Aims: Mineralo-organic nanoparticles (NPs) detected in biological fluids have been described as precursors of physiological and pathological calcifications in the body. Our main objective was to examine the early stages of mineral NP formation in body fluids. Materials & methods: A nanomaterial approach based on atomic force microscopy, dynamic light scattering, electron microscopy and spectroscopy was used. Results: The mineral particles, which contain the serum proteins albumin and fetuin-A, initially precipitate in the form of round amorphous NPs that gradually grow in size, aggregate and coalesce to form crystalline mineral films similar to the structures observed in calcified human arteries. …

Chemoattractant Receptors Blt1 And Cxcr3 Regulate Anti-Tumor Immunity By Facilitating Cd8+ T Cell Migration To Tumors., Zinal S. Chheda

Electronic Theses and Dissertations

Presence of increased numbers of CD8+ T cells in the tumors correspond to better overall survival in the patients. Variety of immuno-therapies have shown considerable efficacy in the clinic, however, a multitude of patients remain unresponsive. Most of these immunotherapies rely on effector T cell responses in the tumor. A major obstacle in the success of these immunotherapies is poor recruitment of CD8+ T cells into tumors despite intact effector responses in the periphery. Therefore understanding the mechanisms that regulate CTL infiltration into tumors becomes essential. Previous studies in our laboratory suggested an important role for BLT1 in …

Determining The Mechanisms Generating Soluble Il-15 Complexes, Scott Anthony 2015 The University of Texas Graduate School of Biomedical Sciences at Houston

Determining The Mechanisms Generating Soluble Il-15 Complexes, Scott Anthony

Dissertations and Theses (Open Access)

A diverse assortment of infectious pathogens and TLR agonists enhance the expression of Interleukin (IL)-15. Additionally, inducing lymphopenia enhances anti-tumor responses in an IL-15-dependent manner. Paradoxically, despite the limited expression of IL-15 during homeostasis, the role of IL-15 during the steady state is well-known, while its roles during inflammation and infections remain largely undefined. IL-15 uses a unique method of production and presentation to support the development and homeostasis of NK and CD8 T cells. IL-15 is produced with its high affinity IL-15Rα and this IL-15Rα/IL-15 complex is shuttled to the cell surface where it is presented in-trans or cleaved …

Treg cells play a crucial role in immune tolerance, but mechanisms that induce Treg cells are poorly understood. We here have described eosinophils in lamina propria (LP) that displayed high aldehyde dehydrogenase (ALDH) activity, a rate-limiting step during all-trans retinoic acid (ATRA) synthesis, and expressed TGF-β1 mRNA and high levels of ATRA. Co-incubation assay confirmed that LP eosinophils induced the differentiation of naïve T cells into Treg cells. Differentiation promoted by LP eosinophils were inhibited by blocked either TGF-β1 or ATRA. Peripheral blood (PB) eosinophils did not produce ATRA and could not induce Treg differentiation. These data identifies LP eosinophils …

Ectopic calcification is associated with various human diseases, including atherosclerosis, cancer, chronic kidney disease, and diabetes mellitus. Although mineral nanoparticles have been detected in calcified blood vessels, the nature and role of these particles in the human body remain unclear. Here we show for the first time that human kidney tissues obtained from end-stage chronic kidney disease or renal cancer patients contain round, multilamellar mineral particles of 50 to 1,500 nm, whereas no particles are observed in healthy controls. The mineral particles are found mainly in the extracellular matrix surrounding the convoluted tubules, collecting ducts and loops of Henle as …

Hirsutella sinensis mycelium (HSM), the anamorph of Cordyceps sinensis, is a traditional Chinese medicine that has been shown to possess various pharmacological properties. We previously reported that this fungus suppresses interleukin-1β and IL-18 secretion by inhibiting both canonical and non-canonical inflammasomes in human macrophages. However, whether HSM may be used to prevent lung fibrosis and the mechanism underlying this activity remain unclear. Our results show that pretreatment with HSM inhibits TGF-β1–induced expression of fibronectin and α-SMA in lung fibroblasts. HSM also restores superoxide dismutase expression in TGF-β1–treated lung fibroblasts and inhibits reactive oxygen species production in lung epithelial cells. Furthermore, …

