact as they return to their occupations (Diener et al., 2021). This capstone research study explored the lived experiences of children with chronic neurological conditions …

Accurate control of nervous system development is critical for normal brain patterning, and defects in this process can lead to neurological disorders such as schizophrenia and Autism Spectrum Disorder. The transcription factor neurogenin is necessary for the development of neural subtypes and is deeply conserved across species. However, the transcriptional targets of neurogenin are poorly understood, creating an imperative for further study. We have used the nematode Caenorhabditis elegans as a model to better understand ngn-1/neurogenin function. Previous work from our lab revealed that ngn-1 plays a role in nerve ring architecture, and neural cell fate specification. In addition, …

Cervical Muscle Strength And Kinematics During An Unanticipated Perturbation In University Aged Male And Female Rugby Athletes, Abigail Ruppert, Lauren J. Lattimer
Journal for Sports Neuroscience
Introduction: Female athletes participating in contact sports demonstrate a concussion injury risk factor that is two times greater than their age-matched male counterparts. Recent literature has outlined the importance of neck muscle strength and neuromuscular characteristics in the mitigation of excessive head kinematics. Rugby is a contact sport in which tackles comprise the mechanism for concussion, which is caused by inertial impacts. Females exhibit higher measurements of acceleration and investigating potential gender differences in dynamic stabilization of the head is warranted in rugby. Methods: Twenty-three (15 female, 8 male) university-aged rugby athletes participated in kinematic analysis; and a series of …

Identification Of Clinical And Behavioral Outcomes Predictive Of Ftld-Tdp Pathology, Nevin Crow, Virginia A. Berry, Eric Febles
Science University Research Symposium (SURS)
Frontotemporal Lobar Dementia (FTLD) is a neurogenerative disease often misdiagnosed as Alzheimer’s Disease (AD), resulting in poor treatment outcomes (Rascovsky et al., 2011). Multi-factorial approaches are increasingly being applied to yield more accurate and earlier diagnoses. These standard clinical outcomes include MRI imaging, biomarkers, and assessments of cognitive ability. While each of these outcomes are partially predictive of FTLD diagnosis, none alone carry enough power to differentiate FTLD patients from other dementias, including AD. One notable variable is TDP-43, a DNA-binding protein involved in regulating the FTLD risk gene UNC13A, suggested as one of the more effective biomarkers for early …

In this dissertation, I aim to advance our understanding of human agency and how it is computed. First, I probe the limits of what behavior we can and cannot produce agentically. Secondly, I explore microdosing with psychedelic drugs as a method of perturbing and studying changes to the sense of agency. Thirdly, I use transcranial magnetic stimulation to induce body movements and investigate how the sense of agency is affected. Finally, I summarise my findings and synthesize insights to further our understanding human agency.

Background: This study examined whether COVID-19-related maternal mental health changes contributed to changes in adolescent psychopathology. Methods: A community sample of 226 adolescents (12 years old before COVID-19) and their mothers were asked to complete COVID-19 surveys early in the pandemic (April–May 2020, adolescents 14 years) and approximately 6 months later (November 2020–January 2021). Surveys assessed pandemic-related stressors (health, financial, social, school, environment) and mental health. Results: Lower pre-pandemic family income-to-needs ratio was associated with higher pre-pandemic maternal mental health symptoms (anxiety, depression) and adolescent internalizing and externalizing problems, and with experiencing more pandemic-related stressors. Pandemic-related stressors predicted increases in …

Causal Inference In Psychology And Neuroscience: From Association To Causation, Dehua Liang
Computational and Data Sciences (PhD) Dissertations
In psychology and neuroscience, inferring causality in non-experimental studies is almost taboo, because data in these studies, e.g., survey data and resting-state neuroimaging data, are often contaminated by unmeasured confounders. Psychologists and neuroscientists are often cautious about their results, and reluctant to make false claims about causality in non-experimental studies. Therefore, they adopt less stringent statistical analysis techniques that can only infer associational relations. However, the ambiguity about causality in traditional statistical analysis creates much confusion in interpreting analytical results - some studies make implicit causal claims about their results using words such as “impacts”, “lead to” and “affects”. This …

Microglia are the primary immune cells of the central nervous system and contribute substantially to both development and maintenance of the brain. Negative alterations to microglia often result in detrimental outcomes and significant effort is being made to understand these powerful cells. The goal of this research is to characterize microglia in the developing and diseased murine dorsal telencephalon. In vivo cell labeling and confocal microscopy identified a transient cell type, amoeboid microglia cells (AMCs), that appear to contribute to the development of microglia. Moreover, the spatial distribution and morphological change of microglia was thoroughly characterized during postnatal development. The …
