Biological Evaluation Of Fda-Approved Drugs As Mirna-31 Inhibitors Using Real Time Qrt-Pcr And A Luciferase Assay, Nicholas Sienkiewicz

Honors Theses

MicroRNAs (miRNAs) are a group of post-transcriptional, negative regulators of gene expression, consisting of ~19-24 nucleotides. Since their discovery in 1993, miRNAs have been found to be involved in a diverse amount of cellular and disease pathways, including certain types of cancer. Overexpression of certain oncogenic miRNAs, including miRNA-31, have been shown to play a role in cancer cell development. Recent findings have suggested that miRNAs play a significant role in every stage of colorectal cancer (CRC) initiation, progression, and development, as well as induce resistance to chemotherapy drugs like 5-FU. In particular, research has shown that increased levels of …

Phosphorus Nmr And Its Application To Metabolomics, Paula Evans, Fatema Bhinderwala, Robert Powers, Martha Morton, Thomas Smith Apr 2020

Phosphorus Nmr And Its Application To Metabolomics, Paula Evans, Fatema Bhinderwala, Robert Powers, Martha Morton, Thomas Smith

UCARE: Research Products

Stable isotopes are routinely employed by NMR metabolomics to highlight specific metabolic processes and to monitor pathway flux. 13C-carbon and 15N-nitrogen labeled nutrients are convenient sources of isotope tracers and are commonly added as supplements to a variety of biological systems ranging from cell cultures to animal models. Unlike 13C and 15N, 31P-phosphourous is a naturally abundant and NMR active isotope that doesn’t require an external supplemental source. To date, 31P NMR has seen limited usage in metabolomics because of a lack of reference spectra, difficulties in sample preparation, and an absence of two-dimensional (2D) NMR experiments. But, 31P NMR …

Computation Of Atmospheric Concentrations Of Molecular Clusters From Ab Initio Thermochemistry, Tuguldur T. Odbadrakh, Ariel G. Gale, Benjamin T. Ball, Berhane Temelso, George C. Shields

Chemistry Publications

The computational study of the formation and growth of atmospheric aerosols requires an accurate Gibbs free energy surface, which can be obtained from gas phase electronic structure and vibrational frequency calculations. These quantities are valid for those atmospheric clusters whose geometries correspond to a minimum on their potential energy surfaces. The Gibbs free energy of the minimum energy structure can be used to predict atmospheric concentrations of the cluster under a variety of conditions such as temperature and pressure. We present a computationally inexpensive procedure built on a genetic algorithm-based configurational sampling followed by a series of increasingly accurate screening …

Hierarchical Plasmon Resonances In Fractal Structures, Isobel C. Bicket, Edson P. Bellido, Danielle M. Mcrae, Francois Lagugne-Labarthet, Gianluigi A. Botton

Chemistry Publications

An equilateral triangular prism is used as the fundamental building block to con- struct additive Sierpinski fractals, enabling new surface plasmon resonances (SPR) in the first three generations of Sierpinski triangles, as well as topological intermediaries between generations. The modes are characterized using electron energy loss spectroscopy accompanied by eigenmode calculations and optical finite-difference time domain simulations. The complex fractal geometries present a predictable hierarchy of new resonances, each arising from the previous generational building blocks used to construct the fractal. Intermediate structures break the polarization degeneracy of the equilateral fractals while maintaining a rich multiband spectral response. Engineering defects …

Identification Of Histone H4-Based Peptoids As Inhibitors Of Prmt1, Megan Demart, Molly Dubose, Sarah Mann, Corey Causey Phd, Bryan Knuckley Phd

Showcase of Osprey Advancements in Research and Scholarship (SOARS)

Protein Arginine Methyltransferases (PRMTs) are a family of 11 mammalian enzymes characterized by the post-translational methylation of arginine residues in the histone tail. The majority of the 11 members of the PRMT family are divided into two main types, Type I and Type II. PRMT1, the major Type I isozyme, catalyzes the formation of asymmetrically dimethylated arginine (ADMA). PRMT1 activates transcription of cancer genes. Peptoids, or poly-N-substituted glycine’s are a class of oligomers whose side chains are appended to the nitrogen atom of the peptide backbone rather than the alpha carbon. Kinetic parameters were conducted for both peptide and peptoid …

Investigating The Substrate Specificity Of Prmt1 Using A Plate Based Screening Method, Tina Sawatzky, Sarah Mann, Bryan Knuckley

Showcase of Osprey Advancements in Research and Scholarship (SOARS)

Enzymes are biological catalysts that speed up the rate of a reaction by lowering the activation energy and converting substrate (reactant) to product more expeditiously. Enzymes have a high degree of specificity and will only catalyze selective reactions by targeting particular substrates. In mammals, there exists a family of 11 enzymes, PRMT’s (Protein Arginine Methyl transferase), that target protein arginine for post translational methylation on the guanidino nitrogen of the residue. They occur naturally as Type I, II and III and can administer asymmetric di-methylation (ADMA), symmetric dimethylation (SDMA) and monomethylation, respectively. The focal point of this project is to …

