This paper introduces a new time-resolved spectral analysis method based on Linear Prediction Coding (LPC) method that is particularly suited to the study of the dynamics of EEG (Electroencephalogram) activity. The spectral dynamic of EEG signals can be challenging to analyse as they contain multiple frequency components and are often heavily corrupted by noise. Furthermore, the temporal and spectral resolution that can be achieved is limited by the Heisenberg-Gabor uncertainty principle [1]. Objective. Understanding how current reaches the brain during transcranial electrical stimulation (tES) underpins efforts to rationalize outcomes and optimize interventions. To this end, computational models of current flow relate applied dose to brain electric field. Conventional tES modeling considers distinct tissues like scalp, skull, cerebrospinal fluid (CSF), gray matter and white matter. The properties of highly conductive CSF are especially important. However, modeling the space between skull and brain as entirely CSF is not an accurate representation of anatomy. Transcranial direct current stimulation (tDCS) is a technique where a weak direct electrical current is applied to the scalp with the goal of stimulating the brain. There is tremendous interest in the use of tDCS for treating brain disorders and improving brain function. However, the effects of tDCS have been highly variable across studies, leading to a debate over its efficacy. About a third of all epilepsy patients may experience seizures that are resistant to medication. Surgical resection may be an option for some of these patients, but localization of the epileptogenic zone is difficult and expensive. For this process, the seizures must be observed and recorded in a clinical setting over several days. High-frequency oscillations (HFOs) have emerged as a possible biomarker for predicting the epileptogenic zone from a brief interictal recording of brain electrical activity without having to wait for seizures. Nanosecond Pulsed Electric Fields (nsPEF) have the potential to treat a variety of cancer types including melanoma, pancreatic and lung squamous cancers. Recent studies show that nsPEF-based cancer therapy may be improved further with the assistance of moderate heating of the target. A feedbacklooped heating system, utilizing a 980-nm fiber optic laser, was integrated into nsPEF electrodes for tumor ablation. The laser beam profile was determined to be Gaussian using a knife-edge technique. Gene electrotransfer (GET) is a safe, reliable, and effective method of delivering plasmid DNA (pDNA) to solid tumors. GET has been previously used to deliver interleukin-15 (IL-15) to mouse melanoma, resulting in long-term tumor regression and the survival of a percentage of treated animals after challenge. To enhance this effect, we evaluated modulating the expression levels of IL-15 and co-expressing its receptor, IL-15Rα. GET was used to deliver plasmids encoding IL-15 and IL-15Rα to established B16.F10 tumors on days 0, 4, and 7. Two delivery protocols that yielded different expression profiles were utilized. Neuromodulation of the primary motor cortex (M1) in pair with physical therapy may be a promising method for improving motor outcomes after spinal cord injury (SCI). Increased excitability of the corticospinal motor pathways (i.e. corticomotor excitability) has shown to be associated with improved motor learning and skill acquisition. Intermittent theta burst stimulation (iTBS) is a form of non-invasive brain stimulation which can increase corticomotor excitability, as measured by an increase in the amplitude of motor evoked potentials (MEPs). This paper attempts to answer the question of if it’s possible to produce a simple, quick, and accurate neural network for the use in upper-limb prosthetics. Through the implementation of convolutional and artificial neural networks and feature extraction on electromyographic data different possible architectures are examined with regards to processing time, complexity, and accuracy. It is found that the most accurate architecture is a multi-entry categorical cross entropy convolutional neural network with 100% accuracy. Impedance spectroscopy (IS) is an important tool for cell detection and characterization in medical and food safety applications. In this thesis, the Cal Poly Biofluidics Lab’s impedance spectroscopy system was re-evaluated and optimized for single-cell impedance spectroscopy. To evaluate the IS system, an impedance spectroscopy bioMEMS chip was fabricated in the Cal Poly Microfabcrication lab, software was developed to run IS experiments, and studies were run to validate the system. Differentiating psychogenic nonepileptic seizures from epileptic seizures is a difficult task that requires timely recording of psychogenic events using video electroencephalography (EEG). Interpretation of video EEG to distinguish epileptic features from signal artifacts is error prone and can lead to misdiagnosis of psychogenic seizures as epileptic seizures resulting in undue stress and ineffective treatment with antiepileptic drugs. The permeabilized condition of the cell membrane after electroporation can last minutes but the underlying mechanisms remain elusive. Previous studies suggest that lipid peroxidation could be responsible for the lasting leaky state of the membrane. The present study aims to link oxidation within the plasma membrane of live cells to permeabilization by electric pulses. We have introduced a method for the detection of oxidation by ratiometric fluorescence measurements of BODIPY-C11 dye using total internal reflection fluorescence (TIRF) microscopy, limiting the signal to the cell membrane. My research focuses on electrokinetic transport. Particularly, in this dissertation, we focus on fabrication and testing of micro electrodes with nanostructured surfaces to minimize the electrode polarization (EP) effects for biosensor applications. In the first study, electrochemical deposition of gold nanoparticles on to planar gold electrodes was used to generate rough surfaces. Dendritic nanostructures that reduced EP up to two orders of magnitude was obtained by optimizing the deposition conditions. These structures also enhanced dielectrophoresis (DEP) response of our bio-chips, making them usable in physiological buffers. In this thesis, the perturbation-based decomposition technique developed by Szlavik [1] was used in an attempt to solve the inverse problem in EEG source localization. A set of dipole locations were forward modeled using a 4-layer sphere model of the head at uniformly distributed lead locations to form the vector basis necessary for the method. Both a two-dimensional and a pseudo-three-dimensional versions of the model were assessed with the two-dimensional model yielding decompositions with minimal error and the pseudo-three-dimensional version having unacceptable levels of error. Background: Reconstruction of bone fractures and defects remains a big challenge in orthopedic surgery. While regenerative engineering has advanced the field greatly using a combination of biomaterial scaffolds and stem cells, one matter of difficulty is inducing osteogenesis in these cells. Recent works have shown electricity’s ability to promote osteogenesis in stem cell lines when seeded in bone scaffolds; however, typical electrical stimulators are either (a) externally housed and require overcomplex percutaneous wires be connected to the implanted scaffold or (b) implanted non-degradable devices which contain toxic batteries and require invasive removal surgeries.
