Training Materials on Antimicrobial Resistance
This is a guide to resources that are available to assist laboratories with the preparation and dissemination of their antimicrobial susceptibility testing data in a format known as an "antibiogram" as an aid to antimicrobial stewardship activities in their hospital. There are three color-coded pathways in the slide deck: Pathway A for laboratories with no experience with antibiograms; Pathway B for laboratories with some experience that want to improve and expand the use of their antibiograms; and Pathway C for experienced laboratories that want to share their data with external surveillance systems.

Respiratory human adenovirus (HAdV) has been closely linked with acute respiratory infections such as pharyngitis and coryza (common cold). While some HAdV serotypes antagonize the immune system leading to meningitis, gastroenteritis, and acute hemorrhagic cystitis among other conditions, HAdV type 3 (HAdV3) predominantly infects the upper respiratory tract. Studies have shown that the release of small, membrane-derived extracellular vesicles (EVs), termed exosomes, may offer a mechanism by which certain viruses, enter cells via receptor-independent entry. In humans and animals, probiotics are widely accepted as crucial for host health and growth. The investigation of the probiotic colonization and expression of probiotics in the host is beneficial for proper usage of probiotics and isolation of indigenous probiotics. COVID-19 has caused over a million deaths in the United States. Preventative measures such as masks, social distancing, and vaccination have been implemented to fight against the disease. Unfortunately, a large subset of Americans opposed these efforts.
SC DHEC reports that as of January 2023, only 53.9 % of eligible South Carolinians have been fully vaccinated against COVID-19. Previous studies have shown vaccine hesitancy is influenced largely by personal concerns about vaccine development, peer vaccination status, and general skepticism of immunization.
Hesitancy towards pediatric vaccination is particularly concerning because unvaccinated children can act as vessels to exponentially accelerate COVID-19 transmission. …

Eosinophilic Esophagitis (EoE), a chronic inflammatory condition of the esophagus, characterized by esophageal dysfunction and eosinophilic inflammation. Current treatments for EoE include dietary elimination and swallowed steroids. Recently, Dupixent was approved for the treatment of EoE, but it has side effects such as eye inflammation and eczema. Traditional Chinese Medicine (TCM) practice has been used to improve GI symptoms in EoE patients. Two key active ingredients of a TCM formula, 7,4’-Dihydroxyflavone (DHF) and Berberine (BBR), have shown remarkable results in treating EoE.

