Effects Of Nanosecond Pulse Electric Fields On Cellular Elasticity, Diganta Dutta, Anthony Asmar, Michael W. Stacey
Effects Of Nanosecond Pulse Electric Fields On Cellular Elasticity, Diganta Dutta, Anthony Asmar, Michael W. Stacey
Bioelectrics Publications
We investigated the effects of a single 60 nanosecond pulsed electric field (nsPEF) of low (15 kV/cm) and high (60 kV/cm) field strengths on cellular morphology and membrane elasticity in Jurkat cells using fluorescent microscopy and atomic force microscopy (AFM). We performed force displacement measurements on cells using AFM and calculated the Young's modulus for membrane elasticity. Differential effects were observed depending upon pulsing conditions. We found that a single nsPEF of low field strength did not induce any apparent cytoskeletal breakdown and had minor morphological changes. Interestingly, force measurements and calculation of Young's modulus showed a significant decrease in …

Bubbles induced by blast waves or shocks are speculated to be the major cause of damages in biological cells in mild traumatic brain injuries. Microbubble collapse was found to induce noticeable cell detachment from the cell substrate, changes in focal adhesion and biomechanics. To better understand the bubble mechanism, we would like to construct a system, which allows us to clearly differentiate the impact of bubbles from that of shocks. Such a generator needs to be low profile in order to place under a microscope. A piezoelectric transducer system was designed to meet the need. The system uses either a …

We tested if picosecond electric pulses (psEP; 190 kV/cm, 500 ps at 50% height), which are much shorter than channel activation time, can activate voltage-gated (VG) channels. Cytosolic Ca2+ was monitored by Fura-2 ratiometric imaging in GH3 and NG108 cells (which express multiple types of VG calcium channels, VGCC), and in CHO cells (which express no VGCC). Trains of up to 100 psEP at 1 kHz elicited no response in CHO cells. However, even a single psEP significantly increased Ca2+ in both GH3 (by 114 +/- 48 nM) and NG108 cells (by 6 +/- 1.1 nM). Trains of …

Ca2+ activation and membrane electroporation by 10-ns and 4-ms electric pulses (nsEP and msEP) were compared in rat embryonic cardiomyocytes. The lowest electric field which triggered Ca2+ transients was expectedly higher for nsEP (36 kV/cm)than forms EP (0.09 kV/cm) but the respective doses were similar (190 and460 mJ/g). At higher intensities, both stimuli triggered prolonged firing in quiescent cells. An increase of basal Ca2+ level by N10 nM in cells with blocked voltage-gated Ca2+ channels and depleted Ca2+ depot occurred at 63 kV/cm (nsEP) or 0.14 kV/cm (msEP) and was regarded as electroporation threshold. These …

Engaging Community College Students Using An Engineering Learning Community, James Maccariella Jr.
Educational Leadership & Workforce Development Theses & Dissertations
The study investigated whether community college engineering student success was tied to a learning community. Three separate data collection sources were utilized: surveys, interviews, and existing student records. Mann-Whitney tests were used to assess survey data, independent t-tests were used to examine pre-test data, and independent t-tests, analyses of covariance (ANCOVA), chi-square tests, and logistic regression were used to examine post-test data. The study found students that participated in the Engineering TLC program experienced a significant improvement in grade point values for one of the three post-test courses studied. In addition, the analysis revealed the odds of fall-to-spring retention were …

CsK2Sb photocathodes with quantum efficiency on the order of 10% at 532 nm, and lifetime greater than 90 days at low voltage, were successfully manufactured via co-deposition of alkali species emanating from an effusion source. Photocathodes were characterized as a function of antimony layer thickness and alkali consumption, inside a vacuum chamber that was initially baked, but frequently vented without re-baking. Photocathode lifetime measured at low voltage is correlated with the antimony layer thickness. Photocathodes manufactured with comparatively thick antimony layers exhibited the best lifetime. We speculate that the antimony layer serves as a reservoir, or sponge, for …

The best capabilities are usually achieved by having the latest technologies in defense systems. However, including the new, usually immature, technologies in a system design does not always easily result in achieving the capabilities at the right level, at an affordable cost, and in a timely manner. Many programs have suffered from immature technologies as cost overruns, late or no deliveries, and poor performance levels. Another impact of technology selection appears as obsolescence after the deployment of systems, or even before the deployment of the system. As the technologies of a system become obsolete, the cost of maintaining the system …

