Nanotechnology & Human Stem Cells : Applications In Cardiogenesis And Neurogenesis, Martin Lyubomirov Tomov

Legacy Theses & Dissertations (2009 - 2024)

Human stem cell research holds an unprecedented promise to revolutionize the way we approach medicine and healthcare in general, moving us from a position of mostly addressing the symptoms to a state where treatments can focus on removing the underlying causes of a condition. Stem cell research can shed light into normal developmental pathways, as we are beginning to replicate them in a petri dish and can also be used to model diseases and abnormal conditions. Direct applications can range from finding cures for single or multigene diseases to demonstrating that we can replace these genes with a normal copy. …

Use Of Lidar In The Design Of Grassed Waterways: Case Study In Agricultural Management In Oklahoma, Annette Sparks 2016 Michigan Technological University

Use Of Lidar In The Design Of Grassed Waterways: Case Study In Agricultural Management In Oklahoma, Annette Sparks

Dissertations, Master's Theses and Master's Reports

Before the advent of Light Detection and Ranging (LiDAR) technology, farm-level infrastructure for sustainable agriculture was designed using topographic maps. LiDAR is a remote sensing method whereby reflected laser pulses are measured to generate high-resolution 3D images of the terrain. This report compares these two methods in the design of grassed waterways for flood drainage and evaluates how the quantitative difference between the two methods can affect the overall hydrologic design. The benefits of LiDAR are expected to be higher accuracy and precision in design, as well as greater reproducibility or consistency, regardless of the designer. To carry out this …

Enhanced Corrosion Protection And Biocompatibility Of A Plga-Silane Coating On Az31 Mg Alloy For Orthopaedic Applications, Sankalp Agarwal, Muhammad Morshed, Marie-Noelle Labour, David Hoey, Brendan Duffy, James Curtin, Swarna Jaiswal

Articles

This paper reports a multi-step procedure to fabricate a novel corrosion resistant and biocompatible PLGA-silane coating on the magnesium (Mg) alloy AZ31. The first step involves alkaline passivation followed by dip coating in a methyltriethoxysilane (MTES) and tetraethoxysilane (TEOS) mixture to produce a cross-linked siloxane coating. The second step is to impart an amine functionalization to the silane modified surface by using 3-aminopropyl-triethoxy silane (APTES) for promoting adhesion of the acid terminated poly-(lactic-co-glycolic) acid (PLGA) as a final coating step. Static contact angle measurements, Fourier transform infrared spectroscopy and scanning electron microscopy analysis confirmed the successful assembly of coatings on …

Bioelectrics Publications

For this systematic review, 203 published reports on effects of electroporation using nanosecond high-voltage electric pulses (nsEP) on eukaryotic cells (human, animal, plant) in vitro were analyzed. A field synopsis summarizes current published data in the field with respect to publication year, cell types, exposure configuration, and pulse duration. Published data were analyzed for effects observed in eight main target areas (plasma membrane, intracellular, apoptosis, calcium level and distribution, survival, nucleus, mitochondria, stress) and an additional 107 detailed outcomes. We statistically analyzed effects of nsEP with respect to three pulse duration groups: A: 1–10 ns, B: 11–100 ns and C: …

Enhancement Of Cancer Vaccine Efficacy Via Nanoparticle Or Molecular-Based Adjuvants, Myunggi An 2016 Wayne State University

Enhancement Of Cancer Vaccine Efficacy Via Nanoparticle Or Molecular-Based Adjuvants, Myunggi An

Wayne State University Theses

Adjuvants are immunomodulators which enhance immune responses to vaccines. However, parenteral administration of unformulated adjuvants fails to reach lymph nodes (LNs), the anatomic organ where the primary functions of immune cells are orchestrated. The LN-targeting delivery plays the key roles in promoting immune activation and has the great potential to transform disease treatment. The main goal of this thesis is to develop efficient vaccine delivery systems to target therapeutics into draining lymph nodes (dLNs) for ensuring their immunostimulatory activity. We introduced therapeutic applications of activating TLR9 with synthetic CpG oligodeoxynucleotide (ODN) agonists in nanoparticle or molecular form to activate immune …

Toward Magnetic Resonance Only Treatment Planning: Distortion Mitigation And Image-Guided Radiation Therapy Validation, Ryan Glen Price

Wayne State University Dissertations

While MR-only treatment planning has shown promise, there are still several well-known challenges that are currently limiting widespread clinical implementation.

