The increasing frequency and intensity of Wildland-Urban Interface (WUI) wildfires, such as the destructive 2025 Pacific Palisades Fire that destroyed over 6,000 structures, have underscored the urgent need for rapid post-fire safety assessment methodologies. This study sought to quantitatively characterize the Thermally Driven Red Hue Effect (TRHE), a unique, irreversible color shift observed in architectural coatings and concrete substrates, to determine its reliability as a permanent visual record of thermal exposure. Triterpenoid saponins are amphiphilic glycosides that exhibit a wide range of biological and pharmacological properties, including antifungal, antiviral, molluscicidal, hypoglycemic, and hypocholesterolemic effects. These biological and pharmacological properties make saponins attractive for diverse applications. In this work, we demonstrate three biological applications: (i) immunostimulant vaccine adjuvant, (ii) protein delivery enhancement, and (iii) gene transfection enhancement. In light of the strong structure dependence of saponin activity, expanding the structural diversity of available scaffolds is essential for establishing structure-activity relationships (SAR) across these applications. Wildland-urban interface (WUI) fires and residential conflagrations present significant challenges for post-fire damage and fire propagation analysis. CAL FIRE observed reddening in architectural coatings during the 2025 Pacific Palisades fire with no determination of the cause. While heat-induced color change of infrastructure coatings has been studied, its potential use as a thermal exposure indicator in residential fire events remains largely unexplored. This dissertation examined the interaction of warfarin with human serum albumin (HSA) by using affinity chromatography, with particular emphasis on ultrafast affinity extraction (UAE) and affinity microcolumns. HSA affinity supports were prepared by immobilizing HSA onto diol-bonded silica via Schiff base chemistry and used to prepare affinity microcolumns for chromatographic studies under physiological-like conditions at pH 7.4 and 37.0 °C. UAE studies were performed to investigate the effects of residence time, flow rate, and column configuration on the measured apparent free fraction of warfarin and on the calculated apparent binding parameters. As tattoos rise in popularity, some marker manufacturers have created temporary tattoo markers so that children and adults can experience tattoos without the permanence of a regular tattoo. These new products are marketed explicitly as ‘temporary tattoo markers’ and intended for direct application to the skin. Henna, a natural dye derived from Lawsonia inermis, is widely used for temporary body art, hair coloring, and cultural decorative practices such as Mehendi. Traditional henna produces a reddish-brown stain through lawsone, a naphthoquinone chromophore that binds to keratin in skin, hair, and nails (Badoni Semwal et al., 2014; de Groot, 2013; Kumar Singh et al., 2015). Bloodstain detection can provide valuable information on the ability of presumptive tests. When perpetrators seek to alter the scene of violent crimes to interfere with investigations or flee from justice, crime scene investigation becomes more complex. In this analysis, scenarios where bloodstain evidence is attempted to be removed by household cleaners and covered up by acrylic or oil-based paint, are tested using luminol and Kastle-Meyer direct testing. Nucleosome core particles (NCP) are the building blocks that form a highly organized and compact chromatin structure. Nucleosomes package DNA in the nucleus of eukaryotic cells. The NCP consists of about 147 base pairs of DNA wrapped around the histone octamer, with 1.65 superhelical turns in a left-handed manner. The histone octamer is composed of two copies of H3, H4, H2A, and H2B. Together with histone H1 and linker DNA, they further assemble into a higher-order chromatin structure. Short peptides can form adaptive supramolecular assemblies, and understanding how minimal sequences organize around neurometabolites or hydrophobic cancer drugs enables the rational design of functional materials. This thesis combines molecular dynamics with experimental validation to establish design rules linking peptide sequence to emergent structure and function. Chapter 1 outlines the molecular determinants governing peptide assembly. Chapter 2 reviews computational workflows that reveal sequence-dependent conformations and supramolecular organization. Solar reflective coatings offer a promising strategy to mitigate the urban heat island effect by reducing absorption of near-infrared solar energy. This thesis project investigates the feasibility of incorporating near-infrared reflective pigments into waterborne direct-to-metal (DTM) latex coating systems while maintaining acceptable color comparison characteristics and coating performance. An all acrylic latex DTM formulation with titanium dioxide as the sole pigment was prepared and tinted with a variety of conventional and solar reflective commercial colorants across the yellow, red, blue, and green color ranges. Background/Objectives: Herbivorous insects feed on plant tissues to obtain nutrients necessary for growth and development while simultaneously ingesting diverse plant secondary metabolites. Understanding the fate of these compounds during digestion is important for advancing knowledge of insect nutritional physiology and diet-associated biochemical processes. Physiological redox homeostasis is a fine balance between prooxidants and antioxidants that are integrated elements of several reduction-oxidation mechanisms at molecular, organellar, cellular and tissue levels. When this equilibrium is disrupted and prooxidants become dominant, the body relies on endogenous and exogenous antioxidants to counterbalance the dysregulation and ultimately prevent the progression of oxidative stress. The major goal of this work was to develop a therapeutic intervention against preeclampsia (PE). Preeclampsia affects up to 5-7% of all pregnancies and has no