The mitochondrial electron transport chain is the major source of reactive oxygen species (ROS) during cardiac ischemia. Several mechanisms modulate ROS production; one is mitochondrial Ca2+ uptake. Here we sought to elucidate the effects of extramitochondrial Ca2+ (e[Ca2+]) on ROS production (measured as H2O2 release) from complexes I and III. Mitochondria isolated from guinea pig hearts were preincubated with increasing concentrations of CaCl2 and then energized with the complex I substrate Na+ pyruvate or the complex II substrate Na+ succinate. We report a novel noncontact diffuse correlation spectroscopy flow-oximeter for simultaneous quantification of relative changes in tissue blood flow (rBF) and oxygenation (Δ[oxygenation]). The noncontact probe was compared against a contact probe in tissue-like phantoms and forearm muscles (n = 10), and the dynamic trends in both rBF and Δ[oxygenation] were found to be highly correlated. However, the magnitudes of Δ[oxygenation] measured by the two probes were significantly different. Cofactor specificities of glycolytic enzymes in Clostridium thermocellum were studied with cellobiose-grown cells from batch cultures. Intracellular glucose was phosphorylated by glucokinase using GTP rather than ATP. Although phosphofructokinase typically uses ATP as a phosphoryl donor, we found only pyrophosphate (PPi)-linked activity. Phosphoglycerate kinase used both GDP and ADP as phosphoryl acceptors. In agreement with the absence of a pyruvate kinase sequence in the C. thermocellum genome, no activity of this enzyme could be detected. Also, the annotated pyruvate phosphate dikinase (ppdk) is not crucial for the generation of pyruvate from phosphoenolpyruvate (PEP), as deletion of the ppdk gene did …

Designing effective antisense sequences is a formidable problem. A method for predicting efficacious antisense holds the potential to provide fundamental insight into this biophysical process. More practically, such an understanding increases the chance of successful antisense design as well as saving considerable time, money and labor. The secondary structure of an mRNA molecule is believed to be in a constant state of flux, sampling several different suboptimal states. We hypothesized that particularly volatile regions might provide better accessibility for antisense targeting. A computational framework, GenAVERT was developed to evaluate this hypothesis. Glass polyalkenoate cements (GPCs) have previously been considered for orthopedic applications. A Zn-GPC (BT 101) was compared to commercial GPCs (Fuji IX and Ketac Molar) which have a setting chemistry analogous to BT 101. Handling properties (working, Tw and setting, Ts times) for BT 101 were shorter than the commercial GPCs. BT 101 also had a higher setting exotherm (Sx - 34 °C) than the commercial GPCs (29 °C). Van Nes rotationplasty may be used for patients with congenital proximal focal femoral deficiency (PFFD). The lower limb is rotated to use the ankle and foot as a functional knee joint within a prosthesis. A small series of cases was investigated to determine the long-term outcome. At a mean of 21.5 years (11 to 45) after their rotationplasty, a total of 12 prosthetic patients completed the Short-Form (SF)-36, Faces Pain Scale-Revised, Harris hip score, Oswestry back pain score and Prosthetic Evaluation Questionnaires, as did 12 age- and gender-matched normal control participants. Summary of background data: Previous work has demonstrated the efficacy of lumbar pedicle screw hook rod (PSHR) techniques and the Buck screw in the stabilization of spondylolysis. The mechanical behavior of lower profile cervical implants used to create PSHR, hybrid cable plate constructs, and titanium miniplating has not previously been described.

Methods: Calf lumbar spines (L2-L6) were utilized for testing (n = 27). Intervertebral rotation was measured in the intact spines across the L4-5 segment before and after creation of bilateral pars interarticularis defects. We tested if small conductance, Ca2 +‐sensitive K+ channels (SKCa) precondition hearts against ischemia reperfusion (IR) injury by improving mitochondrial (m) bioenergetics, if O2‐derived free radicals are required to initiate protection via SKCa channels, and, importantly, if SKCa channels are present in cardiac cell inner mitochondrial membrane (IMM). NADH and FAD, superoxide (O2), and m[Ca2 +] were measured in guinea pig isolated hearts by fluorescence spectrophotometry. Fine wire electromyography (EMG) is commonly used for surgical decision making in equinovarus foot deformity. However, this invasive technique may have the unwanted effect of altering the gait of children with cerebral palsy (CP). The purpose of this study was to determine if fine wire insertion into the posterior tibialis muscle affects temporal-spatial parameters and hindfoot kinematics during gait in children with equinovarus secondary to hemiplegic CP.

Objective

The cardiac workload associated with various types of aortic obstruction was determined using computational fluid dynamic simulations.

Methods

Computed tomography image data were collected from 4 patients with 4 distinct types of aortic arch obstructions and 4 controls. The categorization of arch hypoplasia corresponded to the “A, B, C” nomenclature of arch interruption; a type “D” was added to represent diffuse arch hypoplasia. Vergence eye movement is one of the oculomotor systems which allow depth perception via disconjugate movement of the eyes. Neuroimaging methods such as functional magnetic resonance imaging (fMRI) measure neural activity changes activity in the brain while subjects perform experimental tasks. A rich body of primate investigations on vergence is already established in the neurophysiology literature; on the other hand, there are a limited number of fMRI studies on neural mechanisms behind the vergence system.

Case reports have been published of people experiencing symptoms of traumatic brain injury (TBI) and/or ailments of the vascular tissue around the head. These reports pointed towards G-forces, a scalar measure without a specified direction, which represent the measure of the acceleration of an object divided by acceleration caused by gravity (9.81 m/s2), as a cause of said injuries. Research has been conducted on quantifying the G-forces experienced while on roller coasters, as well as the change in heart rate. However these studies have not been able to show a link between roller-coasters and symptoms.

This work presents a fuzzy logic based HandyMan (Hand gesture commands for grasping and manipulation) system to recognize the operator's hand gestures during task space telemanipulation. A combination of joint positions may be shared between at least two manipulation gestures. To avoid misinterpretation of gestures by the gesture recognition system, six new manipulation gestures were introduced. The gestures are produced by the user wearing a CyberGlove™.

This system replaces the previous HandyMan gesture recognition method. The output of the fuzzy system drives the state machines to implement the gestures with the robot hand.

There is no supporting evidence whether myelin degeneration in diffuse axonal injury (DAI) of the white matter is due to death of myelinating oligodendrocytes or secondary axotomy. Cortical neurons are not able to remain healthy in culture to have their axons myelinated. Dorsal root ganglion (DRG) neurons have been shown to be myelinated by cortical oligodendrocytes in culture. An in vitro stretch injury model was used to analyze the DRG axon pathology during and after injury. Oxidative damage is thought to be a major cause in development of pathologies and aging. However, quantification of oxidative damage is methodologically difficult. Here, we present a robust liquid chromatography-tandem mass spectrometry (LC-MS/MS) approach for accurate, sensitive, and linear in vivo quantification of endogenous oxidative damage in the nematode Caenorhabditis elegans, based on F3-isoprostanes. F3-isoprostanes are prostaglandin-like markers of oxidative damage derived from lipid peroxidation by Reactive Oxygen Species (ROS). Oxidative damage was quantified in whole animals and in multiple cellular compartments, including mitochondria and peroxisomes. Malaria has profoundly influenced human history for over four thousand years and despite numerous attempts at eradication, the prevention, diagnosis, and treatment of malaria have been largely ineffective. More than five hundred million people are affected by malaria every year resulting in over one million deaths. Drug resistance development by the parasite has diminished the effectiveness of numerous treatment options due, in part, to overtreatment of negative patients based on insufficient clinical algorithms and diagnostic methods.