Firstly, MR images are affected by both patient-induced and system-level geometric distortions that can significantly degrade treatment planning accuracy. . In addition, the availability of comprehensive distortion analysis software is currently limited. Also while many groups have been working toward a synthetic CT solution, further study is needed on the implementation of synCTs as the reference datasets for linac-based image-guided radiation therapy (IGRT) to help determine their robustness in an MR-only workflow.

Browse all Theses and Dissertations

Human Replication Protein A (RPA) is a heterotrimeric protein consisting of 70, 32, and 14 kDa subunits. RPA is the predominant single stranded DNA binding protein within the cell. It is involved in all forms of the DNA metabolic pathways, including but not limited to, replication, recombination, damage repair, as well as cell cycle and DNA check point signaling. RPA is phosphorylated (pRPA) during G1-S phase and is dephosphorylated during M phase. Further, RPA is hyperphosphorylated during DNA damage. Through the use of x-ray crystallography and nuclear magnetic resonance, researchers have proposed models and structures based on truncated portions of …

Reference Point Indentation Of Human Trabecular Bone Treated With Bisphosphonates For Varying Durations, Drew Jones

Theses and Dissertations--Biomedical Engineering

Reference point indentation (RPI), a novel form of micro-indentation, quantifies RPI material parameters which correlate with modulus, yield stress, strength, or toughness. Information linking bisphosphonate treatment length with the material properties of osteoporotic trabecular bone is needed to improve patient treatment. The objectives of this study were to: 1) determine if RPI can be used to successfully evaluate human trabecular bone and if so, determine an optimized test method for using RPI on trabecular bone, and 2) use this method to determine if any RPI parameters are related to the duration of bisphosphonate treatment.

Closed-Loop Afferent Nerve Electrical Stimulation For Rehabilitation Of Hand Function In Subjects With Incomplete Spinal Cord Injury, Christopher J. Schildt 2016 University of Kentucky

Closed-Loop Afferent Nerve Electrical Stimulation For Rehabilitation Of Hand Function In Subjects With Incomplete Spinal Cord Injury, Christopher J. Schildt

Theses and Dissertations--Biomedical Engineering

Peripheral nerve stimulation (PNS) is commonly used to promote use-dependent cortical plasticity for rehabilitation of motor function in spinal cord injury. Pairing transcranial magnetic stimulation (TMS) with PNS has been shown to increase motor evoked potentials most when the two stimuli are timed to arrive in the cortex simultaneously. This suggests that a mechanism of timing-dependent plasticity (TDP) may be a more effective method of promoting motor rehabilitation. The following thesis is the result of applying a brain-computer interface to apply PNS in closed-loop simultaneously to movement intention onset as measured by EEG of the sensorimotor cortex to test whether …

Detection Of Electrical Alternans In Ventricular Depolarization Phase Of Human Electrocardiogram, David R. Wasemiller 2016 University of Kentucky

Detection Of Electrical Alternans In Ventricular Depolarization Phase Of Human Electrocardiogram, David R. Wasemiller

Theses and Dissertations--Biomedical Engineering

T-wave Alternans (TWA) in an electrocardiogram (ECG) has received considerable interest as a potential predictor of sudden cardiac death (SCD). However, large clinical trials have shown that while TWA has a very high negative predictive value, its positive predictive value is poor. Results of previous studies suggest that arrhythmia onset can be affected by the phase relationship of alternans of the depolarization and repolarization phase of the action potentials of the ventricles. To assess this relationship, one would first need to establish that depolarization alternans can be detected and then develop methods to determine its relationship with repolarization alternans, which …

Surface-Initiated Polymerizations For The Rapid Sorting Of Rare Cancer Cells, Jacob L. Lilly 2016 University of Kentucky

Surface-Initiated Polymerizations For The Rapid Sorting Of Rare Cancer Cells, Jacob L. Lilly