This study aimed to assess the in vitro antiplasmodial and cytotoxic activities of ethanol extracts from the root barks, leaves and stem barks of seven medicinal plant species growing in Tanzania. The antiplasmodial activities of extracts against chloroquine-sensitive (3D7) and multidrug-resistant (Dd2) strains of Plasmodium falciparum were determined using a SYBR green I-based fluorescence assay, while the cytotoxicity of active extracts was determined against the human HepG2 cell line using a resazurin-based assay. The complex two-membrane organization of the envelope of Gram-negative bacteria imposes an unique biosynthetic and topological constraints that can affect translocation of lipids and proteins synthesized on the cytoplasm facing leaflet of the cytoplasmic (inner) membrane (IM), across the IM and between the IM and outer membrane (OM). Balanced growth of two membranes and continuous loss of phospholipids in the periplasmic leaflet of the IM as metabolic precursors for envelope components and for translocation to the OM requires a constant supply of phospholipids in the IM cytosolic leaflet. Colonization with multidrug-resistant organisms (MDROs) is a risk factor for subsequent infection. Surveillance for MDROs, including methicillin-resistant Staphylococcus aureus, vancomycin-resistant enterococci, extended-spectrum beta-lactamase-producing Enterobacterales, and carbapenemase-producing organisms, is commonly conducted in hospitals to prevent spread of MDROs, in part to reduce the potential for additional infections. Although colonization is a risk factor for infection, data on colonization with various MDROs are often not considered when selecting anti-infective therapy. There are conflicting data on the strength of the positive and negative predictive values of the colonization test results to guide therapeutic strategies. We describe here a genome-wide screening approach to identify the most critical core reaction among a network of many that are supported by an essential gene to establish cell viability. We describe steps for maintenance plasmid construction, knockout cell construction, and phenotype validation. We then detail isolation of suppressors, whole-genome sequencing analysis, and reconstruction of CRISPR mutants. We focus on E. coli trmD, which encodes an essential methyl transferase that synthesizes m1G37 on the 3'-side of the tRNA anticodon. Most organisms have circadian clocks to ensure the metabolic cycle to resonate with the rhythmic environmental changes without “damping,” or losing robustness. Cyanobacteria is the oldest and simplest form of life that is known to harbor this biological intricacy. Its KaiABC-based central oscillator proteins can be reconstituted inside a test tube, and the post-translational modification cycle occurs with 24 h periodicity. KaiC’s two major phosphorylation sites, Ser-431 and Thr-432, become phosphorylated and dephosphorylated by interacting with KaiA and KaiB, respectively. Staphylococcus saprophyticus ICBB 9554 and Citrobacter freundii ICBB 9763 are exoelectrogen bacteria applied as microbial fuel cells (MFC). We selected enrichment media for both these exoelectrogen bacteria, characterized their synergistic traits, and evaluated the growth conditions under different ecological factors. In this study, different enrichment media, such as those containing sugar, molasses, and palm sugar (2, 4, and 6% w/v) were tested for S. saprophyticus ICBB 9554. Aging is accompanied by increased susceptibility to infections including with viral pathogens resulting in higher morbidity and mortality among the elderly. Significant changes in host metabolism can take place following virus infection. Efficient immune responses are energetically costly, and viruses divert host molecular resources to promote their own replication. Objective: To determine if bacteria (Ureaplasma parvum and Sneathia spp.) associated with intra-amniotic infection can trigger the induction of cytokine Thymic stromal lymphopoietin (TSLP) in human amnion epithelial cells (hAECs) in vitro.
Material or subjects: Amniotic fluid and chorioamniotic membrane (CAM) were collected from women with sPTL who delivered at term (n=30) or preterm without intra-amniotic inflammation (n=34), with sterile intra-amniotic inflammation (SIAI, n=27), or with intra-amniotic infection (IAI, n=17). The consumer demand for fresh produce (vegetables and fruits) has considerably increased since the 1980s for more nutritious foods and healthier life practices, particularly in developed countries. Currently, several foodborne outbreaks have been linked to fresh produce. Digital holographic microcopy (DHM) is a label-free technique that has gained attention in recent years as a tool for volumetric imaging. One application of DHM is for the study of microbial motility with the advantage being that organisms may freely move within their environment. Images created from DHM are in the form of holograms. Holograms are time recordings showing XY information with the Z information contained within. Z information can be retrieved from the holograms directly through a variety of numerical techniques or through reconstruction. Individuals experiencing homelessness are at increased risk of suffering from infectious diseases. This is due to a number of social factors and healthcare disparities, as well as the idea of syndemics, by which diseases cluster together to worsen disease burden. Current intervention strategies approach treatment from a post-infection perspective, but reducing transmission rates of infectious diseases within the population of people experiencing homelessness will require a shift in the healthcare framework. Mitochondrial dysfunction is an early event in the pathogenesis of neurologic disorders and aging. Sirtuin 3 (SIRT3) regulates mitochondrial function in response to the cellular environment through the reversible deacetylation of proteins involved in metabolism and reactive oxygen species detoxification. As the primary mitochondrial deacetylase, germline, or peripheral tissue-specific deletion of SIRT3 produces mitochondrial hyperacetylation and the accelerated development of age-related diseases. Given the unique metabolic demands of neurons, the role of SIRT3 in the brain is only beginning to emerge. Fish is one of the most important nutrient sources of the human diet, and its low-cost aquaculture production is advantageous. During processing, large quantities of organic and inorganic nutrients are produced. Discharging fish farming wastewater to the environment without proper treatment can support harmful algal growth. Therefore, aquaculture effluent must be treated prior to discharge to the receiving waters. In this study, six different cyanobacteria and algae species, including Chlorococcumminutum, Porphyridiumcruentum, Chlorella vulgaris, Microcystis aeruginosa, Chlamydomonas reinhardtii, and Fischerellamuscicola were studiedto treatfish aquaculture wastewater, using mix-culture bacteria as the inoccolum. The sensitivity and reliability of Tunable Diode Laser Absorption Spectroscopy (TDLAS) for monitoring the growth of microorganisms in real food matrices was explored. The rapid and non-destructive TDLAS equipment uses laser light to monitor carbon dioxide changes due to microbial growth in the container head space which has the additional advantage of detecting contamination in products and packaging where visual inspection is not possible. TDLAS growth detection of Bacillus fengqiuensis, Candida albicans, Lactococcus lactis, Microbacterium luteolum, Paenibacillus chitinolyticus and Staphylococcus pasteuri was studied in various Ready To Feed (RTF) infant formula products. The dimethyl sulfoxide reductase (or MopB) family is a diverse assemblage of enzymes found throughout Bacteria and Archaea. Many of these enzymes are believed to have been present in the last universal common ancestor (LUCA) of all cellular lineages. However, gaps in knowledge remain about how MopB enzymes evolved and how this diversification of functions impacted global biogeochemical cycles through geologic time. In this study, we perform maximum likelihood phylogenetic analyses on manually curated comparative genomic and metagenomic data sets containing over 47,000 distinct MopB homologs. All microscope imaging is largely affected by inherent resolution limitations because of out-of-focus light and diffraction effects. The traditional approach to restoring the image resolution is to use a deconvolution algorithm to “invert” the effect of convolving the volume with the point spread function. However, these algorithms fall short in several areas such as noise amplification and stopping criterion. In this paper, we try to reconstruct an explicit volumetric representation of the fluorescence density in the sample and fit a neural network to the target z-stack to properly minimize a reconstruction cost function for an optimal result. Neutron activation analysis was used in this research to measure the concentrations of aluminum (Al), bromine (Br), chlorine (Cl), mercury (Hg), arsenic (As), and thorium (Th) in soil and saffron plants as well as their contamination, enrichment, and translocation rates. The edible part of the saffron showed a higher Hg concentration than the WHO/FAO-permitted level in the urban area. The soil was not intoxicated and contaminated by Al, As, and Th except in the urban areas, which were slightly polluted by As. The calculated contamination degree and pollution load index indicated that the soil was almost uncontaminated. The enrichment factors …

Armyworms are polyphagous insect pests of agricultural crops. Their population is usually managed below threshold level using insecticides. However, a single control measure is insufficient to attain a sustainable pest management. Hence, this research was conducted to assess the three species of entomopathogenic fungi as potential biological control agents against Mythimna separata (Walker) or paddy armyworm. Among insects, Wolbachia is an exceedingly common bacterial endosymbiont with a range of consequences of infection. Despite the frequency of Wolbachia infection, very little is known about this bacteria's diversity and role within hosts, especially within ant hosts. In this study, we analyze the occurrence and diversity of Wolbachia across the spiny ants (Polyrhachis), a large and geographically diverse genus. Polyrhachis samples from throughout the host genus' phylogenetic and biogeographical range were first screened for single infections of Wolbachia using the wsp gene and Sanger sequencing.
