Success of newly discovered chemistry in academia is often scored in terms of its novelty and level of scholarship. In industrial settings, cost, safety and quality are often times the ruler by which new processes are measured. Our group has identified that there is a gap between these two measures of success and has sought to develop principles in order formalize an approach to synthetic strategy and developing ready-to-implement manufacturing processes for molecules, simple and complex.

Cyclic-AMP dependent protein kinase (PKA) is a key intracellular signal transduction kinase that is modulated by Gs- and Gi-coupled GPCRs. Under normal physiological conditions, PKA exists as an inactive holoenzyme made up of two catalytic subunits and two regulatory subunits. Upon cAMP binding to the regulatory subunits, the catalytic subunits (PKACa) are released to perform various downstream phosphorylation events. However, aberrant PKA activation can cause various diseases including Cushing’s Syndrome, which is an endocrine disorder caused by the overproduction of cortisol by the hypothalamus-pituitary-adrenal hormone system. The genetic code of all known organisms is comprised of the 20 proteinogenic amino acids that serve as building blocks on a peptide chain to form a vast array of proteins. Proteins are responsible for virtually every biological process in all organisms; however, the 20 amino acids contain a limited number of functional groups that often leaves much to be desired. The pre-synaptic protein α-Synuclein (αS) is often linked to the pathology of Parkinson’s disease (PD), an age-related neurodegenerative disorder. Lewy bodies, the cytopathological hallmarks of PD, are found to be rich in aggregates of misfolded αS protein. Metal dyshomeostasis has also been linked to PD due to the accumulation of iron in the substantia nigra pars compacta, and diminished copper levels reported in this same region. Metal dyshomeostasis in the brain coupled with oxidative stress can enhance the aggregation of αS. Peptides are emerging as promising therapeutics due to their inhibitory affinity towards protein-protein interactions (PPI). However, peptides have been limited mainly by their poor bio-stability and lack of cell permeability. Efforts to generate drug-like peptides have led to the development of macrocyclic peptides, which exhibit improved stability. Yet, most macrocyclic peptides still require the assistance of a cell penetrating peptide (CPP) for cellular entry.

Deep eutectic solvents are a new generation of ionic liquid-like solvents formed by combining hydrogen bond acceptor with hydrogen bond donor which result in the depression of the melting point of the solvent. Like ionic liquids, anions play a critical role in tuning the polarity, physicochemical properties, and thermodynamic behavior of deep eutectic solvent (DES). Cyanide and nitrosoamines (NAs) are ubiquitous compounds, found in our food and water, either by natural process or through man-made activities. The toxicity of cyanide is exerted by its ability to inhibit metalloenzymes such as cytochrome c oxidase, causing concomitant cascades of biochemical effects such as lactic acidosis, inhibition of ATP production, respiratory seizure, and potential death. Nitrosoamines, on the other hand, undergo biotransformation (metabolic activation in cytochrome P450) in the body to produce unstable intermediates that alkylate DNA, causing mutations, and leading to carcinogenesis. The study of cultural heritage objects requires reference materials for comparison in order to determine the identity of chemical compounds in historically/culturally significant artifacts. The reference materials available today can be inaccurately labeled, can contain inauthentic compounds, or can simply be formed differently than what the labeled material should be. Utilizing the laboratory at the Indianapolis Museum of Art at Newfields, three case studies were conducted over three different potential reference materials in order to properly identify each of the three materials being focused on here. Microplastics (or MPs; < 5 mm in size) pollution is largely unstudied in the Philippines. From an environmental sustainability standpoint, it is important to understand the characteristics, abundance, and environmental fate of plastic debris of various sizes, and these include microplastics that are not more easily and readily detected. In this study, we assessed the extent of microplastics contamination in the sediments, waters, oysters, and selected fishes found in the rivers and coastal areas of Ticalan, Batangas, which were identified from water quality parameters as Class C and CS, respectively. The microplastics were extracted from these samples by chemical digestion of the matrix, series of filtration, and separation by flotation through a density gradient to finally isolate the microplastics which were not dissolved by chemical digestion. The isolated samples were imaged by optical microscopy and characterized based on their descriptive attributes. The results showed the presence of microplastics in all the samples tested, which were found mostly in the form of filaments, fragments, films, and pellets – with most showing weathered, degraded, or angular and irregular surfaces. Identification was done through spectral matching of the Fourier transform infrared spectra of isolated fragments with that of known plastics, although identification in some cases is made uncertain by possibility of degradation of the plastics in the environment. The majority of the isolates showed signature absorption bands of the C-H stretching vibrations of polyethylene-based plastics.