Boron-Made N2: Realization Of A B≡B Triple Bond In The B2al3− Cluster, Nikita Fedik, Chaonan Mu, Ivan A. Popov, Wei Wang, Jie Wang, Haopeng Wang, Kit H. Bowen, Alexander I. Boldyrev, Xinxing Zhang

Chemistry and Biochemistry Faculty Publications

Until now, all B≡B triple bonds have been achieved by adopting two ligands in the L→B≡B←L manner. Herein, we report an alternative route of designing the B≡B bonds based on the assumption that by acquiring two extra electrons, an element with the atomic number Z can have properties similar to those of the element with the atomic number Z+2. Specifically, we show that due to the electron donation from Al to B, the negatively charged B≡B kernel in the B2Al3− cluster mimics a triple N≡N bond. Comprehensive computational searches reveal that the global minimum structure of B2Al3− exhibits a …

Tin @ Tio2 Core–Shell Nanoparticles As Plasmon-Enhanced Photosensitizers: The Role Of Hot Electron Injection, Xiaohui Xu, Aveek Dutta, Jacob Khurgin, Alexander Wei, Vladimir M. Shalaev, Alexandra Boltasseva

Department of Chemistry Faculty Publications

Metal–semiconductor heterostructures have attracted a lot of attention due to their ability to enhance photovoltaic and photocatalytic processes via plasmonic effects. Thus far, most of the proposed heterostructures are designed with noble metals and the potential of alternative plasmonic materials, such as titanium nitride (TiN), is not yet well explored. In this work, TiN@TiO2 core–shell nanoparticles (NPs) are synthesized and proposed as plasmon-enhanced photosensitizers for efficient singlet oxygen generation, with the focus on the role of hot electron injection. Excitation of dilute TiN@TiO2 NP dispersions by a 700 nm femtosecond-pulsed laser effectively converts ground-state oxygen into singlet oxygen (1O2), driven …

New Synthetic Methodology Using Base-Assisted Diazonium Salts Activation And Gold Redox Catalysis, Abiola Azeez Jimoh

USF Tampa Graduate Theses and Dissertations

Over the past two decades, homogeneous Au(I) catalysis has been a valuable synthetic tool for activating a series of unsaturated carbon-carbon bonds towards nucleophilic attack. Despite this progress, Au(I/III) redox chemistry has not been well explored. This is largely due to the challenges associated with the high oxidation potential of gold. Hence, the need for an efficient and safe method for Au oxidation.

Herein, a base-assisted diazonium salt activation has been used to facilitate Au redox reactions under mild reaction conditions. This oxidative coupling method afforded functional isoxazolines with good to excellent yields, while the alternative photoactivation method provided trace …

Aldosterone (Aldo), when overproduced, is a cardiotoxic hormone underlying heart failure and hypertension. Aldo exerts damaging effects via the mineralocorticoid receptor (MR) but also activates the antiapoptotic G protein-coupled estrogen receptor (GPER) in the heart. G protein-coupled receptor (GPCR)-kinase (GRK)-2 and -5 are the most abundant cardiac GRKs and phosphorylate GPCRs as well as non-GPCR substrates. Herein, we investigated whether they phosphorylate and regulate cardiac MR and GPER. To this end, we used the cardiomyocyte cell line H9c2 and adult rat ventricular myocytes (ARVMs), in which we manipulated GRK5 protein levels via clustered regularly interspaced short palindromic repeats (CRISPR)/Cas9 and …

Undergraduate Honors Papers

As the demand for renewable energy grows, the investigation of potential battery materials continues to progress hastily. Many devices and machines of present day are operated by battery technology, and their energy requirements warrant which kinds of batteries will power them. The battery in a phone requires hundreds, possibly thousands, of uses and needs to be able to recharge quickly. The battery in the implantable cardiac defibrillator (ICD), the battery by which this product was inspired, is only used once, but it must last years before it is replaced. The cathode of the ICD battery is comprised of a silver …

Direct Arylation Polymerization Of Indophenine-Based Monomers, Sarah Severson Apr 2020

Direct Arylation Polymerization Of Indophenine-Based Monomers, Sarah Severson

Student Research, Papers, and Creative Works

Indophenine, a deep blue dye and a quinoidal compound, has shown potential as a monomer in donor-acceptor copolymer formation for use in energy and device applications. Despite the barrier of low solubility, we previously polymerized an indophenine-based monomer IQ to form PolyIQ-co-thiophene (PIQ) in a 95% yield using Stille coupling. Although PIQ has desirable electronic properties, Stille coupling is a particularly unsustainable method. In contrast, direct arylation polymerization (DArP) is a more sustainable polymerization method that reduces synthetic steps and toxic byproducts. Therefore, we now report the DArP of IQ to form PolyIQ-co-ProDOT (ProPIQ) in a 96% yield. ProPIQ boasts …

Synthesis Of Β-C-Glycosidic Ketones From Unprotected Sugars And Their Use In Aldol Condensations With Hydroxymethylfurfural, Jon Chiaramonte