Chikungunya virus (CHIKV) is a mosquito-transmitted alphavirus that can cause fever and chronic arthritis in humans. CHIKV that is generated in mosquito or mammalian cells differs in glycosylation patterns of viral proteins, which may affect its replication and virulence. Herein, we compare replication, pathogenicity, and receptor binding of CHIKV generated in Vero cells (mammal) or C6/36 cells (mosquito) through a single passage. We demonstrate that mosquito cell derived CHIKV (CHIKVmos) has slower replication than mammalian cell derived CHIKV (CHIKVvero), when tested in both human and murine cell lines. Consistent with this, CHIKVmos infection in both cell lines produce less cytopathic …

An Evolutionary Vaccination Game In The Modified Activity Driven Network By Considering The Closeness, Dun Han, Mei Sun 2015 CUNY City College

An Evolutionary Vaccination Game In The Modified Activity Driven Network By Considering The Closeness, Dun Han, Mei Sun

Publications and Research

In this paper, we explore an evolutionary vaccination game in the modified activity driven network by considering the closeness. We set a closeness parameter p which is used to describe the way of connection between two individuals. The simulation results show that the closeness p may have an active role in weakening both the spreading of epidemic and the vaccination. Besides, when vaccination is not allowed, the final recovered density increases with the value of the ratio of the infection rate to the recovery rate λ/μ. However, when vaccination is allowed the final density of recovered individual first increases and …

Myeloid-Derived Suppressor Cells Contribute To The Subversion Of Innate Immunity During Staphylococcus Aureus Biofilm Infection, Cortney E. Heim 2015 University of Nebraska Medical Center

Myeloid-Derived Suppressor Cells Contribute To The Subversion Of Innate Immunity During Staphylococcus Aureus Biofilm Infection, Cortney E. Heim

Theses & Dissertations

Myeloid-derived suppressor cells (MDSCs) are a heterogeneous population of immature monocytes and granulocytes that are potent inhibitors of T cell activation. A role for MDSCs in bacterial infections has only recently emerged, and our laboratory was the first to demonstrate a functional role for MDSCs during Staphylococcus aureus (S. aureus) biofilm infection. Biofilm infections often lead to significant morbidity due to their recalcitrance to antibiotics and ability to subvert immune-mediated clearance by skewing the immune response toward an anti-inflammatory, pro-fibrotic phenotype. Therefore, we examined whether MDSCs could play a role in this process. CD11b+Gr-1+ MDSCs …

Role Of Macrophages In Muscle Transfection With Pdna/Pluronic Formulation, Vivek Mahajan 2015 University of Nebraska Medical Center

Role Of Macrophages In Muscle Transfection With Pdna/Pluronic Formulation, Vivek Mahajan

Theses & Dissertations

Non-ionic amphiphilic block copolymers of poly(ethylene oxide) (PEO) and poly(propylene oxide) (PPO), Pluronics, arranged in a tri-block structure PEO-PPO-PEO, have raised a considerable interest in skeletal muscle Gene Therapy. Previous studies have demonstrated that co-administration of Pluronics with naked plasmid DNA (pDNA) by direct i.m. injection enhanced transgene expression not only in muscle but also in distal lymphoid organs (spleen and lymph nodes) and this response was strain-dependent; not observed in athymic (BALB/c nu/nu) mouse; suggesting a role of immune cells in gene transfer to skeletal muscles. Therefore, we first evaluated the role of inflammation and inflammatory cells, on muscle …

