Lead (Pb) has long been acknowledged as a systemic toxicant, with pronounced health impacts observed even at low exposure levels, particularly in children. Adverse effects include diminished cognitive function, altered behavior, and developmental delays. Consequently, it is imperative to conduct regular monitoring of Blood Lead Levels (BLLs). In this work, we report on an electrochemical sensor based on screen-printed carbon electrode (SPCE) coated with Nafion and mesoporous carbon (MC). The sensor system uses simple sample preparation (acidification and dilution of whole blood), minimal sample volume (a few blood drops, 200 μl), and swift time-to-results (1 h). A limit of quantitation …

Recent clinical data have identified infant patients with lethal ITPA deficiencies. ITPA is known to modulate ITP concentrations in cells and has a critical function in neural development which is not understood. Polymorphism of the ITPA gene affects outcomes for both ribavirin and thiopurine based therapies and nearly one third of the human population is thought to harbor ITPA polymorphism. In a previous site-directed mutagenesis alanine screen of the ITPA substrate selectivity pocket, we identified the ITPA mutant, E22A, as a gain-of function mutant with enhanced ITP hydrolysis activity. Here we report a rational enzyme engineering experiment to investigate the …

At the turn of the 21st century, the importance of maintaining a balanced microbiome was brought to the forefront of the microbiology, immunology, and physiology research fields. Exploring the complex interactions between vaccine administration, mucosal microbiome, oral tolerance, and enteric inflammation in health and disease is challenging since environmental factors (such as diet and sanitation) have major influences on gut microbiota composition. High enteric pathogen load has been shown to contribute to dampened cell-mediated and humoral immune responses to vaccines in human case studies, either through elevated enteric inflammation or increased tolerance to environmental microbes. Although antibiotic and probiotic interventions …

Epilepsy is a prevalent neurological disorder with a complex pathogenesis and unpredictable nature, presenting limited treatment options in >30 % of affected individuals. Neurometabolic abnormalities have been observed in epilepsy patients, suggesting a disruption in the coupling between neural activity and energy metabolism in the brain. In this study, we employed amperometric biosensors based on a modified carbon fiber microelectrode platform to directly and continuously measure lactate and oxygen dynamics in the brain extracellular space. These biosensors demonstrated high sensitivity, selectivity, and rapid response time, enabling in vivo measurements with high temporal and spatial resolution. In vivo recordings in the …

School of Integrative Biological & Chemical Sciences Faculty Publications
A methodology to access reactive hydride moieties is highly desirable, yet limited. Multimetallic hydride fragments are notable for their heightened reactivity and catalysis, but deliberate access to these species is lacking. In this highlight, we discuss recent developments by our group in the design of a new heterometallic complex that invokes an architecture designed to provide modular access to reactive hydride moieties by leveraging metal hydrides in combination with pendent donors to a model heterotrimetallic Ni–(Al–H)2 complex. An amplification of insertion-based reactivity has been examined in the hydrofunctionalization of quinolines, and our complex substantially outperformed the parent aluminum hydride LAlH …

Molecular recognition plays a crucial role in various biological processes, such as enzymatic reactions, signal transduction, and genetic information processing. Investigating how proteins selectively bind to their partners is an active research area, but there is a lack of experimental details on protein structures and interactions in molecular complexes. Computational techniques based on macromolecular structures offer a way to predict protein-ligand interactions and explore their recognition mechanisms. Estimating binding affinities, particularly through alchemical binding free energy calculations, has become valuable in supporting drug discovery. This work introduces new methodologies, utilizing the Alchemical Transfer Method, to address issues like poor convergence …

Dissertations, Theses, and Capstone Projects
Organic chemistry exists in all aspects of everyday life from polymers to pharmaceutics. Formation of carbon-carbon bonds is essential for the synthesis of complex organic compounds. This goal can be achieved by using transition metal catalysts. Second-row transition metals such as Pd, Pt, Rh, and Ir have shown remarkable efficacy in these reactions. Metal-catalyzed cross-coupling reactions form carbon-carbon bonds between electrophiles and nucleophiles using transition metal catalysts. Pd is mainly used in this reaction.
Transition metal-catalyzed cross-coupling reactions that form a bond between two C(sp2) carbons have been widely studied over the past decades. C(sp2)-C(sp3 …

