Ruthenium complexes are often investigated as potential replacements for platinum-based chemotherapeutics in hopes of identifying systems with improved tolerability in vivo and reduced susceptibility to cellular resistance mechanisms. Inspired by phenanthriplatin, a non-traditional platinum agent that contains only one labile ligand, monofunctional ruthenium polypyridyl agents have been developed, but until now, few demonstrated promising anticancer activity. Here we introduce a potent new scaffold, based on [Ru(tpy)(dip)Cl]Cl (tpy = 2,20:60,200-terpyridine and dip = 4,7-diphenyl-1,10-phenanthroline) in pursuit of effective Ru(II )-based monofunctional agents. The catalytic function of lysyl hydroxylase-2 (LH2), a member of the Fe(II)/αKG-dependent oxygenase superfamily, is to catalyze the hydroxylation of lysine to hydroxylysine in collagen, resulting in stable hydroxylysine aldehyde-derived collagen cross- links (HLCCs). Reports show that high amounts of LH2 lead to the accumulation of HLCCs, causing fibrosis and specific types of cancer metastasis. Some members of the Fe(II)/αKG-dependent family have also been reported to have intramolecular O2 tunnels, which aid in transporting one of the required cosubstrates into the active site. Inhibitors of the Polycomb Repressive Complex 2 (PRC2) histone methyltransferase EZH2 are approved for certain cancers, but realizing their wider utility relies upon understanding PRC2 biology in each cancer system. Using a genetic model to delete Ezh2 in KRAS-driven lung adenocarcinomas, we observed that Ezh2 haplo-insufficient tumors were less lethal and lower grade than Ezh2 fully-insufficient tumors, which were poorly differentiated and metastatic. Using three-dimensional cultures and in vivo experiments, we determined that EZH2-deficient tumors were vulnerable to H3K27 demethylase or BET inhibitors. This dissertation focuses on the design, synthesis, and characterization of well controlled interfaces on nanoparticle surfaces. Reversible addition-fragmentation chain transfer (RAFT) polymerization was used to synthesize block copolymers and polymer grafted silica nanoparticles (PGNs) with precise control over molecular weight, graft density, and architectures.

In the first chapter, polymer molecular weights were determined through diffusion ordered spectroscopy NMR (DOSY NMR) as an alternative to size exclusion chromatography (SEC). Polymer standards of known molecular weight were analyzed through DOSY to obtain their respective diffusion coefficients. Simple Summary: Amino acid metabolism is aberrantly altered in pancreatic cancer (PC). Evidence suggests that circulating histidine (His) levels are low in PC. However, the role of His in modulating the progression of PC remains unknown. We determined the role of His in altering the therapeutic effectiveness of gemcitabine (Gem) against PC. We provide evidence that the expression of histidine ammonia lyase (HAL), an enzyme involved in His catabolism, is greatly increased in mouse and human pancreatic tumors. Metabolomics samples like human urine or serum contain upwards of a few thousand metabolites, but individual analytical techniques can only characterize a few hundred metabolites