This paper examines the challenges and practice implications for Complex System Governance (CSG). CSG is presented as an emerging field focused on the design, execution, and evolution of the higher order (metasystem) functions necessary to provide control, communication, coordination, and integration of a complex system. This paper is focused on three primary objectives. First, we introduce the complex system problem domain that the CSG field is being designed to address. The pervasiveness of this problem domain is demonstrated by a short examination of the water utilities sector. Second, we expound the nature of CSG and an emerging reference model …

Climate change concerns have confronted energy policy makers by unprecedented challenges in the 21st century. Revolution of renewable energy technologies, as well as more efficient energy systems, has been promising in the context of global warming. However, these technologies are not maturing and chaning. Consequently planning for development of these resources requires dealing with various multidisciplinary research questions such as financial feasibility of renewable energy projects. Nevertheless, there is considerable lack of education programs offering multidisciplinary approach for addressing the current energy challenges. Based on the 21st evolving energy landscape, an interdisciplinary graduate certificate course work was designed at Old …

The Space Infrastructure Vulnerability Cadastre: Orbital Debris Critical Loads, Adrian V. Gheorghe, Daniel E. Yuchnovicz
Engineering Management & Systems Engineering Faculty Publications
Orbital debris from over 50 years of human activity in space are threatening the operations of existing and future satellites and the sustainability of high-value satellite orbits. This technical memorandum calls for the development of a cadastre that depicts the vulnerability of critical satellite orbits to accumulating orbital debris. A space infrastructure vulnerability cadastre could serve as a governance tool for use by developers and operators of critical space infrastructures to better communicate the current and future vulnerability of high-value orbits to the accumulation of orbital debris. These high-value orbits are susceptible to "loss" for decades or centuries if generation …

Heating Based Model Analysis For Explosive Emission Intitiation At Metal Cathodes, A. Majzoobi, R. P. Joshi, A. Neuber, J. Dickens
Electrical & Computer Engineering Faculty Publications
This contribution presents a model analysis for the initiation of explosive emission; a phenomena that is observed at cathode surfaces under high current densities. Here, localized heating is quantitatively evaluated on ultrashort time scales as a potential mechanism that initiates explosive emission, based on a two-temperature, relaxation time model. Our calculations demonstrate a strong production of nonequilibrium phonons, ultimately leading to localized melting. Temperatures are predicted to reach the cathode melting point over nanosecond times within the first few monolayers of the protrusion. This result is in keeping with the temporal scales observed experimentally for the initiation of explosive emission.

Particle-in-cell simulations are performed to analyze the efficiency, output power and leakage currents in a 12-Cavity, 12-Cathode rising-sun magnetron with diffraction output (MDO). The central goal is to conduct a parameter study of a rising-sun magnetron that comprehensively incorporates performance enhancing features such as transparent cathodes, axial extraction, the use of endcaps, and cathode extensions. Our optimum results demonstrate peak output power of about 2.1 GW, with efficiencies of ~70% and low leakage currents at a magnetic field of 0.45 Tesla, a 400 kV bias with a single endcap, for a range of cathode extensions between 3 and 6 centimeters.

The Inverse Piezoelectric Effect (IPE) is thought to contribute to possible device failure of GaN High Electron Mobility Transistors (HEMTs). Here we focus on a simulation study to probe the possible mitigation of the IPE by reducing the internal electric fields and related elastic energy through the use of high-k materials. Inclusion of a HfO2 "cap layer" above the AlGaN barrier particularly with a partial mesa structure is shown to have potential advantages. Simulations reveal even greater reductions in the internal electric fields by using "field plates" in concert with high-k oxides

A spark discharge is coupled to a laser multicharged ion source to enhance ion generation. The laser plasma triggers a spark discharge with electrodes located in front of the ablated target. For an aluminum target, the spark discharge results in significant enhancement in the generation of multicharged ions along with higher charge states than observed with the laser source alone. When a Nd:YAG laser pulse (wavelength 1064 nm, pulse width 7.4 ns, pulse energy 72 mJ, laser spot area on target 0.0024 cm2) is used, the total multicharged ions detected by a Faraday cup is 1.0 nC with …