Obesity is associated with low-grade chronic inflammation and intestinal dysbiosis. Ganoderma lucidum is a medicinal mushroom used in traditional Chinese medicine with putative anti-diabetic effects. Here, we show that a water extract of Ganoderma lucidum mycelium (WEGL) reduces body weight, inflammation and insulin resistance in mice fed a high-fat diet (HFD). Our data indicate that WEGL not only reverses HFD-induced gut dysbiosis—as indicated by the decreased Firmicutes-to-Bacteroidetes ratios and endotoxin-bearing Proteobacteria levels—but also maintains intestinal barrier integrity and reduces metabolic endotoxemia. The anti-obesity and microbiota-modulating effects are transmissible via horizontal faeces transfer from WEGL-treated mice to HFD-fed mice. We further …

Leukocyte Responsiveness To Exercise In Hcmv+ Individuals, Jared Nicholas Wilson 2015 University of Nevada, Las Vegas

Leukocyte Responsiveness To Exercise In Hcmv+ Individuals, Jared Nicholas Wilson

UNLV Theses, Dissertations, Professional Papers, and Capstones

Introduction: Emerging research suggests that the lymphocyte immune response during exercise is amplified in individuals who are positive for human cytomegalovirus (HCMV+). However, the responses of monocytes and neutrophils in HCMV+ individuals are unknown. HCMV, a type of herpes virus, infects 50% or more of the adult population in the United States. This virus can become a cause for concern in individuals who have a compromised immune system, which has been shown to occur after high-intensity exercise.

Purpose: The purpose of this study was to characterize the lymphocyte, monocyte, and neutrophil responses to exercise in HCMV+ individuals.

Methods: Participants were …

Characterization Of The Roles Of Carma3 And Bcl10 In Virus-Triggered Rig-I/Mavs Signaling Pathway, Zhicheng Zhou Mr 2015 The University of Texas Graduate School of Biomedical Sciences at Houston

Characterization Of The Roles Of Carma3 And Bcl10 In Virus-Triggered Rig-I/Mavs Signaling Pathway, Zhicheng Zhou Mr

Dissertations and Theses (Open Access)

After RNA virus infection, the innate immunity utilizes RIG-I family of receptors in the cytoplasm to initiate the production of type-I IFNs and proinflammatory cytokines by mitochondrial protein MAVS, which forms a prion-like structure to recruit TBK1and IKK complex to activate IRF3 and NF-κB, respectively. Herein, we revealed the important roles of CARMA3 and BCL10 in RIG-I/MAVS signaling pathway for the first time. CARMA3 or BCL10 deficient cells exhibited partially defective NF-κB activation but hyperactivation of TBK1-IRF3 signaling pathway upon ssRNA virus infection. It led to less production of IL6 but more production of type I IFNs and resulted in …

Parasite Infection Mediates Trait Tradeoffs In Fundulus Heteroclitus, Sarah Dunn 2015 Georgia Southern University

Parasite Infection Mediates Trait Tradeoffs In Fundulus Heteroclitus, Sarah Dunn

Honors College Theses

To be successful, an animal must eat, grow, and reproduce. With limited resources, there are tradeoffs between these critical life history parameters but the direction of the tradeoffs is largely unknown in a changing environment. To determine whether environmental context affects life-history tradeoffs, I surveyed and quantified investment into reproduction, growth, and a proxy for immunity (parasitism), in the mummichog, Fundulus heteroclitus, a common inhabitant of salt marshes in Georgia. Three salt marsh sites along coastal Georgia (Shellman Bluff, Skidaway Island, and Tybee Island) were selected using a proxy for anthropogenic disturbance (impervious surface), which also fell along a …

The Regulation Of Cathepsin L Activity By Estrogen And Bisphenols A & S: Relationship To Systemic Lupus Erythematosus, Dylan T. Cuva

Biology Honors Papers

Systemic Lupus Erythematosus (SLE) is an autoimmune disease that is linked to increased levels of estrogen. Bisphenol A (BPA) is an environmental estrogen that may also play a role in SLE. BPA is used in the production of polycarbonate plastics and epoxy resins, including use in the creation of cans and plastic bottles. This study investigated the role of BPA and other bisphenols on enzyme activity in a murine model for SLE.