at best. The uncertainty in metabolite identification commonly encountered in untargeted metabolomics adds to this low coverage problem. A multiplatform (multiple analytical techniques) approach can improve upon the number of metabolites reliably detected and correctly assigned. This can be further improved by applying synergistic sample preparation along with the use of combinatorial or sequential non-destructive and destructive techniques. งานวิจัยนี้ศึกษาสมรรถนะของตัวทำละลายผสม AMP-PZ-MDEA นำไปเปรียบเทียบกับตัวทำละลายดั้งเดิม 30 wt.% MEA ในเชิงพฤติกรรมการตกตะกอนเพื่อหาค่าความจุการดูดซึมคาร์บอนไดออกไซด์สูงสุดที่ไม่ทำให้ตัวทำละลายผสมตกตะกอน ความหนาแน่น ความหนืด ความจุการดูดซึมคาร์บอนไดออกไซด์ สัมประสิทธิ์การถ่ายโอนมวลรวม ประสิทธิภาพการกำจัดคาร์บอนไดออกไซด์ อัตราการคืนสภาพเริ่มต้น และภาระทางความร้อนในการคืนภาพ จากผลการศึกษาการตกตะกอนพบว่าการเพิ่มความเข้มข้นของ AMP PZ และ MDEA ทำให้มีความจุสูงสุดก่อนตกตะกอนลดลง เมื่อศึกษาความหนาแน่นและความหนืดในช่วงอุณหภูมิ โดยความหนาแน่นมีค่าใกล้เคียงกับ 30 wt.% MEA ในส่วนของความหนืดพบว่าค่าความหนืดที่สูงที่สุดยังอยู่ในช่วงที่สามารถนำไปใช้งานได้ การศึกษาความจุการดูดซึมพบว่า อัตราส่วนความเข้มข้น 5/10/5 AMP-PZ-MDEA มีความจุการดูดซึมคาร์บอนไดออกไซด์สูงสุดสูงกว่าตัวทำละลายดั้งเดิม 30 wt.% MEA 38.46 % การศึกษา KGav และประสิทธิภาพการกำจัดคาร์บอนไดออกไซด์ พบว่า 5/30/5 AMP-PZ-MDEA มีสมรรถนะดังกล่าวสูงที่สุด ซึ่ง KGav สูงกว่า 30 wt.% MEA 2.42 เท่า และประสิทธิภาพการกำจัดคาร์บอนไดออกไซด์สูงกว่า 30 wt.% MEA 14.08 % การศึกษา อัตราการคืนสภาพเริ่มต้น พบว่า 10/5/5 AMP-PZ-MDEA สูงกว่าตัวทำละลายดั้งเดิม 30 wt.% MEA 7.28 เท่า และมีภาระทางความร้อนในการคืนสภาพต่ำกว่า 30 wt.% MEA 48.40 % จากการการประเมินศักยภาพของตัวทำละลายของตัวทำละลายผสมในเชิงเชิงสัมพัทธ์ของตัวทำละลายผสม (SP) พบว่า 5/30/5 AMP−PZ−MDEA เหมาะสมที่สุดและวิธีการหาค่าที่เหมาะสม แบบหลายฟังก์ชั่นจุดประสงค์ (Optimization) พบว่า ทั้ง 4 อัตราส่วน สามารถรำไปใช้งานได้ในความต้องการที่แตกต่างกัน

งานวิจัยนี้นำเสนอการเพิ่มพูนความสามารถในการลดคาร์บอนไดออกไซด์ของวัสดุโครงข่ายโลหะ-อินทรีย์ (Metal-Organic Frameworks, MOFs) ภายใต้รังสีมองเห็นได้โดยเปลี่ยนคาร์บอนไดออกไซด์เป็นสารประกอบไฮโดรคาร์บอน (Hydrocarbon Compounds) โดยวัสดุที่ใช้คือ UiO-66 (Universitetet i Oslo) เป็นตัวเร่งปฏิกิริยาด้วยแสง (Photocatalyst) ซึ่งประกอบด้วยเซอร์โคเนียม (Zr) เป็นแกนโลหะ เเละกรดเทเรฟทาลิก (Terephthalic acid) เป็นลิแกนด์ (ligand) เนื่องจาก UiO-66 มีการดูดซับแก๊สคาร์บอนไดออกไซด์เเละดูดกลืนรังสีมองเห็น (Visible light) ต่ำจึงได้มีการปรับปรุง 3 กลยุทธ์คือ 1. เปลี่ยนชนิดลิแกนด์เป็น เอมีน 2-อะมิโนเทเรฟทาลิก (2-Aminoterephthalic acid, H2ATA) 2. ดัดแปลงโครงสร้างด้วยกรด (acid modification) เเละ 3. รีดิวซ์แกรฟีนออกไซด์/UiO-66(Zr) คอมโพสิต โดย 3 กลยุทธ์ช่วยเพิ่มความสามารถในการเปลี่ยนคาร์บอนไดออกไซด์เป็นสารประกอบไฮโดรคาร์บอน การดูดกลืนรังสีช่วงมองเห็นได้ เเละการดูดซับคาร์บอนไดออกไซด์ โดยในงานวิจัยมีการทดลองวัสดุ 4 รูปแบบคือ NH2-UiO-66(Zr), NH2-UiO-66(Zr) Acid modification, NH2-UiO-66(Zr)/rGO และ NH2-UiO-66(Zr) Acid modification/rGO โดยในการทดสอบการลดคาร์บอนไดออกไซด์ภายใต้รังสีมองเห็นได้พบว่าได้เปลี่ยนคาร์บอนไดออกไซด์เป็นสารประกอบไฮโดรคาร์บอนคือฟอร์เมต (formate) โดยได้ปริมาณดังนี้ 69.5, 60.1, 78.9, เเละ 63.9 µmol g-1 h-1 ตามลำดับ โดยพบว่า NH2-UiO-66(Zr)/rGO ได้ผลิตภัณฑ์สูงที่สุดที่ 78.9 µmol g-1 h-1