Scientists and engineers have observed for some time that tidal amplitudes at many locations are shifting considerably due to nonastronomical factors. Here we review comprehensively these important changes in tidal properties, many of which remain poorly understood. Over long geological time scales, tectonic processes drive variations in basin size, depth, and shape and hence the resonant properties of ocean basins. On shorter geological time scales, changes in oceanic tidal properties are dominated by variations in water depth. Herpes simplex virus type 1 (HSV-1) is the causative agent of oral herpes, and is able to establish lifelong latent infection along with painful, recurring symptoms. Epigallocatechin-3- gallate (EGCG) is the most abundant polyphenol derived from green tea (Camellia sinensis), whose anti-inflammatory, antioxidant, anti-cancer, and antiviral properties have been widely studied. Although EGCG is not chemically stable and has poor hydrophobicity, lipophilic derivatives of EGCG have shown increased stability as well as increased antiviral efficacy against HSV. Methods for the accomplishment of small molecule imaging by mass spectrometry are challenged by the need for sample pre-treatment steps such as cryo-sectioning, dehydration, chemical fixation, or application of a matrix or solvent, that must be performed to obtain interpretable spatial distribution data. According to the results of the Human Genome Project, less than 5% of our genome consists of sequences that code for actual proteins. At the same time, however, ~90% of these non-coding sequences are transcribed into a myriad of classes of ribonucleic acids collectively known as ncRNAs, which have been recognized as essential regulatory factors. The α-crystallins (αA- and αB-crystallin isoforms) from the ocular lens are small heat shock proteins and molecular chaperones, widely believed to play a significant role in protecting the lens from cataract. Mutations and modifications on human αA- and αB-crystallin (HAA and HAB, respectively) are linked to a variety of diseases, including cataract formation, neuropathological protein folding disorders, and many others. This method of identification uses Raman spectroscopy and advanced chemometrics to distinguish between body fluids. The Thrombin Binding Aptamer (TBA), a short DNA strand with G-quadruplex conformation, binds to Thrombin with high affinity and acts as an inhibitor in the blood clotting. For years, fingerprint samples have been believed to be useful only for matching purposes using their unique ridges, shapes, and sizes. The full environmental impacts of microplastic pollution in aquatic environments remain uncertain, but several detrimental effects have been documented, including adverse health effects in multiple species and the susceptibility of microplastics to adsorb toxins and leach plastic additives. Plastic microbeads from rinse-off personal care products are a source of microplastic pollution that is introduced directly into the environment through runoff, including wastewater treatment plant effluent. In December 2015, the federal Microbead-free Waters Act of 2015 banned the manufacture and sale of all rinse-off personal care products containing plastic microbeads in the United States.

A two-step process of protein detection at a single molecule level using Surface Enhanced Raman Spectroscopy (SERS) was developed as a new platform for medical diagnostics in this proof-of-concept study. First, a protein molecule was bound to a linker in the bulk solution and then this adduct was chemically reacted with the SERS substrate. Traut’s Reagent (TR) was used to thiolate Bovine serum albumin (BSA) in solution followed by chemical cross linking to a gold surface through a sulfhydryl group. Natural RNAs utilize extensive chemical modifications to diversify their structures and functions. Geranylated-2-thiouridine is a special hydrophobic tRNA modification that has been discovered very recently in several bacteria, namely E. coli, E. aerogenes, P. aeruginosa and S. typhimurium by Liu group. It has been found as a post-transcription modification on the wobble position of anticodon of tRNAs specific for glutamic acid, glutamine and lysine. Geranylated-tRNA was shown to correct the frameshifting during the translation of E. coli. However, the reason of the existing of the modification is still largely unclear. Blood is one of the principal sources of evidence in crime scene investigations. The level of hemoglobin in human blood is known to hardly fluctuate over time in a healthy adult patient. Using a method like the sodium lauryl sulfate (SLS) method for hemoglobin level determination is beneficial because SLS converts any other derivates of hemoglobin into methemoglobin. The amount of hemoglobin would be determined using the linear correlation between the absorbance of methemoglobin and its amount in blood samples.
