Spotted fever group (SFG) Rickettsia are obligate intracellular bacteria that are carried by ticks. One such tick, Dermacentor variabilis is a vector for the etiologic agent of Rocky Mountain spotted fever, R. rickettsii. These ticks also carry a non-pathogenic R. montanensis, the agent used in this study. Interestingly, field data collected from infected D. variabilis throughout the United States revealed that the majority of Rickettsia in ticks are non-pathogenic species such as R. montanensis. Although ticks serve as both vector and reservoir hosts for SFG Rickettsia, many questions regarding tick-Rickettsia interaction remain unresolved. Therefore, the overall goal of this research …

Unilateral arytenoid lateralization is a commonly performed surgical treatment for laryngeal paralysis in dogs. It involves fixing the moveable arytenoid cartilage to the thyroid (TAL) or cricoid (CAL) cartilage or both (CTAL). This increases the area of the rima glottidis (RGA), to allow reduced airway pressure and laryngeal resistance in vitro and ameliorates clinical signs in vivo. It may also increase the patient’s predisposition for aspiration pneumonia, which occurs in around 20% of clinical patients. Gastrointestinal nematode (GIN) infections affect production systems and exotic hoofstock in zoos, particularly giraffe (Giraffa camelopardalis). Anthelmintic resistance (AR) prevalence is increasing in production systems and zoos. To combat the AR that compounds GIN problems, alternative control methods are used. One such alternative is copper oxide wire particles (COWP), which control the abomasal nematode Haemonchus contortus. COWP was given to seven adult giraffe at Busch Gardens Tampa, at descending dosages: 25 g, 12.5 g, 6.3 g. Treatment administration time was determined by fecal egg count (FEC) being above 600 eggs per gram (EPG), and therefore varied with each animal. FEC …

HSV-1 facilitates virus entry into cells and cell-to-cell spread by mediating fusion of the viral envelope with cellular membranes and fusion of adjacent cellular membranes. Although virus strains isolated from herpetic lesions cause limited cell fusion in cell culture, clinical herpetic lesions typically contain large syncytia, underscoring the importance of cell-to-cell fusion in virus spread in infected tissues. Certain mutations in gB, gK, UL20 and other viral genes, drastically enhance virus-induced cell fusion in vitro and in vivo. Among the soil-transmitted helminths (STH), hookworms are a worldwide problem in both humans and animals. They cause non-specific gastrointestinal symptoms, and in young children and animals, they can cause stunting, malnutrition and anemia. Canine hookworms have significant zoonotic potential as a cause of cutaneous larvae migrans and eosinophilic enteritis in humans. To determine the ecological niche of human hookworm in Brazil, two risk models were developed based on the Growing Degree Day-Water Budget (GDD-WB) concept, one based on accumulation of monthly temperatures above a base temperature of 15oC and threshold WB value >0.4. Herpes simplex virus type 1 (HSV-1) is a DNA virus that is a ubiquitous pathogen of humans. The hallmark of the HSV-lifecycle is infection of a mucosal surface with spread to sensory neurons where the virus establishes a latent infection with periodic recurrences for the life of the host. The most common course of symptomatic disease with HSV-1 is the typical mucocutaneous lesion that is self-limiting. HSV-1 can also cause acute encephalitis and ocular pathology on reactivation. Acute gram-negative bacterial infections are a leading cause of mortality among the nosocomial infections. Increasing numbers of immunosuppressed individuals and growing numbers of antibiotic resistant strains make antibiotic treatment difficult. Neutrophils are the first cells recruited to the site of infection and are critical players in the host defense against gram-negative bacterial pneumonia. Therefore, identification of targets that boost neutrophil-associated host defense in the lung is essential in designing better therapies to control pulmonary infections. Herpes simplex virus type I (HSV-1) is a neurotropic virus that infects primarily mucocutaneous epithelial cells and nervous tissue. Membrane fusion is an important aspect of the HSV-1 lifecycle, that occurs during viral entry (virus-cell fusion), viral spread (cell-to-cell fusion), as well as, during virion morphogenesis (assembly and egress). These membrane fusion steps involve complex interactions between multiple viral glycoproteins and cellular receptors. ABSTRACT Objectives - To compare the osteointegration of plasma sprayed hydroxyapatite (HA) coated and uncoated 5.5 mm cortical screws in equine third metacarpal (MC3) bones in combination with a 4.5mm broad dynamic compression plate (DCP). Study design - In vivo study. Animals – 6 Adult Thoroughbred horses. Methods – For each horse, four HA coated screws were placed unicortically through a 4.5mm broad DCP in the dorsal cortex of a randomly chosen MC3 bone, with 4 uncoated screws placed in an identical manner in the contralateral MC3 bone. Laminitis is an often fatal condition in horses with few available and only moderately effective treatment options. Separation of laminar dermis and epidermis lead to rotation or ventral deviation of the third phalanx inside the hoof capsule. Despite being a modified skin, equine laminar tissue does not completely return to normal after laminitis. The hypothesis tested was that adult progenitor cells in the equine laminar tissue are irreversibly damaged by laminitis. Despite increased recognition of human cases, limited information is available regarding infection of invertebrate and vertebrate hosts for this emerging tick-borne disease. To investigate the role of intraocular leptospiral infections in horses with Equine Recurrent Uveitis (ERU), ocular fluid samples were collected from donated and client-owned horses with a history and ocular findings