โปรตีนเข้มข้นจากโครงไก่: การเตรียมและการใช้ในผลิตภัณฑ์นักเก็ต, ชนากานต์ มณฑาทิพย์กุล

งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาภาวะที่เหมาะสมสำหรับการสกัดโปรตีนจากโครงไก่ เพื่อเตรียมโปรตีนเข้มข้นและนำมาศึกษาสมบัติทางเคมีกายภาพและสมบัติเชิงหน้าที่ของโปรตีนเข้มข้นจากโครงไก่และศึกษาผลของการเติมโปรตีนเข้มข้นต่อคุณภาพและเนื้อสัมผัสของผลิตภัณฑ์นักเก็ตไก่ ผลการศึกษาการสกัดโปรตีนด้วยสารละลายด่างที่ค่าความเป็นกรด-ด่าง 8.5 - 11.5 และระยะเวลาในการสกัด 30 - 300 นาที ด้วยวิธีพื้นผิวตอบสนองและการออกแบบการทดลองแบบส่วนผสมกลางพบว่าสมการการทำนายผลตอบสนองประสิทธิภาพในการผลิตโปรตีนเข้มข้นมีความเหมาะสมในการอธิบายความสัมพันธ์และทำนายค่าของตัวแปรอิสระให้มีความสัมพันธ์กับผลตอบสนองที่ต้องการ โดยค่าสัมประสิทธิ์การถดถอย (Regression coefficient) ของตัวแปรอิสระมีความสัมพันธ์กับประสิทธิภาพในการผลิตโปรตีนเข้มข้นในระบบฐานแห้ง (Dry basis) รวมถึงค่าสัมประสิทธิ์การตัดสินใจมีค่ามากกว่า 0.80 และมีค่า Root Mean Square Error (RMSE) ต่ำหมายความว่าค่าความแตกต่างเฉลี่ยกำลังสองระหว่างค่าที่ได้จากการทำนายกับค่าที่ได้จากการทดลองมีค่าต่ำ เมื่อพิจารณาค่า Adjusted r–Squared พบว่ามีค่าใกล้เคียงกับค่าสัมประสิทธิ์การตัดสินใจ การทำนายภาวะการสกัดโปรตีนจากโครงไก่จากการกำหนดเกณฑ์ของค่าประสิทธิภาพในการผลิตโปรตีนเข้มข้นสูงที่สุดพบว่าภาวะค่าความเป็นกรด-ด่างเท่ากับ 10.15 และระยะเวลาในการสกัดเท่ากับ 185 นาที เป็นภาวะที่เหมาะสมที่สุดสำหรับการสกัดโปรตีนจากโครงไก่ โดยค่าผลตอบสนองที่ได้จากการทำนายจากสมการมีความแตกต่างจากค่าที่ได้จากการทดลองเท่ากับ 12.67% เมื่อนำโปรตีนเข้มข้นที่ผลิตในภาวะที่เหมาะสมที่สุดมาทดสอบความสามารถในการอุ้มน้ำและค่าความสามารถในการเกิดอิมัลชันพบว่ายังมีความสามารถในการอุ้มน้ำต่ำกว่าเนื้อไก่ส่วนขาและส่วนอก (p < 0.05) จึงนำโปรตีนเข้มข้นมาพัฒนาสมบัติเชิงหน้าที่พบว่าการพัฒนาโปรตีนเข้มข้นด้วยกระบวนการปรับปรุงด้วยเกลือ ฟอสเฟตและความดันและการปรับปรุงด้วยเกลือ ฟอสเฟตและส่วนผสม (โปรตีนถั่วเหลืองไอโซเลท แป้งข้าวสาลี แป้งมันสำปะหลัง) และความดันส่งผลให้โปรตีนเข้มข้นมีความสามารถในการเกิดอิมัลชันและความสามารถในการคงตัวของอิมัลชันสูงกว่าโปรตีนเข้มข้นจากโครงไก่ที่ไม่ผ่านการปรับปรุงอยู่ในช่วงระหว่าง 6.03 - 12.27% และ 4.93 - 6.83% ตามลำดับ รวมถึงมีความสามารถในการละลายที่ดีกว่าเนื้อไก่ในช่วง pH เท่ากับ 6 - 7 เมื่อนำโปรตีนเข้มข้นเติมลงในผลิตภัณฑ์นักเก็ตไก่จะเห็นได้ว่าการเติมโปรตีนเข้มข้นที่สัดส่วนเท่ากับ 12.50% ส่งผลทำให้นักเก็ตไก่มีค่า Hardness, Gumminess และ Chewiness สูงกว่าสูตรปกติระหว่าง 3899.44 - 4324.59, 2676.22 - 3243.04 และ 2363.65 - 3011.87 g-force ในขณะที่ปริมาณโปรตีนและค่าสีของตัวอย่างนักเก็ตไม่มีความแตกต่างอย่างมีนัยสำคัญ (p ≥ 0.05) เมื่อเทียบกับสูตรควบคุม นอกจากนี้การใช้โปรตีนเข้มข้นมีส่งผลให้ประสิทธิภาพในการผลิตสูงกว่าสูตรปกติ โดยประสิทธิภาพในการผลิตของสูตรเติมโปรตีนเข้มข้นปรับปรุงด้วยเกลือ ฟอสเฟตและส่วนผสม (โปรตีนถั่วเหลืองไอโซเลท แป้งข้าวสาลี แป้งมันสำปะหลัง) และความดันมีค่าสูงที่สุดเท่ากับ 136.43% แต่อย่างไรก็ตามเมื่อพิจารณาจากต้นทุนในระดับ Commercial production scale พบว่าการเติมโปรตีนเข้มข้นด้วยกระบวนการปรับปรุงด้วยเกลือ ฟอสเฟตและความดันมีต้นทุนการผลิตต่ำที่สุด รวมถึงมีปริมาณโปรตีนและคุณภาพด้านสีไม่แตกต่างจากสูตรปกติ จึงเลือกโปรตีนเข้มข้นด้วยกระบวนการปรับปรุงด้วยเกลือ ฟอสเฟตและความดันเป็นกระบวนการปรับปรุงที่ดีที่สุด โดยในระยะเวลา 1 เดือนจะสามารถผลิตนักเก็ตไก่จากโปรตีนเข้มข้นได้ทั้งหมด 1250 กิโลกรัมและสามารถลดต้นทุนการผลิตได้ที่ 175 บาทต่อเดือน