Theses and Dissertations--Chemical and Materials Engineering

Cancer metastasis directly accounts for an estimated 90% of all cancer related deaths and is correlated with the presence of malignant cells in systemic circulation. This observed relationship has prompted efforts to develop a fluid biopsy, with the goal of detecting these rare cells in patient peripheral blood as surrogate markers for metastatic disease as a partial replacement or supplement to tissue biopsies. Numerous platforms have been designed, yet these have generally failed to support a reliable fluid biopsy due to poor performance parameters such as low throughput, low purity of enriched antigen positive cells, and insufficiently low detection thresholds …

Biventricular Finite Element Modeling And Quantification Of 3d Langragian Strains And Torsion Using Dense Mri, Zhanqiu Liu

Theses and Dissertations--Mechanical and Aerospace Engineering

Statistical data suggests that increased use of evidence-based medical therapies has largely contributed to the decrease in American death rate caused by heart disease. And my studies are about two applications of magnetic resonance imaging (MRI) as a non-invasive approach in evidence-based health care research. In my first study, the achievement of a pulmonary valve replacement surgery was assessed on a patient with tetralogy of Fallot (TOF). In order to evaluate the remodeling of right ventricle, two biventricular finite element models were built up for pre-surgical images and post-surgical images. In my second study, 3D Lagrangian strains and torsion in …

Diffusion-Mediated Deposition Of Proteins, Ruiqian Zhan 2016 University of Kentucky

Diffusion-Mediated Deposition Of Proteins, Ruiqian Zhan

Theses and Dissertations--Mechanical and Aerospace Engineering

Gradients of proteins play a prominent role in many biological processes, from development of multicellular organisms to chemical signaling in the immune system. Deposition of surface gradients is a way to permanently modifying a surface’s properties, resulting in the creation of novel materials which have widespread applications in biologically relevant fields, such as directed cell growth, production of biocompatible implantable materials, and creation of functional biosensors. These types of surfaces can also be used as an ex vivo tool to help understand many biological processes by mimicking the environment in gradient-related phenomena in a controllable way. However, despite the large …

Development And Application Of Gadolinium Free Cardiac Magnetic Resonance Fibrosis Imaging For Multiscale Study Of Heart Failure In Patients With End Stage Renal Disease, Tori A. Stromp 2016 University of Kentucky

Development And Application Of Gadolinium Free Cardiac Magnetic Resonance Fibrosis Imaging For Multiscale Study Of Heart Failure In Patients With End Stage Renal Disease, Tori A. Stromp

Theses and Dissertations--Physiology

Cardiac magnetic resonance (CMR) is a powerful tool to noninvasively image ventricular fibrosis. Late gadolinium enhancement (LGE) CMR identifies focal and, with T1 mapping, diffuse fibrosis. Despite prevalent cardiac fibrosis and heart failure, patients with end stage renal disease (ESRD) are excluded from LGE. Absence of a suitable diagnostic has limited the understanding of heart failure and obstructed development of therapies in the setting of ESRD. A quantitative, gadolinium free fibrosis detection method could overcome this critical barrier, propelling the advancement of diagnostic, monitoring, and therapy options. This project describes the development of a gadolinium free CMR technique and application …

A Software Application For Cardiac-Gated Computerized Tomography Scanning, Stephen Caldwell, Trevor Engelsman 2016 University of Akron

A Software Application For Cardiac-Gated Computerized Tomography Scanning, Stephen Caldwell, Trevor Engelsman

Williams Honors College, Honors Research Projects

Computerized tomography (CT) scans are a common clinical imaging procedure used worldwide. Operating in the X-Ray spectrum, these machines rotate scanners around a stationary body in order to compile two-dimensional images into a unified three-dimensional image. With adjustment to scan frequency and intensity, internal features such as muscles, organs, and tendons can easily be viewed. However, the heart has long evaded CT use due to its near-constant motion. Recently, cardiac-gated scans have entered the market as a technique to image the heart at a specific moment in time when it is nearly still - the quiescent period. Occurring during rapid …

Automated Solid-Substrate Cultivation Of The Anaerobic Bacterium Clostridium Thermocellum, Mathew J. Ruwaya 2016 University of Kentucky

Automated Solid-Substrate Cultivation Of The Anaerobic Bacterium Clostridium Thermocellum, Mathew J. Ruwaya

