Vacuum-assisted headspace solid-phase microextraction (Vac-HS-SPME) is a method used to increase solid-phase microextraction sampling of semi-volatile organic compounds. Three studies were conducted using Vac-HS-SPME via gas chromatography - mass spectrometry (GC-MS) methods for analysis of various sample types. First, a study was performed using Vac- HS-SPME combined with a high-capacity (HC) SPME Arrow to determine the operational fundamentals and workflow necessary to achieve optimal extraction of semi-volatile compounds from a model solid matrix (Ottawa sand). Low-cost biosensors that can rapidly and widely detect viruses are critical for faster diagnosis and treatment decision-making, especially for infections. The commonly used field-effect transistor is sensitive to the biomarker's detection but struggles with precise detection, particularly of nontargets such as ions and proteins. To overcome this limitation, we developed a field-effect transistor biosensor design based on MXene-graphene materials to increase the accuracy and sensitivity of virus detection. Based on the hybridization process between two complementary DNA strands, single-stranded nucleic acids were immobilized on the sensing surface via 3-aminopropyltriethoxysilane and glutaraldehyde to capture the nucleic acids of the target virus. …

Here, we describe a SERS-based vertical flow assay as a platform technology suitable for point-of-care (POC) diagnostic testing. A capture substrate is constructed from filter paper embedded with spherical gold nanoparticles (AuNPs) and functionalized with an appropriate capture antibody. The capture substrate is loaded into a filtration device and connected to a syringe to rapidly and repeatedly pass the sample through the sensor for efficient antigen binding. The antigen is then labeled with a SERS-active detection probe. We show that only a few Raman reporter molecules, exclusively located adjacent to the plasmonic capture substrate, generate detectible signals. To maximize the …

Catalytic RNAs (ribozymes) were central to early stages of life on earth. The first ribozymes probably emerged in the presence of prebiotically generated peptides because amino acids can be generated under abiotic conditions, and amino acids can oligomerize into peptides under prebiotically plausible conditions. Here we tested whether the presence of prebiotically plausible peptides could have aided the emergence of ribozymes, by an in vitro selection of self-triphosphorylation ribozymes from random sequence in the presence of ten different octapeptides. In some sense, chemical graph theory applies graph theory to various physical sciences. This interdisciplinary field has significant applications to structure property relationships, as well as mathematical modeling. In particular, we focus on two important indices widely used in chemical graph theory, the Merrifield-Simmons index and Hosoya index. The Merrifield-Simmons index and the Hosoya index are two well-known topological indices used in mathematical chemistry for characterizing specific properties of chemical compounds. The bacterial stringent response (SR) is a conserved transcriptional reprogramming pathway mediated by the nucleotide signalling alarmones, (pp)pGpp. The SR has been implicated in antibiotic survival in Clostridioides difficile, a biofilm- and spore-forming pathogen that causes resilient, highly recurrent C. difficile infections. The role of the SR in other processes and the effectors by which it regulates C. difficile physiology are unknown. C. difficile RelQ is a clostridial alarmone synthetase. Deletion of relQ dysregulates C. difficile growth in unstressed conditions, affects susceptibility to antibiotic and oxidative stressors and drastically reduces biofilm formation. The obligate anaerobe Clostridioides difficile encodes multiple reductases to detoxify molecular oxygen and reactive oxygen species. Caulat and colleagues have characterized the activity and regulation of four such reductases (L. C. Caulat, A. Lotoux, M. C. Martins, N. Kint, et al., mBio 15:e01591-24, 2024, https://doi.org/10.1128/mbio.01591-24). Each proved critical for clostridial survival in a different range of oxygen concentrations; together, they ameliorate a broad range of oxidative stress levels. Moreover, two previously uncharacterized regulators were found to control reductase gene expression in response to oxidative stress. M-type stars are the most abundant stars in our Galaxy. Their visible spectra are characterized by strong TiO and VO absorption bands with occasional ScO bands near 6000–6100 Å. This region corresponds to the A²Π–X²Σ+ band system and can be used to measure the abundance of ScO. In this work, a new Doppler-limited resolution spectrum of ScO obtained in a hollow-cathode experiment is presented. The A²Π–X²Σ+ electronic transition is analyzed with special care dedicated to fitting the A′²Δ(v+2) ∼ A²Π(v) perturbation. Shock initiation