Cu (In,Ga,Al)Se2 (CIGAS) thin films were studied as an alternative absorber layer material to Cu(InxGa1-x)Se2. CIGAS thin films with varying Al content were prepared by magnetron sputtering on Si(100) and soda-lime glass substrates at 350 °C, followed by postdeposition annealing at 520 °C for 5 h in vacuum. The film composition was measured by an electron probe microanalyzer while the elemental depth profiles were determined by secondary ion mass spectrometry. X-ray diffraction studies indicated that CIGAS films are single phase with chalcopyrite structure and that the (112) peak clearly shifts to higher 2θ …

Multi-Scale Visualization Of Molecular Architecture Using Real-Time Ambient Occlusion In Sculptor, Manuel Wahle, Willy Wriggers
Mechanical & Aerospace Engineering Faculty Publications
The modeling of large biomolecular assemblies relies on an efficient rendering of their hierarchical architecture across a wide range of spatial level of detail. We describe a paradigm shift currently under way in computer graphics towards the use of more realistic global illumination models, and we apply the so-called ambient occlusion approach to our opensource multi-scale modeling program, Sculptor. While there are many other higher quality global illumination approaches going all the way up to full GPU-accelerated ray tracing, they do not provide size-specificity of the features they shade. Ambient occlusion is an aspect of global lighting that offers great …

Modeling, Dynamics And Control Of Spacecraft Relative Motion In A Perturbed Keplerian Orbit, Mohamed Okasha, Brett Newman
Mechanical & Aerospace Engineering Faculty Publications
The dynamics of relative motion in a perturbed orbital environment are exploited based on Gauss' and Cowell's variational equations. The inertial coordinate frame and relative coordinate frame (Hill frame) are used, and a linear high fidelity model is developed to describe the relative motion. This model takes into account the primary gravitational and atmospheric drag perturbations. Then, this model is used in the design of a navigation, guidance, and control system of a chaser vehicle to approach towards and to depart from a target vehicle in proximity operations. Relative navigation uses an extended Kalman filter based on this relative model …

Viscoelastic Effects On Electrokinetic Particle Focusing In A Constricted Microchannel, Xinyu Lu, John Dubose, Sang Woo Joo, Shizhi Qian, Xiangchun Xuan
Mechanical & Aerospace Engineering Faculty Publications
Focusing suspended particles in a fluid into a single file is often necessary prior to continuous-flow detection, analysis, and separation. Electrokinetic particle focusing has been demonstrated in constricted microchannels by the use of the constriction-induced dielectrophoresis. However, previous studies on this subject have been limited to Newtonian fluids only. We report in this paper an experimental investigation of the viscoelastic effects on electrokinetic particle focusing in non-Newtonian polyethylene oxide solutions through a constricted microchannel. The width of the focused particle stream is found NOT to decrease with the increase in DC electric field, which is different from that in Newtonian …

High-occupancy toll (HOT) lanes—an increasingly popular solution for congested roadway networks—give drivers the option to access express lanes. The cost of entry often varies with demand, although no standard method of optimizing these price points exists. Using the principles of a Vickrey auction that incentivizes true-value bids, this paper proposes a tolling system that uses vehicle-to-infrastructure technology to optimize toll operator revenue with HOT lane usage. In the scenario, a roadway network consists of a HOT lane and a general-purpose lane, each with identical physical properties. Drivers can access the HOT lane at the start of the facility or at …

Proton-Electrostatic Localization: Explaining The Bioenergetic Conundrum In Alkalophilic Bacteria, James Weifu Lee
Chemistry & Biochemistry Faculty Publications
The decades-longstanding energetic conundrum of alkalophilic bacteria as to how they are able to synthesize ATP has now, for the first time, been clearly solved using the proton-electrostatics localization hypothesis. This is a major breakthrough advance in understanding proton-coupling bioenergetics over the Nobel-prize work of Peter Mitchell’s chemiosmotic theory. The widespread textbook Mitchellian proton motive force (pmf) equation has now been significantly revised. Use of the newly derived equation results in an overall pmf value (215~233 mV) that is more than 4 times larger than that (44.3 mV) calculated from the Mitchellian equation for the alkalophilic bacteria growing at pH …

