Developing a computational model to model cardiac activity has been increasingly important in recent decades. Accurate cell-level active tension modeling for cardiomyocytes is critical to understanding cardiac functionality on a patient-specific basis and developing an effective in-silico cardiac model. However, cell-level models in the literature fail to account for viscoelasticity and inter-patient variations in active tension. This research proposes a genetic algorithm-optimized, fractional order system to model cell-level active tension by extending Land’s state-of-the-art model of cardiac contraction. The advancement of additive manufacturing technologies has opened new avenues for fabricating biocompatible and structurally functional bone tissue scaffolds, essential in treating osseous fractures, defects, and diseases. This research aims to develop mechanically strong, dimensionally precise, and patient-specific porous bone tissue scaffolds that provide both structural integrity and biological functionality. Through three interconnected studies, several key challenges in the fabrication of these structures using the PME additive manufacturing process are addressed. An accurate representation of cellular mechanobiology necessitates the inclusion of subcellular elements characterized by minute masses and dimensions. These minute objects yield multiscale dynamic models with disproportionate terms, which require inordinate amounts of computational time to simulate. The computational requirements limit the time span of the simulation to time histories shorter than one second, even when employing supercomputers. This work presents a high-speed approach to simulating the mechanobiology of stem cells. Hydrogels, with physical characteristics resembling biological soft tissues, hold great potential for bioinspired and biomedical applications. However, using them in additive manufacturing systems has challenges because of their poor mechanical and rheological properties. In response to these challenges, we introduce a fugitive carrier method, a novel and generalizable approach for materials incompatible with pneumatic-based extrusion three-dimensional (3D) printers. Our developed fugitive carrier method allows the 3D printing of a broad spectrum of hydrogel precursors with low viscosity while retaining their inherent properties. Electrospun polymeric biodegradable scaffolds are essential in tissue engineering, particularly for Engineered Tissue Vascular Grafts (ETVGs), which promise advancements in treating coronary artery disease, peripheral arterial disease, congenital cardiovascular defects, and renal disease. These scaffolds present a solution to issues with autologous graft availability and durability. While large-diameter grafts in low-pressure environments have seen success, small-diameter grafts in high-flow scenarios remain challenging. Understanding polymeric scaffold degradation and behavior during incubation, especially under dynamic mechanical loading, is vital for clinical translation of small-caliber ETVGs.

The organization of myofibers and extra cellular matrix within the myocardium plays a significant role in defining cardiac function. When pathological events occur, such as myocardial infarction (MI) or hypertrophic cardiomyopathy (HCM), this organization can become disrupted, leading to alteration in pumping performance. The current study proposes a multiscale finite element (FE) framework to determine realistic fiber distributions in the left ventricle (LV). Contemporary total knee arthroplasty (TKA) has not fully restored natural patellofemoral (P-F) mechanics across the patient population. Previous experimental simulations have been limited in their ability to create dynamic, unconstrained, muscle-driven P-F articulation while simultaneously controlling tibiofemoral (T-F) contact mechanics. The purpose of this study was to develop a novel experimental simulation and validate a corresponding finite element model to evaluate T-F and P-F mechanics. Non-enzymatic glucose sensing holds promise to overcome limitations associated with glucose oxidase, such as oxygen dependence and short shelf life. This study explores the potential sensing capabilities of borophene and graphene through direct interaction with various compounds, including β-glucose, uric acid, ascorbic acid, fructose, and acetaminophen. Using Density Functional Theory (DFT), we calculated binding energies and the respective Density of States (DOS) for these adsorbates on both graphene and borophene surfaces. Preliminary results suggest that borophene might exhibit nearly twice the affinity for β-glucose compared to graphene. Concussion assessment in athletes has gained prominence due to potential long-term consequences of traumatic brain injuries. Evaluating balance alterations is crucial for understanding post-injury motor control strategies. This study introduces a novel approach to understanding center of pressure (COP) dynamics during quiet stance tasks for assessing balance impairments in athletes recovering from concussion. Concussed athletes often experience impaired motor function and cognitive deficits, increasing the risk of orthopedic injury. Traditional balance assessments focus on total COP (COPt), overlooking nuanced hip and ankle mechanisms. This dissertation focused on modeling specimen-specific soft tissue structures in the context of joint replacement surgery. The research addressed four key aspects. The first study involved developing a workflow for creating finite element models of the hip capsule to replicate its torque-rotational response. Experimental data from ten cadaveric hips were used to calibrate the models, resulting in improved accuracy and relevance for surgical planning and implant design.