Theses and Dissertations--Biosystems and Agricultural Engineering

The organism Clostridium thermocellum grows on cellulosic substrates and produces ethanol, acetate, lactate, formic acid, and CO2. The organic acids produced alter the growth environment in which the bacteria grows and ultimately inhibit bacterial growth. One method which has been used successfully to maintain the system at acceptable growth conditions is to intermittently flush out the spent media and metabolic products and replace with new fermentation media. Our goal was to design and build an automated system that will automatically flush the spent media from the growing culture and resupply new media without manual intervention. An automated control …

Spatio-Temporal Progression Of Cortical Activity Related To Continuous Overt And Covert Speech Production In A Reading Task, Jonathan S. Brumberg, Dean J. Krusienski, Shreya Chakrabarti, Aysegul Gunduz, Peter Brunner, Anthony L. Ritaccio, Gerwin Schalk 2016 Old Dominion University

Spatio-Temporal Progression Of Cortical Activity Related To Continuous Overt And Covert Speech Production In A Reading Task, Jonathan S. Brumberg, Dean J. Krusienski, Shreya Chakrabarti, Aysegul Gunduz, Peter Brunner, Anthony L. Ritaccio, Gerwin Schalk

Electrical & Computer Engineering Faculty Publications

How the human brain plans, executes, and monitors continuous and fluent speech has remained largely elusive. For example, previous research has defined the cortical locations most important for different aspects of speech function, but has not yet yielded a definition of the temporal progression of involvement of those locations as speech progresses either overtly or covertly. In this paper, we uncovered the spatio-temporal evolution of neuronal population-level activity related to continuous overt speech, and identified those locations that shared activity characteristics across overt and covert speech. Specifically, we asked subjects to repeat continuous sentences aloud or silently while we recorded …

Automated Image Analysis And Spatial Computational Modeling Of Nf-Kb In Cerebrovascular Endothelial Cells, Kasey Catalfomo

Theses and Dissertations

Cerebrovascular endothelial cells play a key part in the inflammatory response of the blood-brain barrier in pathological conditions such as Alzheimer’s disease. Specifically, the NF-κB signaling pathway plays a central role. Better understanding of the factors in inflammatory disease progression can lead to more effective treatments for such devastating illnesses like Alzheimer’s, asthma, arthritis, cancer, diabetes and many more inflammatory diseases. The proposed approach analyzes spatial NF-κB distribution contained in multispectral stacked micrograph images of cerebrovascular endothelial cells indexed based on dose of the activating protein and the length of activation. Image analysis code identifies the location of nuclear boundaries …

Knee Internal Forces In Moderate Squat Exercise, Dumitru I. Caruntu, Jose Mario Salinas 2016 The University of Texas Rio Grande Valley

Knee Internal Forces In Moderate Squat Exercise, Dumitru I. Caruntu, Jose Mario Salinas

Mechanical Engineering Faculty Publications

This paper deals with internal forces of human knee during moderate squat exercise. The moderate squat exercise consists of a descending phase from standing to the lowest position (largest flexion angle) in which no significant contact between thigh and calf occurs, and an ascending phase back to standing position. This research predicts the internal forces such as muscle forces, contact forces, and ligamentous forces. The ligamentous structures in this research consist of Anterior Cruciate Ligament (ACL), Posterior Cruciate Ligament (PCL), Lateral Collateral Ligament (LCL), and Medial Collateral Ligament (MCL). The ligaments are modeled as nonlinear elastic strips (they do not …

A New Technique To Improve The Operation Of Prosthetic Limbs During Muscle Fatigue, H. Albunashee, G. Rasool, K. Iqbal, G. White 2016 University of Arkansas at Little Rock

A New Technique To Improve The Operation Of Prosthetic Limbs During Muscle Fatigue, H. Albunashee, G. Rasool, K. Iqbal, G. White

Journal of the Arkansas Academy of Science

Prosthetic limbs hold a promise to renew the quality of life for the amputee. Neural commands are decoded via a classifier to generate control signals for the prosthetic devices. In the literature, many challenges and limitations have been identified that affect the prosthesis operation. One such drawback is muscle fatigue which degrades the surface electromyogram (sEMG) signals, and consequently, the performance of the deployed classification algorithm declines from 90% to 50% of average accuracy. We used a new technique using the Linear Discrimination Analysis (LDA) algorithm and the muscle synergy-based task discrimination (MSD) algorithm to improve the classification accuracy. In …