causative treatments. It is associated with oxidative stress caused by a prolonged ischemic state and increase in reactive oxygen species (ROS) in the placenta. The cardinal symptoms include high blood pressure, kidney dysfunction, and in severe cases, eclampsia. With no treatment for the underlying causes available, we offer small molecule antioxidants which may scavenge ROS in the placenta arising from the initiating event of PE, the oxidative stress. Tissue engineering is widely used in research for investigating cellular proliferation, behavior, and responses to various stimuli. However, the predictive value of preclinical studies using cell culture plates is limited by the inability to recapitulate the complexity of the physiological microenvironment. Since the industrial revolution, widespread fossil fuel combustion has significantly increased atmospheric pollution. The combustion of fossil fuels releases nitrogen oxides (NOx) and volatile organic compounds (VOCs), which form tropospheric (ground-level) ozone when exposed to sunlight. Ozone directly harms plants by disrupting their ability to absorb carbon dioxide, release oxygen, and carry out photosynthesis. High-voltage n-type organic cathode materials are critical for constructing zinc-organic batteries (ZOBs) with high energy density and long cycle life. However, the intrinsically unfavorable electronic structures and relatively high LUMO energy levels of most n-type materials often lead to sluggish kinetics, high solubility, and suboptimal discharge voltages (< 0.8 V). Here, we design a small molecule, quinoxalino[2’,3’:5,6]pyrazino[2,3-f][1,10]phenanthroline (DPQP), as a ZOB cathode by introducing locally electron-deficient motifs into the conjugated backbone of aromatic compounds. The linearly fused pyrazine units extending the pyrazine–benzene framework effectively optimize the electronic structure, thereby significantly enhancing the discharge voltage. Meanwhile, the expanded π-conjugated plane suppresses dissolution and accelerates charge-transfer kinetics. Benefiting from these features, the DPQP electrode exhibits an exceptional increase in average operating voltage from 0.61 V to 1.07 V (vs. Zn2+/Zn) at 0.1 A·g–1, with an overpotential of only 140 mV. Notably, no discernible voltage decay occurs as the current density increases, indicating rapid and highly reversible redox kinetics. High-entropy oxides (HEOs) present significant scientific challenges in both design and synthesis due to their multielement and high-entropy nature, which involves complex combinations of multiple metal cations and oxygen anions, typically arranged in equimolar ratios to achieve structural stability. Herein, one-dimensional (Co,Ni,Mn,Cu,Zn)O high-entropy oxide nanotubes (HEO-NTs) are fabricated by means of a gradient electrospinning strategy with a tailored polyvinyl alcohol (PVA) molecular weight distribution and controlled pyrolysis. Surface functionalization of colloidal gold nanoparticles with capping ligands is critical for controlling their dispersive stability and for introducing application-specific moieties. Peptoids (N-substituted glycine oligomers) are useful candidates for study as capping ligands owing to their straightforward preparation and their biocompatibility. In this work, we have prepared a library of five-residue peptoids and evaluated them as capping ligands for 10 nm gold nanospheres. Our findings revealed important sequence features of peptoids that stabilize nanosphere colloidal dispersions in a range of aqueous conditions that vary in ionic strength and pH. Roselle (Hibiscus sabdariffa L.) is rich in phenolic compounds and anthocyanins, but the extraction method may influence its chemical composition and biological activity. This study compared hot-water infusion, ethanolic maceration, and maltodextrin-assisted drying. It aimed to examine the relationship between the extraction method, chemical profile, and bioactivity. The resulting extracts (EI, EM, and EP) were analyzed for total phenolic, flavonoid, and anthocyanin, LC-HRMS profiles, and antioxidant capacity by the FRAP assay. Methane (CH4) is a potent greenhouse gas, and tropical forests account for roughly one–third of global atmospheric CH4 uptake by soils. Projected warming and more frequent hurricanes in these ecosystems may alter soil CH4 sink strength, as warmer and wetter soils enhance methanogenesis activity. Non-heme Fe(II)/2-oxoglutarate (2OG)-dependent halogenases catalyze highly selective C-H halogenation. BesD is a non-heme Fe(II)/2OG halogenase that performs regio- and stereoselective chlorination of the l-lysine (l-Lys) substrate. Understanding the mechanism by which halogenation is favored over canonical hydroxylation is essential for guiding enzyme engineering efforts aimed at converting hydroxylases into halogenases. Here, we combine molecular dynamics (MD) and hybrid quantum mechanics/molecular mechanics (QM/MM) calculations to elucidate the origin of chlorination selectivity in BesD and variants derived from a homologous hydroxylase. This honors thesis examines the biochemical, ethical, and public health consequences of insufficient post-operative follow-up care for undocumented immigrants injured in border falls. Discussing pathways of inflammation resolution, wound healing, and bone remodeling, this thesis argues that recovery depends on tightly regulated molecular and cellular processes that are highly vulnerable to disruption without continued monitoring and rehabilitation (Loi et al., 2016; Maruyama et al., 2020). Substituted aromatic rings are present in industry in a plethora of organic molecules such as pharmaceuticals, agrochemicals, materials and beyond. Therefore, methods for broadly installing aryl rings into other compounds to create substituted aromatic rings are valuable to many organic chemists. Historically these transformations are achieved through methods such as nucleophilic aromatic substitution and metal catalyzed cross-coupling. Nucleophilic aromatic substitution is a user-friendly method in this space but is limited by reliance on specific electronics.