Student Scholars Day Posters

C-glycosides are a growing interest in synthetic chemistry for their use in the pharmaceutical industry as important drug pharmacophores and their potential use in chemical industry as surfactants. Previous literature describes the synthesis of C-glycosides using many steps and toxic reagents, often involving the use of protecting groups. The extra steps result in lower yields, require more reagents and produce more waste, which limits the environmental sustainability of the overall synthesis. This study aimed to reproduce the literature synthesis of β-C-glycosidic ketones from acetylacetone and monosaccharides without the use of protecting groups. These ketones were further reacted in aldol condensations …

Testing The Ancestral Function Of Divergent Candida Albicans Virulence-Associated Proteins Via Expression In Saccharomyces Cerevisiae Mutants, Curtis Mack

Student Scholars Day Posters

Candida albicans is an opportunistic fungal pathogen and a member of the normal human microbiota. It is closely related to the harmless fungus, Saccharomyces cerevisiae. Some genes associated with the virulent nature of C. albicans are similar to genes in S. cerevisiae. Three of these genes, PHO13, NRG1, and TUP1 are known to repress the change from yeast to filamentous growth in C. albicans, an important virulence trait. However, similar genes in S. cerevisiae are known to have functional differences. Our goal was to see if the ancestral function of these genes have drifted apart by examining …

Structural And Functional Characterization Of New Clinically Relevant Enzymes Involved In Antibiotic Resistance, Micah Fernando

Student Scholars Day Posters

Resistance to β-lactam drugs such as penicillins and cephalosporins has become a worldwide health problem. The World Health Organization has listed multi-drug resistant Acinetobacter baumannii as requiring “top priority for new antibiotic development.” Such antibiotic resistance derives primarily from β-lactamases, including Acinetobacter-derived cephalosporinase (ADC), which bind and destroy antibiotics. A recent study of clinical A. baumannii infections identified the most prevalent ADC β-lactamases conveying antibiotic resistance. This research study seeks to determine the structure and function of two clinical ADC variants: ADC-30 and ADC-162. Using X-ray crystallography and enzyme kinetics, we aim to relate the small changes in structure to …

Monitoring And Identifying The Rhodamine 6g-Hydroxide Ion Reaction Using In-Situ, Surface-Enhanced Raman Spectroscopy, Ryan Lamb

Masters Theses & Specialist Projects

An effective method for monitoring chemical reactions is necessary to better understand their mechanisms and kinetics. Effective reaction monitoring requires a spectroscopy technique with fast data acquisition, high sensitivity, structure-to-spectrum correlation, and low solvent interference. Surface-enhanced Raman spectroscopy (SERS) provides these features, which makes it a valuable tool for monitoring reactions. To obtain the Raman enhancement, metallic nanostructures typically made of silver or gold are aggregated using a salt. The nanoparticles aggregates must then be stabilized using a surfactant to use this method in situ due to eventual nanoparticle precipitation. In this study, gold nanoparticles stabilized with sodium dodecyl sulfate …

To Probe Full And Partial Activation Of Human Peroxisome Proliferator-Activated Receptors By Pan-Agonist Chiglitazar Using Molecular Dynamics Simulations., Holli-Joi Sullivan, Xiaoyan Wang, Shaina Nogle, Siyan Liao, Chun Wu

College of Science & Mathematics Departmental Research

Chiglitazar is a promising new-generation insulin sensitizer with low reverse effects for the treatment of type II diabetes mellitus (T2DM) and has shown activity as a nonselective pan-agonist to the human peroxisome proliferator-activated receptors (PPARs) (i.e., full activation of PPARγ and a partial activation of PPARα and PPARβ/δ). Yet, it has no high-resolution complex structure with PPARs and its detailed interactions and activation mechanism remain unclear. In this study, we docked chiglitazar into three experimentally resolved crystal structures of hPPAR subtypes, PPARα, PPARβ/δ, and PPARγ, followed by 3 μs molecular dynamics simulations for each system. Our MM-GBSA binding energy calculation …

Crystallization At Droplet Interfaces For The Fabrication Of Geometrically Programmed Synthetic Magnetosomes, Matthew Gromowsky, Michael Stoller, Maddee Rauhauser, Marcus Judah, Abhiteja Konda, Christopher Jurich, Stephen Morin

UCARE: Research Products

Many organisms rely on the precise growth, assembly, and/or organization of inorganic crystals to achieve vital functions, for example, three-dimensional structural support (i.e., skeletal systems based on calcite) or environmental sensing (i.e., magnetosomes based on magnetite). Mimicking the production of the complex products observed in these biomineralization processes, synthetically, remains challenging. Herein, a method for the synthesis of artificial magnetosomes with programmable magnetic domains was developed. Specifically, precursors were compartmentalized inside different surfactant-stabilized aqueous-phase droplets suspended in oil and microfluidic technologies were implemented to control their interactions precisely. When reactive droplets were brought into contact with one another, a lipid …
