Decellularization Of Porcine Cartilage Promotes Chondrogenic Differentiation Of Human Chondrocytes, Roxanne Nicole Stone
Boise State University Theses and Dissertations
Knee osteoarthritis (knee OA) is the most common type of osteoarthritis (OA) and accounts for 70% of arthritis-related hospital admissions and 23% of clinical visits. Major limitations in both the current non-surgical and surgical methods are that they only relieve pain and show no evidence for restoring natural tissue anatomy. Leaders in the field propose that a stem cell treatment approach holds promise for the regeneration of a greater proportion of hyaline-like tissue at the repair site. (Cross et al., 2014; Escobar Ivirico, Bhattacharjee, Kuyinu, Nair, & Laurencin, 2017; Helmick et al., 2008; Toh, Foldager, Pei, & Hui, 2014).

Designing Silk Protein-Based Composite Materials With Tunable Heat Transfer And Magnetic Properties, Ye Xue
Theses and Dissertations
Renewable and biocompatible silk protein materials with desired physical properties show promising applications in biomedical field. In this work, a set of protein-based composites with desired thermal and magnetic properties, enhanced by the appropriate distribution of nanofillers in the protein matrix in both 2D-film and 1D-fiber forms was investigated. Results indicate that secondary structures of silk protein materials regenerated by a formic acid-calcium chloride method are different from those of their respective natural silk fibers. Intramolecular beta-sheet structures were found to dominate these silk films, causing these regenerated samples to be water-insoluble but more flexible than traditional silk films with …

Effects Of Hard-To-Soft Segment Ratios On The Synthesis And Physico-Mechanical Properties Of Polyurethane Films, Aaron C. Wilson
Mechanical Engineering Theses
Blood-contacting cardiovascular stents often induce a secondary clotting event due to unrestricted enzymatic activities. The use of hemocompatible polyurethane coatings on these implantable devices is one of the most promising methods to reduce device rejection. In this study, four commercial polyurethane films of various thicknesses and compositions were evaluated for their anticoagulation properties. Results suggested that these films exhibited excellent thermal and physico-mechanical properties while capable of increasing contact time with blood plasma by over a thousand-fold as compared to a control surface. Due to the unknown structure and composition of these commercial films, polyurethane samples were synthesized from toluene …

Electronic Theses and Dissertations
Celecoxib nanoemulsion (CXB-NE) has been developed as a macrophage targeted analgesics by Dr. Janjic and her team at Duquesne University, (Janjic et al, 2018; Liu et al, 2020; Saleem et al, 2019b; Vasudeva et al, 2014). The CXB-NE nanoemulsion carrying a Nonsteroidal Anti-inflammatory (NSAID) inhibitor of COX-2 activity result in a reduction in PGE2 expression in macrophages. Using CXB-NE in rats that have peripheral nerve injury constricting the sciatic nerve relieves hypersensitivity, a pain-like behavior. The treatment also decreases inflammation associated with this chronic constriction injury (Janjic et al, 2018; Saleem et al, 2019b; Stevens et al, 2019). In this …

Design Of A 3d Printed Bioreactor For Bone Cancer Research, Brooklyn K. Vanderwolde, Katelyn Hillson
The Journal of Undergraduate Research
Bone cancer is an aggressive disease and has peak occurrence during physiological stimulation of growth and aging. Astronauts who undergo long-term space missions also acquire an increased risk of bone tissue degeneration and cancer. Few in-vitro models currently exist capable of reproducing the complex microenvironment of bone tissue to support multicellular activity in a three-dimensional structure. This limits the ability to understand disease progression and develop suitable treatment strategies. Therefore, developing a bone tissue model and an associated bioreactor is critical to understand the risks associated with cancer progression and improve treatment and preventions related to those risks. The goal …

The ninth leading cause of death in the United States is kidney disease, and hemodialysis is the process most commonly prescribed for treatment. It utilizes a selectively permeable membrane filter to remove toxins such as urea from the blood and retain necessary protein levels. However, traditional filters, such as cellulose triacetate, used during dialysis can be inefficient in terms of separation performance and reduction of fouling. Recent exploration of nanoparticles has resulted in the creation of Oxone Mediated TEMPO-Oxidized Nano Cellulose which has properties that are believed to increase hydrophilicity, increase tensile capacity, decrease membrane resistance and lower fouling, making …

Modified Stent Design For A Coronary Bifurcation Lesion, Abigail Nowell
Biological Sciences Undergraduate Honors Theses
Currently, 18.2 million adults aged 20 and older are diagnosed with Coronary Artery Disease (CAD) (Benjamin et al., 2019). Stenosis is the most common intervention. However, when a patient has a bifurcated artery, treatment becomes more difficult and is often unsuccessful. This project created a new stent and balloon complex that was tested in vitro using a gel phantom artery model. Two separate prototypes have been created and tested so far, with improvements made upon each. Testing is still underway with Prototype 2.

