Cloning And Transcriptional Activation Of Thevitamin D Receptor (Amphibians)1, Meral Güzey Jan 1999

Cloning And Transcriptional Activation Of Thevitamin D Receptor (Amphibians)1, Meral Güzey

Turkish Journal of Medical Sciences

The vitamin D receptor (VDR), a member of the nuclear receptor superfamily, is an integral part of the body's calcium regulatory system. Mammalian and avian VDR genes were isolated, sequenced and characterized. In this study we cloned the vitamin D receptor from amphibians. We received a cDNA library prepared from amphibian (frog) tissues and prepared a cDNA probe based on the avian VDR. We screened the library and located the positive clones. Complete structure of VDR gene has been compared with other species and concluded that it shares 79%, 73%, 73% and 75% identity at the aminoacid level with the …

Characterization Of Catalytic Phenotype Of \Beta-Galactosidasefrom Laci Mutant, E. Coli Csh-36,As A Tool For The Management Of Lactoseintolerance, Ayşegül Ayyildiz

Turkish Journal of Medical Sciences

Objectives: Lactose intolerance is a common public health problem with 5-100% prevalance among different populations of the world. Use of \beta-galactosidase to reduce the lactose content in food or consumption of special products of milk or exogeneous lactase enzyme are among the approaches taken to manage lactose intolerance. Recombinant \beta -galactosidase as well as \beta -galactosidase derived from different sources have variances in their bioefficiency. Thermodynamic properties of \beta -galactosidase determines the industrial efficiency. In this \beta -galactosidase from E. coli CSH-36, a constitutive expressor of enzyme was investigated for enzyme biofunctionality. Methods: The enzyme was purified by cell lysis, …

The Relationship Between Reactivity Of Metabolites Of Pyrrolizidine Alkaloids And Extrahepatic Toxicity, R. A. Cooper, R. J. Huxtable Dec 1998

The Relationship Between Reactivity Of Metabolites Of Pyrrolizidine Alkaloids And Extrahepatic Toxicity, R. A. Cooper, R. J. Huxtable

Roland A. Cooper

Pyrrolizidine alkaloids (PAs) are a large group of structurally similar toxins. In animals, including man, they are hepatotoxic and in some cases pneumo- and neurotoxic. PAs are metabolized by the liver P450 system to reactive dehydroalkaloid (DHA) intermediates. PA toxicity is a result of alkylation of macromolecules by DHAs. We have measured the relative reactivity of a series of semi-synthetic DHAs by recording the rate at which they alkylate a model nucleophile, 4-(p-nitrobenzyl)pyridine. Rate data fit mono- or biexponential equations. Rank order of reactivity for the macrocyclic and open ester DHAs was the same as those measured for DHA hydrolysis. …

Factors Affecting The Circumcision Decision, Jeffrey D. Tiemstra Md Dec 1998

Factors Affecting The Circumcision Decision, Jeffrey D. Tiemstra Md

Jeffrey Tiemstra, MD, FAAFP

BACKGROUND: Studies from more than 10 years ago have shown that most parents choose circumcision for their infant sons for nonmedical reasons. Since then a wealth of data has accumulated on the relative risks and benefits of the procedure, although the medical community remains divided on the appropriateness of the procedure. Whether the ongoing research effort and medical debate have had an effect on parental decision making is unknown.

METHODS: A survey of parents of 55 male infants was conducted to determine when the parents made their decision regarding circumcision, whether discussions with providers affected their decisions, and what the …