New Discoveries On Protein Recruitment And Regulation During The Early Stages Of The Dna Damage Response Pathways, Kelly Waters, Donald E. Spratt
Chemistry
Maintaining genomic stability and properly repairing damaged DNA is essential to staying healthy and preserving cellular homeostasis. The five major pathways involved in repairing eukaryotic DNA include base excision repair (BER), nucleotide excision repair (NER), mismatch repair (MMR), non-homologous end joining (NHEJ), and homologous recombination (HR). When these pathways do not properly repair damaged DNA, genomic stability is compromised and can contribute to diseases such as cancer. It is essential that the causes of DNA damage and the consequent repair pathways are fully understood, yet the initial recruitment and regulation of DNA damage response proteins remains unclear. In this review, …

The Effect Of Variation Of N-Substituents On Oxidopyridinium Ions In (4+3) Cycloadditions, Zahria Patrick, Madison Clark
Undergraduate Research Symposium
The aim of this research project is to expand the scope of 4 + 3 cycloadduct chemistry by varying functional groups attached to the prerequisite oxidopyridinium ion for each respective cycloadduct product. While N-substitution of the pyridinium precursor is known to proceed smoothly if alkylated by a lone methyl group, we evaluated the effect a larger alkyl group would have on the overall yield of the 4 + 3 cycloadduct product.
Isobutyl triflate, generated from the known reaction between isobutyl alcohol and triflic anhydride, was reacted with ethyl 5-hydroxy-nicotinate to generate the respective N-isobutyl oxidopyridinium ion in quantitative …

The Roaring Lion Of Berlin: The Life, Thought, And Influence Of Eugen Dühring, Arden Roy
Undergraduate Research Symposium
The life and influence of 19th-century German polymath Eugen Dühring remain but a mere footnote in the history of ideas, being primarily relegated to the status of little more than a theoretical rival to Marxism in the German socialist movement and the occasional object of Freidrich Nietzsche's rhetorical flogging. Despite the current consensus on the subject, Eugen Dühring was a scholar of vast, remarkable learnedness, contributing greatly to philosophy, economics, and the natural sciences. The aim of this talk will be to clear the fog surrounding the life and work of the controversial blind scholar and give an account of …

Determination Of Antibody Affinity For The Alzheimer’S Amyloid-Β Protein, Bree Carlton
Undergraduate Research Symposium
Alzheimer's Disease (AD) is a common form of dementia that is characterized by inflammation, loss of brain mass, and loss of motor function and memory. One of the pathological hallmarks of AD is the accumulation of Amyloid-β (Aβ) in the brain as senile plaques. Aβ is formed from a large protein called Amyloid Precursor Protein (APP) when specific enzymes cut a segment of the protein into a shorter polypeptide chain, Aβ. Different aggregation species were used in this project, specifically monomers and protofibrils. Monomers are naturally occurring and nontoxic, while protofibrils are neurotoxic and neuroinflammatory. One way that Aβ can …

Synthesis Of Template-Free Flower-Like Zno Nanorods Using A Simple Chemical Bath Technique, Benard S. Mwankemwa
Tanzania Journal of Science
Flower-like ZnO nanorods were successfully synthesised using a template-free, low-temperature chemical bath deposition method. The effects of growth time on the structural, morphological, and vibrational properties of flower-like zinc oxide were investigated. X-ray diffraction (XRD) and scanning electron microscopy (SEM) indicated that the crystallite size and density of flower-like ZnO nanorods increased with increasing growth time. Raman spectroscopy confirmed the existence of specific Raman modes that correspond to the wurtzite structure of ZnO. In addition, Raman measurements revealed that increased growth time increases the Raman modes intensity of the flower-like ZnO nanorods. Moreover, the measurements of the Au/ZnO nanorods/FTO Schottky …

Aromatic Residue Positioning Influences Helical Peptoid Structure In Aqueous Solution, Jwwad M. Javed, Katherine Scukas, Michelle T. Nguyen, Amelia A. Fuller
Chemistry and Biochemistry
Water-soluble peptidomimetics, including peptoids, are promising functional surrogates for biologically relevant, amphiphilic, helical peptides. Twenty amphiphilic peptoid hexamers with predicted helical structures were designed, prepared, and studied using circular dichroism (CD) spectroscopy. The site-specific contributions of aromatic and charged residues to the helical structure of peptoid hexamers in aqueous solution was evaluated, revealing that aromatic residue positioning most significantly impacts structure.