consistent with chronic ERU. Additionally, eyes were harvested from horses with normal ophthalmic examinations as a control group. Blood samples were obtained for Leptospira serology using microscopic agglutination test (MAT). Aqueous and vitreous humor samples were aseptically obtained and submitted for aerobic culture and Leptospira culture, PCR and MAT. The objectives of this study were to determine what effects iodixanol would have on total and progressive motility, plasma membrane integrity (viability), acrosome integrity and DNA structure when present during cryopreservation of equine spermatozoa,. We hypothesized that the addition of iodixanol would improve post-thaw values for measured parameters. Ejaculates from six stallions were collected, centrifuged at 900 x g for ten minutes to remove supernatant, and suspended to 200 x 106 cells/ml with 0%, 2.5% and 5% iodixanol in an egg-yolk based extender and cryopreserved. Arrhythmogenic cardiomyopathy (AC), also known as arrhythmogenic right ventricular cardiomyopathy (ARVC), is a disease characterized by fatty or fibro-fatty myocardial replacement, predominantly in the right ventricle and to a lower extent the left ventricle. It is recognized as a disease affecting the cardiac intercalated disc. Clinically, it is associated with ventricular arrhythmias, although atrial arrhythmias and atrial histopathological changes characteristic of AC have occasionally been reported. Extrinsic and environmental factors are known to affect the transmission of arthropod-borne viruses (arboviruses), including variations in the arthropod vector populations. Differences among these factors have been associated with differential transmission and are sometimes used to control the spread of an arbovirus through a vertebrate population in an effort to prevent or disrupt an outbreak. Objective -To compare the compression produced in reduction of experimentally created medial condylar fractures using lag screw fixation with medial vs. lateral approach, and to determine the maximum torque at screw failure. Materials and Methods- Twelve (12) pairs (left and right) 3rd metacarpal bones (MC3) were collected from adult (2-7 years) Thoroughbreds euthanized for reasons unrelated to orthopedic disease. Accurately defining disease distributions and calculating disease risk is an important step in the control and prevention of diseases. This study used geographical information systems and remote sensing technologies within the MaxEnt ecological niche modeling program to create predictive risk maps for leprosy and Schistosomiasis in Brazil and Chagas disease in both Brazil and Bolivia. New disease cases were compiled for leprosy, Schistosomiasis, and Chagas disease from the Brazilian ministry of Health for 2001 to 2009 and the data was stratified to a 10,000 population for each municipality. There are many contradictory reports in the literature involving the culture of Spiroplasma mirum, its resistance to disinfectants and antibiotics, and its potential role in Transmissible Spongiform Encephalopathies (TSEs). Edwardsiella ictaluri encodes a urease operon and an arginine decarboxylase (AdiA) that are required for virulence in head kidney derived macrophages (HKDM). The urease produces ammonia in amounts sufficient to alter environmental pH from acid to neutral. A hypothetical model was proposed, involving arginine metabolism in E. ictaluri infected HKDM, focusing on bacterial urease, AdiA, a second arginine decarboxylase (SpeA), and agmatinase (SpeB). Using fluorescence based ratiometric pH determination of E. ictaluri in live HKDM, it was shown that E. ictaluri modulates HKDM phagosome pH to above six. Lyme disease is a multisystem disorder caused by the tick vector Borrelia burgdorferi. During its life cycle between the tick vector and the mammalian host, Borrelia up- and down-regulates the expression of its surface lipoproteins. Of its many surface lipoproteins, the Outer surface protein C (OspC) is crucial for initial mammalian infection. OspC has a common role shared with other lipoproteins of protection against host innate defences and a unique function in facilitating the dissemination of B. burgdorferi in the murine host. Methadone is a mu-opioid receptor agonist which is a very effective analgesic used to treat moderate to severe acute and chronic pain in humans. Due to methadone’s minimal undesirable side-effects in people, we believed it could be of use in horses as an analgesic agent. As found with the majority of lipophilic drugs, absorption of methadone occurs primarily in the small intestine via transcellular transport and its absorption is regulated by P-glycoprotein. P-glycoprotein is a transmembrane transporter protein encoded by the multidrug resistance gene, which is constitutively expressed in the apical membrane of enterocytes of various species. To compare insertion temperatures and torques of Hydroxyapatite (HA) coated and uncoated 5.5 mm AO cortical screws in equine third metatarsal bones (MTIII) in vitro, and to compare insertion and extraction torques of HA coated and uncoated screws after 4, 8, 12 and 16 weeks of healing in equine third metacarpal bones (MCIII) in vivo. No significant temperature differences were recorded in cadaveric bones for AO and HA coated screws. Insertion torques were significantly higher for HA coated implants compared to uncoated screws. Objectives 1) To measure conductance and permeability of canine colonic mucosa exposed to increasing concentrations of carprofen. 2) To compare conductance and permeability of canine colonic mucosa exposed to carprofen or 2,4-dinitrophenol (DNP) and tempol blockade. Design In vitro randomized block design Animal 20 mixed breed dogs Methods Conductance, mannitol flux, and histology were evaluated in colonic mucosa mounted in Ussing chambers. Mucosa was first exposed to increasing concentrations of carprofen. Mucosa was then exposed to either carprofen (200 μg/ml) or DNP (0.25mM) +/- tempol (1mM) pretreatment.
