The purpose of this project is to find the effects of staining toxic cyanobacteria with SYTOXGreen after lysis by freeze-thaw and lysis by ethanol. This project will focus on two strains of toxic cyanobacteria known as Aphanizomenon - A. flos-aquae and Planktothrix - P. agardhii. The live and dead cells are observed under blue and red fluorescent light to show the effectiveness of each lysis. It is important that this topic is researched because it is significantly affecting our waterways and sometimes drinking water. Previous research investigating the speleogenesis of iron-ore caves (IOC) in the Quadrilátero Ferrífero, or “Iron Quadrangle,” of Brazil suggests that microbial iron reduction and subsequent dissolution of the surrounding Fe(III) rich rocks is responsible for cave formation. A soft intra-wall substance (sub muros) containing iron-reducing bacteria (FeRB) was discovered underneath the durable crusts of cave walls. The goal of the study was to determine if reduction rates were comparable between sub muros samples, while additionally observing how different electron donors affect microbial iron reduction. The acquisition of microbial partners is a strategy used by a diverse group of arthropods to overcome ecological barriers that might normally make certain niches uninhabitable. The unique phylogenetic opportunities attainable from the natural experiment of the Sodalis-allied clade allow for better understanding of how molecular structures evolve through time. Here, we focus on the evolution of the flagellar synthesis pathway, due to its complexity and ability to diverge in response to ecological pressures. Ergot alkaloids are fungal tryptophan derived toxins which affect mammalian circulation and neurotransmission. These compounds are biosynthesized by a conserved genetic pathway, known as the ergot alkaloid synthesis (EAS) pathway by fungi belonging to the ascomycete families Trichocomaceae and Clavicipitaceae. Several Ipomoea species and related plants in the morning glory family harbor vertically transmitted symbiotic fungi in the genus Periglandula, also members of Clavicipitaceae, that produce ergot alkaloids. Metabolomic analysis of seeds identified a previously uncharacterized glycoside form of the pharmaceutically important ergot alkaloid, ergonovine. Microbial eukaryotes are key components of the ocean plankton. Yet, our understanding of their community composition and activity in different water layers of the ocean is limited, particularly for picoeukaryotes (0.2–3 µm cell size). Here, we examined the picoeukaryotic communities inhabiting different vertical zones of the tropical and subtropical global ocean: surface, deep chlorophyll maximum, mesopelagic (including the deep scattering layer and oxygen minimum zones), and bathypelagic. Quorum quenching (QQ) has been suggested as an alternative to antibiotic treatments for pathogenic infections as it does not affect the viability of the pathogen and imposes minimum selection pressure promoting drug resistance. During an ecological survey for quorum quenching (QQ) bacteria from soil collected in Lagoon Island (Ryder Bay, Adelaide Island, Antarctica), a bacterial strain was isolated and identified as Planococcus species, L10.15T, based on 16S rRNA gene analysis. The genomic, phylogenetic, chemotaxonomic and phenotypic data obtained showed that L10.15T represents a novel species for which the name Planococcus versutus sp. nov. is proposed. The McMurdo Dry Valleys (MDV) of Antarctica are on the threshold of widespread landscape scale change due to increasing temperature and solar radiation and altered hydrology: buried ice is melting, the soil active layer is thickening, thermokarst features are developing along streams, water tracks are expanding, and lake levels are rising. The current work extends the phenotypic characterization of a bacterial culture collection obtained from white, yellow, grey or pink microbial cave wall colonies that are common in the caves of Slovenian Karst. We have determined antibiotic resistance to 22 natural and synthetic antibiotics in 69 isolates from the microbial mats. Thirty-eight isolates (52%) were resistant to 1-5 antibiotics; another 27 isolates (37%) were resistant to 6-10 antibiotics; and 7 isolates (0.1%) were resistant to 11-17 antibiotics. This study was conducted to evaluate the interaction between disease incidence of both Fusarium head blight (FHB) and crown rot (FCR) and physiochemical parameters for soil samples and environmental factors in 14 selected wheat fields in the North of Basra province. The results showed that both diseases were occurred in all surveyed fields. The level of FCR incidence was higher than FHB. The incidence of both diseases was increased with favourable weather conditions (high humidity and temperature ˃15 °C), reaching the highest levels in crucial periods before and during the ripening stage. Legionella spp. are gram-negative bacteria that are found in a wide range of freshwater environments such as lakes, streams, soil, and manmade water systems. The Legionella genera contains 58 species of bacteria and of them approximately half have been linked to human disease. Infection due to Legionella spp. is known as Legionellosis, which consists of Legionnaires’ disease and Pontiac fever. Legionnaires’ disease cases have been increasing in recent years and the CDC report 7,500 cases in 2017. White-nose syndrome (WNS), a disease caused by the fungus Pseudogymnoascus destructans, is responsible for the extensive mortality of bats in the United States. In an effort to develop tools to reduce bat mortality attributed to WNS, an in vitro experiment was conducted to quantify the inhibitory effects of select volatile organic compounds (VOCs) as well as explore potential synergistic activities. Microbial communities, or microbiomes, are the major drivers of global biogeochemical cycles, acting as primary producers and decomposers across the water column in the oceans. Thus, they reflect changes in physicochemical properties and nutrient composition of the ocean. However, this correlation between ecological changes and the function of marine microbiomes is poorly understood. Ciguatera fish poisoning (CFP) is a type of illness that causes human intoxication after consuming contaminated coral fishes. The neurotoxins, ciguatoxins, are produced by species of Gambierdiscus, one of the harmful benthic dinoflagellates. Identification of harmful dinoflagellate species is important to gain further insights into the global dispersion. In this study, morphological characteristics of species in the genus Gambierdiscus and their habitat preferences were investigated. Field sampling was undertaken at Pulau Rawa, Terengganu and Pulau Sibu, Johor. Single cells of Gambierdiscus were isolated and clonal cultures were established.