of energetic materials produces cations and anions that can contribute to initial endothermic chemical reactions which lead to detonation. Hence, knowledge of the initial dissociation pathways of ionized energetic molecules is important for advancing the understanding of initiation mechanisms and meeting longstanding goals of the Department of Defense. Despite their potential importance, these transient charged species are difficult to detect in detonation experiments and are often ignored in molecular dynamics modeling of shocked energetic materials. Silicon and silicon-germanium alloy nanostructures have garnered increasing attention in the past 25 years due to their favorable optical properties, visible to near-IR luminescence, and inherently low toxicity. While many synthetic routes have been discussed in the literature, the high-temperature, solid-state disproportionation of hydrogen silsesquioxane (HSQ) has become the most popular way to produce highly crystalline, size-tunable Si and Si-based nanostructures (NCs). The cytotoxic and immunogenic-activating properties of a cobalt(III)-cyclam complex bearing the non-steroidal anti-inflammatory drug, flufenamic acid is reported within the context of anti-cancer stem cell (CSC) drug discovery. The cobalt(III)-cyclam complex 1 displays sub-micromolar potency towards breast CSCs grown in monolayers, 24-fold and 31-fold greater than salinomycin (an established anti-breast CSC agent) and cisplatin (an anticancer metallopharmaceutical), respectively. Strikingly, the cobalt(III)-cyclam complex 1 is 69-fold and 50-fold more potent than salinomycin and cisplatin towards three-dimensionally cultured breast CSC mammospheres. Mechanistic studies reveal that 1 induces DNA damage, inhibits cyclooxygenase-2 expression, and prompts caspase-dependent apoptosis. Breast CSCs treated with 1 exhibit …

Microbial electrosynthesis (MES) is a rapidly growing technology at the forefront of sustainable chemistry, leveraging the ability of microorganisms to catalyze electrochemical reactions to synthesize valuable compounds from renewable energy sources. The reduction of CO2 is a major target application for MES, but research in this area has been stifled, especially with the use of direct electron transfer (DET)-based microbial systems. Accumulation of arsenic (As) in rice grain was reported in many regions of the world, including the United States, which has been a threat to human health. This field research investigated the grain as accumulation and its relationship with the uptake of selenium (Se), molybdenum (Mo), and cadmium (Cd) in soils with and without monosodium methane arsonate (MSMA) amended, as effects of selected rice cultivars and water management. Results indicated that MSMA increased the accumulation of as and Se but decreased Mo for all six cultivars under four irrigation management. Perchlorate, which derives from both anthropogenic and natural sources in the current environment, poses a substantial health hazard to humans as it competes with iodine uptake in the thyroid gland. Consequently, there has been considerable concern about minimizing human exposure to environmental perchlorate by restricting its release from man-made sources. However, the absence of a clear understanding regarding the respective contributions of man-made and natural sources has hindered widespread regulation efforts. Designing highly active and robust catalysts for the oxygen evolution reaction is key to improving the overall efficiency of the water splitting reaction. It has been previously demonstrated that evaporation induced self-assembly (EISA) can be used to synthesize highly porous and high surface area cerate-based fluorite nano catalysts, and that substitution of Ce with 50% rare earth (RE) cations significantly improves electrocatalyst activity. An Improved Design Is Described For Ferroelectric Crystals And Implemented With The "Methoxyphenyl Series" Of Acetophenone Azines, (MeO−Ph, Y)-Azines With Y=F (1), Cl (2), Br (3), Or I (4). The Crystal Structures Of These Azines Exhibit Polar Stacking Of Parallel Beloamphiphile Monolayers (PBAMs). Azines 1, 3, And 4 Form True Racemates Whereas Chloroazine 2 Crystallizes As A Kryptoracemate. Azines 1–4 Are Helical Because Of The N−N Bond Conformation. This study explores the potential to predict the drug-loading and release profiles of aerogels based on their morphologies: a milestone in drug delivery research, which can help save time and cost in formulating new aerogel drug carriers and cut-down evaluation of the drug delivery capabilities of aerogels to a few experimental runs. Polyurea (PUA) and poly(isocyanurate-urethane) (PIR-PUR) aerogels were used as model systems, while 5-fluorouracil (5-FU) and paracetamol (PM) were used as model drugs. Accumulation of arsenic (As) in rice grain was reported in many regions of the world, including the United States, which has been a threat to human health. This field