Even though antituberculous drugs are available, treatment is challenging due to antibiotic resistance associated with point mutations in the bacterial genome. The process of extracting and refining crude oil is both expensive and environmentally hazardous. The synthesis of biodiesel sourced from vegetable oils is a renewable process and less hazardous to the environment. Therefore, we seek to understand the pyrolysis procedure at an atomic level in hopes of optimizing future fuel viability. Herein, I analyze methyl stearate (a component of biodiesel) using an in-house database of ab initio trajectories, each simulating 1.0 ps (with 1.0 fs resolution). Preformed particle gels (PPG) have been widely applied in oilfields to control excessive water production. However, PPG has limited success in treating opening features because the particles can be flushed readily during post-water flooding. We have developed a novel micro-sized Re-crosslinkable PPG (micro-RPPG) to solve the problem. The microgel can re-crosslink to form a bulk gel, avoiding being washed out easily. This paper evaluates the novel microgels' transport and plugging performance through super-permeable channels. Micro-RPPG was synthesized and evaluated for this study. Metal-organic supercontainers (MOSCs), a breakthrough development in our laboratory since 2012, have emerged as promising enzyme mimics, showcasing their prowess in encapsulating guest molecules and catalyzing confined chemical reactions. The intrigue surrounding MOSCs stems from their extraordinary traits, encompassing multiple binding domains, high stability, large surface area, tunable pore size, and structural modularity. Acetyl CoA synthetases (ACSs) are Acyl-CoA/NRPS/Luciferase (ANL) superfamily enzymes that couple acetate with CoA to generate acetyl CoA, a key component of central carbon metabolism in eukaryotes and prokaryotes. Normal mammalian cells are not dependent on ACSs, while tumor cells, fungi, and parasites rely on acetate as a precursor for acetyl CoA. Consequently, ACSs have emerged as a potential drug target. This dissertation comprehensively studies the materials and interfaces of electrochemical energy storage and conversion for lithium-sulfur (Li-S) batteries as well as aqueous zinc-manganese dioxide (Zn-MnO2) batteries to fundamentally meet the renewable energy demands.This dissertation is divided into 2 sections. Particulate phases transport trace metals (TM) and thereby exert a major control on TM distribution in the ocean. Particulate TMs can be classified by their origin as lithogenic (crustal material), biogenic (cellular), or authigenic (formed in situ), but distinguishing these fractions analytically in field samples is a challenge often addressed using operational definitions and assumptions. These different phases require accurate characterization because they have distinct roles in the biogeochemical iron cycle. Interactions of lectins with glycoconjugate-terbium(III) self-assembly complexes lead to sensing through enhanced lanthanide luminescence. This glycan-directed sensing paradigm detects an unlabelled lectin (LecA) associated with pathogen P. aeruginosa in solution, without any bactericidal activity. Further development of these probes could have potential as a diagnostic tool.