The Effects Of Digestion On Innate Immunity, Rachel L. Luoma 2015 Georgia Southern University

The Effects Of Digestion On Innate Immunity, Rachel L. Luoma

Honors College Theses

Following a meal, an animal can exhibit dramatic shifts in its physiology that can result in rapid growth of the gut and heart, as well as a massive (>40-fold) increase in metabolic rate associated with the energetic costs of processing the meal. However, little is known about the effects of digestion on an important physiological trait: immune function. Thus, I tested the following competing hypotheses. First, digesting animals may upregulate their immune systems due to increased microbial exposure from ingested food. This hypothesis predicts that animals will exhibit greater immune function during digestion. Second, digesting animals may downregulate their …

The cytolethal distending toxin (Cdt) is produced from a number of bacteria capable of causing infection and inflammatory disease. Our previous studies with Actinobacillus actinomycetemcomitans Cdt demonstrate not only that the active toxin subunit functions as a phosphatidylinositol-3,4,5-triphosphate (PIP3) phosphatase but also that macrophages exposed to the toxin were stimulated to produce proinflammatory cytokines. We now demonstrate that the Cdt-induced proinflammatory response involves the activation of the NLRP3 inflammasome. Specific inhibitors and short hairpin RNA (shRNA) were employed to demonstrate requirements for NLRP3 and ASC as well as caspase-1. Furthermore, Cdt-mediated inflammasome activation is dependent upon upstream signals, including reactive …

The Role Of Batf2 In Lps/Ifnγ Polarized Macrophages, Marie A. Gehman 2015 University of Kentucky

The Role Of Batf2 In Lps/Ifnγ Polarized Macrophages, Marie A. Gehman

Theses and Dissertations--Microbiology, Immunology, and Molecular Genetics

Transcription factors regulate distinct macrophage functions by regulating gene expression in response to micro-environmental cues. This functional plasticity is critical for regulating innate and adaptive immune responses during infection and during chronic disease processes including inflammatory diseases and cancer. Microarray analysis of macrophages polarized to a pro-inflammatory (M1) phenotype with LPS and IFNγ revealed that basic leucine zipper transcription factor ATF-like 2 (Batf2), a member of the AP1 transcription factors, is selectively upregulated in M1 macrophages compared to anti-inflammatory IL-4-polarized (M2) macrophages. The initial hypothesis was that Batf2 is a master regulator of gene expression that orchestrates M1 polarization. To …

Zymosan Activation Of Tlrs: Stimulation Of Innate Immunity And Nitric Oxide Production, Sarah Loeser 2015 The University of Akron

Zymosan Activation Of Tlrs: Stimulation Of Innate Immunity And Nitric Oxide Production, Sarah Loeser

Williams Honors College, Honors Research Projects

Chronic inflammation can result in pathophysiological changes to numerous organs of the body such as heart tissue (atherosclerosis) and damage to bone. Sources of inflammation can include autoimmune disease, cancer, and chronic infections such as those triggered by HIV or Mycobacterium tuberculosis. Most of the damage associated with chronic inflammation can be associated with chemical mediators, cytokines, given off by cells of the innate immune system. One measure of an active innate immune system can be assessed by quantifying nitric oxide (NO) production by bone marrow (BM) cells.

The purpose of these experiments is to determine if zymosan, a …

The Effect Of Statins On Il-33 Mediated Mast Cell Function, Marcela Taruselli 2015 Virginia Commonwealth University

The Effect Of Statins On Il-33 Mediated Mast Cell Function, Marcela Taruselli

Theses and Dissertations

This study demonstrates original findings of statin effects on IL-33 stimulated mast cells. Statins are a class of drugs used to lower cholesterol production by targeting HMG CoA reductase. These commonly prescribed drugs have been shown to be immunomodulatory. In this study, we have found that pretreatment with statins has a variety of effects on IL-33 stimulated mast cells. Atorvastatin suppresses TNF and IL-6 production, while fluvastatin significantly enhances release of these proinflammatory cytokines in BMMCs. Although they have differing effects on cytokine production, both statins lowered ST2 expression on the cell surface, decreased cell viability, and enhanced expression of …