Poly(Vinyl Alcohol)/Α-Chitin/Nanocellulose Composites Developed From Oil Palm Empty Fruit Bunch (Opefb) For Removal Of Cationic Dyes, Chun Fah Mok
Student Works (2020-2029)
In search of an eco-friendly polymeric adsorbent for dye effluent treatment application, poly(vinyl alcohol) (PVA)/chitin/nanocellulose composites were synthesized. The composites were reinforced by oil palm empty fruit bunch fibers (OPEFB)-derived nanocrystalline cellulose (NCC). With addition of 1 wt.% NCC, a maximum improvement of 57.64% and 50.66% in tensile strength and Young’s Modulus were achieved, respectively. However, the PVA-based composites exhibited low resistance in aqueous medium. Hence, maleic acid (MA) was incorporated, in view of modifying PVA-based composites through cross-linking reactions. The effect of varying chitin content (10 wt.%-30 wt.%) and MA content (10%-50% based on total solid content of composite) …

Design Of An Affordable Rotating Drum Electrospinner For Classroom Education, Peder Solberg
The Journal of Undergraduate Research
Electrospinning is a technology used to generate small fibers down to nano-scale size. This method of fiber creation has been around for many years. However, in recent years electrospinning has found increased applications, especially in the area of tissue engineering due to its ability to create fibers with properties similar to the extracellular matrix in tissue. An electrospinning platform can illustrate concepts of engineering, electro-mechanical system design, manufacturing, and biomedical applications in one single package. Hence, it provides an excellent opportunity to integrate into secondary (middle and high school) and post-secondary (undergraduate) technology education.
Furthermore, just as integration of 3D …

Role Of Nanoparticle–Polymer Interactions On The Development Of Double-Network Hydrogel Nanocomposites With High Mechanical Strength, Andrew Chang, Nasim Babhadiashar, Emma Barrett-Catton, Prashanth Asuri
Bioengineering
Extensive experimental and theoretical research over the past several decades has pursued strategies to develop hydrogels with high mechanical strength. Our study investigated the effect of combining two approaches, addition of nanoparticles and crosslinking two different polymers (to create double-network hydrogels), on the mechanical properties of hydrogels. Our experimental analyses revealed that these orthogonal approaches may be combined to synthesize hydrogel composites with enhanced mechanical properties. However, the enhancement in double network hydrogel elastic modulus due to incorporation of nanoparticles is limited by the ability of the nanoparticles to strongly interact with the polymers in the network. Moreover, double-network hydrogel …

Dendrimer-Based Therapeutic Delivery Vehicle For The Treatment Of Ocular Diseases, Remy C. Cooper
Theses and Dissertations
The treatment of ocular diseases poses a significant challenge to drug delivery design, due to the eye’s complex anatomy, with a semi-permeable blood-ocular barrier and associated immune privilege. To address some of the delivery challenges, polycationic dendrimers are explored as vehicle for the treatment of posterior chamber disease. Dendrimers are a class of polymeric nanoparticles that are highly branched, multivalent and monodisperse, whose three-dimensional globular architecture has given them the nickname of “synthetic proteins”. Dendrimers are synthesized by reacting monomers in a step-wise fashion to produce generationally and radially increasing structures, with core, interior and surface chains that are highly …

Poor oral hygiene is a severe global health issue, with about 44% of the Earth’s population suffering from untreated tooth decay, accounting for over $100 billion dollars spent on dental services per year in the United States, alone. Manual treatments, such as brushing, flossing, or swishing with antiseptic rinse are the main ways to alleviate harmful bacteria that cause these oral health problems. Streptococcus mutans (S. mutans) is a group of highly antibiotic, Gram-positive bacteria that heavily contribute to oral cavities, plaque build-up, and potential loosening or loss of dental implants if a biofilm develops. Currently, the mechanism …

Gravity-Drawing Flexible Silicone Filaments As Fiber Optics And Model Foldamers, Katherine Snell
CMC Senior Theses
Here, we present a method of gravity-drawing polydimethylsiloxane (PDMS) silicone fibers with application as fiber optics and as model foldamers. Beginning as a viscous liquid, PDMS is cured using heat until its measured viscosity reaches 4000 mPa•s. The semi-cured elastomer is then extruded through a tube furnace to produce thin (diameters on the order of hundred micrometers) filaments with scalable lengths. PDMS is biocompatible, gas-permeable, flexible, and hydrophobic. Additionally, the PDMS surface hydrophobicity can be modified via UV exposure, O2 plasma, and corona discharge. We demonstrate the patternibility (i.e patterns of hydrophobicity) of PDMS fibers, adding complexity to potential foldamer …

Mussel adhesive proteins contain catechol moiety, which allows the protein to crosslinked, solidify, and adhere to surrounding surfaces even under wet conditions. Incorporating the catechol moiety into polymeric adhesive resulted in bioadhesive, which still functions under wet conditions. However, the adhesive must be oxidized in order to crosslink and adhere to surfaces. The oxidation process of catechol adhesive was proven to produce singlet oxygen, superoxide, and hydrogen peroxide (H2O2). These reactive oxygen species, with lack of control, can wreak havoc to the biological system causing poor healing and undesired biological response.
Here, we studied the silica …

The mechanotransduction of cells is the intrinsic ability of cells to convert the mechanical signals provided by the surrounding matrix and other cells into biochemical signals that affect several distinct processes such as tumorigenesis, wound healing, and organ formation. The use of biomaterials as an artificial scaffold for cell attachment, differentiation and proliferation provides a tool to modulate and understand the mechanotransduction pathways, develop better in vitro models and clinical remedies. The effect of topographical cues and stiffness was investigated in fibroblasts using polycaprolactone (PCL)- Polyaniline (PANI) based scaffolds that were fabricated using a self-assembly method and electrospinning. Through this …