The growth of diatoms in the Southern Ocean, especially the region surrounding the West Antarctic Peninsula, is frequently constrained by low dissolved iron and other trace metal concentrations. This challenge may be overcome by mutualisms between diatoms and co-occurring associated bacteria, in which diatoms produce organic carbon as a substrate for bacterial growth, and bacteria produce siderophores, metal-binding ligands that can supply diatoms with metals upon uptake as well as other useful secondary compounds for diatom growth like vitamins. Constraining the role of dust deposition in regulating the concentration of the essential micronutrient iron in surface ocean waters requires knowledge of the flux of seawater-soluble iron in aerosols and the replacement time of dissolved iron (DFe) in the euphotic zone. Here we estimate these quantities using seasonally resolved DFe data from the Bermuda Atlantic Time-series Study region and weekly-scale measurements of iron in aerosols and rain from Bermuda during 2019. Marine diazotrophs convert dinitrogen (N2) gas into bioavailable nitrogen (N), supporting life in the global ocean. In 2012, the first version of the global oceanic diazotroph database (version 1) was published. Here, we present an updated version of the database (version 2), significantly increasing the number of in situ diazotrophic measurements from 13 565 to 55 286. Data points for N2 fixation rates, diazotrophic cell abundance, and nifH gene copy abundance have increased by 184 %, 86 %, and 809 %, respectively. The work in this dissertation computationally examines four areas involving variations on archetype molecules and how they affect reactivity and physical properties. Studies in Chapters 2 and 3 assess substituent effects on cyclic systems, while those in Chapters 4 and 5 explore both partial and complete substitution of ring component atoms as well as the chemical implications of the substitution.The effect of substituents on molecular reactivity is a critical concept that affects every branch of chemistry. Chemotherapy is like taking poison and hoping it kills the cancer cells before it kills you. Dihydropyridines are a practical organic scaffold commonly used for their pharmaceutical properties. Nucleophilic dearomatization of pyridines has proven to be a useful method in obtaining dihydropyridines. By using N-heterocyclic carbene (NHC) organocatalysts, dihydropyridines can be synthesized with high regioselectivity of the 1,4 regioisomer over the 1,2- regioisomer but often with low enantioselectivity. By incorporating non-covalent interaction contact points, we hypothesize that enantioselectivity can be enhanced. While several improvements have been made in recent years to optimize the recovery of ‘touch’ DNA, relatively little research has been conducted to understand the relationship between ‘touch’ DNA and the binding affinity of that DNA to metal surfaces, specifically those with a significant copper presence. Furthermore, characterization of cell-free DNA (cfDNA) and its contribution to ‘touch’ samples and those cfDNA-metal interactions from objects commonly identified at crime scenes (cartridge casings, knives, doorknobs) have been lacking.