Several modified versions of the Monod model have been proposed to simulate algal dynamics in lakes by keeping the parent model's advantages of simplicity and low data requirement. This study evaluated the performance of a widely-used modified Monod model in predicting algal dynamics at various time scales in Lake Tai, a typical shallow lake in east China, using multiple time series. Chlorophyll-a (Chl-a) concentration was used as a surrogate for algal (CyanoHABs: cyanobacterial harmful algal blooms) growth and the independent variables were total nitrogen (TN), total phosphorus (TP), and either water temperature or air temperature. The evaluation indicated that the …

Failure Modes For I-Section Gfrp Beams, Mamadou Konate, Zia Razzaq
Civil & Environmental Engineering Faculty Publications
This paper presents calculations for the failure modes for I-section Glass Fiber Reinforced Polymer (GFRP) beams with single mid-span web brace. Theoretical predictions are made using ASCE-LFRD Pre-Standard for FRP structures. For the member length considered, it is found that for small and medium I-sections the failure mode is governed by lateral-torsional buckling and for bigger I-sections the failure mode is governed by material rupture. The outcome of the predicted lateral-torsional buckling mode is compared with that observed experimentally.

Obtaining exascale performance is a challenge. Although the technology of today features hardware with very high levels of concurrency, exascale performance is primarily limited by energy consumption. This limitation has lead to the use of GPUs and specialized hardware such as many integrated core (MIC) co-processors and FPGAs for computation acceleration. The Intel Xeon Phi co-processor, built upon the MIC architecture, features many low frequency, energy efficient cores. Applications, even those which do not saturate the large vector processing unit in each core, may benefit from the energy-efficient hardware and software of the Xeon Phi. This work explores the energy …
Adaptive Double Chain Quantum Genetic Algorithm For Constrained Optimization Problems, Kong Haipeng, Li Ni, Yuzhong Shen
Adaptive Double Chain Quantum Genetic Algorithm For Constrained Optimization Problems, Kong Haipeng, Li Ni, Yuzhong Shen
Computational Modeling & Simulation Engineering Faculty Publications
Optimization problems are often highly constrained and evolutionary algorithms (EAs) are effective methods to tackle this kind of problems. To further improve search efficiency and convergence rate of EAs, this paper presents an adaptive double chain quantum genetic algorithm (ADCQGA) for solving constrained optimization problems. ADCQGA makes use of double-individuals to represent solutions that are classified as feasible and infeasible solutions. Fitness (or evaluation) functions are defined for both types of solutions. Based on the fitness function, three types of step evolution (SE) are defined and utilized for judging evolutionary individuals. An adaptive rotation is proposed and used to facilitate …

Counting And Classification Of Highway Vehicles By Regression Analysis, Mingpei Liang, Xinyu Huang, Chung-Hao Chen, Alade Tokuta
Electrical & Computer Engineering Faculty Publications
In this paper, we describe a novel algorithm that counts and classifies highway vehicles based on regression analysis. This algorithm requires no explicit segmentation or tracking of individual vehicles, which is usually an important part of many existing algorithms. Therefore, this algorithm is particularly useful when there are severe occlusions or vehicle resolution is low, in which extracted features are highly unreliable. There are mainly two contributions in our proposed algorithm. First, a warping method is developed to detect the foreground segments that contain unclassified vehicles. The common used modeling and tracking (e.g., Kalman filtering) of individual vehicles are not …

System errors, both mechanical and human in nature, can have a grave effect on aircrew judgement in flight. The effects of these errors can be massively compounded during emergency situations. Crew Resource Management (CRM) is an important process aircrews can utilize to minimize risks and enhance assessments. The employment of this technique can be validated by aviation mishaps over the last three decades and how system errors increased the probability of the incident occurring. Suggestions can be made to further prevent similar accidents from occurring in the future utilizing historical aeronautical records. This paper outlines an approach by which systems …

We present in this paper a simulation-based approach to evaluate the risk associated with supply chain disruptions caused by failures in some supply chains nodes and measure the impact of such disruptions on supply chain key performance measures (KPIs) of interest. The proposed framework enables analysts and managers to repeatedly assess the risk to their supply chains based on various simulated scenarios and identify the most critical nodes whose disruption will have the highest impact on the KPIs of interest. As a result, companies can focus on the most critical supply chain assets and develop targeted mitigation plans that minimize …