research investigated the grain As accumulation and its relationship with the uptake of selenium (Se), molybdenum (Mo), and cadmium (Cd) in soils with and without monosodium methane arsonate (MSMA) amended, as effects of selected rice cultivars and water management. Results indicated that MSMA increased the accumulation of As and Se but decreased Mo for all six cultivars under four irrigation management. MSMA also increased grain-Cd in some cultivars. In this article, we demonstrated carbon aerogel-silica composites derived from polyurea-cross-linked silica xerogel powders (C-PUA@silica) as anodes for Na-ion batteries. These xerogel-derived hard carbons with embedded silica showed a stable high capacity. Of the several hard carbon samples derived at different temperatures, the pyrolyzed sample at 800 °C (C-PUA@silica-800) showed a high capacity of 236 mAh/g at a current density of 10 mA/g and a stable cycle-life at 200 mAh/g. The galvanostatic charge-discharge curves displayed a sloping voltage profile reminiscent of the adsorption mechanism of Na+ storage. Porous polymers, known for their high internal surface area and robust chemical stability, are promising candidates for applications in gas storage, water purification, energy storage, and catalysis. The Yamamoto-type Ullmann reaction, known for yielding high porosity polymers, serves as a benchmark in this study. However, its limitations as a homocoupling reaction restricts the synthesis of ordered A-B type copolymers. This research addresses this challenge by investigating the use of cross-coupling reactions, particularly the Negishi reaction, to synthesize A-B type copolymers with reduced defectivity and enhanced surface area. The dissertation research focused on understanding the relationship between the chemical structure of organic molecules and their photophysical properties, particularly examining coherent electronic and vibrational coupling within and between molecules, with implications for next-generation energy technologies. In Chapter 2, we employed both steady-state and transient optical and magnetic spectroscopic techniques to understand the electronic delocalization in both singlet and triplet exciton delocalization in weakly bound porphyrin dimeric molecules. Our results highlight that the delocalization of singlet exciton delocalization does not guarantee the delocalization of triplet excitons between weakly bound porphyrin macromolecules. Histone deacetylase (HDAC) proteins are linked to disease states, including cancer, and have become important targets for therapeutic development. Many inhibitors have been developed to target HDAC proteins, including FDA-approved inhibitors Vorinostat (SAHA), Belinostat, Romidepsin, and Panobinostat. These inhibitors non-specifically influence most of the 11 metal-dependent HDAC isoforms, which might contribute to the side effects that patients experience during treatment. The lack of selectivity also diminishes the use of HDAC inhibitors as biomedical research tools. To address these drawbacks, the development of selective inhibitors has been heavily studied. The Indian Ocean is an understudied region for marine phosphorus (P) biogeochemistry. Emerging evidence indicates marine phytoplankton can adapt to decreasing supplies of inorganic P, a required nutrient for growth, by alternatively utilizing organic forms of P. Here we investigate the dissolved organic phosphorus concentration ([DOP]) distribution across the southern Indian Ocean using observations collected on the I05 US GO-SHIP Cruise (2023) across ~33ºS latitude. We quantify the longitudinal variability of [DOP] in the upper 350 m of the Indian Ocean and infer its rate of biological consumption from surface waters, contributing as an organic nutrient to sustain marine autotrophs …

Plasmids are mobile genetic elements known to carry secondary metabolic genes that affect the fitness and survival of microbes in the environment. Well-studied cases of plasmidencoded secondary metabolic genes in marine habitats include toxin/antitoxin and antibiotic biosynthesis/resistance genes. Here, we examine metagenome-assembled genomes (MAGs) from the permanently-stratified water column of the Cariaco Basin for integrated plasmids that encode biosynthetic gene clusters of secondary metabolites (smBGCs). We identify 16 plasmid-borne smBGCs in MAGs associated primarily with Planctomycetota and Pseudomonadota that encode terpenesynthesizing genes, and genes for production of ribosomal and non-ribosomal peptides. Pathogenic microsatellite RNA expansions are toxic and can cause neuronal degeneration and dysfunction. Therapeutics that eliminate or neutralize these RNA repeats hold great promise for the treatment of many neurodegenerative diseases because they are uniformly fatal but there is no effective treatment available today. New compounds targeting these pathogenic RNA repeat sequences represent new hope to treat these diseases.
